The Central European flora is an important source pool of plant species introduced to many regions throughout theworld. In this study,we identified a total of 759 plant species of the Central European flora that are currently recognized as alien species in Australia. We explored temporal patterns of introduction of these species to Australia in relation to method of introduction, growth form, naturalization status and taxonomy. Across all species, substantially larger numbers of species were introduced between 1840 and 1880 as well as between 1980 and the present, with a small peak of introductions within the 1930s. These patterns reflect early immigration patterns to Australia, recent improvements in fast and efficient transportation around the globe, and emigration away from difficult conditions brought about by the lead up to the Second World War respectively. We found that the majority of species had deliberate (69%) rather than accidental (31%) introductions and most species have not naturalized (66% casual species, 34% naturalized species). A total of 86 plant families comprising 31 tree species, 91 shrub species, 533 herbaceous species and 61 grass species present in Central Europe have been introduced to Australia. Differential patterns of temporal introduction of species were found as a function of both plant family and growth form and these patterns appear linked to variation in human migration numbers to Australia.
Plant volatiles can synergize the response to moth pheromone. Synthetic pheromone analogs, in turn, have the opposite effect in reducing pheromone attractiveness. To determine how these two types of stimuli interact and influence male moth behaviour, we performed wind tunnel experiments on the grapevine moth, Lobesia botrana. We noticed that a blend of host plant volatiles [(E)-β-caryophyllene, 1-hexanol, (Z)-3-hexenyl acetate and 1-octen-3-ol in a 100:20:10:5 ratio] significantly increased the response of males to an optimized blend of sex pheromone [(7E,9Z)-dodeca-7,9-dienyl acetate (E7,Z9-12:Ac), (7E,9Z)- dodeca-7,9-dienol (E7,Z9-12:OH) and (Z)-9-dodecenyl acetate (Z9-12:Ac)] in a 100:10:2 ratio. However, the response of males to the natural attractant was significantly reduced by two analogs [(9E,11Z)-tetradeca-9,11-dien-2-one (MK 2) and [(9E,11Z)-1,1,1-trifluoro-tetradeca-9,11-dien-2-one (TFMK 3)], of the major component of the sex pheromone of the insect (E7,Z9-12:Ac). When both stimuli were tested on males at pheromone:analog:plant volatile blend 1:100:1000 ratio, the plant blend offset the inhibitory effect induced by TFMK 3 but not that of MK 2. A greenhouse experiment was conducted to study the indirect effects of soil salinity on a caterpillar that induces gall formation on a non-halophilic plant. Larvae of Epiblema scudderiana (Clemens) were allowed to feed on potted goldenrods (Solidago altissima L.) treated with 3 concentrations of NaCl (0, 8,000, 16,000 PPM). Experiments were also carried out with the larvae of two species of leaf beetles, Trirhabda borealis Blake, a leaf-chewer, and Microrhopala vittata F., a leaf-miner, to determine the influence of feeding guild. Adding salt to the soil affected both the plant and insect herbivores. The biomass of roots and shoots as well as root/shoot ratios of salt-stressed plants were lower, relative to controls. The biomass of the fully grown larvae and galls were decreased for the plants treated with the highest salt concentration. The percentage of biomass allocated to the gall was increased by soil salinity. All gall-inducing larvae completed their development (from second to final instar) even though their biomass was significantly reduced in the 16,000 PPM treatment. Soil salinity increased nitrogen concentrations in both gall and stem (normal) tissues but the levels were always higher in the gall. The salt treatments also increased sodium and potassium concentrations in galls and stems. Interestingly, sodium concentrations as well as the ratio of sodium ions to potassium ions increased more rapidly in the stem compared with the gall. Responses of folivorous insects to salt-stressed plants varied. Leaf-chewing larvae ate smaller amounts of plant tissue with high salt content compared with control, which also resulted in shorter feeding periods. The performance of the leaf-mining insect was not affected. However, it was able to complete its larval development within a smaller portion of the leaves. This study showed that soil salinity has a strong negative effect on S. altissima, especially on root development. Conversely, salt stress effects seemed to be progressively decreasing from the stem to the gall to the gall-inducer, which suggests that the gall tissue might act as a buffer against drastic changes in the mineral balance of the host plant. Nevertheless, it seems that unless the host plant dies, larvae of E. scudderiana can always produce a gall in which they can complete their development. On the other hand, leaf-chewing insects appeared to be sensitive to salt-rich tissues since they were deterred by them. Leaf-miners could complete their development with fewer food without any effect on their growth, suggesting that the peculiar tissues on which they feed within leaves became more abundant or nutritious in salt-treated plants.
Cardiovascular and neuroendocrine responses to exercise in a physically fit and an untrained group of young healthy subjects were compared to study the significance of physical fitness for performance in a discipline for which the athletes were not trained. Ten wrestlers of national rank prepared for an international competition (age 18 years) and 9 untrained healthy males (age 21 years). Exercise consisted of 27-min swimming, freestyle, in water of 29 °C, with last 3 min increased to maximal effort. The blood pressure, heart rate and sublingual temperature were measured and blood samples were withdrawn before exercise, immediately after and after a 30 min period of rest. Catecholamines were analyzed by radioenzymatic method and plasma renin activity (PRA) using commercial kits. Systolic blood pressure and heart rate after swimming were increased comparably in the two groups, diastolic pressure was unchanged in the controls and decreased in the wrestlers. Plasma cortisol remained unchanged. Plasma glucose tended to increase in the controls and so decrease in wrestlers, with a significant difference between them after swimming (p<0.05). However, plasma adrenaline was concomitantly increased in both groups (p<0.01). Noradrenaline and PRA were increased after swimming in both the control and trained group. The increments of noradrenaline and PRA in wrestlers were significantly reduced compared to the control group (p<0.01, p<0.05, respectively). Higher physical fitness in athletes significantly reduced plasma noradrenaline and angiotensin responses to maximal exercise demanding special skill in work performance which had not been included in their training program. Training of wrestlers did not cause an exaggerated plasma adrenaline response to exercise.
Clusterin is a heterodimeric glycoprotein with wide range of functions. To further explore its possible regulatory role in energy homeostasis and in adipose tissue, we measured plasma clusterin and its mRNA expression in subcutaneous adipose tissue (SCAT) of 15 healthy lean women, 15 obese women (OB) and 15 obese women with type 2 diabetes mellitus (T2DM) who underwent a 2-week very low-calorie diet (VLCD), 10 obese women without T2DM who underwent laparoscopic sleeve gastrectomy (LSG) and 8 patients with T2DM, 8 patients with impaired glucose tolerance (IGT) and 8 normoglycemic patients who underwent hyperinsulinemic euglycemic clamp (HEC). VLCD decreased plasma clusterin in OB but not in T2DM patients while LSG and HEC had no effect. Clusterin mRNA expression in SCAT at baseline was increased in OB and T2DM patients compared with controls. Clusterin mRNA expression decreased 6 months after LSG and remained decreased 12 months after LSG. mRNA expression of clusterin was elevated at the end of HE C compared with baseline only in normoglycemic but not in IGT or T2DM patients. In summary, our data suggest a possible local regulatory role for clusterin in the adipose tissue rather than its systemic involvement in the regulation of energy homeostasis., J. Kloučková, Z. Serum adipocyte fatty acid-binding protein (FABP-4) concentrations are linked to human obesity and other features of metabolic syndrome. Patients with Cushing's syndrome (CS) develop numerous features of metabolic syndrome due to chronic cortisol excess. Here we tested the hypothesis that chronically increased cortisol levels in CS patients may alter circulating levels of FABP-4. Fourteen patients with CS, 19 patients with simple obesity (OB) and 36 healthy control subjects (C) were included in the study. Serum FABP-4 concentrations were significantly higher in both CS and OB patients relati ve to C group, but they did not differ between CS and OB groups. In a combined population of all groups, serum FABP-4 levels correlated positively with BMI, body fat content, serum glucose, triglycerides, HbA1c and HOMA index and were inversely relate d to HDL-cholesterol, resting energy expenditure and freeT3 levels. We conclude that FABP-4 levels are significantly increased in both patients with simple obesity and obese patients with Cushing's syndrome. We suggest that increased FABP-4 concentrations in CS patients are rather due to their excessive fat accumulation and related metabolic abnormalities than due to a direct effect of cortisol on FABP-4 production., V. The objective of this study was to measure plasma fibroblast growth factor 21 and 19 (FGF21 and FGF19) levels in patients with Cushing's syndrome (CS) and to compare it with those of lean control subjects (C) and patients with obesity (OB). Fourteen untreated patients with CS, 19 patients with OB and 36 controls were included in the study. Plasma FGF21 and FGF19 levels were measured by ELISA kits, other hormonal and biochemical parameters were measured by standard laboratory methods. Plasma FGF19 did not significantly differ among the studied groups. Plasma FGF21 levels were significantly higher in both CS and OB groups relative to C group but they did not differ between CS and OB groups. In a combined population of all three groups FGF21 levels positively correlated with BMI, waist circumference and percentage of total and truncal fat mass. Less prominent inverse relationship with these parameters was found for FGF19. Neither FGF21 nor FGF19 were significantly related to cortisol concentrations. Increased FGF21 concentrations in both patients with CS and OB relative to lean subjects suggest that excessive body fat and/or related metabolic abnormalities rather than direct effects of cortisol are responsible. The impact of anesthetic agen ts on endocrine and metabolic factors is an important issue. The present study has compared the effects of a short-term exposure to diethyl ether, isoflurane, or CO2 on plasma corticosterone, insulin and glucose concentrations since the duration of anesthetic exposure may have an effect on those factors. Male rats were divided into fed and fasted groups. The experimental rats were briefly exposed to diethyl ether, isoflurane, or CO2 (the degree of anesthesia was identical), while a control group was not exposed to the anesthetics. In the fed rats, diethyl ether exposure increased the levels of plasma glucose. CO2 exposure decreased plasma corticosterone and increased plasma glucose levels. Isoflurane exposure caused no changes in plasma corticosterone, glucose, or insulin levels. In the fasted rats, diethyl ether exposure increased plasma corticosterone and reduced plasma insulin levels. The plasma corticosterone and insulin levels were significantly increased by CO2 exposure. Isoflurane exposure decreased plasma insulin levels. A brief exposure to either diethyl ether or CO2 changed the plasma corticosterone, glucose, and insulin levels in fed and/or fasted rats. Tissue betaine is an intracellular osmolyte that also provides a store of labile methyl groups. Despite these important biological roles, there are few data regarding tissue betaine content. We measured the betaine concentration of plasma and various tissues (brain, heart, lungs, liv er, kidney, spleen, intestine, reproductive tissues, skeletal muscle and skin) in male and female rats and assessed whether there were any gender-specific differences in betaine content or distribution and whether there was any relationship between tissue accumulation and plasma levels. Betaine was highest in the liver and kidney with values ranging from 1.6 to 9.5 mmol/l and 2.0 to 5.4 mmol/l, respectively. Plasma betaine concentrations were significantly lower than tissue levels except in the brain ( ≈ 25 % of plasma) and skeletal muscle (similar to pl asma). Regression analysis of the combined male and female data revealed a significant plasma-related accumulation of betaine in the heart, skin and skeletal muscle, while the lung, liver, kidney, spleen, and intestine showed significant plasma-related and plasma- independent accumulations of be taine. The betaine content of the skin, liver and kidney was no t significantly different between males and females, but in plasma and all tissues analyzed it was significantly higher in males (P<0.01)., S.
