The present study describes the anatomy and surface topography of the metacercaria of Microphallus primas (Jägerskiöld, 1909) infecting the shore crab Carcinus maenas (L.) in Aveiro estuary, northern Portugal. The metacercaria species identification resulted from the combined use of morphological and molecular data, particularly the 28S rDNA gene. The metacercariae encysted preferentially in the host's hepatopancreas and also in the gonads. Isolated cysts were present in two distinct forms, spherical and oval, and were shown to be the identical species by the internal transcribed spacer 1 (ITS1) sequence. Chemically excysted metacercariae were studied by light (LM) and scanning electron microscopy (SEM). Their specific characteristics observed include the particular aspect of the vesiculo-prostatic pouch surrounded by a very thin membrane, the presence of a prominent muscular papilla, and an obvious metraterm. The dorsal and ventral tegumental surfaces of the metacercaria were densely packed with similar squamous spines, which decreased in number and size towards the hindbody. The edges of the posterior and ventral face of the body were coated with numerous microvilli, whose function remains unknown. In order to identify the species of metacercariae, we compared a 28S partial rDNA sequence of the two forms of cysts with the same 28S partial region of M. primas available in GenBank. With this comparison, we determined that the sequences had a 100% similarity and therefore belonged to the same species, i.e., M. primas.
This study aimed to investigate the effects of waterlogging on the growth and photosynthetic characteristics of paired near-isogenic lines of waterlogging-tolerant (Zz-R) and waterlogging-sensitive
(Zz-S) waxy corn inbred line seedlings. All plants were grown until the fifth leaves were fully expanded. Subsequently the plants in the pots were submerged in water for 4 d. During the waterlogging period, morphological and photosynthetic parameters related to waterlogging tolerance were examined. After 4 d, a significant decrease was observed in shoot and root fresh mass, net photosynthetic rate, stomatal conductance, transpiration, water-use efficiency, light-saturation point, maximal photosynthetic rate, apparent quantum yield, maximal quantum yield of PSII, and effective quantum yield of PSII photochemistry in waterlogged plants of both genotypes. The Zz-R genotype showed lesser reduction in all mentioned indices when compared to the Zz-S genotype. The inhibition of photosynthesis under waterlogging occurred due to the reduction in stomatal conductance, fluorescence parameters, and chlorophyll content. This study was performed to evaluate the ecophysiological acclimation of Catalpa bungei plantlets to different light conditions. We hypothesized that the acclimation of old and newly developed leaves to both increasing and decreasing irradiance should follow different patterns. The growth, photosynthesis, chlorophyll (Chl) content, and Chl fluorescence response were examined over a range of light treatments. The plants were grown under fixed light intensities of 80% (HH), 50% (MM), 30% (LL) of sun light and transferring irradiance of 80% to 50% (HM), 80% to 30% (HL), 30% to 50% (LM) and 30% to 80% (LH). For old leaves, light-saturation point, photosynthetic capacity, dark respiration rate of LH were lower than that of HH, while HL were higher than LL, indicating that light-response parameters were affected by the original growth light environment. Initial fluorescence increased and variable fluorescence decreased in LH and LM after transfer, and the PSII damage was more serious in LH than that in LM, and could not recover within 30 d. It suggested that the photoinhibition damage and recovery time in old leaves was related to the intensity of light after transfer. For the newly emerged leaves with leaf primordia formed under the same light environment, a significant difference was observed in leaf morphology and pigment contents, suggesting that previous light environment exhibited carry-over effect on the acclimation capacity to a new light environment. Thermophilic unicellular cyanobacterium Synechococcus elongatus Näg. var. thermalis Geitl. strain Kovrov 1972/8 was cultivated in continuous flow reactor to simulate conditions occurring in nature in regions with low iron concentration. Two degrees of iron deprivation were established: (a) low iron (LI) conditions (9.0 µM Fe) when cells still maintained maximal growth rate but already exhibited changes in photosynthetic apparatus, and (b) iron deficient (ID) conditions (0.9 µM Fe) when cell growth rate decreased and extensive morphological and functional changes were observed. A decrease in the cellular content of phycobilin antenna was observed in both ID and LI cells and an increase of carotenoid concentration only in the ID culture. Morphologically, ID cells showed a decrease in the amount of phycobilins and in the number of thylakoid membranes. This suggests that S. elongatus responds to decrease in iron availability by substitution of the phycobilisomes by antennae containing chlorophyll (Chl) and carotenoids. Photochemical activity of photosystem (PS) 2, determined as Fv/Fm ratio was similar in high iron (HI) and LI cultures and approximately five times lower in ID culture. On the other hand, the activity of the whole electron transport chain showed the opposite tendency: the relative rates of the CO2-dependent oxygen evolution in HI : LI : ID cultures were approximately 1 : 2 : 4. Thus in nutrient stress the photosynthetic apparatus preserved its activity despite the decrease in the amount of both Chl-binding complexes and thylakoid membranes. and J. Benešová ... [et al.].
Chloromyxum leydigi Mingazzini, 1890, the type species of Chloromyxum Mingazzini, 1890, is redescribed based on material found in the gall bladder of the cartilaginous fish Torpedo marmorata Risso collected from the Portuguese Atlantic coast and its sporogonic development is described. Plasmodia and mature spores were floating free in the bile. Plasmodia are polysporic and highly polymorphic in shape and organization. Mature spores are spherical to subspherical with a pointed anterior end, measuring 12.3 ± 0.5 μm in length and 9.0 ± 0.5 μm in width. The spore wall is composed of two asymmetric shell valves, each bearing 4-5 elevated surface ridges. A bundle of 40-50 tapering caudal filaments extends from the basal portion of the shell valves. Four pyriform equal-sized polar capsules, measuring about 5.3 × 3.2 μm, are observed at the same level in the anterior pole of the spores, each containing a polar filament coiled in 8-9 (rarely 10) turns. Spore morphology, tissue tropism, host species and sequences of the SSU rRNA gene supported species identification. Since its discovery, this species has been dubiously reported from several cartilaginous hosts, namely due to the poor description of its features.
Using histochemical analysis (NADPH-diaphorase, Fluoro-Jade B dye and bis-benzimide 33342 Hoechst) we studied the influence of intraperitoneal administration of nicotine (NIC), kainic acid (KA) and combination of both these substances on hippocampal neurons and their changes. In experiments, 35-day-old male rats of the Wistar strain were used. Animals were pretreated with 1 mg /kg of nicotine 30 min prior to the kainic acid application (10 mg/kg). After two days, the animals were transcardially perfused with 4 % paraformaldehyde under deep thiopental anesthesia. Cryostat sections were stained to identify NADPH-diaphorase positive neurons that were then quantified in the CA1 and CA3 areas of the hippocampus, in the dorsal and ventral blades of the dentate gyrus and in the hilus of the dentate gyrus. Fluoro-Jade B positive cells were examined in the same areas in order to elucidate a possible neurodegeneration. In animals exposed only to nicotine the number of NADPH-diaphorase positive neurons in the CA3 area of the hippocampus and in the hilus of the dentate gyrus was higher than in controls. In contrast, KA administration lowered the number of NADPH-diaphorase positive cells in all studied hippocampal areas and in both blades of the dentate gyrus. Massive cell degeneration was observed in CA1 and CA3 areas of the hippocampus and in the hilus of the dentate gyrus after kainic acid administration. Animals exposed to kainic acid and pretreated with nicotine exhibited degeneration to a lesser extent and the number of NADPH-diaphorase positive cells was higher compared to rats, which were exposed to kainic acid only., V. Gastrointestinal form is the second stage of the Acute Radiation Syndrome (ARS) with a threshold dose of 8 Gy. It represents an absolutely lethal clinical-pathological unit, enteritis necro-hemorrhagica (duodenitis, jejunitis, ileitis, respectively) with unknown causal therapy. The purpose of our study has been to evaluate the morphological changes in a model of radiation-induced enteritis in rats and estimate the significance of changes in biodosimetry. Wistar rats were randomly divided into 21 groups, 10 animals per group. Samples of the jejunum were taken 24, 48, 72, and 96 h after the whole-body γ-irradiation with the doses of 1, 5, 10, 15, and 20 Gy, and routinely stained with hematoxylin and eosin. Five morphometric markers – intercryptal distance, enterocytal height on the top and base of villus, length of basal lamina of 10 enterocytes and enterocytal width – in irradiated rat jejunum were examined. The results were compared with sham-irradiat ed control group. After lethal doses of irradiation, all morphometric parameters of jejunum significantly changed. With the exception of intercryptal distance, they might be considered as suitable biodosimetric markers under these experimental conditions. Our morphometry results in radiation-induced jejunitis are in accordance with those in other studies. We were the first who quantified morphological post-irradiation changes in animal jejunum. Some of them might be used under experimental conditions. This experimental study is a predecessor of the clinical assessment of a specific marker. Under clinical practice, the sensitive biodosimetric parameter could serve as one of the guidance for evaluation of the absorbed dose in irradiated troops as well as rescue workers. The taxonomical status of Paranoplocephala bairdi (Schad, 1954)-like cestodes (Anoplocephalidae) in heather voles Phenacomys spp. and tree voles Arborimus spp. (Muridae: Arvicolinae) and their discrimination from five related species of Paranoplocephala is assessed using uni- and multivariate morphometrics. The analyses support the independent status and conspecificity of specimens from Phenacomys spp. and Arborimus spp., and P. bairdi is therefore suggested to be a host-specialist species of heather and tree voles with a wide geographical distribution in North America. A redescription is presented for P. bairdi.
To date, three species of Isospora Schneider, 1881 have been described from lizards of the genus Acanthodactylus Wiegmann. Two of these, although representing separate species parasitizing two different hosts, Acanthodactylus boskianus Daudin in Egypt and A. schmidti Haas in Saudi Arabia, were described under the name Isospora acanthodactyli. The third species is Isospora abdallahi Modrý, Koudela et Šlapeta, 1998 from A. boskianus in Egypt. In the present study, Isospora alyousifi nom. n. is proposed to accommodate Isospora acanthodactyli Alyousif et Al-Shawa, 1997 (homonym of I. acanthodactyli Sakran, Fayed, El-Toukhy et Abdel-Gawad, 1994) and its redescription based on newly collected material is provided.
The external morphology of two bucephalid digenean parasites of Conger conger (Linnaeus) (Congridae, Anguilliformes) caught northwest of the Iberian Peninsula, Prosorhynchus crucibulum (Rudolphi, 1819) Odhner, 1905 and P. aculeatus Odhner, 1905, were studied using scanning electron microscopy (SEM). SEM techniques elucidated new external morphological details, mainly relating to the tegument and protruding organs, such as, in P. crucibulum, a papilla-like structure associated with the pharynx and, in P. aculeatus, the cirrus. The tegument bears scale-like spines, which in both species are arranged quincuncially. The spines of P. crucibulum are wider than long and cover the major part of the body and rhynchus. However, no spines were found in either the central apical depression of the rhynchus or in the middle of the ventral indentation. Also, spines were rarely seen on the tegument around mouth, around the genital aperture or close to the excretory pore. P. aculeatus has spines of a different shape, as wide as they are long and with a rounded margin. They cover the whole body and almost the entire rhynchus, but none were found in the middle of the rhynchus or on its neck region.
