Chlorophyll (Chl) a fluorescence parameters and rapid light curves of soybean [Glycine max (L.) Merrill] were measured by pulse amplitude modulation fluorometry. Measurements were taken during different stages of soybean growth under field conditions with 20% enhancement in ultraviolet-B (UV-B) radiation. Results showed that supplemental UV-B radiation decreased Chl contents by 5.5% (P=0.048), 8.7% (P=0.046), and 10.5% (P=0.005) in seedling, in branching-flowering, and in pod-setting stages, respectively. In the branching-flowering and pod-setting stages, maximum quantum yield of photosystem (PS) II photochemistry (Fv/Fm) decreased by 6.1% (P=0.001) and 3.0% (P=0.009), respectively. Supplemental UV-B radiation significantly decreased the effective quantum yield (Y). The photosynthetic capacity at light saturation (Pm) also decreased in both the seedling and branching-flowering stages by 28.9% (P=0.007) and 15.5% (P=0.041), respectively. However, Y and Pm showed no significant difference in the trefoil and pod-setting stages with and without the UV treatment. The light saturation parameter (E k) decreased by 21.1% (P=0.000) and 23.2% (P=0.029) in the trefoil and seedling stages, respectively. Moreover, the initial slope (α) decreased by 21.1% (P=0.001) in the branching-flowering stage. branching-flowering stage decreased significantly under UV-B treatments. Gas exchange, chlorophyll (Chl) fluorescence, and contents of photosynthetic pigments, soluble proteins (ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO), and antioxidant enzymes were characterized in the fully expanded 6th leaves in rice seedlings grown on either complete (CK) or on nitrogen-deficient nutrient (N-deficiency) solutions during a 20-chase period. Compared with the control plants, the lower photosynthetic capacity at saturation irradiance (Pmax) was accompanied by an increase in intercellular CO2 concentration (Ci), indicating that in N-deficient plants the decline in Pmax was not due to stomatal limitation but due to the reduced carboxylation efficiency. The fluorescence parameters ΦPS2, Fv'/Fm', electron transport rate (ETR), and qP showed the same tendency as Pmax in N-deficient plants. Correspondingly, a higher qN paralleled the rise of the ratio of carotenoid (Car) to Chl contents. However, Fv/Fm was still diminished, suggesting that photoinhibition did occur in the photosystem 2 (PS2) reaction centres. In addition, the activities of antioxidant enzymes on a fresh mass basis were gradually lowered, leading to the aggravation of membrane lipid peroxidation with the proceeding N-deficiency. The accumulation of malonyldialdehyde resulted in the lessening of Chl and soluble protein content. Analyses of regression showed PS2 excitation pressure (1 - qP) was linearly correlated with the content of Chl and inversely with soluble protein (particularly RuBPCO) content. There was a lag phase in the increase of PS2 excitation pressure compared to the decrease of RuBPCO content. Therefore, the increased excitation pressure under N-deficiency is probably the result of saturation of the electron transport chain due to the limitation of the use of reductants by the Calvin cycle. In the mammalian autonomic nervous system, tonic and phasic neurons can be differentiated on firing patterns in response to long depolarizing current pulse. However, the similar firing patterns in the somatic primary sensory neurons and their functional significance are not well investigated. Here, we identified two types of neurons innervating somatic sensory in rat dorsal root ganglia (DRG). Tonic neurons fire action potentials (APs) in an intensity-dependent manner, whereas phasic neurons typically generate only one AP firing at the onset of stimulation regardless of intensity. Combining retrograde labeling of somatic DRG neurons with fluorescent tracer DiI, we further find that these neurons demonstrate distinct changes under inflammatory pain states induced by complete Freund’s adjuvant (CFA) or bee venom toxin melittin. In tonic neurons, CFA and melittin treatments significantly decrease rheobase and AP durations (depolarization and repolarization), enhance amplitudes of overshoot and afterhyperpolarization (AHP), and increase the number of evoked action potentials. In phasic neurons, however, the same inflammation treatments cause fewer changes in these electrophysiological parameters except for the increased overshoot and decreased AP durations. In the present study, we find that tonic neurons are more hyperexcitable than phasic neurons after peripheral noxious inflammatory stimulation. IgA nephropathy (IgAN) is the most common glomerulonephritis worldwide. Current studies have shown that the Th17/Treg immune balance may be involved in the occurrence of IgAN, but the exact mechanism is still unclear. Indoleamine 2,3-dioxygenase (IDO) is an enzyme that catalyses degradation of tryptophan (Trp) through the kynurenine (Kyn) pathway; it can control inflammation and immune response by inducing Trp starvation. IDO may be a key molecule in regulating the Th17/Treg immune balance. However, it is not clear whether IDO is involved in the IgAN disease occurrence by regulating the Th17/Treg immune balance. In this study, an IgAN mouse model was established. The mice were intraperitoneally inoculated with IDO inhibitor 1-MT or agonist
ISS-ODN to observe whether the IDO signalling pathway participates in the occurrence and development of IgAN by regulating the Th17/Treg immune balance. The results showed that IDO inhibitor 1-MT significantly increased renal injury and glomerular IgA accumulation and up-regulated Th17/Treg and Th17-related cytokine expression in IgAN mice, while ISS-ODN significantly decreased renal injury and glomerular IgA

We investigated the role of cyclic electron flow around photosystem 1 (CEF1) and state transition (ST) in two soybean cultivars that differed in salt tolerance. The CEF1 and maximum photochemical efficiency (Fv/Fm) were determined under control and NaCl (50 mM) stress and the NaCl-induced light-harvesting complex 2 (LHC2) phosphorylation in vitro was analysed in light and dark. NaCl induced the increase of CEF1 more greatly in wild soybean Glycine cyrtoloba (cv. ACC547) than in cultivated soybean Glycine max (cv. Melrose). The Fv/Fm was reduced less in G. cyrtoloba than in G. max after 10-d NaCl stress. In G. cyrtoloba, the increase of CEF1 was associated with enhancement of LHC2 phosphorylation in thylakoid membrane under both dark and light. However, in G. max the NaCl treatment decreased the LHC2 phosphorylation. Treatment with photosynthetic electron flow inhibitors (DCMU, DBMIB) inhibited LHC2 phosphorylation more in G. max than in G. cyrtoloba. Thus the NaCl-induced up-regulation in CEF1 and ST might contribute to salt resistance of G. cyrtoloba. and K. X. Lu ... [et al.].
The combined effects of water stress (WS) and low irradiance (LI) on growth, photosynthesis, osmotic adjustment, and lipid peroxidation were studied in dragon spruce (Picea asperata Mast.) seedlings grown under two water treatments (well watered, 100 % of field capacity, and water stressed, 30 % of field capacity) and two irradiances (HI, 100 % of full sunlight and LI, 15 % of full sunlight). WS reduced growth, chlorophyll (Chl) a and b contents, net photosynthetic rate, transpiration rate, stomatal conductance, and effective quantum yield of photosystem 2 (Y) but increased free proline and malondialdehyde contents. LI increased Chl contents and decreased Y, photochemical quenching (qP), and non-photochemical quenching (qN) under both water treatments. To uncover adaptation capacities of two flooding-tolerant plant species, Pterocarya stenoptera (a native species) and Pinus elliottii (an exotic species from southeastern USA), to alternating submergence and drought, we investigated their physiological and growth responses to water stress. Water treatments, including control, continuous flooding (CF), and periodic flooding and drought (PF), were applied to seedlings in order to simulate water level fluctuation in the hydrofluctuation zone of the Three Gorges Reservoir Region. Results showed that net photosynthetic rate (P N), stomatal conductance, and intrinsic water-use efficiency of both plant species were negatively affected under CF and PF compared with the corresponding controls. The P N of both species under PF was comparable to that under CF. At the end of the experiment, the ratio of intercellular to ambient CO2 concentration was not statistically different between water treatments, while that of P. elliottii was significantly higher than that of P. stenoptera. Although P. stenoptera formed lenticels under flooding conditions, P. elliottii seedlings allocated more mass to leaves and increased the relative growth rate of height to enhance the photosynthetic efficiency. Our results illustrated that P. stenoptera and P. elliottii seedlings developed different adaptive strategies in response to flooding, both CF and PF.
