- For companies doing business in mining mineral deposits, ensuring safe work is one of the key tasks (Safety First!). One of the important trends in this area is prevention and endeavour to forestall risk situations. Risks need to be searched, technically described, spatially defined, evaluated and categorized by degree of risk. Complex geological and stability conditions can be one of the sources of persistent and significant risks, which are mainly landslides and rockslides threatening both mining equipment and employees. The problem described in this article and its solution concerns the Most Basin (formerly the North Bohemian Lignite Basin). This is a tertiary basin that was founded in the Oligocene. The main mineral is lignite and mining takes place on the surface. 22853. The time-course of agonist-induced solubilization of trimetric Gqα/G11α proteins resolved by two-dimensional electrophoresis

- Prolonged agonist stimulation results in specific transfer of activated Gα subunits of Gqα/G11α family from particulate membrane fraction to soluble (cytosol) cell fraction isolated as 250 000 x g supernatant. In this study, we have used 2D electrophoresis for more defined resolution of Gα subunits of Gqα/G11α family and followed the time course of solubilization effect. The small signal of soluble G proteins was already detected in control, hormone-unexposed cells. Hormone stimulation resulted in a slow but continuous increase of both intensity and number of immunoreactive signals/spots of these G proteins (10, 30, 60, 120 and 240 min). At longer times of agonist exposure (>2 hours), a marked increase of Gqα/G11α proteins was detected. The maximal level of soluble Gqα/G11α proteins was reached after 16 hours of continuous agonist exposure. At this time interval, eight individual immunoreactive signals of Gqα/G1 α proteins could be resolved. The relative proportion among these spots was 15:42:10:11:7:7:2:5. Solubilization of this class of Gα proteins was thus observed after prolonged agonist stimulation only, induced by ultra high concentration of hormone and in cells expressing a large number of GPCRs. Sleduji dále jejich vliv na současné praktiky modelování a zdůrazňuji, že materiální modely, jež tradičně stály za hranicemi deduktivní, pozitivistické vědy, jsou nyní v těchto oblastech chemie akceptovány jako způsob vědeckého uvažování., Chemical visualizations and models are special kinds of media for thinking. In this paper, I examine several historical case studies-an archive of images from museums, special collections, and popular magazines- as examples of emergent practices of physical modeling as theoretical play which became the basis for molecular biology and structural chemistry. I trace a legacy of visualization tools that starts with Archibald Scott Cooper and Friedrich Kekulé in the late 1800s, crystallizes as material manipulatives in Van’t Hoff and his folded paper “toys”, is legitimized in the California lab of Linus Pauling, and is glorified in the popular imaginary with James Watson and Francis Crick’s model of DNA. My tracing then follows several threads into contemporary modeling practices. I ultimately argue that modeling play, originally outside of the boundary of deductive, positivist science, is now an accepted mode of reasoning in these related chemical fields., and Kate McKinney Maddalena.

Mráčkové se zabývá problematikou přejímání středověkých hymnických melodií, ilustrovanou na konkrétním zvoleném příkladu., The hymn, one of the most frequently encountered songs in Western European sources, poses many questions with regard to the conflict between common and local traits. The Office hymns on which I focus were transmitted mainly in fourteenth- and fifteenth-century sources, which contain huge collections of hymns for different liturgical occasions, including those in honour of local saints. We still do not have a reasonable explanation of how or why the same tunes were adapted for different texts (or vice versa). The ‘Franciscan’ hymn melody Stäblein 752 (originally devoted to St Francis), for example, which occurs with different hymn texts in Austria, Germany, Bohemia, Poland and Spain, has many melodic variants reflecting regional characteristics. Why was this particular melody transferred to and adopted in other parts of Europe, where there was certainly no shortage of alternative hymn melodies? Is it a question of the adoption of favourite melodies in the Middle Ages? Were the same tunes used for hymns in honour of both male and female saints? - A strongly recommended conclusion in sociology about trends in class inequality has been summarised by Goldthorpe as a high degree of 'temporal constancy and cross-national communality'. This conclusion, here called 'the stability thesis', was first challenged by Ringen in 1987 and again, on more methodological grounds, by Ringen and Hellevik in two papers published in 1997. These challenges resulted in a process of debate and reassessment. It is now possible to sum up and conclude. The stability thesis rests on empirical results from odds-ratio readings of mobility table data. The authority of this methodology is re-examined in terms of normative significance and statistical validity. Mobility table data which have generated stability thesis findings are reanalysed with the standard gini-index methodology in the study of inequality, then yielding different findings which contradict the stability thesis. - BACKGROUND: The presence of several risk factors (genetic and non-genetic) has greater impact on the risk of premature coronary artery disease (CAD) than single risk factor. OBJECTIVE: The aim of the study was to establish possible relations between genotypes and alleles of 677C>T polymorphism of MTHFR gene and some traditional risk factors e.g. elevated levels of lipid parameters and smoking in development of premature CAD. METHODS: The groups comprised 152 patients with angiographically documented premature CAD (aged 42.9 +/- 5.5) and 121 age-matched blood donors (aged 42.3 +/- 6.5) were studied. The MTHFR 677C>T polymorphism was genotyped with Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method. RESULTS: Patients with TT genotype who simultaneously smoked had increased risk of premature CAD compared to non-smoking cases with CC genotype (OR = 24.62). We also found that individuals with TT genotype and elevated LDL-cholesterol (LDL-chol.) level had significantly higher risk of CAD (OR = 9.92) than individuals with normal LDL-chol. level and CC genotype. CONCLUSIONS: The present study shows that simultaneous presence of MTHFR TT genotype and smoking or elevated levels of LDL-chol. influences the risk of premature CAD. Na rozdíl od nich, tento příspěvek zdůrazňuje význam kontextu (imitační hry Turingova testu), který je klíčem k interpretaci univerzálního Turingova stroje jako mládeneckého stroje a potenciálně jako autoportrétu., Since the beginning of the twenty-first century there has been an increasing awareness that software represents a blind spot in new media theory. the growing interest in software also influences the argument in this paper, which sets out from the assumption that Alan M. Turing’s concept of the universal machine, the first theoretical description of a computer program (software), is a kind of bachelor machine (Carrouges). Previous writings based on a similar hypothesis (Daniels, Baudrillard, Turkle, Ascott) have focused either on a comparison of the universal machine and the bachelor machine in terms of the similarities of their structural features, or they have taken the bachelor machine as a metaphor for a man or a computer (artificial intelligence). Unlike them, this paper stresses the importance of the context (the imitation game of the Turing test) as a key to interpreting the universal Turing machine as a bachelor machine and, potentially, as a self-portrait., and Jana Horáková.

- The anterior jejunum from common vole naturally infected with Giardia microti (Kofoid et Christiansen, 1915) was examined by ТЕМ and compared with the anterior jejunum from control (metronidazole-treated, Giardia-free) common voles (Pallas, 1778). Giardia microti infection resulted in significant diffuse shortening of microvilli and significant greater microvillous diameters. In addition, deformations of the microvilli were observed at the margin of the ventral disc. The microvilli attached to the lateral crest of the ventral disc were vesiculated with a disorganised filamentous core and contained whorled structures resembling “myelin-like figures”. The findings are discussed in context of the Giardia-epithelial cell interaction.
