The cotton aphid, Aphis gossypii Glover (Hemiptera: Aphididae), is a serious pest of citrus in northern Iran, both because of the damaged caused by its feeding and as a vector of several viruses. The genetic structure of populations of A. gossypii on citrus trees at eight localities in Iran was surveyed using seven polymorphic microsatellite loci. Of 240 individuals tested, 142 multilocus genotypes (MLGs) were identified. The presence of multicopy genotypes and negative FIS values revealed that the major mode of reproduction in northern Iran is obligate parthenogenesis. The genotypic diversity of populations ranged between 0.24 and 0.93. Considerable genotypic diversity and a high frequency of unique MLGs, confirmed there is some cyclical parthenogenesis in the region. The analysis of molecular variance revealed high intrapopulation and weak interpopulation genetic differentiation (overall FST = 0.036) among the different populations. The UPGMA dendrogram of eight populations based on Nei's genetic distance indicated two clusters: genotypes from West of Mazandaran and Guilan provinces and those from East of Mazandaran. The same results were also obtained from the STRUCTURE analysis of these populations. This information on the genetic diversity of populations of A. gossypii in northern Iran could be useful for improving the Integrated Pest Management of this aphid.
Brazilian native meliponines are currently threatened by increased human impacts. The assessment of their genetic variation by microsatellite DNA markers can assist in the conservation of populations and help in the planning and establishment of efficient management strategies. The purpose of this study was to develop the first set of microsatellite markers for Melipona fasciculata, selected from partial genome assembly of Illumina paired-end reads. Primer pairs were designed for each detected locus at their flanking regions. Bee samples were genotyped from two different populations of Northeastern Brazil for marker characterization and validation. A total of 17 microsatellite loci displayed polymorphism. Mean HE and HO heterozygosities were 0.453 and 0.536, respectively. PIC across all loci ranged from 0.108 to 0.714. A genetic diversity analysis revealed high values for population differentiation estimates (FST = 0.194, RST = 0.230, and Dest = 0.162) within the investigated region. PCoA and Bayesian clustering showed a separation of the species into two distinct clusters. These microsatellite markers have demonstrated strong potential for population-level genetic studies. Moreover, the preliminary analysis of the genetic diversity in M. fasciculata provides provisional evidence of significant population differentiation between the two studied populations., Geice Ribeiro Da Silva, Isis Gomes De Brito Souza, Fabia De Mello Pereira, Bruno De Almeida Souza, Maria Teresa Do Rego Lopes, Paul Bentzen, Fabio Mendonça Diniz., and Obsahuje bibliografii
Increased plasma cholesterol levels are listed between the major
atherosclerosis risk factors. The final plasma cholesterol levels
result from the interplay between the genetic and environmental
(diet, physical activity) factors. Little is known, how dietary
factors influence epigenetics. We have analyzed, if an overgeneration feeding of rat with cholesterol influences total liverDNA methylation, and if total liver-DNA methylation differ
between the different rat strains (Prague hereditary
hypercholesterolemic rats, Prague hereditary hypertriglyceridemic
rats and Wistar Kyoto rats). The animals were feed with high fat
(additional 5 % over normal capacity) high cholesterol (2 %) diet
for 14 days. DNA methylation in the liver tissue in different
generations was analyzed using the liquid chromatography
coupled with tandem mass spectrometry. We have not observed
any significant changes in total liver-DNA methylation over the
9 generations of animals feed by fat/cholesterol enriched diet.
Additionally, there were no differences in DNA methylation
between different rat strains. In animal model, the dietary
changes (hypercholesterolemic diet) not significantly influence
the total DNA methylation status within the liver.
The study explores a latent structure of perceived impacts on the country. Perceived controllability of the context shapes individual plans and forms resource for dealing with changes. The direct and indirect exposure to global challenges (e.g., terrorist attacks) can change individual views of the controllability of the context and lead to negative psychological consequences. On the basis of the analysis, a set of perceived impacts was included in the investigation – impacts on a person, people of Latvia, its political institutions, and global players and threats – as factors representing different levels of ecological systems. Six hundred forty-seven university students from Riga, Valmiera, and Daugavpils participated in the study. The Future of Country Questionnaire was modified and used for the assessment of perceived impacts on Latvia. Exploratory and confirmatory factor analyses provided evidence for four main factors representing perceived impacts: personal control, political power, globalization, and global threats. The latter factor involved military tension and terrorism. Revealed links between control beliefs confirmed the interconnectedness of sub- and super-ordinated ecological systems. Cooperation with other people can facilitate the sense of controllability of the macro-societal context. and Tato studie zkoumá latentní strukturu vnímaných dopadů na stát. Exploratorní a konfirmační faktorová analýza podpořila čtyři hlavní faktory reprezentující vnímané dopady: osobní kontrolu, politickou moc, globalizaci a globální hrozby, přičemž poslední faktor zahrnoval vojenské napětí a terorismus. Zjištěné souvislosti přesvědčení o kontrolovatelnosti potvrdily provázanost mezi podřízenými a nadřízenými vnějšími systémy. Spolupráce s ostatními lidmi může posílit pocit vlivu (kontroly).
Through their receptors at each level of hypothalamo-pituitarygonadal axis glucocorticoid excess, either endogenous or administered or stress-induced, could affect steroid production in the testis and thus male fertility. The main ways by which glucocorticoids act are as follows: 1) Affecting gonadoliberin and LH synthesis and release through glucocorticoid receptors in hypothalamic neurons and pituitary gonadotropes. 2) By so far not clearly evidenced reduction of the number of LH receptors on the membrane of Leydig cells. 3) By affecting expression and function of steroidogenic enzymes in the testis. 4) By regulation of in situ access of glucocorticoid to its target cells in the testis. 5) By promotion Leydig cell apoptosis. The review provides a survey of physiological and molecular mechanisms staying behind these effects. It does not deal with the clinical effects of glucocorticoid treatment which would substantially exceed the scope of the pater., Richard Hampl, Luboslav Stárka., and Obsahuje bibliografii
Knowledge of blood parasites in Brazilian chelonians is limited, since they have been recorded in only six species. Mesoclemmys vanderhaegei (Bour) is a freshwater turtle with a wide geographic distribution in Brazil, but there is little information about its natural history. This paper reports on a study of the prevalence and infection intensity of a haemogregarine in two subpopulations of M. vanderhaegei. The study was conducted in two areas of Cerrado in the Upper Paraguay River basin in the state of Mato Grosso, Brazil, between November 2010 and August 2013. Ninety-five (53%) of the 179 turtles captured were positive for haemogregarine parasites. The parasitic forms observed were two morphotypes of intraerythrocytic gametocytes. The prevalence differed between size classes, increasing significantly according to the animals' body size. There was no significant difference between prevalence and sex, or between sampling periods. The mean parasite intensity was 9 parasites/2,000 erythrocytes (0.45%) and the parasite population presented an aggregated distribution, with an aggregation index of 19 and discrepancy of 0.772. This is the first record of a hemoparasite in the freshwater turtle M. vanderhaegei., Vinícius C. Goes, Elizângela S. Brito, Rafael M. Valadão, Camila O. Gutierrez, Amanda M. Picelli, Lúcio A. A parasitic nematode from the stomach of Japanese eel Anguilla japonica Temminck et Schlegel in western Japan, previously identified as Heliconema longissimum (Ortlepp, 1922), was morphologically re-examined and compared with the previous descriptions. In addition, the third-stage larva of this nematode is described, based on the specimens of encapsuled larvae found in musculature of two crabs, Hemigrapsus sp. and Perisesarma bidens (De Haan), caught from the upper-intertidal zone of the same locality. As a result of the morphological observation, seven pairs of postcloacal papillae in adult males are confirmed. This matches with the character of H. longissimum, but the shape of the fifth postcloacal papillae differs between the present material and H. longissimum; the former possesses pedunculate papillae in the fifth pair whereas the latter has sessile papillae. Since the pedunculate papillae can be found in the original description and the syntype specimens of H. anguillae Yamaguti, 1935 that has been synonymised with H. longissimum, we thus here resurrect H. anguillae as an accepted species. For the life-cycle of the present nematode, littoral crabs, including the two infected species, are likely to be the source of infections for Japanese eels, acting as intermediate hosts., Hirotaka Katahira, Kazuya Nagasawa., and Obsahuje bibliografii
