2252. Isolation and characterisation of the first microsatellite markers for the European stag beetle, Lucanus cervus (Coleoptera: Lucanidae)

- The European stag beetle, Lucanus cervus, is recognised as a flagship species for biodiversity conservation. Although the species is widely distributed across Europe declines have led to it being granted protected or endangered status in a number of countries and regarded as "near threatened" by the IUCN. The integration of genetic approaches into conservation efforts is urgently needed but has been impeded to date by the lack of appropriate genetic markers. To provide such a resource the development of the first microsatellite loci for stag beetle is described. Loci were identified using two methods (i) enriched library cloning (ELC) and (ii) Restriction enzyme Associated DNA sequencing (RAD-Seq). Inefficient microsatellite detection using the ELC method suggests that RAD-Seq, or other Next Generation Sequencing based methods, may ultimately be more cost effective for obtaining informative suites of markers for this and other coleopteran species. 18 loci were characterised by genotyping 42 UK specimens collected as prey leftover/roadkill. All loci produced unambiguous genotypes and were polymorphic. Though preliminary, estimates of genetic variability suggest UK populations may be genetically depauperate. - In this study, we report the development of a set of 15 polymorphic microsatellite markers for the box tree moth, Cydalima perspectalis (Walker), a highly invasive insect in Europe causing significant damage to natural and ornamental Buxus trees. The markers were characterized for four distant populations in both its native (China, two populations) and invasive ranges (Czech Republic and Turkey, one population each). The number of alleles ranged from 2 to 12. No marker significantly deviated from the Hardy-Weinberg equilibrium for all the populations sampled. 2254. Isolation and characterization of cytosolic malate dehydrogenase from Trichomonas vaginalis

- Malate dehydrogenase (EC 1.1,1,37.) (MDH) was purified to apparent homogeneity from the cytosolic fraction of the protozoan Trichomonas vaginalis Donné. The four step purification included ion-exchange chromatography (DEAE-Sephacel and Q-Sepharose, elution with NaCl) and affinity chromatography (Reactive Red Agarose, elution with NADH and NaCl). The enzyme was purified about 132-fold (30.6% yield) to a specific activity of 352 units mg~'. The Km values determined at pH 7.8 (pH optimum from 7.5 to 8.3) for oxaloacetate and NADH were 16.2 pM and 10.6 μΜ, respectively. The MDH activity was inhibited by the substrate, decreasing to 50% at about 1 mM concentration of oxaloacetate. The reverse reaction from malate to oxaloacetate showed a pH optimum around pH 9.5. The Km for malate and NAD* (determined at pH 7.8) were 1220 pM and 69.9 pM, respectively. SDS-PAGE analysis of the purified MDH revealed a single band with an apparent size of 34.5 kDa. The native molecular weight was estimated by HPLC gel filtration to be 60 kDa, which indicates that the T. vaginalis MDH exists as a dimer.

2255. Isolation and characterization of novel microsatellite loci in the genome of the dragonhead sap beetle Thymogethes norvegicus (Coleoptera: Nitidulidae)

- Here we describe 16 novel microsatellite loci in the genome of the dragonhead sap beetle Thymogethes norvegicus, which were identified using a next-generation sequencing approach. In 40 dragonhead sap beetles from a population in southern Norway, we found 2-4 alleles per locus. Expected heterozygosity ranged from 0.18-0.65. Three of the loci showed significant deviation from Hardy-Weinberg equilibrium. After correcting for multiple tests, one pairwise locus combination displayed significant linkage disequilibrium. - Prolonged cultivation of separated rat lung mast cells (LMC) in vitro is necessary to better investigate a possible role of LMC in different stages of tissue remodeling induced by hypoxia. Rat lung mast cells (LMC) were sepa rated using a protocol including an improved proteolytic extracti on and two subsequent density gradient separations on Ficoll-P aque PLUS and a new generation of Percoll, i.e. Percoll PLUS. Instead of usual isotonic stock Percoll solution, an alternative “asymptotically isotonic” stock solution was more successful in our density separation of LMC on Percoll PLUS. Separated cells were cultivated for six days in media including stem cell factor, interleu kins IL-3 and IL-6, and one of two alternative mixtures of antibi otics. These cultivations were performed without any contaminatio n and with only rare changes in cell size and morphology. Model co-cultivation of two allogenic fractions of LMC often caused considerable rapid changes in cell morphology and size. In contrast to these observations no or rare morphological changes were found after cultivation under hypoxic conditions. In conclusions, we modified separation on Percoll PLUS to be widely used, altered LMC separation with respect to purposes of long-lasti ng cultivation and observed some model morphological changes of LMC., J. 2259. Isospora abdallahi sp. n. (Apicomplexa: Eimeriidae), a new coccidium from the lacertid lizard Acanthodactylus boskianus (Sauria: Lacertidae) from Northern Egypt

- Seven of 12 lacertid lizards Acanthodactylus boskianus (Daiidin, 1802), passed oocysts of an Isospora species. Comparison with other species of the genus Isospora Schneider, 1881 indicated that found coccidium represented a new species, for which the name /. abdallahi is proposed. Sporulated oocysts of /. abdallahi are spherical or subspherical, 25.8 (24.5-29.0) x 23.9 (23.0-25.5) pm, shape index (length/width) being 1.07 (1.00-1.16), with a smooth, bilayered oocyst wall that is slightly yellowish, about 2 pm thick. Micropyle, oocyst residuum and polar granule are absent. Sporocysts are ovoidal, 15.4 (14.0-16.0 x 9.4 (9.0-10.0) pm, with smooth and colorless sporocyst wall, shape index 1.6 (1.5-1.8). Stieda body is dome-like, substieda body spherical to subspherical. Sporocyst residuum is composed of numerous granules of different size, scattered among sporozoites. Most oocysts are passed unsporulated; sporulation was completed within 12 h at 25'C. Endogenous development occurs inside nuclei of enterocytes in the small intestine.

- Despite the state borders, and the different socioeconomical and cultural contexts, productions of Italian opera in Prague and Dresden become considerably interconnected due the activity of the impresario Giuseppe Bustelli. During his directorship (1764-1777 in Prague, 1765-1777 in Dresden) and even in the next decade, more than 50% of the repertoire was shared. Furthermore, some of the artists performed in both cities, and the same or similar adaptations of operas were used. The main difference in repertoire consisted in opera seria productions in Prague until 1777, whereas in Dresden only opera buffa was staged since 1765.
