Safranal and crocin are biologically active compounds isolated from Crocus sativus L., commonly known as saffron. Clinical trials confirm that saffron has antidepressant effect, thus being a potential valuable alternative in the treatment of depression. The aim of the present study was to determine, whether systemic administration of safranal and crocin can influence the metabolic activity of CYP3A, CYP2C11, CYP2B, and CYP2A in rat liver microsomes (RLM). The experiments were carried out on male Wistar albino rats intragastrically administered with safranal (4, 20, and 100 mg/kg/day) or with intraperitoneal injections of crocin (4, 20, and 100 mg/kg/day). Our results demonstrate the ability of safranal and crocin to increase the total protein content and to change the metabolic activity of several CYP enzymes assessed as CYP specific hydroxylations of testosterone in RLM. Crocin significantly decreased the metabolic activity of all selected CYP enzymes, while safranal significantly increased the metabolic activity of CYP2B, CYP2C11 and CYP3A enzymes. The eye nematode Thelazia callipaeda Railliet et Henry, 1910 (Spirurida: Thelaziidae) is a vector-borne zoonotic nematode infecting a range of wild and domestic carnivores as well as humans. It is considered to be a causative agent of emerging and neglected disease and currently invades central part of Europe. Nematodes were collected from the eye of a dog living in Prague, which never travelled outside the Czech Republic. The nematodes were identified based on their morphology and partial sequence of the cox1 gene as T. callipaeda haplotype 1. This finding represents the northernmost record of autochthonous canine thelaziosis in Europe. The insufficient control of imported animals as well as free movement of dogs and wild carnivores within Europe probably facilitates spreading of T. callipaeda throughout the continent. We observed instances of cannibalism (intraspecific predation) among intra-instar larvae of Culex pipiens Linnaeus, 1758 while performing a bioassay of Lysinibacillus sphaericus (formerly named Bacillus sphaericus) larvicide, when the larvae were exposed to the larvicide for 48 h in the absence of food. Larvae without symptoms of poisoning attacked and devoured those visibly affected. Cannibalism was more prevalent in 1st-2nd instar larvae than in 3rd-4th instar. This phenomenon should be taken into account when interpreting the results of larvicide bioassays, especially when the exposure lasts over 24 h. A large number of songs have been preserved in the Strahov Codex (ca. 1465/67-1470), an important collection of polyphony from the latter half of the fifteenth century. These songs were once generally regarded as cantiones in Latin or as instrumental compositions, but a detailed study of this repertoire shows that the manuscript is an important source of secular compositions, and especially of chansons. Most of these songs have been preserved only in this manuscript. Capsazepine is a competitive antagonist of capsaicin, a TRPV1 agonist responsible for the spicy taste of pepper. TRPV1 agonists and antagonists are known to affect mammalian body temperature, but their action on thermoregulation in insects is poorly known. In this study we evaluated the effect of capsazepine on the thermal preference of the American cockroach, Periplaneta americana using a thermal gradient. Our results revealed that capsazepine in submicromolar concentrations induces a preference for higher ambient temperatures when compared to the control insects. To assess whether capsazepine may act also as an antagonist of capsaicin in insects, we determined this insects' thermal behaviour when capsazepine was applied before capsaicin. The hypothermic response to capsaicin was clearly blocked by pre-treatment with capsazepine only in female American cockroaches. Angiotensin converting enzyme (ACE) inhibition has been reported to induce regression of hypertrophy in several models of hemodynamic pressure overload. The aim of the present study was to determine whether the ACE inhibitor captopril can reduce hypertrophy of the left ventricle induced by a chronic volume overload and modify collagen composition of the hypertrophied myocardium. Rabbits with four months lasting aortic insufficiency were divided into two groups: treated with captopril (20 mg/kg/day) for five weeks and treated with placebo. The respective control groups were represented by sham-operated animals. Aortic insufficiency induced a decrease of diastolic pressure, an increase of systolic and pulse pressure, hypertrophy of the left and right ventricle, and an increase of hydroxyproline content in the left ventricle without a change of hydroxyproline concentrations in either ventricle. Captopril treatment further enhanced pulse pressure by decreasing diastolic blood pressure. Hypertrophy of the left ventricle, hydroxyproline content and concentration in both ventricles were unaffected by captopril treatment. It is concluded that ACE inhibition did not reverse the left ventricular hypertrophy developed as a result of overload induced by aortic insufficiency. We suggest that mechanisms different from activation of the renin-angiotensin system may play a decisive role in the maintenance of hypertrophy in this particular model of volume hemodynamic overload., F. We studied the effects of the H2S donor Na2S on the mean arterial blood pressure (MAP) and heart and breathing rates of anesthetized Wistar rats in the presence and absence of captopril. Bolus administration of Na2S (1-4 μmol/kg) into the right jugular vein transiently decreased heart and increased breathing rates; at 8-30 μmol/kg, Na2S had a biphasic effect, transiently decreasing and increasing MAP, while transiently decreasing heart rate and increasing and decreasing breathing rate. These results may indicate independent mechanisms by which H2S influences MAP and heart and breathing rates. The effect of Na2S in decreasing MAP was less pronounced in the presence of captopril (2 μmol/l), which may indicate that the renin-angiotensin system is partially involved in the Na2S effect. Captopril decreased H2S-induced NO release from S-nitrosoglutathione, which may be related to some biological activities of H2S.
