Morphometric data from spores of ten myxosporean species were statistically analysed to explore myxosporean intraspecific variation in measurements when obtained from a sample from: (1) the same plasmodium, (2) different plasmodia from the same host and (3) different host individuals and localities. In some cases, significant differences in spore dimensions were found between samples from the same plasmodium, highlighting the difficulty of obtaining representative measurements of myxosporean spore. In addition, significant differences in spore dimensions were found when plasmodia from the same site of infection were compared, suggesting that measurements of spores should come from several different plasmodia of the sampling to increase the reliability of the morphology data. Moreover, significant differences in spore dimensions were observed for most spore dimensions when data were compared between localities. In all cases, there was clear overlap in ranges of dimensions even when means differed significantly. Avian haemosporidians are common vector-borne blood parasites that have been reported in birds all over the world. Investigations of avian haemosporidian parasites are conducted mainly on passerine birds. However, studies that focus on non-passerine avian hosts are important for our understanding of the true diversity, host specificity and genetic variability among these widespread parasites. In the present study, blood samples from a total of 22 raptor birds belonging to two orders, two families and six species from the Central Anatolia Region of Turkey were investigated for three genera of avian haemosporidians (Plasmodium Marchiafava et Celli, 1885, Haemoproteus Kruse, 1890 and Leucocytozoon Sambon, 1908) using a combination of microscopic examination of blood films and nested PCR targeting the parasite mitochondrial cytochrome b gene (cyt-b). In total, six individual raptor birds identified positive for species of Plasmodium or Leucocytozoon and one individual was found co-infected with all three haemosporidian genera. We identified five parasite cyt-b haplotypes, three of which were reported for the first time. Among these, one Plasmodium haplotype is linked to a corresponding morphospecies (P-TURDUS1, Plasmodium circumflexum Kikuth, 1931). All haplotypes were clearly distinguishable in phylogenetic analyses. As one of the first studies to investigate blood parasites from non-passerine birds in the Central Anatolia Region of Turkey, this study provides important new information on the phylogenetic relationships and genetic diversity of avian haemosporidian parasites from raptor birds. The European stag beetle, Lucanus cervus, is recognised as a flagship species for biodiversity conservation. Although the species is widely distributed across Europe declines have led to it being granted protected or endangered status in a number of countries and regarded as "near threatened" by the IUCN. The integration of genetic approaches into conservation efforts is urgently needed but has been impeded to date by the lack of appropriate genetic markers. To provide such a resource the development of the first microsatellite loci for stag beetle is described. Loci were identified using two methods (i) enriched library cloning (ELC) and (ii) Restriction enzyme Associated DNA sequencing (RAD-Seq). Inefficient microsatellite detection using the ELC method suggests that RAD-Seq, or other Next Generation Sequencing based methods, may ultimately be more cost effective for obtaining informative suites of markers for this and other coleopteran species. 18 loci were characterised by genotyping 42 UK specimens collected as prey leftover/roadkill. All loci produced unambiguous genotypes and were polymorphic. Though preliminary, estimates of genetic variability suggest UK populations may be genetically depauperate. In this study, we report the development of a set of 15 polymorphic microsatellite markers for the box tree moth, Cydalima perspectalis (Walker), a highly invasive insect in Europe causing significant damage to natural and ornamental Buxus trees. The markers were characterized for four distant populations in both its native (China, two populations) and invasive ranges (Czech Republic and Turkey, one population each). The number of alleles ranged from 2 to 12. No marker significantly deviated from the Hardy-Weinberg equilibrium for all the populations sampled. Here we describe 16 novel microsatellite loci in the genome of the dragonhead sap beetle Thymogethes norvegicus, which were identified using a next-generation sequencing approach. In 40 dragonhead sap beetles from a population in southern Norway, we found 2-4 alleles per locus. Expected heterozygosity ranged from 0.18-0.65. Three of the loci showed significant deviation from Hardy-Weinberg equilibrium. After correcting for multiple tests, one pairwise locus combination displayed significant linkage disequilibrium. Steroid hormone 20-hydroxyecdysone and the sesquiterpenoid juvenile hormone are the main regulators of insect development; however, it is unclear how they interact in the regulation of metamorphic events. Using the silkworm, Bombyx mori, we show that the juvenile hormone analogue fenoxycarb affects the cascade of ecdysone regulated genes that control the programmed cell death in the larval midgut. Morphological changes that occur during cell death were investigated by studying cross-sections of the midgut stained with hematoxylin and eosin. Apoptosis-specific DNA fragmentation was detected using TUNEL assay. Expression patterns of genes ATG8 and ATG12, which were used as indicators of autophagy, and genes of the ecdysone-regulated gene cascade were examined using real-time quantitative polymerase chain reaction. Fenoxycarb application on day 0 of the 5th larval instar extended the feeding period and postponed programmed cell death in mature larval midgut. This effect was probably due to a delay in ecdysone secretion and associated changes in gene expression were mostly not a direct response to the fenoxycarb. The K13 propeller domain mutation and pfmdr1 amplification have been proposed as useful molecular markers for detection and monitoring of artemisinin resistant Plasmodium falciparum Welch, 1897. Genomic DNA isolates of P. falciparum was extracted from 235 dried blood spot or whole blood samples collected from patients with uncomplicated falciparum malaria residing in areas along the Thai-Myanmar border during 2006-2010. Nested polymerase chain reaction (PCR) and sequencing were performed to detect mutations in K13 propeller domain of P. falciparum at codon 427-709. Pfmdr1 gene copy number was determined by SYBR Green I real-time PCR. High prevalence of pfmdr1 multiple copies was observed (42.5% of isolates). The presence of K13 mutations was low (40/235, 17.2%). Seventeen mutations had previously been reported and six mutations were newly detected. The C580Y was found in two isolates (0.9%). The F446I, N458Y and P574L mutations were commonly detected. Seven isolates had both K13 mutation and pfmdr1 multiple copies. During a survey the occurrence of Kudoa quraishii Mansour, Harrath, Abd-Elkader, Alwasel, Abdel-Baki et Al Omar, 2014, recently identified in the muscles of the Indian mackerel, Rastrelliger kanagurta (Cuvier), a species of Kudoa Meglitsch, 1947 infecting oocytes of mature females of the same host fish was found. The new species, for which the name Kudoa saudiensis sp. n. is proposed, infects oocytes that are enlarged with a whitish colour. The parasite develops in vesicular polysporous plasmodia within the oocyte. Infection occurs with a mean prevalence of 20% (7/35) of examined females. Mature spores are quadratic in shape in apical view, having four equal valves and four symmetrical polar capsules. Fresh spores are 2.4-3.6 µm long (mean ± SD 3.1 ± 0.3 µm), 4.3-5.4 µm (4.7 ± 0.3 µm) wide and 3.4-4.3 µm (3.8 ± 0.3 µm) in thickness and long. The smaller size of the new Kudoa species was the distinctive feature that separates it from all previously described species. Molecular analysis based on the SSU rDNA sequences shows that the highest percentage of similarity of 98.5% was observed with K. ovivora Swearer et Robertson, 1999, reported from oocytes of labroid fish from the Caribbean coasts of Panama. The percentage of similarity was 98% with K. azevedoi Mansour, Thabet, Chourabi, Harrath, Gtari, Al Omar et Ben Hassine, 2013 and 89% with K. quraishii. Phylogenetic analysis of the SSU and LSU rDNA data revealed a consistent of the new species with K. azevedoi and K. ovivora. Setothosea asigna van Eecke is a dominant defoliator pest in oil palm plantations. To control this pest, a generalist predatory bug, Sycanus annulicornis Dohrn, was used as it is easy to rear on several different species of prey. In this study, we evaluated the influence of different prey on the biology and the ability of S. annulicornis to attack and kill the nettle caterpillar pest S. asigna. Based on laboratory rearing, the larvae of Crocidolomia pavonana F. (Lepidoptera: Crambidae) is a suitable prey for both the growth and development of S. annulicornis, as its nymphal development is shorter (74.0 ± 7.3 days) and adult longevity longer (81.0 ± 9.0 days for male and 64.8 ± 12.4 days for female, respectively) than when reared on the larvae of Tenebrio molitor L. (Coleoptera: Tenebrionidae) (44.0 ± 16.7 days for male and 52.6 ± 14.4 days for female). However, S. annulicornis reared on T. molitor larvae attacked 2.0-2.2 larvae of S. asigna per day, which is more than the 1.6-1.7 larvae per day of those reared on C. pavonana, which indicates that the larvae of T. molitor are a more effective diet for rearing S. annulicornis as biocontrol agent for the S. asigna., Abdul Sahid, Wahyu D.
