The optimization of functions subject to partial differential equations (PDE) plays an important role in many areas of science and industry. In this paper we introduce the basic concepts of PDE-constrained optimization and show how the all-at-once approach will lead to linear systems in saddle point form. We will discuss implementation details and different boundary conditions. We then show how these system can be solved efficiently and discuss methods and preconditioners also in the case when bound constraints for the control are introduced. Numerical results will illustrate the competitiveness of our techniques.
More than 60 neuropeptides that inhibit juvenile hormone synthesis by the corpora allata have been isolated from the brains of various insect species. Most of them are characterized by a common C-terminal pentapeptide sequence Y/FXFGL/I/V (allatostatin A family, allatostatin superfamily). Besides the allatostatin A family, allatostatic neuropeptides belonging to other two peptide families (W2W9)-allatostatins or allatostatin B family; lepidopteran allatostatin) were reported. So far, only one allatotropin has been identified. Four peptides with allatostatic activity were isolated from brains of the Mediterranean field cricket, Gryllus bimaculatus. Three of them (Grb-AST A3: AGMYSFGL-NH2; Grb-AST A4: SRPFGFGL-NH2; Grb-AST A5: GPDHRFAFGL-NH2) belong to the wide-spread family of Y/FXFGL/I-amide peptides, the fourth (Grb-AST B5: AWDQLRPGW-NH2) is a member of the W2W9 - amide family of neuropeptides. All of these peptides are potent inhibitors of juvenile hormone (JH) biosynthesis by cricket corpora allata in vitro, causing 50% inhibition of JH biosynthesis at 0.4-3 × 10-8 M. The two peptides Grb-AST A5 and Grb-AST B5 have virtually the same potency and efficacy in inhibiting JH biosynthesis in vitro. No synergistic effect of the two peptide families with respect to the inhibition of JH biosynthesis could be observed. Peptides of both families decrease the accumulation of methylfarnesoate, the direct precursor of JH, within CA that have been incubated in farnesol-rich medium. Epidemiological and clinical studies suggest that asthma is associated with adverse cardiovascular outcomes, but its mechanism is uncertain. 5-Hydroxytryptamine (5-HT) is a mediator involved in asthma and in cardiovascular functioning. Thus, in the present study, we explored whether allergic sensitization in guinea pigs modifies 5-HT-induced contractile responses and 5-HT2A receptor expression in thoracic aorta rings. We found that sensitization produced a significant increase of 100 µM 5-HT-induced contractions of aorta rings (~27 % greater contraction than in non-sensitized animals, p<0.05). Preincubation with 10 nM ketanserin (a 5-HT2A receptor antagonist) reduced by ~30 % (p=0.003) and ~36 % (p=0.005) the area under the curve of 5-HT-induced contractions in aortas from non-sensitized and sensitized animals, respectively. There were no differences between sensitized and non-sensitized animals with respect to mRNA (qPCR) and protein (Western blot) expression of 5-HT2A receptor in thoracic aortas. We concluded that in this guinea pig model of asthma, allergic sensitization is not confined to airways, but also affects arterial contractile responses to 5-HT; changes in the expression of the 5-HT2A receptor appear not to be involved in this phenomenon.
Fundamentals of the theory of system alliances are briefly reviewed. An accent is put on interfaces (IFs). The model of IFs consisting of a pair of finite deterministic automata sharing a part of their internal state space is introduced. The presented model of alliance interface can be successfully implemented for the study of typical phenomena in complex heterogeneous objects with a significant degree of uncertainty.
The spatial distribution of cytotypes can provide valuable insights into the evolution of polyploid complexes. Previously, only tetraploid Allium oleraceum was reported from Slovakia. Analysing 863 individuals from 93 populations from Slovakia revealed an extensive variation in the DNA ploidy levels of Allium oleraceum (3x, 4x, 5x and 6x). Of the main cytotypes, the penta- and tetraploids had strongly overlapping distributions, although the pentaploids exhibited a tendency to occur more frequently in the southern and the tetraploids had a tendency to occur in the northern regions of Slovakia. A triploid cytotype was found in one population in the southern part of Slovakia, which is the third locality worldwide for this cytotype. The hexaploid cytotype was rare and sparsely occurred in western and southern Slovakia. Sixteen per cent of the populations sampled consisted of more than one ploidy level; the most common was a combination of penta- and tetraploids. The cytotypes differed with respect to altitude; the tetraploids were found significantly more frequently at higher altitudes than the penta- and hexaploids. When compared with reanalysed altitudinal distribution data from the Czech Republic divided into two geographic areas (Carpathian and Herzynian) the pattern found in the Carpathian part of the Czech Republic was similar to that in Slovakia, with tetraploids at the higher altitudes. The distribution in the Herzynian part (Bohemian Massif) was just the opposite: the tetraploids were more often found at lower altitudes than the penta- and hexaploids. Both tetra- and pentaploid cytotypes occurred in a wide and similar spectrum of habitats, while hexaploids were limited to human-influenced habitats. A local-scale distribution of cytotypes analysed in detail in the Slovak Karst area, showed surprising differences in the distribution of cytotypes on particular karst plains, which can be related to different land uses. Concerning the contrasting altitudinal differentiation of tetraploids in the regions compared, the results suggest that at least two different types of tetraploids occur in Central Europe. The apparent cytotype diversity in the surrounding Slovak Karst area may suggest the existence of a primary contact zone.
Neutral trehalase 1 (Nth1) from Saccharomyces cerevisiae
catalyzes disaccharide trehalose hydrolysis and helps yeast to
survive adverse conditions, such as heat shock, starvation or
oxidative stress. 14-3-3 proteins, master regulators of hundreds
of partner proteins, participate in many key cellular processes.
Nth1 is activated by phosphorylation followed by 14-3-3 protein
(Bmh) binding. The activation mechanism is also potentiated by
Ca2+ binding within the EF-hand-like motif. This review
summarizes the current knowledge about trehalases and the
molecular and structural basis of Nth1 activation. The crystal
structure of fully active Nth1 bound to 14-3-3 protein provided
the first high-resolution view of a trehalase from a eukaryotic
organism and showed 14-3-3 proteins as structural modulators
and allosteric effectors of multi-domain binding partners.
Allozyme polymorphism was investigated in adult males of the stenotopic peat bog (tyrphobiontic) noctuid moth, Coenophila subrosea Stephens, from three isolated peat bog localities in Austria (Styria) and Czech Republic (South and North Bohemia). Of the eighteen enzyme loci examined, twelve were polymorphic and six monomorphic. Significant deviations of genotype frequencies from Hardy-Weinberg equilibrium were observed at about one third of polymorphic loci within the populations. The average heterozygosities for the populations from three geographically distinct localities ranged from 0.192 to 0.245, and 61% of the loci were polymorphic. The FST mean value of 0.0675 was higher than that found in most other Lepidoptera. The genetic distances based on allozyme heterozygosity ranged from 0.019 to 0.051, with the population from South Bohemia being the most distant. The genetic distances and FST values do not reflect the geographic distances between the populations. Morphometric analysis revealed a difference between the Austrian Purgschachen Moor and Bohemian populations. These isolated relict peat bogs are habitat islands inhabited by unique "geographical races" of tyrphobiontic taxa., Jan Šula, Karel Spitzer, 4 obrázky, 5 tab., and Lit.
In this paper we establish some conditions for an almost $\pi $-domain to be a $\pi $-domain. Next $\pi $-lattices satisfying the union condition on primes are characterized. Using these results, some new characterizations are given for $\pi $-rings.
Generalizing the notion of the almost free group we introduce almost Butler groups. An almost $B_2$-group $G$ of singular cardinality is a $B_2$-group. Since almost $B_2$-groups have preseparative chains, the same result in regular cardinality holds under the additional hypothesis that $G$ is a $B_1$-group. Some other results characterizing $B_2$-groups within the classes of almost $B_1$-groups and almost $B_2$-groups are obtained. A theorem of stating that a group $G$ of weakly compact cardinality $\lambda $ having a $\lambda $-filtration consisting of pure $B_2$-subgroup is a $B_2$-group appears as a corollary.
