The insect growth regulator NC-184, a juvenile hormone mimic, prevents moulting to the adult stage in the desert locust, Schistocerca gregaria (Forskål) (Orthoptera: Acrididae). Male nymphs treated in the penultimate or final nymphal instar with NC-184 exhibit precocious mating behaviour in the final instar. We examined whether this chemical affects the development of the internal reproductive organs of crowded nymphs. In treated males, both accessory glands and seminal vesicles were underdeveloped, and no sperm was found in the seminal vesicle, whereas these organs in control individuals had greatly increased in size 10 days after treatment, when all the insects had moulted to adults. Testis size in treated males was similar to that in controls, regardless of their smaller body size due to the inhibition of moulting. Oogenesis and development of spermatheca in females treated with NC-184 continued to some degree, but no eggs matured, unlike what occurred in the control. In conclusion, treatment of S. gregaria nymphs with NC-184 resulted in changes in the reproductive organs in both sexes.
The new Hungarian Labour Code came into effect on the 1st of July 2012. The main objective of the reform is flexibilisation of labour law in order to increase the employment rate by promoting competitiveness of employers. This new orientation of labour law policy has been widely criticised, since the immediate effect of the new Act is the reduction of employee protection. Firstly, the paper describes the main objectives and background of the labour law reform. Secondly, we elucidate the main points of the reform, with a special emphasis on the situation of trade unions. The main question is, what effects may be expected as a consequence of flexibilised employment protection and diminished trade union rights.
The metabolic turnover in the isolated in vitro perfused and superfused rat skeletal muscle (musculus gracilis cranialis) was enhanced by increasing the medium flow rate under relaxed conditions. In a recent study we have measured the tissue concentrations of second messengers: cyclic adenosine 3 ́5 ́- monophosphate (cAMP), cyclic guanosine 3 ́5 ́-
monophosphate (cGMP), and D-myo-inositol 1, 4, 5-trisphosphate (IP3) under similar experimental conditions to analyze their potential role in the described stimulation of metabolic rate by changes of perfusion flow rate. The tissue levels of the two second messengers’ cAMP and cGMP were not significantly changed after increasing the perfusion
flow rate and they probably have no transduction role in the induced alteration of skeletal muscle metabolism. However, the IP3 content was extremely reduced after increasing flow rate. This decrease in the tissue concentration of IP3 induced by increasing the flow rate indicates the possible role of IP3 in this signal transduction, leading to changes in
the cellular metabolic pathways.
The aim of this study was to investigate the effect of 7-oxo-DHEA (dehydroepiandrosterone) on the serum levels of steroid sexual hormones, gonadotropins, lipids and lipoproteins in men. 7-oxo-DHEA was applied onto the skin as a gel to 10 volunteers aged 27 to 72 years for 5 consecutive days. The single dose contained 25 mg 7-oxo-DHEA. Serum concentrations of testosterone, estradiol, cortisol, androstenedione, luteinizing hormone (LH), follicle-stimulating hormone (FSH), sex hormone binding globulin (SHBG), total cholesterol, HDL- and LDL-cholesterol, triglycerides, apolipoprotein A-I and B and lipoprotein(a) were measured before the beginning and shortly after the end of the steroid application. After the treatment, we noted the following significant changes: a decline of testosterone and estradiol levels, increase of LH, HDL-cholesterol and apolipoprotein A-I levels. The decrease of total cholesterol levels was of the borderline significance. A slight but significant increase was found in apolipoprotein B and lipoprotein(a). The most expressive was the fall of the atherogenic index. In order to ascertain the kinetics of absorption and metabolism of transdermally administered dehydroepiandrosterone (DHEA), 10 men 29-72 years old (mean 52.4±14.5) received 50 mg DHEA/day in a gel applied onto the skin of the abdomen for 5 consecutive days. The objective was to establish the extent to which DHEA influences the levels of gonadotropins, sex hormone-binding globulin and lipids. It was found that DHEA is well absorbed and rapidly metabolized to its sulfate (DHEAS), androstenedione, and consequently to testosterone and estradiol. The DHEA levels that markedly increased after the first doses gradually declined already during the application, and this decline proceeded even after it was discontinued, reaching levels significantly lower than the original ones. On the other hand, the levels of DHEA metabolites (with the exception of DHEAS) rose during the application and reached values significantly higher than the basal ones within 5 weeks. This effect was accompanied by significantly decreased levels of LH. The serum levels of lipids, namely of cholesterol (both HDL and LDL cholesterol), triglycerides, apolipoproteins A-I and B and lipoprotein(a) after DHEA application were not changed significantly, and the atherogenic index (AI) remained unaltered. However, some correlations between hormones and lipids were found. Negative correlations concerned the following indices: DHEA/Lp(a); DHEAS/cholesterol; DHEA, DHEAS, testosterone/TG; testosterone/AI. On the other hand, LH, FSH/cholesterol, FSH, SHBG/LDL cholesterol, FSH/Apo B, Lp(a) correlated positively. It can be concluded that transdermal short-time application of DHEA results in a decrease of endogenous DHEA after finishing the treatment, with a parallel marked increase in the levels of sex hormones. The spontaneously hypertensive rat (SHR is the most widely used animal model of essential hypertensio and left ventricular hypertrophy. Catecholamines play an important role in the pathogenesis of both essential hypertension in humans and in the SHR. Recently, we obtained evidence that the SHR harbors a variant in the gene for dopamine beta hydroxylase (Dbh) that is associated with reduced adrenal expression of Dbh mRNA and reduced DBH enzymatic activity which correlated negatively with blood pressure. Dbh expression predisposes the SHR to hypertension and that augmentation of Dbh expression would reduce blood pressure. We derived 2 new transgenic SHR-Dbh lines expressing Dbh cDNA under control of

hrine. The observed changes in catecholamine metabolism were associated with increased blood pressure and left ventricular mass in both transgenic lines. We did not observe any consistent changes in brainstem levels of catecholamines or of mRNA levels of Dbh in the transgenic strains. Contrary to our initial expections, these findings are
consistent with the possibility that genetically determined decreases in adrenal expression and activity of DBH do not represent primary determinants of increased blood pressure in the SHR model.
Exposure to chronic stress stimulates the hypothalamic-pituitaryadrenal (HPA) axis and then simultaneously inhibits hypothalamic-pituitary-gonadal axis (HPG) axis activity. The inhibition formed by the HPA axis is the main mechanism of action of stress on reproductive function. HPG axis activity is known to be changed by various factors, including exercise. Exercise has been found to have a number of positive effects on sexual behavior, reproductive hormones, and sperm parameters in studies with animal models for many years. The main aim of this study is to investigate the effects of chronic treadmill exercise on chronically stressed-male rats’ sexual behavior, reproductive hormones, and sperm parameters. A total of 40 sexually adult male rats were randomly and equally divided into four groups as control, stress, exercise, and stress+exercise. Animals in the exercise group were subjected to the chronic treadmill exercise (moderate intensity) for 33 days with a periodic increase in speed and duration. Animals in the stress group were exposed to restraint stress for 1 h, 2 h, and 3 h during the first, second and third 15 days respectively. Sexual behavior parameters, hormone measurements, and sperm parameters were evaluated. The main effects of chronic exercise on sexual behavior were centered on a significant increase in the ejaculation frequency (EF) in the stress+exercise group. Also, sperm concentration and motility in the stress group significantly decreased, and then sperm motility was improved by exercise in the stress+exercise group. The in vivo effects of recombinant human interleukin-1 receptor antagonist (rhIL-1Ra) administration on endogenous IL-1 levels in the circulation and conditioned media (CM) from different immunohematopoietic organ/tissues were studied in CBA mice under steady state and postirradiation conditions. In normal mice, constitutive IL-1 levels were demonstrated in the plasma, CM of peritoneal exudate cells and full-thickness skin explants with low or undetectable levels in CM of splenic and bone marrow cell suspensions. In irradiated mice (2 Gy, X rays) on day 3 post exposure a significant increase of IL-1 levels was seen in the circulation and CM of peritoneal exudate cells, with no significantly different levels in postirradiation bone marrow, spleen and skin. After rhIL-1Ra treatment of the animals (2 x 50 mg/mouse, i.p.), significantly elevated IL-1 levels were observed in the skin and CM of peritoneal exudate cells in normal mice, whereas slightly increased levels were detected in CM of splenic cells. The rhIL-1Ra administration in irradiated mice led to decreased IL-1 concentrations in the circulation, and CM of peritoneal exudate cells and skin. The results pointed out the importance of IL-1 secretion and receptor expression in the maintenance of homeostasis in steady state, as well as during recovery after irradiation. The maize weevil, Sitophilus zeamais (Motschulsky), is a major pest of stored grain kernels. Irradiation is an established technique for controlling insects in stored grain and is a major stress factor affecting these insects. Since heat shock protein (hsp) genes respond to this stress, we proposed that hsps may be associated with irradiated stress tolerance in S. zeamais. The responses of the maize weevil to exposure to ultraviolet-C (UV-C) and microwave irradiation were assessed at four developmental stages: egg, larva, pupa and adult. The results revealed that exposure to UV-C (254 nm, < 8 h) did not affect the survival of the maize weevils; however, Szhsp70, Szhsc70 and Szhsp90 mRNA levels significantly increased during the first 1 h of UV-C exposure. The median lethal time (LT50) of exposure to microwave irradiation indicated that the adult stage was more tolerant of microwave irradiation than the other developmental stages. Microwave irradiation enhanced the expression of the three hsps, but the intensity of up-regulation differed among the three genes, with Szhsp70 the most highly up-regulated. Our experiments revealed that UV-C and microwave irradiation influenced the expression profile of hsp genes in S. zeamais. Soil salinization and alkalinization frequently co-occur in naturally saline and alkaline soils. To understand the characteristics of mixed salt-alkali stress and adaptive response of Medicago ruthenica seedlings to salt-alkali stress, water content of shoots, growth and photosynthetic characteristics of seedlings under 30 salt-alkaline combinations (salinity 24-120 mM and pH 7.03-10.32) with mixed salts (NaCl, Na2SO4, NaHCO3, and Na2CO3) were examined. The indices were significantly affected by both salinity and pH. The interactive effects between salt and alkali stresses were significant, except for photosynthetic pigments. Water content of shoots, relative growth rates of shoots and roots and pigment concentrations showed decreasing trends with increasing salinity and alkalinity. The root activity under high alkalinity and salinity treatments gradually decreased, but was stimulated by the combined effects of low alkalinity and salinity. The survival rate decreased with increased salinity, except at pH 7.03-7.26 when all plants survived. Net photosynthetic rate, stomatal conductance and intercellular CO2 concentration decreased with increased salinity and pH. M. ruthenica tolerated the stress of high salt concentration when alkali concentration was low, and the synergistic effects of high alkali and high salt concentrations lead to the death of some or all seedlings. M. ruthenica appeared to be salt-alkali tolerant. Reducing the salt concentration or pH based on the salt components in the soil may be helpful to abate damage from mixed salt-alkaline stress. and J.
