The Northern pine processionary moth, Thaumetopoea pinivora (Treitschke, 1834) shows a highly scattered distribution with fragmented populations across Europe. A previous study exploring the postglacial history of T. pinivora defined it as a cold-tolerant relict species and concluded that a progressive reduction of suitable habitats after the postglacial expansion from refugia in the southern Iberian peninsula best explained the distribution and genetic structure of populations of this species. However, recent records, both by us and others, challenge this view. Surprisingly, some of the newly found populations from southern Spain use black pine, Pinus nigra J.F. Arnold as a host plant despite the fact that the typical host of the species, Scots pine, Pinus sylvestris L. occurs in the area. We provide genetic data for one of these recently found southern populations where the larvae feed on P. nigra, and compare this with previously published data on individuals collected on P. sylvestris. This data reveals that populations from different host trees are no more genetically differentiated than populations sharing the same host plant. The findings of a wider diet breadth open the way to widen the search for the still unidentified glacial refugium of T. pinivora, and as such may contribute to a better understanding about how the species has spread across Europe., José A. We have assessed the phylogenetic status of the Leishmania genome project Friedlin reference strain by MLEE and multiprimer RAPD including a set of 9 stocks representative of the main Leishmania species and of the whole genetic diversity of the Leishmania genus. To our knowledge, the detailed genetic characterization of the Friedlin strain has never been published before. As previously recorded (Tibayrenc et al. 1993), MLEE and RAPD data gave congruent phylogenetic results. The Friedlin reference strain was definitely attributed to Leishmania (Leishmania) major Yakimoff et Schokhor, 1914. Five specific RAPD patterns made it possible to distinguish between the Friedlin strain and the 2 other L. (L.) major stocks included in the study. Various specific MLEE and RAPD characters permitted to distinguish between the Leishmania species included in the study. All these characters are usable to detect accidental laboratory mix-ups involving the Friedlin reference strain. In confirmation with previous studies involving a more limited set of genetic markers, the general genetic diversity of the Leishmania genus proved to be considerable. It must be made clear that only one strain cannot be considered as representative of the whole genetic variability of the genus Leishmania. In the future, it is therefore advisable to complement the results obtained in the framework of the Leishmania genome project with data from other strains that should be selected on a criterion of important genetic differences with the Friedlin strain.
During the past four decades, several species of aphidophagous Coccinellidae became established in North America, including Coccinella septempunctata, Harmonia axyridis, Hippodamia variegata, and Propylea quatuordecimpunctata. After their
establishment, unknown circumstances favoured a rapid increase in population densities and distribution of H. axyridis and C. septempunctata at localities hundreds and thousands of kilometers from their release sites. Propylea quatuordecimpunctata and Hippodamia
variegata have spread more slowly after becoming established in northeastern North America. Comparative studies based
upon allozyme variation in these four introduced species and in six native North American species of ladybird beetles revealed no
significant differences in genetic diversities. Genetic variation, assessed by allelic diversity and heterozygosity, was uncorrelated
with the establishment and spread of these predatory species in North America. All ladybirds studied show a remarkable degree of
dispersion with little detectable population subdivision.
Physical disturbance by large herbivores can affect species diversity at the community level and concurrently genetic diversity at the species level. As seedling establishment is rarely observed in clonal plants, short-term experiments and demographic studies are unlikely to reveal the response of clonal plants to disturbances. A long-term (30-year) field experiment and the availability of molecular markers allowed us to investigate the clonal structure of populations of Elytrigia atherica subjected to different management regimes. The long-term field study provided us with five replicated blocks that had been subjected to three different management regimes, grazing by cattle, mowing and abandonment. In this study we examined the effects of herbivore grazing and mowing on clonal richness and genetic diversity of populations in salt marshes using multilocus microsatellite genotypes. In addition, phenotypic traits and spatial positions of E. atherica ramets were determined for 20 samples in a 5 × 10m plot in each of the blocks. Abundance and phenotypic traits were affected by the management regimes, resulting in a higher abundance in abandoned fields and plants having shorter and narrower leaves in managed fields. Biomass removal did affect the clonal structure of populations and increased the genetic diversity compared to that in abandoned fields. However, no distinct difference was found between the two management regimes, mowing and grazing. Although seedling recruitment has rarely been observed, the present study shows that such rare events have occurred within the populations studied. Thus, molecular tools can greatly increase our understanding of vegetation dynamics and processes within populations growing under different conditions.
The utility of inter simple sequence repeat-PCR (ISSR-PCR) assay in the genetic characterisation and elucidation of the phylogenetic relationship of different microsporidian isolates infecting tropical tasar silkworm, Antheraea mylitta Drury, is demonstrated. A total of 22 different microsporidians collected from the diseased tasar silkworms from Jharkhand state of India were analysed using morphological characters and ISSR-PCR. Observations spores under phase contrast microscope revealed oval to elongate in shape with length ranging from 3.8 μm to 5.1 μm and width from 2.6 μm to 3.3 μm. All the microsporidian isolates except MIJ-1gC showed gonadal infection and transovarial transmission in infected tasar silkworms. Fourteen out of 20 ISSR primers tested generated reproducible profiles and yielded a total of 281 fragments, of which 273 were polymorphic (97%). The degree of banding pattern was used to evaluate genetic distances and for phylogenetic analysis. We examined the phylogeography and the variation of the mitochondrial DNA (mtDNA) cytochrome c oxidase subunit 1 gene (cox1) of the Chinese liver fluke Clonorchis sinensis (Cobbold, 1875) in two geographic localities in the Russian southern Far East and compared them with those from different geographical regions (China, Korea, Japan and Vietnam). The Russian samples differed from those of the other regions in haplotype frequencies, haplotype and nucleotide diversities, and AT/GC ratios. Only 4 of the 18 haplotypes were common to Russian and Chinese samples, and two haplotypes were common to Russia and other regions. The intraspecific genetic distances ranged from 0 to 1.58% for the entire dataset studied and from 0 to 1.25% among the samples from Russia. Phylogenetic trees revealed no significant genealogical clades of samples corresponding to sampling localities and no strong isolation by distance was estimated with Mantel test. Neutrality test analysis suggested a relatively recent population expansion for C. sinensis, whereas goodness-of-fit tests indicated deviation from the strict model of uniform expansion. Therefore, the sequences of the mtDNA cox1 gene provide useful genetic markers for evaluating intraspecific diversity and generating phylogeographic reconstructions for this fish-borne trematode.
The genetic structure and phenotypic diversity of two populations of Cheilosia aff. longula (Diptera: Syrphidae) in Lapland, Finland, were examined using DNA sequencing, protein electrophoresis, and geometric morphometrics. The morphological identification of the species were verified using partial sequences of mitochondrial cytochrome c oxidase subunit I (COI mtDNA), and the nuclear ribosomal internal transcribed region 2 (ITS2 rDNA), and comparing the corresponding sequences of Cheilosia aff. longula and the closely related C. longula. Two and three haplotypes of the genes COI mtDNA and ITS2 rDNA were identified in the two populations. Analysis of 12 isozyme loci showed an extremely low genetic variability in the populations originating from Utsjoki and Kevo. Discriminant analysis combined with canonical variate analysis revealed inter-population divergence in wing shape. Variation among genetically diverse individuals, both within- and among studied populations was studied, and directional (DA) and fluctuating asymmetry (FA) estimated using landmarks in the framework of geometric morphometrics. It is likely that the documented DA and FA asymmetry in both wing shape and size reflects the developmental instability of the individuals studied. By using Procrustes ANOVA the locations of particular landmarks responsible for the variation in shape were determined. The decomposition of the components of variance accorded to each landmark showed that the landmarks differed in the percentage of variation they accounted for (DA, FA and variation among individuals). In the discussion the implications of the reduced genetic diversity and asymmetry in wing traits for conservation is considered.
In this study, we report the development of a set of 15 polymorphic microsatellite markers for the box tree moth, Cydalima perspectalis (Walker), a highly invasive insect in Europe causing significant damage to natural and ornamental Buxus trees. The markers were characterized for four distant populations in both its native (China, two populations) and invasive ranges (Czech Republic and Turkey, one population each). The number of alleles ranged from 2 to 12. No marker significantly deviated from the Hardy-Weinberg equilibrium for all the populations sampled. Here we describe 16 novel microsatellite loci in the genome of the dragonhead sap beetle Thymogethes norvegicus, which were identified using a next-generation sequencing approach. In 40 dragonhead sap beetles from a population in southern Norway, we found 2-4 alleles per locus. Expected heterozygosity ranged from 0.18-0.65. Three of the loci showed significant deviation from Hardy-Weinberg equilibrium. After correcting for multiple tests, one pairwise locus combination displayed significant linkage disequilibrium. The longhorn crazy ant, Paratrechina longicornis (Latreille), is a ubiquitous agricultural and urban pest that has invaded most tropical and subtropical regions. Although P. longicornis has been found worldwide for more than a century, the genetic structure, origin, and invasion history of this species have not yet been extensively studied, partially because of the limited number of genetic markers currently available. In the present study, we developed 36 polymorphic microsatellite markers for P. longicornis and characterized these markers by genotyping P. longicornis workers from 74 colonies in East and Southeast Asia. All loci were polymorphic, with the number of alleles per locus ranging from 3 to 18 (8.5 on average). Extremely high levels of heterozygosity were found in all populations, suggesting that workers are invariably produced from the mating of divergent queen and male lineages. Queens and males possess non-overlapping allele size ranges at 18 loci, indicating the potential resolving power of the subset of markers in inferring the history of queen and male lineages. Genetic differentiation among three studied populations was low yet significant and may likely reflect their close association with human activities. Overall, the new microsatellite markers developed in the present study serve as a practical tool to reconstruct routes of invasion and assess the population genetics of this invasive ant.
