The aim of our study was to determine the effect of selected cytostatics on a human ovarian cancer cell line A2780 as a model system for ovarian cancer treatment. This cell line is considered cisplatin-sensitive. Panel of tested cytostatics included cisplatin, paclitaxel, carboplatin, gemcitabine, topotecan and etoposide. These cytostatics have a different mechanism of action. To evaluate cytotoxic potential of the tested compounds, the methods measuring various toxicological endpoints were employed including morphological studies, MTT assay, dynamic monitoring of cell proliferation with xCELLigence, cell cycle analysis, caspase 3 activity and expression of proteins involved in cell cycle regulation and cell death. Mantle cell lymfom (MCL) neboli lymfom z plášťových buněk patří mezi vzácnější B-lymfoproliferace s podílem asi 7–8 % na celkovém množství všech nehodgkinových lymfomů. Klinicky se MCL vyznačuje neblahou kombinací vlastností indolentních (rekurentní průběh) a agresivních (vysoká proliferační aktivita, chemorezistence) lymfomů. Prognóza onemocnění zůstává stále neuspokojivá s mediánem přežití 4–5 let. V současné době se předpokládá, že pro vznik klinicky manifestního MCL je nutný souběh několika patologických událostí na genové úrovni. Nezbytným předpokladem je porucha regulace buněčného cyklu řízené cyklinem D1, za další krok se považuje defekt v buněčné odpovědi na poškození DNA. Prvotní a zásadní genovou přestavbou je u MCL pravděpodobně translokace t(11;14), při níž vzniká patologický fuzní gen BCL1/IGH. Produktem této přestavby je nadměrná produkce cyklinu D1, která však ke vzniku maligního lymfomu sama o sobě nestačí. U MCL je také častá delece 11q22–23. Na tomto lokusu je umístěn gen pro ATM kinázu. V případě delece ATM genu se výrazně zvyšuje genomická nestabilita a vzrůstá tak frekvence dalších mutací. Na základě studií genové exprese byly identifikovány další patogenetické mechanizmy. Jedním z nich je například konstitutivní aktivace transkripčního faktoru NF-kappaB majícího antiapoptotický účinek nebo aktivace systému mTOR zvyšujícího translaci mRNA. Je důležité zmínit možnost cíleného terapeutického ovlivnění výše zmíněných drah (bortezomib, temsirolimus). Dalšími sekundárními aberacemi je rekurentní somatická mutace NOTCH1 a SOX 11 (neuronální transkripční faktor), jejich význam v prognóze a patogenezi zůstává nejednoznačný., Mantle cell lymphoma represents about 7-8 % of all non-Hodgkin lymphomas. This entity is characterized by a combination of mostly aggresive clinical course and frequent relapses. The prognosis remains poor with a median overall survival of 4-5 years. It is hypothesized that a coincidence of several pathological events is necessary for arise of clinically manifest MCL. The primary oncogenic mechanism is considered to be a chromosomal translocation t(11;14) with juxtaposition of BCL1 gene to the IGH gene. It results in overexpression of cyclin D1, which positively affects the G1-S transition of cell cycle. Another frequent mutation is a deletion 11q22-23 leading to the inactivation of the ATM gene responsible for the DNA damage response. In such case, the genomic instability and subsequently the frequency of another mutations is highly increased. The gene expression profiling revealed many additional genetic alterations, such as constitutive activation of trancription factor NF-kappaB with anti-apoptotic effect or activation of mTOR system. Importantly, these pathways can be influenced by new targeted drugs (bortezomib, temsirolimus). Another secondary aberrations found in MCL are mutations in NOTCH1 gene and overexpression of neuronal transcription factor SOX11.
