We measured plasma concentrations, adipose tissue and placental mRNA expression of hepatokines fetuin A, fetuin B and fibroblast growth factor 21 (FGF21) in 12 healthy pregnant women (P group), 12 pregnant women with gestational diabetes (GDM) and 10 healthy non-pregnant women (N group) to explore their potential role in the etiopathogenesis of GDM. GDM and P group had comparable BMI, C-reactive protein (CRP) and glycated hemoglobin levels while IL-10 and TNF-α levels were higher in GDM group. Fetuin A and fetuin B levels were higher in pregnancy as compared to N group and decreased after delivery with no apparent influence of GDM. In contrast, the pattern of changes of circulating FGF21 levels differed between GDM and P group. Fetuin A concentrations positively correlated with CRP, TNF-α mRNA expression in adipose tissue and IL-6 mRNA expression in placenta. Fetuin B positively correlated with CRP. FGF21 levels correlated positively with IFN-γ mRNA in adipose tissue and inversely with IL-8 mRNA in the placenta. Taken together, fetuin A and fetuin B levels were increased during pregnancy regardless of the presence of GDM. In contrast, FGF21 patterns differed between healthy pregnant women and GDM patients suggesting a possible role of this hepatokine in the etiopathogenesis of GDM., P. Šimják, A. Cinkajzlová, K. The changes in shape of the marginal coenobial cells in an experimentally cultured population of Pediastrum duplex Meyen var. duplex were investigated. The methods of landmark-based geometric morphometrics, including sliding landmarks registration, were used. The populations were cultured at 11 different pH levels and the changes in shape related to pH were studied using multivariate regression. The results of relative warps analysis revealed that morphological trends are related to both size and pH. The potential application of the results of this geometric morphometrical analysis of Pediastrum for biomonitoring and palaeoecological studies are discussed.
Because nutrients accrued during larval stages represent the major limiting factor for egg production, the use of adult feeding to enhance the reproductive output in moths is considered to be largely weight-dependent. It is hypothesized, however, that feeding by adults could be adaptive and an effective means of increasing their reproductive success. In order to test this, the calling behaviour of Spodoptera littoralis females that differed in body weight and whether they had fed or not were recorded. Two experiments were carried out. In the first, the calling behaviour of food-deprived females of different body weights was recorded. A strong positive correlation was found between body weight at emergence and the total duration of calling of females on the second to the fifth night after emergence. In the second experiment, groups of female moths that varied in body weight were given access to water or sucrose. Feeding on sucrose significantly reduced the pre-calling period and increased the total time spent calling on the six nights after emergence. The increase in time spent calling associated with ingesting sucrose were proportionately similar for both small and large females, implying that feeding by adults can result in an increase in the time spent calling by moths irrespective of larval nutritional status. Female longevity was also correlated with moth weight at emergence and/or sucrose availability. It is concluded that it is advantageous for female S. littoralis to be large and/or have access to sucrose-rich food in the adult stage as they can spend more time attracting a mate, which increases their chances of mating in early adult life, and their longer adult life may indirectly result in an increase in fecundity., Medhat M. Climatic conditions can modify the life history traits, population dynamics and biotic interactions of species. Therefore, adaptations to environmental factors such as temperature are crucial for species survival at different altitudes. These adaptive responses, genetically fixed or plastic (phenotypic plasticity), can be determined by physiological thresholds and might vary between sexes. The objective of this study was to determine whether the life history traits of the European Map butterfly (Araschnia levana) differ at different altitudes. A field experiment was carried out along an altitudinal gradient from 350 to 1010 m a.s.l. in a low mountainous region (Bavaria, Germany). 540 butterfly larvae were placed at different altitudes in 18 planted plots of their larval host plant, the stinging nettle (Urtica dioica). After three weeks the larvae were collected and reared under laboratory conditions. Developmental traits of the butterflies, mortality and percentage parasitism were measured. Larval development was generally slower at higher altitudes and lower temperatures and larval weight decreased with increasing altitude and decreasing temperature. However, there were no significant differences in pupation, adult lifespan and percentage mortality at the different altitudes and temperatures. Female larvae were heavier than those of males, and the pupal and adult lifespans were longer in females than in males. However, male and female butterflies reacted similarly to altitude and temperature (no significant interactions). None of the 188 larvae collected were parasitized. In conclusion, the phenotypic plasticity of European Map butterfly has enabled it to adapt to different temperatures, but the strategies of the sexes did not differ. and Kathrin D. Chromosomes of the males of five species of Odontura, belonging to the subgenera Odontura and Odonturella, were analyzed. Intensive evolution of the karyotype was recorded, both in terms of changes in the numbers of chromosomes (from 2n = 31 to 27) and the sex chromosome system (from X0 to neo-XY and X0 to neo-X1X2Y). Karyotype evolution was accompanied by tandem autosome fusions and interspecific autosomal and sex chromosome differentiation involving changes in the locations of nucleolar organizer regions, NORs, which were revealed by silver impregnation and confirmed by FISH using an 18S rDNA probe. O. (Odonturella) aspericauda is a polytypic species with X0 and neo-X1X2Y sex determination. The latter system is not common in tettigoniids. It possibly originated by a translocation of a distal segment of the original X chromosome onto a medium sized autosome, resulting in a shortened neo-X1 and a metacentric neo-Y. The remaining autosome homologue became the neo-X2 chromosome. This shift from X0 to neo-X1X2Y is supported by the length of the X chromosome and location of the NOR/rDNA. and Elżbieta Warchałowska-Śliwa, Anna Maryańska-Nadachowska, Beata Grzywacz, Tatjana Karamysheva, Arne W. The invasion of Austria by the alien vascular plant Ambrosia artemisiifolia (Asteraceae) is analysed in detail, based on a survey of available records. In total, 697 records were collated. The first record for Austria is a herbarium specimen collected in 1883. Up to the end of the 1940s, records were rare and only of casual populations resulting from long-distance dispersal. Since the 1950s, the number of records has increased exponentially, and more than one third of all records (242) were collected in the last 5-year period (2001–2005) included in the survey. The first naturalized population was recorded in 1952, nearly 70 years after the first record of a casual population. Recently, the number of naturalized populations increased considerably faster than that of casual populations. Several pathways (contaminated crops and bird seed, agricultural machines, transport of soil) have contributed to the high levels of propagule pressure and this successful invasion. Ambrosia artemisiifolia has undergone a niche expansion during the invasion process. Up to 1950, most records were from sites along railway routes, whereas in the period 1950–1974 itwas mostly ruderal habitats, not associated with traffic infrastructure, which were colonized. Since the 1970s, records from roadsides have increased strongly and now dominate. Fields were colonized first in the 1970s and since then have gained in importance. The distribution of naturalized populations was related to environmental and climatic variables by means of a generalized linear model. Their distribution in Austria is closely related to temperature. Landscape variables, describing aspects of habitat availability (topography, land use, major street density) also significantly explain the current distribution of A. artemisiifolia. Suitable habitats currently occur mainly in the eastern and southeastern lowlands. We conclude that global warming will disproportionally enhance the invasion success of A. artemisiifolia in Austria, even if there is only a slight increase in temperature, as significant areas of agricultural land in Austria are currently only slightly too cool for A. artemisiifolia. The widespread occurrence of this species will have serious consequences for human health and agriculture.
The mixture of volatile compounds emitted by Brassica oleracea var. sabauda changed significantly in response to feeding and/or oviposition by Murgantia histrionica (Heteroptera: Pentatomidae). Volatiles were collected from (1) healthy plants and those with (2) feeding punctures, (3) a combination of feeding punctures and oviposition, (4) feeding punctures and one hatched egg mass and (5) plants bearing only an egg mass. In the case of plants with feeding punctures or feeding punctures plus an egg mass, the volatiles were also collected at different time intervals after plants were subjected to these two treatments (0-24 h, 24-48 h and 48-72 h). Gas chromatographic and gas chromatograph-mass spectrometric analysis showed that the percent emission of several compounds changed significantly from plants subjected to the feeding and oviposition or just oviposition. Percentage of terpenes generally decreased after feeding and oviposition, although the percentage emission of (E)-β-caryophyllene from these plants and those with just feeding punctures significantly increased. Plants with just an egg mass emitted linalool de novo but not (E)-β-ocimene. The emission of jasmonates, mainly methyl jasmonate, increased from plants with feeding punctures plus an egg mass compared to those with only an egg mass. Higher percentages of the volatile glucosinolate derivatives (VGSs), mainly 4-methoxy-3-indolylacetonitrile, were emitted by plants with feeding punctures and an egg mass. The percentage emission of most of these compounds increased during the first 24 h after the treatment and then decreased over the next 24 h, except for methyl jasmonate, which remained high also 48-72 h later. The possible ecological roles of such volatiles in plant interactions with the second and third trophic levels are discussed.
Tissue differentiation and proliferation throughout fetal development interconnect with changes in the oxidative phosphorylation system (OXPHOS) on the cellular level. Reevaluation of the expression data revealed a significant increase in COX4 and MTATP6 liver transcription levels after the 22nd gestational week (GW) which inspired us to characterize its functional impact. Specific activities of cytochrome c oxidase (COX), citrate synthase (CS), succinate-coenzyme Q reductase (SQR) and mtDNA determined by spectrophotometry and RT-PCR were studied in a set of 25 liver and 18 skeletal muscle samples at 13th to 29th GW. Additionally, liver hematopoiesis (LH) was surveyed by light microscopy. The mtDNA content positively correlated with the gestational age only in the liver. The activities of COX, CS and SQR in both liver and muscle isolated mitochondria significantly decreased after the 22nd GW in comparison with earlier GW. A continuous decline of LH, not correlating with the documented OXPHOS-specific activities, was observed from the 14th to the 24th GW indicating their exclusive reflection of liver tissue processes. Two apparently contradictory processes of increasing mtDNA transcription and decreasing OXPHOS-specific activities seem to be indispensable for rapid postnatal adaptation to high energy demands. The inadequate capacity of mitochondrial energy production may be an important factor in the mortality of children born before the critical developmental point of the 22nd GW., H. Kolarová, J. Brain perfusion is reduced early after subarachnoid hemorrhage (SAH) due to intracranial hypertension and early vasospasm. The contribution of these two mechanisms is unknown. By performing a prophylactic decompressive craniectomy (DC) in a rat model of SAH we aimed to study brain perfusion after the component of intracranial hypertension has been eliminated. We used 2x2 factorial design, where rats received either decompressive craniectomy or sham operation followed by injection of 250 μl of blood or normal saline into prechiasmatic cistern. The cortical perfusion has been continually measured by laser speckle-contrast analysis for 30 min. Injection of blood caused a sudden increase of intracranial pressure (ICP) and drop of cerebral perfusion, which returned to baseline within 6 min.
DC effectively prevented the rise of ICP, but brain perfusion after SAH was significantly lower and took longer to normalize compared to non-DC animals due to increased cerebral vascular resistance, which lasted throughout 30 min experimental period. Our findings suggest that intracranial hypertension plays dominant role in the very early hypoperfusion after SAH whilst the role of early vasospasm is only minor. Prophylactic DC effectively maintained cerebral perfusion pressure, but worsened cerebral perfusion by increased vascular resistance.
