We studied the diet of 10 species of grasshopper belonging to the family Pamphagidae over a period of 3 years at 6 localities in North Eastern Algeria. The species of plants consumed by the grasshoppers was determined by comparing slide mounted specimens of the pieces of plant epidermis in their faeces with those in a reference collection of identified plants collected from the same localities. The percentages of occurrence of the different species of plants in the faeces of the grasshoppers were not related to the abundance of the plants at the sites studied. All the grasshoppers were polyphagous but differed in the percentage of Poaceae in their diets. The diet of Tmethis and Ocneridia contained a higher percentage of Poaceae than the other species and are considered to be ambivores. The three species in the Pamphagus gr. djelfensis complex differ in their diets but all tend to avoid consuming Poaceae and are categorized as forbivores. We also compared the frequency of occurrence of Fabaceae in the faeces and in the field and O. volxemii is the only species that avoided consuming this plant family. The number of sensilla on the labrum was also studied in both sexes of each species. Once one corrects for differences in the size of the labrum, the forbivores have higher numbers of sensilla in groups A1, A2 and A3 (but not A10) than the ambivores. The present paper reports some cytogenetic peculiarities observed in the Ag-NORs of Pamphagus ortolaniae chromosomes, the unusual behaviour of ribosomal sites after silver staining and the intense Ag-positive reaction of centromeric regions at spermatogonial metaphase and spermatocyte metaphase I and II. Moreover, a conclusive identification and localization of all the ribosomal clusters is provided by using heterologous rDNA FISH on spermatocyte chromosomes. 18S-28S rDNA mapped on a single chromosome pair and resulted multiclustered along the chromosomal body in three distinct serial regions, r1, r2 and r3. Surprisingly, these areas were scarcely (r1) or never (r2 and r3) detectable by silver impregnation. As in other Orthoptera and many groups of arthropods, FISH with the pentamer (TTAGG)n as the probe labelled the telomeres of all chromosomes.
The corpora cardiaca (CC) of the two grasshopper species Zonocerus elegans (Pyrgomorphidae) and Lamarckiana sparrmani (Pamphagidae) contain (a) substance(s) that cause(s) hyperlipaemia in the migratory locust and hypertrehalosaemia in the American cockroach. Isolation of neuropeptides belonging to the adipokinetic hormone (AKH) family was achieved by single-step reversed-phase high performance liquid chromatography of CC extracts from both species and monitoring tryptophan fluorescence. The material of both species showed three distinct fluorescence peaks with adipokinetic activity in the migratory locust. The peptides were identifid by at least two of the following methods: (1) sequencing by Edman degradation, (2) sequencing by tandem fast atom bombardment mass spectrometry, (3) mass determination by matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry and (4) co-elution of the native and synthetic peptides. Both species were found to have three AKH peptides stored in the CC, but unlike in other grasshoppers, none of those peptides were decapeptides. In Z. elegans the following three octapeptides occur: Schgr-AKH-II (pELNFSTGWamide), Peram-CAH-II (pELTFTPNWamide) and Phymo-AKH-III (pEINFTPWWamide), whereas L. sparrmani contains the octapeptides Grybi-AKH (pEVNFSTGWamide), Pyrap-AKH (pELNFTPNWamide) and also Phymo-AKH-III. Conspecific bioassays show no adipokinetic and only a weak (not significant) hypertrehalosaemic effect (in the pamphagid grasshopper). Some explanations are offered on the possible role of these peptides in the species investigated by interpreting their life style.
