During a regular veterinary inspection of fishes from Lake Balaton, Hungary, echinostomatid metacercariae (Digenea), with collar spines characteristic of species of the genera Petasiger Dietz, 1909 and Paryphostomum Dietz, 1909, were found in the lateral line scales of a roach Rutilus rutilus (Linnaeus), an apparently unique site. In a subsequent examination of 586 fishes from 20 different species, similar infections were found in 11 species. The infection was virtually restricted to the lateral line scales, other scales being infected only incidentally. These encysted metacercariae had 27 collar spines, including eight larger angle spines and 19 smaller dorsal spines arranged in two rows. Two types of metacercarial cyst were found. One type had a cyst diameter of 138-171 µm × 105-120 µm and three central dorsal spines that were larger than the remainder and tended to resemble the angle spines. The second type of metacercarial cyst had a diameter of 128-157 µm × 105-115 µm and all 19 dorsal spines of the metacercaria were of a similar size. ITS sequences of the second type of metacercaria exhibited a 100% similarity to sequences of two adult Petasiger phalacrocoracis (Yamaguti, 1939) specimens collected from the gut of Phalacrocorax carbo (Linnaeus) in Hungary and to P. phalacrocoracis deposited in the GenBank database. Sequences obtained from two metacercariae of the first type showed a 2.8-2.9 % difference from sequences of the second type of metacercaria and from those of adult specimens of P. phalacrocoracis from cormorants. Based on these results, the second type metacercaria is considered to be a larval stage of P. phalacrocoracis, but the identity of the first type is uncertain. The importance of the institution of family in housing practices has deep historical roots in Greece, and families tend to follow certain housing strategies such as late emancipation from the parental home, intergenerational house transfers and financial support for housing. Providing and maintaining a housing solution for young members is one of the top worries in this geographical region, and it is relieved via intergenerational micro-solidarities. Moreover, today’s crisis and austerity are threatening, through indirect budgetary cuts and rising taxation, the housing well-being of the citizenry which is supported only by family welfare. Nonetheless, the family still constitutes the main shock absorber of social and economic turbulence, but at what price?
Anatomical variations and congenital anomalies of the uterine tubes (UTAVsCAs) are rare conditions, which are often undiagnosed, or accidentally diagnosed upon imaging, laparotomy, laparoscopy for unrelated condition, or during the Cesarean section. UTAVsCAs are often asymptomatic, but their clinical relevance lies in their possibly adverse impact on fertility. Since their rare occurrence, they are usually published as case reports. The most typically described are: agenesis of the uterine tubes (UTs), accessory UT (UT duplication), accessory UT ostium, and paratubal cysts (e.g. the hydatid cyst of Morgagni). UTAVsCAs are classified into an umbrella category of Müllerian duct anomalies (MDAs) which comprises anomalous development of all the organs developing from the paramesonephric (Müllerian) ducts, i.e., UTs, uterus and upper portion of the vagina. Interestingly, most of the classification systems of MDAs discuss solely the uterine and vaginal anomalies, while the UTs are often utterly ignored. This probably originates from the fact that UTs are no longer interesting for many clinicians as they think of UTs as superfluous organs whose function can be easily replaced in the in vitro fertilization (IVF) laboratory. Indeed, the modern reproductive medicine has been helping enormously with the conception of infertile couples. In many instances, the UTs are in fact successfully bypassed and a “test-tube” baby is born. Nevertheless, the UTs are still absolutely unique in providing suitable environment for fertilization and early embryo development - processes that has not been still completely understood. This fact could partially explain why the success rate of IVF is “only” around 30-50 % depending on age. Therefore, the research of the UTAVsCAs is still clinically relevant in the context of reproductive medicine and should not be omitted from research endeavors.
Objectives. The aim of this study was to create a Czech translation of the Perceived Stress Scale (PSS), to assess its psychometric properties on a representative sample of the Czech general adult population, and to compare the original 14-item version (PSS-14) with the shortened 10-item (PSS-10) and four-item (PSS-4) versions. Sample and setting. Two pilot studies were conducted to create the final Czech translation of the scale (n = 365 and n = 420). The final version of the Czech PSS was administered to a sample of the Czech general adult population (n = 1725 of whom 981 were women, M = 44.32,SD = 12.8). The Beck Depression Inventory (BDI-II) or the State-Trait Anxiety Inventory (STAI) were administered alongside the PSS to a part of the sample. A retest measurement after 14 days was conducted (n = 159). Statistical analysis. Using the confirmatory factor analysis, the one-factor, two-factor and bifactor models were compared. The internal consistency, stability in time, and convergent validity of the scale, as well as the known-group differences were assessed. The three versions of the PSS were compared. Results. The confirmatory factor analysis sup-ported the bifactor model of the PSS-14 and PSS-10, and the two-factor model of the PSS-4. All versions of the scale showed good internal consistency and stability in time. There was a moderate to strong positive correlation between the PSS and the BDI-II and STAI. Differences based on age, sex, education level, and situ-ational factors were found. Overall, the PSS-10 showed the best psychometric properties of all three versions of the scale. Study limitation. Výzkumný soubor a metody získávání dat. Při tvorbě českého překladu škály byly provedeny dvě pilotní studie (n = 365 a n = 420). Finální překlad škály byl administrován vzorku běž-né české populace (n = 1725, z toho 981 žen, M = 44,32, SD = 12,8). Části vzorku byla také administrována Beckova škála deprese (BDI-II) nebo Dotazník na měření úzkosti a úzkostlivosti (STAI). Po 14 dnech proběhl retest, kterého se zúčastnilo 159 respondentů. Analýza dat. Pomocí konfirmační faktorové analýzy autoři porovnali jednofaktorový, dvou-faktorový a bifaktorový model škály. Ověřili vnitřní konzistenci škály, její stabilitu v čase, konvergentní validitu a rozdíly mezi známými skupinami. Porovnali vlastnosti jednotlivých forem PSS. Výsledky. Konfirmační faktorová analýza pod-pořila bifaktorový model PSS-14 i PSS-10 a dvoufaktorový model PSS-4. Všechny tři formy škály dosahovaly dobré hodnoty vnitřní konzis-tence i stability v čase. Korelace PSS s BDI-II a STAI ukázaly středně silný až silný pozitiv-ní vztah. Byly nalezeny rozdíly v závislosti na věku, pohlaví, vzdělání a řadě situačních fakto-rů. Nejlepších psychometrických vlastností do-sahovala desetipoložková verze škály. Limity studie. Ve zkoumaném vzorku dominují respondenti s vysokoškolským vzděláním.
The Chinese white pine beetle, Dendroctonus armandi Tsai and Li is a native species of bark beetle and one of the most destructive in much of western China. Little is known about the characterization of the pheromones trans-verbenol and verbenone, and their functions in D. armandi are unknown. Electroantennogram tests (EAG) and olfactory assays (Y-tube assays) in the laboratory revealed that (1) trans-verbenol may be an anti-aggregation pheromone for male and an aggregation pheromone for female D. armandi and (2) female beetles are more attracted to controls (hexane) than low concentrations of verbenone and male beetles more attracted to controls than high concentrations of verbenone. Field trials indicated that the addition of verbenone to bait used to trap D. armandi remarkably decreased the efficiency of field trapping. These results indicate that verbenone is an anti-aggregation pheromone for male D. armandi. This research provides evidence of the role of verbenone among the different types of pheromones. Development of a new dug is a very lengthy and highly expensive process since only preclinical, pharmacokinetic, pharmacodynamic and toxicological studies include a multiple of in silico, in vitro, in vivo experimentations that traditionally last several years. In the present review, we briefly report some examples that demonstrate the power of the computer-assisted drug discovery process with some examples that are published and revealing the successful applications of artificial intelligence (AI) technology on this vivid area. Besides, we address the situation of drug repositioning (repurposing) in clinical applications. Yet few success stories in this regard that provide us with a clear evidence that AI will reveal its great potential in accelerating effective new drug finding. AI accelerates drug repurposing and AI approaches are altogether necessary and inevitable tools in new medicine development. In spite of the fact that AI in drug development is still in its infancy, the advancements in AI and machine-learning (ML) algorithms have an unprecedented potential. The AI/ML solutions driven by pharmaceutical scientists, computer scientists, statisticians, physicians and others are increasingly working together in the processes of drug development and are adopting AI-based technologies for the rapid discovery of medicines. AI approaches, coupled with big data, are expected to substantially improve the effectiveness of drug repurposing and finding new drugs for various complex human diseases.
Proliferative kidney disease (PKD) is a widespread temperature-dependent disease in salmonids caused by the myxozoan parasite, Tetracapsuloides bryosalmonae (Canning, Curry, Feist, Longshaw et Okamura, 1999) (Tb). Tb has a two-host life cycle, involving fish as an intermediate host and freshwater bryozoans as the definitive host. Although salmonids are acknowledged as hosts for the parasite, it is less clear which fish species are active hosts in the life cycle of Tb. Differences in infection dynamics have been observed between some fish species, which are thought to be related to the existence of two main Tb-strains, the American and European. Iceland, having three species of indigenous salmonids and positioned geographically between Europe and North America, is an ideal location to study the natural development of Tb in wild fish. The main aim of this study was to determine the genetic origin of Tb in Iceland and confirm whether mature spores are produced in Icelandic salmonids. In this study, Icelandic salmonids were infected with the European Tb-strain. In situ hybridisation revealed that intraluminal sporogonic stages, including mature spores, were commonly observed in all three salmonid species. The presence of intraluminal stages has previously been confirmed in brown trout Salmo trutta Linnaeus and Atlantic salmon S. salar Linnaeus in Europe, but they have only been observed in Arctic charr Salvelinus alpinus (Linnaeus) in North America, infected by the local strain. This is, therefore, the first time that sporogonic stages have been observed in Arctic charr in Europe, where fish are infected with the European Tb-strain. Our data strongly suggest that all the three salmonid species inhabiting Icelandic waters serve as active hosts in the life cycle of Tb. However, for full confirmation, transmission trials are needed.
The purpose of this study was to investigate the possible relationship between Toxoplasma gondii (Nicolle et Manceaux, 1908) and stuttering. We investigated the seropositivity rate for anti-T. gondii IgG and antibodies by enzyme-linked immunosorbent assay (ELISA) in stuttering children to ascertain a possible relationship between T. gondii infection and stuttering. We selected 65 stuttering children and 65 control children (non-stutterers) to investigate the seropositivity rate of anti-T. gondii antibodies by ELISA. Cranial magnetic resonance imaging (MRI) and scalp electroencephalography (EEG) were also performed in stuttering children. The seropositivity rate of anti-T. gondii IgG antibodies among stuttering children (28%) was significantly higher than in control group (5%; p = 0.001). No abnormality was detected in cranial MRI's of stutttering children and their EEG recordings were also normal. There was no significant difference in seropositivity rate regarding age, genders and residence area. The association between seroprevalence of infection with T. gondii and stuttering may be due to hyperdopaminergic state in brains of patients who are T. gondii-seropositive. An excessive, irritable, productive or non-productive coughing
associated with airway inflammation belongs to pathological
cough. Increased activation of airway vagal nociceptors in
pathological conditions results from dysregulation of the neural
pathway that controls cough. A variety of mediators associated
with airway inflammation overstimulate these vagal airway fibers
including C-fibers leading to hypersensitivity and hyperreactivity.
Because current antitussives have limited efficacy and unwanted
side effects there is a continual demand for the development of
a novel more effective antitussives for a new efficacious and safe
cough treatment. Therefore, inhibiting the activity of these vagal
C-fibers represents a rational approach to the development of
effective antitussive drugs. This may be achieved by blocking
inflammatory mediator receptors or by blocking the generator
potential associated with the specific ion channels. Because
voltage-gated sodium channels (NaVs) are absolutely required for
action potentials initiation and conduction irrespective of the
stimulus, NaVs become a promising neural target. There is
evidence that NaV1.7, 1.8 and 1.9 subtypes are predominantly
expressed in airway cough-triggering nerves. The advantage of
blocking these NaVs is suppressing C-fiber irrespective to stimuli,
but the disadvantage is that by suppressing the nerves is may
also block beneficial sensations and neuronal reflex behavior. The
concept is that new antitussive drugs would have the benefit of
targeting peripheral airway nociceptors without inhibiting the
protective cough reflex.
Mangiferin is a kind of polyphenol chemical compound separated from these herbal medicines of Mangifera indica L., Anemarrhena asphodeloides Bge. and Belamcanda chinensis L., which has anti-inflammatory, anti-virus, and other physiological activities without toxic effects. Osteoarthritis (OA) is a chronic disease that is also a kind of arthritis disease in which articular cartilage or bones under the joint is damaged. In addition, artificial replacements are required in severe cases. At present, there are not too much researches on the potential biological activities of mangiferin that plays a protective role in the treatment of OA. In this study, we evaluated the protective effect of mangiferin on osteoarthritis (OA) in vitro and in vivo. First, the effect of different concentrations of mangiferin on rat chondrocytes was determined by MTT assay. Second, the effects of mangiferin on the expression levels of matrix metalloproteinase (MMP)-13, TNF-α, Collagen II, Caspase-3, and cystatin-C in interleukin-1β (IL-1β)-induced rat chondrocytes were examined by the real-time polymerase chain reaction in vitro, meanwhile the effects of mangiferin on the nuclear factor kappa-B (NF-κB) signaling pathway were also investigated by Western Blot. Finally, the antiosteoarthritic protective effect of mangiferin was evaluated in the rat model by anterior cruciate ligament transection (ACLT) combined with bilateral ovariectomy-induced OA in vivo. The results showed that the mangiferin was found to inhibit the expression of MMP-13, TNF-α, and Caspase-3 which also increased the expression of Collagen II and cystatin-C in IL-1β-induced rat chondrocytes. In addition, IL-1β-induced activation of nuclear factor kappa-B (NF-κB) and the degradation of inhibitor of κB (IκB)-α were suppressed by mangiferin. For the in vivo study in a rat model of OA, 100 μl of mangiferin was administered by intra-articular injections for rats, the results showed that the cartilage degradation was suppressed by mangiferin through Micro CT and Histological Examination. According to both in vitro and in vivo results, mangiferin has a protective effect in the treatment of OA which may be a promising therapeutic agent for OA.
