When cells get metals in small excess, mechanisms of avoidance occur, such as exclusion, sequestration, or compartmentation. When the excess reaches sub-lethal concentrations, the oxidative stress, that toxic metals trigger, leads to persistent active oxygen species. Biomolecules are then destroyed and metabolism is highly disturbed. At the chloroplast level, changes in pigment content and lipid peroxidation are observed. The disorganized thylakoids impair the photosynthetic efficiency. The Calvin cycle is also less efficient and the photosynthetic organism grows slowly. When an essential metal is given together with a harmful one, the damages are less severe than with the toxic element alone. Combined metals and phytochelatins may act against metal toxicity. and M. Selected light wavebands promote plant development and/or the biosynthesis of targeted metabolites. This work offers new insights on the effects of red (R), green (G), blue (B), and white (W - R:G:B; 1:1:1) LED light supplementation on physiochemical traits of strawberry leaves. Gas exchange and chlorophyll fluorescence parameters, photosynthetic pigments, and superoxide anion (*O2-) content were analysed in plants grown for 1 (T1) and 17 (T17) d with light supplementations. At T1, light supplementations resulted in the enhancement of the de-epoxidation state of xanthophylls and nonphotochemical quenching, but no changes were observed in maximal photosynthetic rate (PNmax), irrespective of light spectra. At T17, xanthophyll contents remained higher only in R-supplemented plants. Overall, W light resulted in higher photosynthesis, whilst R and B light depressed PNmax values and promoted *O2- formation at T17. G light did not induce variations in photosynthetic traits nor induced oxidative stress at both T1 and T17.
Elevated plasma concentration of apolipoprotein B-48 (apoB-48) is an independent risk factor of cardiovascular disease. Stearoyl-CoA desaturase-1 (SCD1) is a rate-limiting lipogenic enzyme and a key regulator of fuel metabolism. The aim of this study was to analyse associations between clinical, biochemical, and genetic factors and different apoB-48 levels in subjects at increased cardiometabolic risk. We examined 220 subjects exhibiting at least one metabolic syndrome (MetS) component. In conjunction with basic clinical, anthropometric and laboratory measurements, we analysed various polymorphisms of stearoyl-CoA desaturase-1 (SCD1). Subjects were divided into two groups according to the median apoB-48 level: (1) high apoB-48 (≥ 7.9 mg/l, N = 112) and (2) low apoB-48 (< 7.9 mg/l, N = 108). Neither group differed significantly in anthropometric measures. High plasma apoB-48 levels were associated with increased systolic blood pressure (+3 %; P < 0.05), MetS prevalence (59.8 vs. 32.4 %; P < 0.001), small-dense LDL frequency (46.4 vs. 20.4 %; P < 0.001), triglycerides (+97 %; P < 0.001), non-HDL-cholesterol (+27 %; P < 0.001), and lower concentrations of HDL-cholesterol (-11 %; P < 0.01). This group was further characterized by a higher HOMA-IR index (+54 %; P < 0.001) and increased concentrations of conjugated dienes (+11 %; P < 0.001) and oxidatively modified LDL (+ 38 %; P < 0.05). Lower frequencies of SCD1 minor genotypes (rs2167444, rs508384, P < 0.05) were observed in subjects with elevated plasma concentrations of apoB-48. Elevated plasma concentrations of apoB-48 are associated with an adverse lipid profile, higher systolic blood pressure, insulin resistance, and oxidative stress. Excessive production of oxygen free radicals has been regarded as a causative common denominator of many pathological processes in the animal kingdom. Hydroxyl and nitrosyl radicals represent the major cause of the destruction of biomolecules either by a direct reaction or by triggering a chain reaction of free radicals. Scavenging of free radicals may act preventively or therapeutically. A number of substances that preferentially react with free radicals can serve as scavengers, thus increasing the internal capacity/activity of endogenous antioxidants and protecting cells and tissues against oxidative damage. Molecular hydrogen (H2) reacts with strong oxidants, such as hydroxyl and nitrosyl radicals, in the cells, that enables utilization of its potential for preventive and therapeutic applications. H2 rapidly diffuses into tissues and cells without affecting metabolic redox reactions and signaling reactive species. H2 reduces oxidative stress also by regulating gene expression, and functions as an anti-inflammatory and anti-apoptotic agent. There is a growing body of evidence based on the results of animal experiments and clinical observations that H2 may represent an effective antioxidant for the prevention of oxidative stress-related diseases. Acute lung injury (ALI) caused by lipopolysaccharide (LPS) is a common, severe clinical syndrome. Injury caused by inflammation and oxidative stress in vascular endothelial and alveolar epithelial cells is a vital process in the pathogenesis of ALI. Toll-like receptor 9 (TLR9) is highly expressed in LPS-induced ALI rats. In this study, Beas-2B human pulmonary epithelial cells and A549 alveolar epithelial cells were stimulated by LPS, resulting in the upregulation of TLR9 in a concentrationdependent manner. Furthermore, TLR9 overexpression and interference vectors were transfected before LPS administration to explore the role of TLR9 in LPS-induced ALI in vitro. The findings revealed that inhibition of TLR9 reduced inflammation and oxidative stress while suppressing apoptosis of LPS-induced Beas-2B and A549 cells, whereas TLR9 overexpression aggravated these conditions. Moreover, TLR9 inhibition resulted in downregulated protein expression of myeloid differentiation protein 88 (MyD88) and activator activator protein 1 (AP-1), as well as phosphorylation of nuclear factor-κB (NF-κB), c-Jun N-terminal kinase (JNK), and p38 mitogen-activated protein kinase (MAPK). The phosphorylation of extracellular-regulated protein kinases 1/2 was upregulated compared to that of cells subjected to only LPS administration, and this was reversed by TLR9 overexpression. These results indicate that inhibition of TLR9 plays a protective role against LPS-induced inflammation and oxidative stress in Beas-2B and A549 cells, possibly via the MyD88/NF-κB and MyD88/MAPKs/AP-1 pathways.
Chronic kidney disease (CKD) represents a serious public health problem with increasing prevalence and novel approaches to renal protection are continuously under investigation. The aim of this study was to compare the effect of melatonin and angiotensin II type 2 receptor agonist compound 21 (C21) to angiotensin converting enzyme inhibitor captopril and angiotensin II type 1 receptor blocker olmesartan on animal model of doxorubicin nephrotoxicity. Six groups of 3-month-old male Wistar rats (12 per group) were treated for four weeks. The first group served as a control. The remaining groups were injected with a single dose of doxorubicin (5 mg/kg i.v.) at the same day as administration of either vehicle or captopril (100 mg/kg/day) or olmesartan (10 mg/kg/day) or melatonin (10 mg/kg/day) or C21 (0.3 mg/kg/day) was initiated. After four week treatment, the blood pressure and the level of oxidative stress were enhanced along with reduced glomerular density and increased glomerular size. Captopril, olmesartan and melatonin prevented the doxorubicin-induced increase in systolic blood pressure. All four substances significantly diminished the level of oxidative burden and prevented the reduction of glomerular density and modestly prevented the increase of glomerular size. Pulmonary hypertension resulting from chronic hypoxia is at least partly caused by the increased production of reactive oxygen species (ROS). The goal of the presented study was to investigate the dynamics and the site of production of ROS during chronic hypoxia. In our study Wistar rats were kept for 1, 4 and 21 days in an isobaric hypoxic chamber (FiO2=0.1), while controls stayed in normoxia. We compared NO production in expired air, plasma and perfusate drained from isolated rat lungs and measured superoxide concentration in the perfusate. We also detected the presence of superoxide products (hydrogen peroxide and peroxynitrite) and the level of ROS-induced damage expressed as the concentration of lipid peroxydation end products. We found that the production and release of ROS and NO during early phase of chronic hypoxia has specific timing and differs in various compartments, suggesting the crucial role of ROS interaction for development of hypoxic pulmonary hypertension., D. Damage of alveolar-capillary barrier, inflammation, oxidative
injury, and lung cell apoptosis represent the key features of acute
lung injury (ALI). This study evaluated if selective
phosphodiesterase (PDE)-4 inhibitor roflumilast can reduce the
mentioned changes in lavage-induced model of ALI. Rabbits with
ALI were divided into 2 groups: ALI without therapy (A group)
and ALI treated with roflumilast i.v. (1 mg/kg; A+R group). One
group of healthy animals without ALI served as ventilated
controls (C group). All animals were oxygen-ventilated for further
4 h. At the end of experiment, total and differential counts of
cells in bronchoalveolar lavage fluid (BALF) and total and
differential counts of white blood cells were estimated. Lung
edema formation was assessed from determination of protein
content in BALF. Pro-inflammatory cytokines (TNFα, IL-6 and
IL-8) and markers of oxidation (3-nitrotyrosine, thiobarbituricacid reactive substances) were detected in the lung tissue and
plasma. Apoptosis of lung cells was investigated
immunohistochemically. Treatment with roflumilast reduced leak
of cells, particularly of neutrophils, into the lung, decreased
concentrations of cytokines and oxidative products in the lung
and plasma, and reduced lung cell apoptosis and edema
formation. Concluding, PDE4 inhibitor roflumilast showed potent
anti-inflammatory actions in this model of ALI.
Increased oxidative stress in the brain during the course of Alzheimer’s disease (AD) leads to an imbalance of antioxidants and formation of free radical reaction end-products which may be detected in blood as fluorescent lipofuscin-like pigments (LFPs). The aim of this study was to evaluate and compare LFPs with plasma selenium concentrations representing an integral part of the antioxidant system. Plasma samples from subjects with AD dementia (ADD; n=11), mild cognitive impairment (MCI; n=17) and controls (n=12), were collected. The concentration of selenium was measured using atomic absorption spectroscopy. LFPs were analyzed by fluorescence spectroscopy and quantified for different fluorescent maxima and then correlated with plasma selenium. Lower levels of selenium were detected in MCI and ADD patients than in controls (P=0.003 and P=0.049, respectively). Additionally, higher fluorescence intensities of LFPs were observed in MCI patients than in controls in four fluorescence maxima and higher fluorescence intensities were also observed in MCI patients than in ADD patients in three fluorescence maxima, respectively. A negative correlation
between selenium concentrations and LFPs fluorescence was observed in the three fluorescence maxima. This is the first study focused on correlation of plasma selenium with specific lipofuscin-like products of oxidative stress in plasma of patients with Alzheimer´s disease and mild cognitive impairment.
n this study we addressed a question of whether experimental manipulations that increase life span also reduce physical activity and molecular oxidative damage. We used three phenotypes of male and female Pyrrhocoris apterus that survive for different lengths of time, diapausing insects, reproductive insects and insects from which the corpus allatum, the source of juvenile hormone, was surgically removed. Protein carbonyl content of the thoracic muscles was used as an index of molecular oxidative modification. Diapause or ablation of the corpus allatum (allatectomy) was associated with an extended life span of both sexes, but only those individuals that were in diapause were less active. The carbonyl content, both relative (per protein unit) and absolute (per thorax) increased with age in reproductive insects of both sexes. However, the associations between the carbonyl content and diapause and allatectomy differed in males and females. In males, the carbonyl content was not associated with either diapause or allatectomy. There was no age-related increase in the relative and absolute carbonyl levels in diapausing females, while only the increase in the relative carbonyl level was absent in allectomized females. Overall, the results indicate that both allatectomy and diapause prolonged life span, but had different and sex-specific effects on locomotor activity and carbonyl content.
