Pulmonary pathology is common in HIV-infected individuals, but the possible role of the parasitic protist Toxoplasma gondii (Nicolle et Manceaux, 1908) is not completely known. The present account reports result of a retrospective cohort study. Medical cards of 907 HIV-positive people, which included 120 deceased patients, were analysed. During a three-year follow-up, the pulmonary pathology was diagnosed in 306 patients (33.7 ± 1.6%): pneumocystis pneumonia in 124 (13.7 ± 1.1%), primary pulmonary tuberculosis in 113 (12.5 ± 1.1%), bacterial pneumonia in 58 (6.4 ± 0.8%) toxoplasmosis pneumonia in two (0.2 ± 0.2%), and others. All patients were divided into two cohorts: 531 individuals seropositive for T. gondii and 376 seronegative ones. It has been found out that general lung pathology is more common in patients with seropositivity to T. gondii than in seronegative ones (43.3 ± 2.2% vs. 20.1 ± 2.0%, p < 0.001). The diagnosis of pneumocystis pneumonia was made ten times more often in the cohort of seropositive patients than in the cohort of seronegative ones (21.9 ± 1.8% vs. 2.1 ± 0.7%, respectively, p < 0.001) and in deceased patients of these cohorts it was 5.5 times more (45.1 ± 5.9% vs. 8.2 ± 3.9, respectively, p < 0.001). In patients with fatal outcome and seropositivity to T. gondii, the incidences of pneumocystis pneumonia increased by 23.2% (p < 0.001) and bacterial pneumonia by 12.4% (p < 0.05), whereas in seronegative individuals only pulmonary tuberculosis increased by 13.1% (p < 0.05) сompared with corresponding whole cohorts. Pearson's contingency coefficient showed the mean strength association between infection with T. gondii and incidence of pneumocystis pneumonia both in whole cohort (C = 0.272) and in patients with fatal outcomes (C = 0.368). In сonclusion, significantly increasing rate of pneumocystis pneumonia in patients with HIV/AIDS and T. gondii infection can be caused by certain synergism between T. gondii and Pneumocystis jirovecii and in some cases overdiagnosis pneumocystis pneumonia due to undiagnosed toxoplasmosis pneumonia.
Ichthyophthirius multifiliis Fouquet, 1876, a ciliate parasite, is a cosmopolitan and problematic parasite of cultured freshwater fish. Each geographical isolate of I. multifiliis has variations in life cycle timing under different abiotic water conditions, such as temperature and salinity. We assessed the effects of salinity and temperature on the development and the preferred settlement site of a temperate Australian isolate of I. multifiliis. The time until theront release was significantly different between each temperature; development time was longest at 5 °C with a mean time of 189 h and decreased to a mean time of 11.7 h at 30 °C. At 5 °C our isolate produced a mean of 267 theronts per tomont, which increased to a mean of 493 theronts at 25 °C and reduced to a mean of 288 theronts at 30 °C. Theront length showed an inverse relationship to temperature; mean length was 62 μm at 5 °C and 41 μm at 30 °C. Our isolate reproduced faster at all temperatures and a greater sensitivity to salinity than all reported profiles for temperate isolates. Parasite abundance was highest on the dorsal region of the fish. An accurate understanding of temperature-life cycle information and optimal region to sample for surveillance will aid in the development of specific management plans for the Australian isolate of I. multifiliis, facilitating the strategic timing of treatments., James M. Gnathostomes are of interest because of their unique appearance and medical importance. Among 13 valid species of the genus Gnathostoma Owen, 1836 (Nematoda: Spirurida), two species, G. doloresi Tubangui, 1925 and G. hispidum Fedtschenko, 1872, are parasites of pigs but their larvae can infect humans to cause gnathostomiasis. In this study, we collected adults of Gnathostoma sp. from the stomach of domestic pigs (Sus scrofa domesticus Linnaeus) from Dien Bien Province, northern Vietnam. Morphologically, nematodes found here are most similar to G. doloresi with a slight difference in the spicules of males. In contrast, they are genetically distinct from G. doloresi and other species of Gnathostoma in both ITS2 and cox1 sequences. The findings of the present study suggest that specimens of Gnathostoma sp. found in Dien Bien are likely a new species and emphasise the need of further studies on the taxonomy and phylogenetic relationship of species of Gnathostoma. We encountered two cases of infection with large female nematodes of the genus Philometra Costa, 1845 in the body cavity of a map puffer Arothron mappa (Lesson) caught off Okinawa, Japan, and a blackspotted puffer Arothron nigropunctatus (Bloch et Schneider) caught off Queensland, Australia, both reared in aquariums in Japan. No morphological difference was observed between the nematodes from A. mappa and A. nigropunctatus. We identified the nematodes as Philometra pellucida (Jägerskiöld, 1893) based on their morphology. The sequences of the nematodes from both hosts were identical to each other (1,643 bp) and formed a clade with other 17 nematodes belonging to the genera Philometra and Philometroides Yamaguti, 1935 with high bootstrap value (bp = 100). It is the first time that the genetic data on P. pellucida are provided. Philometra robusta Moravec, Möller et Heeger, 1992 is synonymised with the former species.
Species of the cnidarian genus Henneguya Thélohan, 1892 (Myxosporea: Myxobolidae) are histozoic parasites commonly found in freshwater and, more rarely, in marine fish. The development of these parasites in fish tissues includes the formation of plasmodia within which occurs the sporogony originating spores with two caudal processes, which are usually randomly distributed within the plasmodia. In this report the authors present some cases of non-random distribution of the spores of six species of Henneguya within their plasmodia. Two different patterns of non-random distribution were found based on a literature survey. These patterns and their origin are discussed. Apparently this non-random distribution of the spores is due to both internal and external factors.
Abiotic and biotic factors determine success or failure of individual organisms, populations and species. The early life stages are often the most vulnerable to heavy mortality due to environmental conditions. The deer ked (Lipoptena cervi Linnaeus, 1758) is an invasive insect ectoparasite of cervids that spends an important period of the life cycle outside host as immobile pupa. During winter, dark-coloured pupae drop off the host onto the snow, where they are exposed to environmental temperature variation and predation as long as the new snowfall provides shelter against these mortality factors. The other possible option is to passively sink into the snow, which is aided by morphology of pupae. Here, we experimentally studied passive snow sinking capacity of pupae of L. cervi. We show that pupae have a notable passive snow sinking capacity, which is the most likely explained by pupal morphology enabling solar energy absorption and pupal weight. Phylogenetic relationships within the suborder Trypanosomatina were inferred from the kinetoplast DNA minicircle conserved region sequences. Trees built using distancc-matrix (Neighbor-Joining) and maximum parsimony methods showed that the minicircle conserved regions (CRs) provide a sensitive and specific molecular marker suitable for phylogenetic analyses of subspecies and strains of trypanosomalid flagellates, as testified by the subdivision of the genus Leishmania into the subgenera Leishmania, Viannia and Sauroleishmania. However, since Phytomonas and monogenetic parasites of insects represent the earliest diverging groups, the CRs do not seem to be useful for inference of relationships among major lineages of the order Kinetoplastida.
Some free-living amoebae are a potential threat to human health. The best known species are those of the genus Acanthamoeba Volkonsky, 1931, which cause Acanthamoeba keratitis, granulomatous amoebic encephalitis and other forms of tissue inflammation. The aim of the present study was to search for potential pathogenic genotypes of free-living amoeba in the sand in children's playgrounds. Our results confirmed that free-living amoebae were present in all examined playgrounds. Sequences of the 18S rDNA have shown that all isolated potentially pathogenic strains of amoebae belong to genotype T4 of Acanthamoeba. The potential pathogenicity of isolates was confirmed on mice. Faecal samples from 162 wild animals were collected from 32 distinct sites of Łęczyńsko-Włodawskie Lakeland (eastern Poland). The presence of Giardia duodenalis (Stiles, 1902) was assessed by a Direct Fluorescence Assay (DFA) and by Polymerase Chain Reaction (PCR) and sequencing of a fragment of the beta-giardin gene. DFA showed the presence of cysts of G. duodenalis in 12 of 162 faecal samples (7%), namely in four wild boars (15%), four foxes (19%), two roe deer (4%), and two wolves (29%). PCR identified 34 of the 162 (21%) samples as positive, including 11 wild boars (41%), five red deer (18%), 11 roe deer (23%), four moose (17%), two wolves (29%) and a single sample from the European badger. Thus, PCR detected a significantly higher number of infection than DFA (P = 0.0005). However, 14 of 34 PCR products could not be sequenced because of their insufficient amount; the low number of cysts, poor conservation of the faeces or presence of PCR inhibitors may have contributed to weak DNA amplification. Sequence analysis of the remaining 20 products showed the presence of assemblage B in wild boars, red deer and roe deer, whereas samples from wolves were identified as assemblage D. This is the first detection of assemblage B in wild boars and deer. In the present study, we critically revised the recently proposed classification of the subfamily Leishmaniinae Maslov et Lukeš in Jirků et al., 2012. Agreeing with erection of the genus Zelonia Shaw, Camargo et Teixeira in Espinosa et al., 2017 and the subgenus Mundinia Shaw, Camargo et Teixeira in Espinosa et al., 2017 within the genus Leishmania Ross, 1908, we argue that other changes are not well justified. We propose to: (i) raise Paraleishmania Cupolillo, Medina-Acosta, Noyes, Momen et Grimaldi, 2000 to generic rank; (ii) create a new genus Borovskyia gen. n. to accommodate the former Leptomonas barvae Maslov et Lukeš, 2010 as its type and only species; (iii) leave the subfamily Leishmaniinae as originally defined, but establish two infrafamilies within it: Leishmaniatae infrafam. n. and Crithidiatae infrafam. n., Alexei Y.
