Patch clamp method developed more than 30 years ago is widely used for investigation of cellular excitability manifested as transmembrane ionic current and/or generation of action potentials. This technique could be applied to measurement of ionic currents flowing through individual (single) ion channels or through the whole assembly of ion channels expressed in the whole cell. Whole cell configuration is more common for measurement of ion currents and the only one enabling measurement of action potentials. This method allows detailed analysis of mechanisms and structural determinants of voltagedependent gating of ion channels as well as regulation of channel activity by intracellular signaling pathways and pharmacological agents., M. The long QT syndrome (LQTS) is a monogenic disorder characterized by prolongation of the QT interval on electrocardiogram and syncope or sudden death caused by polymorphic ventricular tachycardia (torsades de pointes). In general, mutations in cardiac ion channel genes (KCNQ1, KCNH2, SCN5A, KCNE1, KCNE2) have been identified as a cause for LQTS. About 50-60 % of LQTS patients have an identifiable LQTS causing mutation in one of mentioned genes. In a group of 12 LQTS patients with no identified mutations in these genes we have tested a hypothesis that other candidate genes could be involved in LQTS pathophysiology. SCN1B and KCND3 genes encode ion channel proteins, ANK2 gene encodes cytoskeletal protein interacting with ion channels. To screen coding regions of genes SCN1B, KCND3, and 10 exons of ANK2 following methods were used: PCR, SSCP, and DNA sequencing. Five polymorphisms were found in screened candid ate genes, 2 polymorphisms in KCND3 and 3 in SCN1B. None of found polymorphisms has coding effect nor is located close to splice sites or has any similarity to known splicing enhancer motifs. Polymorphism G246T in SCN1B is a novel one. Moravské zemské muzeum v Brně bylo vždy nositelem Mendelova badatelského odkazu, a to prostřednictvím bývalé vědecké akademie pro Moravu a Slezsko, tzv. Hospodářské společnosti (Ackerbaugesellschaft) a jejího Přírodozpytného spolku (Naturforschender Verein). Jako druhou největší a nejstarší muzejní instituci jej založila Moravskoslezská hospodářská společnost, jejímž členem se roku 1854 stal i Gregor Johann Mendel (1822-1884). Zapojil se nejen do organizační práce ve většině sekcí společnosti, ale také do výzkumu. Disorders of ATP synthase, the key enzyme of mitochondrial energy provision belong to the most severe metabolic diseases presenting as early- onset mitochondrial encephalo- cardiomyopathies. Up to now, mutations in four nuclear genes were associated with isolated deficiency of ATP synthase. Two of them, ATP5A1 and ATP5E encode enzyme’s st ructural subunits α and ε , respectively, while the other two ATPAF2 and TMEM70 encode specific ancillary factors that facilitate the biogenesis of ATP synthase. All these defects share a similar biochemical phenotype with pronounced decrease in the content of fully assembled and functional ATP synthase complex. However, substantial differences can be found in their frequency, molecular mechanism of pathogenesis, clinical manifestation as well as the course of the disease progression. While for TMEM70 the number of reported patients as well as spectrum of the mutations is steadily increasing, mutations in ATP5A1, ATP5E and ATPAF2 genes are very rare. Apparently, TMEM70 gene is highly prone to mutagenesis and this type of a rare mitochondrial disease has a rather frequent incidence. Molekulární genetika vstoupila na antropologické kolbiště koncem 60. let, ale teprve přímá analýza archaické DNA (aDNA) z fosilních pozůstatků od 80. let umožnila přesnější vhled do evoluce našeho druhu ve středním a mladém pleistocénu. Navzdory různým „Jurským parkům“ totiž DNA po smrti organismu rychle degraduje a časové okno její možné analýzy je poměrně omezené. Navíc jsou vzorky kontaminovány DNA okolních organismů. Nicméně velice záhy se pozornost paleogenetiků soustředila na naše příbuzné - neandertálce. Analýza jejich genomu ukázala, že ~2 % jejich DNA se vyskytuje v genomu anatomicky moderního člověka s výjimkou subsaharské Afriky a celkový rozsah tohoto přenosu může dosahovat až 20 %. Křížení s neandertálci lidem zřejmě umožnilo snadnější adaptaci na chladnější podmínky eurasijského kontinentu, současně však přineslo i výskyt některých chorob. Překvapení přinesla sekvence aDNA izolovaná z článku prstu nalezeného v Denisově jeskyni na Altaji. Ukázalo se, že tento jedinec patřil k neznámému druhu odlišnému jak od moderních lidí, tak i od neandertálců. I tito hominini přispěli až 6 % svojí DNA do genomu některých současných populací člověka (JV Asie, Oceánie). Podle nejnovějších poznatků byl tok genů mezi homininy středního a mladého paleolitu poměrně složitý, byla např. detekována příměs neandertálské DNA v genomu denisovců, kteří navíc získali další sekvence od dalšího, blíže neurčeného druhu hominina. Posledním příspěvkem paleogenetiky do obrazu naší evoluce je sekvence mitochondriální DNA získaná ze zhruba 400 tisíc let starých fosilních pozůstatků heidelberského člověka (Homo heidelbergensis) ze Sima de los Huesos (Šachty kostí) z krasové oblasti Atapuerca ve Španělsku, která ukazuje na příbuznost tohoto druhu., Molecular genetics entered the arena of anthropology at the end of the 1960s, but only direct analysis of ancient DNA (aDNA) from fossils since the 80s has permitted a better insight into the evolution of our own species. Despite the rapid decomposition of DNA starting immediately after death, molecular geneticists are now able to retrieve and sequence aDNA tens or even hundreds of thousands years old. Paleogenetic studies of ancient humans and their relatives have revealed a rather complex picture of Middle and Upper Pleistocene hominins (Neanderthals, Denisovans, ante-Neanderthals etc.) and gene flow among them. The objective of the current study was to search for genetic determinants associated with antihypertensive effects of angiotensin-converting enzyme (ACE) inhibitor captopril. Linkage and correlation analyses of captopril-induced effects on blood pressure (BP) with renal transc riptome were performed in the BXH/HXB recombinant inbred (RI) strains derived from spontaneously hypertensive rat (SHR) and Brown Norway (BN-Lx) progenitors. Variability of blood pressure lowering effects of captopril among RI strains was continuous suggesting a polygenic mode of inheritance. Linkage analysis of captopril- induced BP effects revealed a significant quantitative trait locus (QTL) on chromosome 15. This QTL colocalized with cis regulated expression QTL (eQTL) for the Ednrb (endothelin receptor type B) gene in the kidney (SHR allele was associated with increased renal expression) and renal expression of Ednrb correlated with captopril-induced BP effects.
