The mitochondrial DNA (mtDNA) amount in cells as the basis for mitochondrial energy generating system, which produces ATP, plays an important role in the fetal development and postnatal morbidity. Isolated human cord blood leukocytes (HCBL) contribute very little to the overall metabolic turnover, but they may serve as easily available marker cells for the study of the mtDNA amount changes in cord blood during fetal development. The aim of our study was to analyze the mtDNA amount in HCBL. HCBL were isolated from cord blood samples of 107 neonates born between the 25th and 41st week of gestation. The mtDNA amount was analyzed by the real-time PCR method. The significant negative correlations were found between the relative mtDNA amount in HCBL and gestational age (r = -0.54, p<0.01) and birth weight (r = -0.43, p<0.01), respectively. The results revealed that the mtDNA content per cell decreases in HCBL with progressing fetal development. This may be explained by gradual shift of the hematopoiesis from fetal liver to bone marrow during the second half of pregnancy presumably accompanied by decreasing cell volume of HCBL as it was shown similarly in red blood cells., M. The light response changes of oxygen evolutíon in the isolated spinách (Spinacia oleracea L.) thylakoids associated with the photoinhibition, influenced by the temperature and radical protection, were determined. Photoinhibition was performed at 20 ®C with or without an addition of antioxidant mixture consisting of glutathione, ascorbate, superoxide dismutase and catalase, oř, altematívely, at 0 oC without the antioxidants. Beside the effects on decline of atrazine binding sites and variable room temperature fluorescence, the three variants of photoinhibition differed with respect to their effects on the yield of oxygen evolutíon. Quantum yield of oxygen evolutíon (<{>02) flecline preceded Úie decrease of electron transport capacity at the beginning, while similar low values were found at the end of the treatment. In the presence of antioxidants the electron transport capacity remained high, whereas marked decline of (|»o^ occurred with the equally treated thylakoids. Comparable deviating behaviour of (t»Oj and the electron transport capacity was not found following photoinhibition at 0 oC; both were affected to the same degree, regardless if ferricyanide or the Qg- independent electron acceptor siUcomolybdate were ušed. With isolated thylakoids the quotient FyF,„, which is often ušed as a measure for photochemical efficiency of open photosystem 2 reaction centres in whole plant studies, did not decline to the same extent as (|)Oj under the dififerent photoinhibitoiy conditions applied. The main conclusion is that in thylakoid photoinhibition the independent mechanisms are associated with the changes of both the and electron transport capacity, and that the (j>02-related mechanism is inhibited at 0 oC.
Anguillicola australiensis (Johnston et Mawson, 1940) is widespread and common in Anguilla reinhardtii Steidachner in rivers and dams of eastern Queensland, Australia, having been found in nine out of ten localities. Overall prevalence was 50% and maximum local prevalence reached 77.7%. The parasite never attained high levels of abundance and maximum adult abundance never exceeded 3.22 or intensity 10. Adults were overdispersed throughout the eel populations and abundance was unrelated to eel or swimbladder size. The greater part of the adult population was composed of immature parasites. The occurrence of larvae in the swimbladder wall was erratic and unrelated to the size of the adult population. Larvae were never abundant and the great majority were damaged by a host response. It appears that parasites either pass through the swimbladder wall rapidly and moult to adults or if delayed are destroyed. There was no indication that a paratenic host was involved in the life cycle. There was no evidence that adult parasites had any local pathogenic effects on their hosts. The population biology of Anguillicola australiensis in its natural host Anguilla reinhardtii appears to be far more similar to those of other Pacific species of Anguillicola in Anguilla japonica in China and Japan than to A. crassus in Anguilla anguilla in Europe or Japan. This latter host-parasite combination appears to be the exception not the rule. It is suggested that the lack of pathogenicity of A. australiensis may reflect a long period of host-parasite co-evolution and/or lower transmission rates resulting in lower parasite population densities.
Quantitative data are presented on the spatial distribution of metacercariae of the digenean trematode Diploslomum phoxini (Faust, 1918) in the brains of minnows, Phoxinus phoxinus (Linnaeus, 1758), from two Scottish populations. Sequential examination of serial histological sections revealed metacercariae to be unevenly distributed throughout the brain, aggregating in specific regions including the cerebellum, the medulla oblongata and the optic lobes. In addition, a number of metacercariae were found in the anterior part of the spinal cord. The inferior lobe of the cerebellum, pituitary, olfactory lobes and olfactory bulbs were largely free of metacercariae. Reasons for the uneven distribution of metacercariae within the brains of infected minnows are discussed, including the possibility that the parasite may have evolved to enhance its transmission to subsequent hosts by aggregating in regions known to be important in the control of the host’s antipredator responses.
Food webs are of crucial importance for understanding any ecosystem. The accuracy of food web and ecosystem models rests on the reliability of the information on the feeding habits of the species involved. Water boatmen (Corixoidea) is the most diverse superfamily of water bugs (Heteroptera: Nepomorpha), frequently the most abundant group of insects in a variety of freshwater habitats worldwide. In spite of their high biomass, the importance of water boatmen in aquatic ecosystems is frequently underestimated. The diet and feeding habits of Corixoidea are unclear as published data are frequently contradictory. We summarise information on the feeding habits of this taxon, which exemplify the difficulties in evaluating published data on feeding habits in an invertebrate taxon. It is concluded that Corixoidea are, unlike other true bugs, capable of digesting solid food, but their feeding habits are still insufficiently known. The dominant feeding strategy in this taxon is zoophagy, but several species consume other foods, particularly algae and detritus. Only members of the subfamily Cymatiainae seem to be exclusively predators. In other subfamilies, the diet of different species and different sexes or populations of a single species may vary depending on the food available or is still unknown. Increased colonic Cl - secretion was supposed to be a causative factor of diarrhea in inflammatory bowel diseases. Surprisingly, hyporesponsiveness to Cl - secretagogues was later described in inflamed colon. Our aim was to evaluate changes in secretory responses to cholinergic agonist ca rbachol in distal and proximal colon during colitis development, regarding secretory activity of enteric nervous system (ENS) and prostaglandins. Increased responsiveness to carbachol was observed in both distal and proximal colon after 3 days of 2 % dextran sodium sulfate (DSS) administration. It was measured in the presence of mucosal Ba2+ to emphasize Cl - secretion. The described increase was abolished by combined inhibitory effect of tetrodotoxin (TTX) and indomethacin. Indomethacin al so significantly reduced TTX- sensitive current. On the 7 th day of colitis development responsiveness to carbachol decreased in distal colon (compared to untreated mice), but did not change in proximal colon. TTX- sensitive current did not change during colitis development, but indomethacin-sensitive current was significantly increased the 7 th day. Decreased and deformed current responses to serosal Ba 2+ were observed during colitis induct ion, but only in proximal colon. We conclude that besides inhibitory effect of DSS on distal colon responsiveness, there is an early stimulatory effect that manifests in both distal and proximal colon., M. Hock ... Two hundred and forty brown trout (Salmo trutta) and 49 rainbow trout (Oncorhynchus mykiss), obtained from 21 locations in Central Scotland between October 1990 and August 1993, were examined for endoparasitic helminth infections. Crepidostomum farionis (Digenea) was the most widely distributed helminth species, followed by Eubothrium crassum (Cestoda), Diphyllobothrium dendriticum and D. ditremum (Cestoda), Neoechinorhynchus rutili (Acanthocephala), Echinorhyn-chus truttae (Acanthocephala), Eustrongylides sp. (Nematoda), Capillaria salvelini (Nematoda), Cyalhocephalus truncatus (Cestoda), Raphidascaris acus (Nematoda) and Cystidicola farionis (Nematoda), in that order. The prevalences and intensities of each helminth infection were recorded. No evidence was found to indicate that even fish with the highest worm burdens (e.g. 339 plerocercoids of Diphyllobothrium spp.) were experiencing any obvious morbidity. An analysis of pairs of associations between species of helminths revealed a significantly positive association between N. rutili and C. farionis (P < 0.01). The results are discussed in terms of patterns in helminth communities in freshwater fish.
Herbivory on liverworts is rarely reported. We studied the effects of feeding by larvae of the sciarid fly Scatopsciara cunicularius on the growth of the thalloid liverwort Marchantia polymorpha at two different constant temperatures, 12°C and 22°C. Larvae reared at the lower temperature fed slower and over a longer period of time, which resulted in more damage and a greater reduction in the growth of the liverwort than that caused by those reared at the higher temperature. The reduction in growth of the liverwort was positively density-dependent in terms of number of larvae at both temperatures. Attention has recently been focused on endothelial function after a single high-fat meal, i.e. on the anticipated direct atherogenic effect of triglyceride-rich lipoproteins. Our study was designed to investigate the effect of a low-fat diet given for four weeks followed by a high-fat diet for another four weeks. At the end of each dietary period, a non-invasive ultrasound investigation of endothelial function of the brachial artery was performed along with laboratory tests. Endothelial function was measured immediately before the dietary load and after three and six hours in 11 healthy volunteers. The results were expressed as percentage of the changes in artery diameter at rest and during hyperemia; the data were processed using computer technology. When compared to the low-fat regimen, the total cholesterol content rose after the high-fat diet from 4.28 mmol/l to 5.15 mmol/l (p<0.05) in the whole group of volunteers. There was no difference between both dietary regimens in baseline triglycerides. The brachial artery dilatation under basal conditions was 5.26±2.88 mm after the high-fat diet compared with the value of 3.13±3.01 mm (p<0.05) after the low-fat diet. When measured individually endothelial function in the whole group of volunteers in the course of the day, the degree of arterial dilatation after one month on low-fat diet was 3.13±3.0 %, 3.88±2.5 % and 5.23±3.3 % at single measurement. When comparing arterial dilatation at two closest measurements, a non-significant trend, p>0.05 was seen in either case. The following values were obtained after one month on the high-fat diet: 5.26±2.9 %, 4.47±1.7 %, and 6.2±3.6 %; again showing a non-significant trend of p>0.05. Contrary to clinical trials, experimental studies revealed that diabetes mellitus (DM) may initiate, besides increased myocardial vulnerability to ischemia-reperfusion injury (I/R) and pro/antioxidant dysbalance, development of adaptation leading to an enhanced tolerance to I/R. The aims were to characterize 1) susceptibility to ischemia-induced ventricular arrhythmias in the diabetic rat heart 2) its response to antioxidant N-acetylcysteine (NAC ) and a NOS inhibitor L-NAME, and 3) the effect of DM on endogenous antioxidant systems. Seven days after streptozotocin injection (65 mg/kg, i.p.), Langendorff-perfused control (C) and DM hearts were subjected to 30-min occlusion of the LAD coronary artery with or without prior 15-min treatment with L-NAME (100 μM) or NAC (4 mM). Total number of ventricular premature beats (VPB), as well the total duration of ventricular tachycardia (VT) were reduced in the DM group (from 533±58 and 37.9±10.2 s to 224.3±52.6 and 19±13.5 s; P<0.05). In contrast to the antiarrhythmic effects of L-NAME and NAC in controls group (VPB 290±56 and 74±36, respectively; P<0.01 vs. control hearts), application of both drugs in the diabetics did not modify arrhythmogenesis (L-NAME: VPB 345±136, VT 25±13 s; NAC: VPB 207±50, VT 12±3.9 s; P>0.05 vs non-treated diabetic hearts). Diabetic state was associated with significantly elevated levels of CoQ 10 and CoQ 9 (19.6±0.8 and 217.3±9.5 vs. 17.4± 0. 5 and 185.0±5.0 nmol/g, respectively, in controls; P<0.05), as well as α-tocopherol (38.6±0.7 vs. 31.5±2.1 nmol/g in controls; P<0.01) in the myocardial tissue. It is concluded that early period of DM is associated with enhanced resistance to ischemia-induced arrhythmias.
