The heteronormative order is based on the categorisation of people into men and women. The categories of man and woman are understood as given, fixed, ahistorical, and universal. Trans* identities represent a ‘border area’ where categories of woman, man, and binary differentiation between ‘male’ and ‘female’ bodies are redefined and re-established. This article is based on an analysis of 18 semi-structured interviews with trans people in the Czech republic. The first part of the article is focused on the ways in which trans* people relate to their bodies. The second part discusses the gender aspects of hormonal body modification. The third part raises the question of passing and legibility in the context of body modification and the representation of their own gender identity.
High salt concentration is a major abiotic stress limiting plant growth and productivity in many areas of the world. Elaeagnus angustifolia L. adapts to adverse environments and is widely planted in the western region of China as a windbreaker and for landscape and soil stabilization. High salt concentrations inhibited photosynthesis of E. angustifolia, but the mechanism is not known. In this paper, RNA-sequencing was used to investigate effects of salt stress on the photosynthetic characteristics of the species. In total, 584 genes were identified and involved in photosynthetic pathways. The downregulation of genes that encode key enzymes involved in photosynthesis and genes correlated to important structures in photosystem and light-harvesting complexes might be the main reason, particularly, the downregulation of the gene that encodes magnesium chelatase. This would decrease the activity of enzymes involved in chlorophyll synthesis and the downregulation of the key gene that encodes Rubisco, and thereby decreases enzyme activity and the protein content of Rubisco., J. Decades of liver regeneration studies still left the termination phase least elucidated. However regeneration ending mechanisms are clinicaly relevant. We aimed to analyse the timing and transcriptional control of the latest phase of liver regeneration, both controversial. Male Wistar rats were subjected to 2/3 partial hepatectomy with recovery lasting from 1 to 14 days. Time-series microarray data were assessed by innovative combination of hierarchical clustering and principal component analysis and validated by real-time RT-PCR. Hierarchical clustering and principal component analysis in agreement distinguished three temporal phases of liver regeneration. We found 359 genes specifically altered during late phase regeneration. Gene enrichment analysis and manual review of microarray data suggested five pathways worth further study: PPAR signalling pathway; lipid metabolism; complement, coagulation and fibrinolytic cascades; ECM remodelling and xenobiotic biotransformation. Microarray findings pertinent for termination phase were substantiated by real-time RT-PCR. In conclusion, transcriptional profiling mapped late phase of liver regeneration beyond 5th day of recovery and revealed 5 pathways specifically acting at this time. Inclusion of longer post-surgery intervals brought improved coverage of regeneration time dynamics and is advisable for further works. In order to obtain basic information on the transport properties of differentiating embryonic nephrons, we examined the 7-day-old chick mesonephros by measuring the transtubular epithelial potential difference (TPD) and by histochemical detection of Na,K-ATPase activity. TPD as an indicator of the electrogenic transport was measured in individual segments of superficial nephrons in vivo. Their electric polarity was always lumen-negative. TPD was reduced by addition of 10 mM KCN applied to the mesonephric nephrons from the outside. In the proximal tubules, TPD was significantly lower (mean±SD: -1.0±0.5 mV) than in the distal and collecting tubules (-2.2±1.0 mV, pŁ0.05). Activity of the sodium pump was evaluated histochemically by detection of ouabain-sensitive potassium-dependent p-nitrophenyl phosphatase in cryostat sections of the mesonephros. The enzyme activity was demonstrated only in distal tubules and in the collecting ducts, but not in the proximal tubules. These findings have revealed significant differences between embryonic nephron segments: the distal tubule, in contrast to the proximal one, is supplied by the sodium pump and is able to generate higher TPD. Studie shrnuje současné poznatky o teorii transformačního vedení. Zabývá se tématy, která jsou důležitá pro další výzkum transformačního vedení a pro využití teorie transformačního vedení v psychologické praxi v České republice a na Slovensku. Zařazuje transformační teorii do kontextu dalších teorií vůdcovství, představuje pozitivní důsledky transformačního vedení na úrovni organizací, týmů i jednotlivců, věnuje se také kritice teorie a jejím slabým stránkám. Všímá si metody měření transformačního vedení a její validity a možností rozvoje vedoucích v organizacích. Závěr je věnován aktuálním trendům ve výzkumu transformačního vedení., The overview study sums up the contemporary knowledge concerning the transformational leadership theory. It deals with topics important for further study of transformational leadership and for utilization the theory of transformation leadership in psychological praxis in the Czech Republic and Slovakia. It puts the transformational theory into the context of further leadership theories, presents the positive impacts of transformational leadership on levels of organizations, teams, and individuals, deals also with the critics of the theory and its weak aspects. It also takes notice of the method of transformational leadership measurement and its validity, and possibilities of development of leaders in organizations. Blood monocytes (BMs) from 139 subjects (70 malignant melanoma patients, 31 breast cancer patients, 38 healthy controls) were cultured for at least 7 days. The formation of multinucleated giant cells (MGCs), which was checked during the whole time of culture, was observed in all cases. By the seventh day MGCs represented 25-50 % and during the second and third month more than 90% of all cells. Lymphokines and/or concanavalin A stimulation (16-34 cases respectively) of BMs was performed as well. This stimulation greatly accelerated MGC formation. There were no differences either in spontaneous or in stimulated fusion between the different groups compared.
To reveal and evaluate the mechanism of transforming rainfall into runoff in the region, where the subsurface flow plays a dominant role in the runoff formation, a continuous hydrological and climatic data monitoring has been set-up in the experimental catchment Uhlířská (the Jizera Mountains, CR). The soil profile (Dystric Cambisol), formed on the weathered granite bedrock, is shallow and highly heterogeneous. Beside a standard catchment data observation a hillslope transect was instrumented to control the flow dynamics in the soil profile. From three soil horizons, the subsurface outflow is recorded in the subsurface trench. Adjacent to the trench the soil water suction is scanned by triplets of automatic tensiometers. Within the soil profile the unsaturated regime prevails, nevertheless the soil keeps almost saturated. Nearly simultaneous reaction of suction on a rainfall in all soil horizons implies a rapid vertical flow. Local preferential flow paths are conducting infiltrating water at significantly variable rates when saturation is reached. Groundwater table, soil moisture and subsurface runoff measured at the hillslope transect and the total outflow from the catchment, are correlated. The outflow from the catchment is dominantly controlled by soil moisture however the mechanism of its generation is not yet fully understood. and V oblasti s dominantním podpovrchovým odtokem bylo započato s kontinuálním hydrologickým a klimatickým monitoringem s cílem popsat a vyhodnotit transformaci srážky na odtok. Experimentální povodí Uhlířská se nachází v severní části České republiky v Jizerských horách. Půdní profil, klasifikovaný jako dystrická kambizem na zvětralém žulovém podloží, je mělký a velmi heterogenní. Svahový transekt byl vystrojen pro sledování dynamiky podpovrchového odtoku. Ve třech půdních horizontech je monitorován odtok a půdní sací tlak. V půdním profilu převládá nenasycený stav, ačkoliv je půdní vlhkost dlouhodobě blízko nasycení. Rychlé vertikální proudění je indikováno téměř současnou odezvou půdního sacího tlaku na srážku ve všech půdních horizontech. Po dosažení nasycení infiltrující voda protéká preferenčními cestami s výrazně odlišnými lokálními rychlostmi. Závislost hladiny podzemní vody, půdní vlhkosti, podpovrchového odtoku ve svahovém transektu na odtoku z povodí je významná. Odtok vody z povodí, které leží na zvětralém žulovém podloží, je dominantně určován půdní vlhkostí. Přes tato zjištění není mechanismus tvorby odtoku zatím jednoznačně popsatelný.
Rice being the major food crop for more than half of the world population is severely affected by drought stress starting from the establishment of the seedling. We focused on the UV-B priming mediated transgenerational drought tolerance of a drought-tolerant rice variety (Vaisakh) towards polyethylene glycol (PEG) 6,000 (20%)-induced drought. Results showed that priming in F0 generation and re-priming in F1 generation with UV-B enhanced the PEG stress tolerance potential of rice seedlings with increased expression of genes encoding antioxidant enzymes and stress-related proteins offering better protection to primed plants. UV-B priming maintained oxidative homeostasis of the plant cell thus ensuring uninterrupted mitochondrial and photosynthetic activities. Cumulatively, our results suggest that the transgenerational priming memory retained in the seeds is transferred to offspring without any loss. Moreover, re-priming in F1 generation further boosted the innate tolerance potential of a tolerant variety resulting in stable cellular redox homeostasis.
