Dark respiration rate in the night (R D) was measured in five-year-old Scots pine (Pinus sylvestris L.) trees grown for two years under ambient (AC) and elevated (AC + 400 µmol mol-1 = EC) CO2 concentrations in open top chambers. Two needle age classes (i.e., current-year and one-year-old) were measured at AC and EC in both AC- and EC-grown pines. Additionally different chemical characteristics were determined on the needles, such as nitrogen (N), carbon (C), starch, and soluble sugar concentrations as well as specific leaf area. The direct, short-term and indirect, long-term effects of EC on R D for the two needle age classes were examined. R D was expressed on a per needle area, needle mass, N, C, and C/N bases. Direct effects were only pronounced in the AC treatment where inhibition of R D was found at EC in both current- and one-year-old needles. Indirect effects were only significant in one-year-old needles where a decrease was found in the EC grown trees as compared with AC ones when R D was expressed per unit needle mass, C, or C/N. R D per unit needle area and needle N were not sensitive to long-term EC, in any needle age class. Long-term EC treatment also influenced the response of the two needle age classes. One-year-old needles from the EC treatment had significantly lower R D than current-year needles, but no such response was observed in the AC treatment. Our experiment re-emphasised the importance of expressing R D on different bases for a correct interpretation of the responses to EC. Glucagon-like-peptide 2 (GLP-2) is an endogenous enteroendocrine physiological trophic peptide. Glepaglutide is a novel long-acting GLP-2 analog under development for the treatment of patients with Short Bowel Syndrome (SBS). The objective of this work was to compare the small intestinal trophic effects in both genders following short (1 week) versus long-term (26-39 weeks) GLP-2 treatment in Wistar rats and Beagle dogs. Following both short- and long-term treatment with glepaglutide, a significant dose-dependent intestinotrophic effect was seen in both genders and species. At all doses increased length and weight of the small intestine as well as macroscopic thickening and villous hypertrophy were noted in all segments of the small intestine, without any differences between genders. The findings were still present following a 6-week recovery period, indicating long-acting intestinotrophic effects of glepaglutide. These studies demonstrate that a long-acting GLP-2 analog (glepaglutide) has a fast onset and long duration of intestinotrophic action with similar profile in both genders and species (rat and dog).
As wine polyphenols were show n to possess many positive effects in mammals, including improvement of vascular function, this study investigated the effect of the Slovak Alibernet red wine extract (AWE) on blood pressure and vascular function in young normotensive Wistar-Kyoto (WKY) and spontaneously hypertensive (SHR) rats. Six weeks old, male, WKY and SHR were treated with AWE for three weeks at the dose of 24.2 mg/kg/day. Blood pressure (BP), determined by tail-cuff plethysmography, was significantly elevated in SHR vs. WKY and AWE failed to affect it. Lipid peroxidation was evaluated by determination of thiobarbituric acid-reactive substances. Vascular function was assessed in rings of the femoral artery using Mulvany-Halpern’s myograph. Maximal endothelium-dependent acetylcholine (ACh)-induced rela xation was reduced in control SHR vs. WKY rats by approximatel y 9.3 %, which was associated with a significant decrease of its NO-independent component. AWE failed to affect maximal AC h-induced relaxation, both its NO-dependent and independen t components, compared to controls of the same genotype. AWE however reduced lipid peroxidation in the left ventricle of both WKY and SHR and in the liver of SHR. Arterial compliance (C) is a complex parameter influencing ventricular-arterial coupling depending on structural (arterial wall remodeling) and functional (blood pressure, smooth muscles tone) changes. Based on Windkessel model, C can be calculated as the ratio of a time constant Tau characterizing diastolic blood pressure decay and total peripheral resistance (TPR). The aim of this study was to assess changes of C in the context of systolic arterial pressure (SAP) perturbations during four physiological states (supine rest, head-up tilt, supine recovery, mental arithmetic). In order to compare pressure independent changes of C a new index of C120 was proposed predicting C value at 120 mm Hg of SAP. Eighty-one healthy young subjects (48 f, average age 18.6 years) were examined. Hemodynamic parameters were measured beat-to-beat using volume-clamp photoplethysmographic method and impedance cardiography. We observed that C was strongly related to SAP values on the beat-to-beat time scale. Interestingly, C120 decreased significantly during stress phases. In conclusion, potential changes of SAP should be considered when measuring C. Arterial compliance changes in the opposite direction to TPR pointing towards influence of vascular tone changes on its value.
The aim of this study was the histochemical characterization of NADPH diaphorase-positive neuronal pools in the rabbit lumbosacral segments using a model of single, repeated and multiple sublethal spinal cord ischemia. Following a single 8-min sublethal spinal cord ischemia and 1-hour reperfusion, the staining of NADPH diaphorase-exhibiting neurons in the dorsal horn, pericentral region, dorsal gray commissure and sacral parasympathetic nucleus was comparable with the control sections. In contrast to the foregoing sublethal ischemia, a regionally different somatic NADPH diaphorase (NADPHd) staining was found after multiple sublethal spinal cord ischemia. Whereas an almost complete loss of the staining of large NADPHd-exhibiting somata in the pericentral region was detected, the staining of the NADPHd-exhibiting neuronal pools in the deep dorsal horn and sacral parasympathetic nucleus was fully preserved. Physiological responses of two duckweed species, Lemna gibba and Lemna minor, to hexavalent chromium [Cr(VI)] were studied in axenic cultures using short-term (48 h) treatments by K2Cr2O7 (0-200 μM). Chlorophyll (Chl) fluorescence parameters and photosynthetic pigment composition of plants were screened to determine the effects of Cr(VI) exposures. The two duckweed species exhibited different sensitivity in the applied Cr(VI) concentration range. Chl fluorescence parameters of dark-adapted and light-adapted plants and electron transport inducibility were more sensitive to Cr(VI) in L. minor than in L. gibba. We also found fundamental differences in quantum yield of regulated, Y(NPQ), and nonregulated, Y(NO), non-photochemical quenching between the two species. As Cr(VI) concentration increased in the growth medium, L. minor responded with considerable increase of Y(NPQ) with a parallel significant increase of Y(NO). By contrast, in L. gibba only 200 μM Cr(VI) in the growth medium resulted in elevation of Y(NPQ) while Y(NO) remained more or less constant within the regarding Cr(VI) concentration range during 48 h. Photosynthetic pigment content did not change considerably during the short-term Cr(VI) treatment but decrease of Chl a/b and increase of Car/Chl ratios were observed in good accordance with the changes in Chl fluorescence parameters. The data suggest that various duckweed species respond with different sensitivity to the same ambient concentrations of Cr(VI) in the growth medium, and presumably to other environmental stresses too, which may have an influence on their competitive relations when heavy metal pollution occurs in aquatic ecosystem. and V. In our previous study, healthy volunteers showed considerable short-term dynamics and patterns of the coherence of high time resolution between respiratory movements (RESP), heart rate fluctuations (HRF), and arterial blood pressure fluctuations (BPF). These are physiological indicators of autonomic short-term coordination mediated mainly by the brainstem which could be impaired in severe brain disorders. We hypothesized a direct or indirect impairment of these functions by these disorders and examined these patterns in 16 patients suffering from severe brain disorders. We calculated partial and ordinary coherence sequences and found almost the same patterns of coherence sequences as in healthy volunteers, but a distinctly reduced frequency of pattern incidence in patients (2.8*1.5/10 min/patient and 9.5*2.8/10 min/subject, P<0.05). Furthermore, there is a significantly smaller frequency of HRF-related patterns in patients with poor outcome, compared with those in patients with good outcome (1.8*0.8/10 min/patient and 4.5*2.7/10 min/patient, P<0.05). The possible linear short-term coordination between respiratory movements (RESP), heart rate fluctuations (HRF), and arterial blood pressure fluctuations (BPF) in conscious human beings has not yet been investigated because of the restricted time resolution of conventional time series analysis. At present, this short-term dynamics as an expression of relative coordination can be quantified by newly developed adaptive autoregressive modeling of time series using Kalman filtering. Thus, in 6 conscious healthy volunteers, RESP, HRF, and BPF were recorded during 10 min in the supine position, at rest and during paced breathing. A considerable part of calculated ordinary and partial coherence sequences of short-term resolution between RESP and HRF, RESP and BPF, and partially between HRF and BPF showed patterns varying in time that could be correlated to changes between gradual coordinations (coherence changing between 0.40 and 0.95). They were more seldom complete or absent. There were mostly opposite changes between partial coherence sequences RESP-HRF/BPF and RESP-BPF/HRF demonstrating competitive behavior between these coordinations. Paced breathing did not essentially affect any observed characteristics. Therefore, these coherence dynamics are not essentially dependent on voluntary breathing movements. We conclude that to a different extent these linear and changing couplings between RESP, HRF, and BPF in conscious human beings exhibit properties of short-term complete and more frequently gradual coordinations showing dynamics that can not be determined by conventional methods., U. Environmental factors that influence stomatal conductance (gs) interact through a complex network of signal transduction and have therefore highly interdependent effect.
In the present study we examined how plant water status affects stomatal sensitivity to the change of CO2 concentration ([CO2]). We investigated the short-term dynamic of stomatal response to a sudden [CO2] increase (from 400 to 700 µmol(CO2) mol-1) in maize supplied with different amounts of water (resulting ψw = -0.35, -0.52 and -0.75 MPa). Gas exchange measurements were performed in short logging intervals and the response was monitored under two different levels of water vapour pressure deficit (VPD) of 1 and 2 kPa in order to observe the impact of air humidity. Generalized logistic curves were fitted to standardized stomatal response data, which enabled us to objectively estimate the level (relative decrease of g s) and the dynamics of the response.
Soil water stress and high VPD significantly decreased relative stomatal closure in response to [CO2] rise, but simultaneously accelerated stomatal response to [CO2], as revealed by shorter half life (t1/2). VPD significantly affected the response of well-watered plants. In contrast, a fast stomatal reaction of water-deprived plants was predetermined by a low xylem water potential (ψw) of the leaf and the influence of air humidity was minor. and J. Hladnik ... Despite the increasing interest in applying composts as soil amendments worldwide, there is a lack of knowledge on short-term effects of compost amendments on soil structural and hydraulic properties. Our goal was to study the effect of compost and vermicompost-based soil amendments on soil structure, soil water retention characteristics, aggregate stability and plant water use efficiency compared to that of mineral fertilizers and food-waste digestate and examine if these effects are evident within a short time after application. We set up a pot experiment with spring wheat using a sandy and a loamy soil receiving either mineral fertilizer (MF); dewatered digestate from anaerobic digestion of food waste (DG), vermicomposted digestate (VC_DG); sewage sludge-based compost (C_SS) and sewage sludge-based vermicompost (VC_SS). We then monitored and calculated the soil water balance components (irrigation, outflow, evaporation, transpiration, and soil water content). At harvest, we measured shoot biomass, soil texture, bulk density, water retention characteristics and aggregate stability. The irrigation use efficiency (IE) and the plant water use efficiency (WUE) were calculated for each treatment by dividing the transpiration and the dry shoot biomass with the amount of water used for irrigation, respectively. For the sandy soil, we used X-Ray computed tomography to visualise the pore system after applying organic amendments and to derive metrics of the pore-network such as its fractal dimension, imaged macroporosity and critical pore diameter. X-Ray tomography indicated that composting and vermicomposting resulted in more complex and diverse porous system and increased soil macroporosity. The increased fractal dimensions also indicated that compost and vermicompost can contribute to structure formation and stabilization within a short time after their application. Despite the small application rate and short incubation time, the application of organic amendments to the two different soil types resulted in improved soil water holding capacity and water use efficiency. Composting and vermicomposting appeared to have the best effect at reducing the irrigation demand and evaporation losses and increasing the water use efficiency of the plant, likely through their effect on soil structure and the pore-size distribution.
