The intertidal collembolan Anurida maritima can endure periods of twice-daily submergence by seawater. The air-breathing terrestrial apterygote insect has developed specific adaptations to prevent respiratory failure during hypoxic stress. When submerged, the animal is initially enclosed by an air-bubble. This bubble lasts three times longer than the small amount of stored oxygen would allow. The air bubble acts not only as an oxygen store but also as a compressible gas gill. This was demonstrated by an O2-needle electrode technique which allowed recordings of pO2 changes in the watery medium close to the animal. PURPOSE: A variety of anti-glaucomatous shunt designs have been proposed so far. This study evaluates the feasibility of a novel shunt design, the intra-scleral parathalamus implant (IPI), in a rabbit eye model. METHODS: Ten healthy albino rabbits were included. Measurements of the IOP, using TonoPenXL, as well as ocular rigidity (OR) and aqueous outflow facility (AO), using a previously described methodology, were performed in both eyes of each animal. Subsequently, the IPI was implanted at the left eye of all animals whereas the right eye served as control. Measurements of IOP were repeated at weekly intervals for 2 months postoperatively whereas measurements of OR and AO were repeated at the 1st and 2nd postoperative week. RESULTS: The IOP decreased significantly whereas the AO increased significantly postoperatively at the operated eyes. A significant postoperative decrease in OR was also recorded at the operated eyes. Respective differences at the fellow eyes were statistically not significant. Two animals developed postoperative complications and were excluded from follow-up. Larval Manduca prothoracic gland cells in vitro responded to prothoracicotropic hormone (PTTH) from neurosecretory cells of the brain with an increase of intracellular free calcium. This effect is reversible and dose-dependent. Preincubation of the glands with TMB-8 and dantrolene, which inhibit the release of calcium from intracellular stores, did not decrease the PTTH-stimulated increase in calcium, indicating that intracellular calcium stores are not involved in the control of ecdysteroidogenesis. Pharmacological studies of the PTTH effect with calcium channel blockers revealed that the increase in calcium was totally blocked by cadmium, partially inhibited by nickel and lanthanum and by amiloride, an antagonist of T-type calcium channels. All other inhibitors tested were ineffective, suggesting that the increase in cytosolic calcium is induced by opening of calcium channels, presumably of the T-type, in response to PTTH. The action of PTTH on these channels may be mediated by a G-protein as shown by the effect of mastoparan, a G-protein activator, which increased the concentration of cytosolic calcium comparable to that evoked by PTTH., Heiner Birkenbeil, and Lit
Nanocarriers bearing anticancer drugs are promising candidates to improve the efficacy of cancer therapy and minimize side effects. The most potent cytostatics used in the treatment of various cancers are anthracyclines, e.g. doxorubicin or pirarubicin. Recently, polymer therapeutics carrying anthracyclines have been intensively studied. The precise characterization of in vitro nanocarrier biological behavior brings a better understanding of the nanocarrier characteristics and enables prediction of the behavior of the nanocarrier during in vivo application. Advanced fluorescence detection methods, e.g. fluorescence lifetime imaging microscopy (FLIM), were successfully exploited to describe the properties of various polymeric nano-systems and contributed to a complex view of anthracyclines’ intracellular transport and DNA intercalation. Here, we report the application of a specific technique for processing FLIM images, called fluorescence pattern decomposition, to evaluate early events after doxorubicin or pirarubicin treatment of cells. Intramurální hematom jícnu je vzácně se vyskytující diagnóza. Prezentuje se retrosternální bolestí, dysfagií či hematemezou. Je způsoben barotraumatem či přímým poraněním jícnu, nebo se může objevit spontánně bez zjevné příčiny, což je častější u pacientů užívajících antiagregační či antikoagulační preparáty. Ve většině případů je léčba konzervativní, v ojedinělých případech endoskopická, endovaskulární či chirurgická. Tento článek prezentuje kazuistiku pacientky se spontánním hematomem jícnu řešeným konzervativně a diskuzi o dalších možnostech průběhu a léčby onemocnění., Intramural hematoma of the oesophagus is a rare diagnosis presenting with retrosternal pain, dysphagia and haematemesis. It is caused by barotrauma or direct injury to the esophagus, or it may occur spontaneously without apparent cause, which is more common in patients taking antiplatelet or anticoagulant medications. In most cases, treatment is conservative. Rarely, endoscopic, endovascular or surgical treatment is effective. Morphometric data from spores of ten myxosporean species were statistically analysed to explore myxosporean intraspecific variation in measurements when obtained from a sample from: (1) the same plasmodium, (2) different plasmodia from the same host and (3) different host individuals and localities. In some cases, significant differences in spore dimensions were found between samples from the same plasmodium, highlighting the difficulty of obtaining representative measurements of myxosporean spore. In addition, significant differences in spore dimensions were found when plasmodia from the same site of infection were compared, suggesting that measurements of spores should come from several different plasmodia of the sampling to increase the reliability of the morphology data. Moreover, significant differences in spore dimensions were observed for most spore dimensions when data were compared between localities. In all cases, there was clear overlap in ranges of dimensions even when means differed significantly. The present statistical analysis shows that intraspecific morphometric variation of myxosporean species commonly occurs, highlighting the importance of reporting ranges of measurements for a species, not just the mean dimensions, and taking into account all evidence when assigning or describing myxosporean species., Yanhua Zhai, Christopher M.
