The effects of water stress on leaf surface morphology (stomatal density, size, and trichome density of both adaxial and abaxial surfaces) and leaf ultrastructure (chloroplasts, mitochondria, and cell nuclei) of eggplant (Solanum melongena L.) were investigated in this study. Higher stomata and trichome densities were observed on abaxial surface compared with the adaxial surface. Compared with well watered (WW) plants, the stomata and trichome density of the abaxial surface increased by 20.39% and 26.23% under water-stress condition, respectively. The number of chloroplasts per cell profile was lesser, the chloroplasts became round in a shape with more damaged structure of membranes, the number of osmiophilic granules increased, and the number of starch grains decreased. The cristae in mitochondria were disintegrated. The cell nuclei were smaller and the agglomerated nucleoli were bigger than those of WW plants. A natural population of Psorophora ferox (Humbold, 1820) infected with the microsporidium Amblyospora ferocis García et Becnel, 1994 was sampled weekly during a seven-month survey in Punta Lara, Buenos Aires Province, Argentina. The sequence of development of A. ferocis in larvae of P. ferox leading to the formation of meiospores followed the developmental pathway previously reported for various species of Amblyospora. The natural prevalence of A. ferocis in the larval population of P. ferox ranged from 0.4% to 13.8%. Spores were detected in the ovaries of field-collected females of P. ferox and were shown to be responsible for transovarial transmission of A. ferocis to the next generation of mosquito larvae in laboratory tests. These spores were binucleate and slightly pyriform in shape. The prevalence of A. ferocis in the adult population ranged from 2.7% to 13.9%. Data on effects of the infection on female fecundity showed that infected field-collected adults of P. ferox laid an average of 47.6 ± 6.5 eggs of which 35.8% ± 4.1% hatched. Uninfected field-collected adults of P. ferox laid 82.8 ± 6.8 eggs of which 64.1% ± 5.5% hatched. Six species of copepods living together with P. ferox were fed meiospores from field-infected larvae but none became infected. Horizontal transmission of A. ferocis to P. ferox larvae remains unknown.
Ileterosporous (polymorphic) microsporidia in mosquitoes are characterized by intricate life cycles involving multiple spore types responsible for horizontal (per os) and vertical (transovarial) transmission. They affect two generations of the mosquito and some involve an obligate intermediate host. Heterosporous microsporidia are generally very host and tissue specific with complex developmental sequences comprised of unique stages and events. Full details on the intricate relationships between heterosporous microsporidia and their mosquito hosts have only recently been elucidated. Edhazardia aedis (Kudo, 1930) and Culicospora magna (Kudo, 1920) have developmental sequences in larvae that involve gametogony followed by plasmogatny and nuclear association to form diplokarya. These diplokaryotic stages then undergo karyogamy and form binucleate spores responsible for transovarial transmission. In the filial generation, haplosis occurs as a result of nuclear dissociation to produce uninucleate spores infectious to larval mosquitoes. Amblyospora cali-fornica (Kellen et Lipa, 1960) has similar sequences except that haplosis is by meiosis to produce spores infectious for a copepod intermediate host. A third spore type is formed in the intermediate host responsible for infection in a new generation of the mosquito host.
Two kinds of cecidomyiid galls induced by Daphnephila on Machilus thunbergii Sieb. & Zucc. leaves at various developmental stages, i.e., young, growing, and mature, were analyzed for their biochemical composition of photosynthetic pigment-protein complexes located in thylakoid membranes using the Thornber and MARS electrophoretic fractionation systems. Both kinds of galls were totally deficient in the pigment-protein complexes CP1, and A1, AB1, and AB2 through the whole period of gall formation. Immunoblotting of antibody against light-harvesting complex 2b (LHC2b) apoprotein confirmed this deficiency in gall's lifetime, which never recovered under any condition. Electron microscopy demonstrated that already at the early developmental stage the gall chloroplasts had thylakoid morphology like that in a normal leaf. and C. M. Yang ... [et al.].
Alterations in geometry and structure of coronary arteries have marked consequences on blood flow to the respective area. We evaluated long-term effect of losartan on blood pressure (BP), heart weight/body weight (HW/BW), geometry and structure of septal branch of coronary artery (RS) of young SHR and Wistar rats. Four-week-old Wistar rats and SHR were used. Losartan was administered (20 mg/kg/day) in drinking water by gavage for 5 weeks. BP was measured by plethysmographic method. Cardiovascular system was perfused with a fixative (120 mm Hg). RS was processed for electron microscopy. Wall thickness of intima + media (WT), inner diameter (ID), cross-sectional area of intima + media (CSA), volume dens ities (VD) of endothelial cells (EC), extracellular matrix (ECM) of intima, smooth muscle cells (SMC) and ECM of media were eval uated. BP of 4-week-old SHR did not differ from that of Wistar rats. BP, HW/BW, WT, CSA, WT/ID, CSAs of SMC, ECM of media were increased in 9-week- old SHR, whereas their VD and CSA of EC were decreased. Losartan administration decreased BP and HW/BW in both groups. Geometry of RS was affected only in SHR (reduction of WT, CSA, WT/ID and increased of ID, circumferential tension, VD and CSA of EC). Two of 98 native littleneck clams, Protothaca staminea Conrad, from Cooper’s Cove, Sooke Basin were infected with an eimeriorin coccidian parasite. Merogonic, gamontogonic and sporogonie development were observed in renal tubular epithelial cells. Sporulation of the oocysts occurred within the host. Mature oocysts were spherical mean 41 pm (range 30-44), and contained about 32 subspherical sporocysts (9 x 10 pm), each of which contained 4 sporozoites. Spherical 19 pm (18-20), cyst-like structures and smaller multinucleate bodies, some of which resembled sporocysts, were also seen.A review of the coccidian parasites of bivalves led to the erection of the new genus, Margolisiella (family Eimeriidae Minchin, 1903) to accommodate M. kabatai sp. n., the parasite in Protothaca staminea described herein. Four previously described monoxenous species (Pseudoklossia patellae Debaisieux, P. chitonis Debaisieux, P. tellinovum Buchanan and P. halintis Friedman, Gardner, Hedrick, Stephenson, Cawthom et Upton) were also transferred to the new genus. The 2 remaining possibly heteroxenous species (P. pelseneeri Léger and P. glomerata Léger et Duboscq) were retained in the genus Pseudoklossia Léger et Duboscq (family Aggregatidae Labbé, 1899).
In the mating behaviour of Aphidius ervi Haliday the antennae play a pivotal role in partner recognition and acceptance. Mating failure was always observed when antennal contact was experimentally prevented. The male of A. ervi has filiform antennae, consisting of scape, pedicel and 18-20 cylindrical antennomeres (flagellar segments), which bear numerous types of sensory structures and, interspersed among the multiporous plate sensilla, especially on the 1th and 2nd flagellar segments, scattered pores. A secretion oozes from these pores in virgin males exposed to conspecific females. Transmission electron microscopy revealed that these pores are the external openings of integumentary glands. Behavioural and morpho-functional observations indicated that a double step sex recognition mechanism is present in A. ervi, as in other parasitic Hymenoptera. Basically, female recognition by males appears to be mediated by a volatile sex pheromone, that triggers the behavioural sequence leading to mounting. Then, the female recognizes and accepts the male after antennal contact. This is mediated by the secretion that oozes from the male antennal glands, which acts as a contact pheromone.
Marssoniella elegans Lemmermann, 1900, a parasite of ovarial tissues of the copepod Cyclops vicinus Uljanin, 1875, was studied as a representative of aquatic-clade microsporidia which form ''heteroinfectious spores'' (spores not infective to the original host as opposed to ''homoinfectious spores'' which are infective for the original host) and which thus should require an alternate host. Several structural characters of this microsporidian are similar to those of copepod morphs of microsporidia infecting mosquitoes. However, small subunit ribosomal DNA phylogeny indicates that caddis flies (Insecta, Trichoptera) might be the alternate hosts of Marssoniella. Ultrastructural data obtained are used to redefine the genus Marssoniella Lemmermann, 1900 and its type species Marssoniella elegans.
Previous data suggest that type 1 diabetes mellitus leads to the deterioration of myocardial intercellular communication mediated by connexin-43 (Cx43) channels. We therefore aimed to explore Cx43, PKC signaling and ultrastructure in non -treated and omega-3 fatty acid (omega-3) treated spontaneously diabetic Goto-Kakizaki (GK) rats considered as type 2 diabetes model. Four-week-old GK and non-diabetic Wistar-Clea rats were fed omega -3 (200 mg/kg/day) for 2 months and compared with untreated rats. Realtime PCR and immunoblotting were performed to determine Cx43, PKC- epsilon and PKC-delta expression. In situ Cx43 was examined by immunohistochemistry and subcellular alterations by electr on microscopy. Omega-3 intake reduced blood glucose, triglycerides, and cholesterol in diabetic rats and this was associated with improved integrity of cardiomyocytes and capillaries in the heart. Myocardial Cx43 mRNA and protein levels were higher in diab etic versus non- diabetic rats and were further enhanced by omega-3. The ratio of phosphorylated (functional) to non-phosphorylated Cx43 was lower in diabetic compared to non- diabetic rats but was increased by omega-3, in part due to up -regulation of PKC-epsilon. In addition, proapoptotic PKC-delta expression was decreased. In conclusion, spontaneously diabetic rats at an early stage of disease benefit from omega-3 intake due to its hypoglycemic effect, upregulation of myocardial Cx43, and preservation of cardiovascular ultrastructure. A new species, Sphaeromyxa noblei sp. n., is described from Heteroclinus whiteleggii (Perciformes: Clinidae), a marine fish from the coast of New South Wales in Australia. This raises the number of Sphaeromyxa species to 38; their list is presented. The species is characterised by a layer of branched glycostyles, which is about 2.4 µm thick and is a feature rather unique in Myxosporea. Pansporoblasts form one or two spores. The study of ultrastructure of this species and of those described to date result in recognition of a combination of patterns characterising the genus: plasmodia have marked surface projections, the endoplasm is full of vacuoles larger than in any other myxosporean genus, and contains a special kind of cells, the lobocytes. Sections through polar capsule reveal different appearance of subsequent stretches of the polar filament unlike in other Myxosporea.
