The aim of this study was to investigate the effects of silicon in alleviating cadmium stress in maize plants grown in a nutrient solution and to evaluate the potential of the spectral emission parameters and the ratio of red fluorescence (Fr) to far-red fluorescence (Ffr) in assessing the beneficial effects of Si. An experiment was carried out using a nutrient solution with a toxic dose of Cd and six doses of Si; biomass, Cd, Si, and photosynthetic pigments of the plants were measured. Chlorophyll (Chl) a fluorescence analysis demonstrated that Si alleviated Cd toxicity in plants. Chl fluorescence measurements were sensitive in detecting such effects even when significant changes in biomass production and concentrations of photosynthetic pigments were not observed. Under optimal conditions, most of the light energy is used to drive electron transport. However, when the light energy exceeds the capacity of photosynthesis, the overall photosynthetic efficiency drops down. The present study investigated the effects of high light on rice photooxidation-prone mutant 812HS, characterized by a mutation of leaf photooxidation 1 gene, and its wild type 812S under field conditions. Our results showed no significant difference between 812HS and 812S before exposure to high sunlight. However, during exposure to high light, shoot tips of 812HS turned yellow and their chlorophyll (Chl) content decreased. Transmission electron microscopy showed that photooxidation resulted in significant damage of chloroplast ultrastructure. It was confirmed also by inhibited photophosphorylation and reduced ATP content. The decreased coupling factor of ATP, Ca2+-ATPase and Mg2+-ATPase activities also verified these results. Further, significantly enhanced activities of antioxidative enzymes were observed during photooxidation. Malondialdehyde, hydrogen peroxide, and the superoxide generation rates also increased. Chl a fluorescence analysis found that the performance index and maximum quantum yield of PSII declined on August 4, 20 days after high-light treatment. Net photosynthetic rate also decreased and substomatal CO2 concentration increased in 812HS at the same time. In conclusion, our findings indicated that excessive energy triggered the production of toxic reactive oxygen species and promoted lipid peroxidation in 812HS plants, causing severe damage to cell membranes, degradation of photosynthetic pigments and proteins, and ultimately inhibition of photosynthesis., J. Our present work showed that the expression of genes encoding PTOX (terminal oxidase of chlororespiration) and PGR5 (one essential component of cyclic electron transfer) were stimulated by red and blue light, but the stimulation under red light was soon reversed by subsequent far-red light. The expression levels of PTOX and PGR5 under simulated light quality conditions in line with maize-soybean relay strip intercropping (SRI) were obviously lower than those under simulated soybean monocropping (SM), since the lower red:far-red ratio under SRI. Measurements on photosynthetic and chlorophyll fluorescence parameters suggested a decline of assimilatory power supply and a lower nonphotochemical quenching under SRI as compared to SM. In this case, weaker PGR-dependent cyclic electron transfer and chlororespiration under SRI, suggested by lower expression levels of PGR5 and PTOX, could be considered as means of reducing excitation energy dissipation to allocate more power toward CO2 assimilation., X. New spectral absorption photometry methods are introduced to estimate chlorophyll (Chl) content of corn leaves by smart phones. The first method acquires light passing through a leaf by smartphone camera, compensating for differences in illumination conditions. In order to improve performance of the method, spectral absorption photometry (SAP) with background illumination has been considered as well. Data were acquired by smartphone camera in Iowa State University maize fields. Various indices were extracted and their correlation with Chl content were examined by Minolta SPAD-502. Hue index in SAP reached R2 value of 0.59. However, with light-aided SAP (LASAP), R2 of 0.97 was obtained. Among traits, the vegetation index gave the most accurate indication. We can conclude that the high performance of LASAP method for estimating Chl content, leads to new opportunities offered by smart phones at much lower cost. ‘Hass‘ and ‘Fuerte‘ avocado plants were grown under well-watered or waterlogged conditions. Results indicated significant effects on the majority of the allometric parameters in waterlogged plants, with ‘Fuerte‘ displaying a more pronounced growth inhibition. Waterlogged conditions caused a progressive and simultaneous decline in net photosynthetic rate and stomatal conductance, earlier in ‘Fuerte‘ than in ‘Hass‘. Maximal potential quantum yield of PSII was unaffected by the soil water regime and/or variety and leaf water potential values in waterlogged plants were not more negative compared with control plants. ‘Fuerte‘ waterlogged plants exhibited increased contents of thiobarbituric acid reactive substances, whereas oxidative injury was not detected in ‘Hass‘. Leaf senescence can be induced by numerous factors. In order to explore the relationship between root respiration and leaf senescence, we utilized different types of phloem girdling to control the root respiration of Alhagi sparsifolia and its physiological response. Our results showed that both girdling and inhibition of root respiration led to a decline of stomatal conductance, photosynthesis, transpiration rate, chlorophyll (Chl) a, Chl b, carotenoid (Car) content, Chl a/b, Chl/Car, water potential, and Chl a fluorescence, as well as to an increase of abscisic acid (ABA), proline, and malondialdehyde content in leaves and to upregulation of senescence-associated gene expression. Our present work implied that both inhibition of root respiration and girdling can induce leaf senescence. In comparison with phloem girdling, the leaf senescence caused by inhibition of root respiration was less significant. The reason for girdling-induced senescence was ABA and carbohydrate accumulation. Tropical rainforest trees adjust leaf traits during ontogeny to cope with changes in the physical environment and maximize their carbon uptake. The aim of this study was to determine the plasticity index (PI) of leaf traits in understory and canopy leaves of six Amazonian tree species. In four of the six species the PI of leaf traits varied within species, and in four of the ten leaf traits assessed, the PI differed between species. The greatest PI values were found for stomatal density (Ds) and CO2-saturated photosynthesis, and the lowest ones were found for stomatal size, and leaf thickness. Despite the differences in PI values within species, the mean PI was similar in all the six species. Na práci Julia Sachse navázali v českém prostředí již v 19. století svými pokusy František Farský a Julius Stoklasa. Velký význam pro uchování paměti na Julia Sachse měl také profesor botaniky na české univerzitě v Praze Ladislav Josef Čelakovský, který stejně jako Sachs žil v domácnosti Jana E. Purkyně. Nejvýznamnějším pokračovatelem v díle J. Sachse a zároveň osobností, která udržovala povědomí o této osobnosti v českém prostředí i ve 20. století, byl Bohumil Němec. Ten se inspiroval Sachsovými výzkumy např. i při svém objevu rostlinných statolitů. Zdůrazňoval také Sachsovy zásluhy o prosazení experimentální metodologie a vysoce ocenil stylistiku jeho prací. S J. Sachsem ho spojoval rovněž zájem o dějiny oboru. S rozpaky ovšem vnímal okolnosti soužití J. Sachse s J. E. Purkyněm v Praze, když Sachsovo jednání vůči českému přírodovědci, který ho učinil prakticky členem své rodiny, interpretoval jako nevděčnost a následek problematických stránek Sachsova charakteru., Several distinguished scientists of the 19th century built on the experimental work of J. Sachs, including František Farský, Julius Stoklasa, and Ladislav Josef Čelakovský. His most prominent successor was Bohumil Němec who emphasised the importance of Sachs’s experimental approach and admired his art of writing. Němec also shared his interest in the history of the field but was disappointed by Sachs’s attitude to his former friend Jan Evangelista Purkyně., and Martin Franc.
Calmodulin (CaM) is a highly conserved calcium sensor protein associated with chilling tolerance in living organisms. It has four EF-hand domains for binding of four Ca2+, two of them located in the N-terminus, and the other two in the C-terminus. A notothenioid CaM gene fragment (CaMm), which only codes for N-terminus of CaM (with two EF-hand domains), was introduced into Nicotiana benthamiana. Effects of its overexpression on chilling tolerance in plants were explored. During 4◦C or 0◦C chilling treatment, both CaMm and CaM transgenic plants showed higher PSII maximum quantum yield, actual quantum yield, and soluble protein content, lower electrolyte leakage and malondialdehyde content than that of the control. The changes in these physiological indices were comparable between the CaMm and CaM transgenic plants during the treatments. Available water stored in deep soil layers could increase the photosynthetic capacity of cotton. It was hypothesized that the photosynthesis of cotton would be enhanced by changing the fertilizer application depth under different deep-layer water conditions. We examined two deep-layer water levels, i.e.,
well-watered (W80) and not watered (W0), combined with surface application (F10) and deep application (F30) of basal fertilizer. Compared to W0, W80 resulted in increased leaf area (LA), photosynthetic pigment contents, maximal PSII efficiency (Fv/Fm), effective quantum yield of PSII (YII) and PSI (YI), electron transport rate of PSII (ETRII) and PSI (ETRI). W80 also increased the aboveground and root dry mass by 39 and 0.6%, respectively, and decreased the root/shoot ratio by 40-73%. Under the W0 condition, higher values of Fv/Fm, YII, YI, ETRII, and ETRI were measured for F10 compared to F30 after 69 d from emergence. Under the W80 condition, cotton plants with F10 showed higher LA, Fv/Fm, YII, YI, ETRII, and ETRI, but there were no significant differences in the photosynthetic pigments compared to F30. Our results suggest that sufficient water in deeper soil layers and the surface application of basal fertilizer could increase photosynthetic activity and efficiency, which promoted aboveground dry mass accumulation and partitioning towards reproductive organs., Z.
