A new genus and species, Avitocaligus assurgericola gen. et sp. n., of the family Caligidae is established based on an ovigerous female collected from a razorback scabbardfish, Assurger anzac (Alexander), caught off New Caledonia. This is the first record of any parasitic copepod from this fish. The new genus is placed in the Caligidae since it possesses the caligid cephalothorax incorporating the first to third pedigerous somites. It also exhibits biramous first and fourth swimming legs but lacks dorsal plates on the fourth pedigerous somite. This combination of characters serves to differentiate the new genus from existing genera. In addition, it possesses loosely coiled, uniseriate egg sacs, concealed between the middle lamellar plates on the genital complex and the ventral plates on the abdomen. Preliminary phylogenetic analysis with a restricted matrix suggests that the new genus represents the earliest offshoot from the main caligid lineage since it does not exhibit the dominant exopod on the fourth swimming leg found in all other members of the Caligidae, including the genus Euryphorus. It strongly supports the newly recognized monophyletic status of the Caligidae, incorporating the genera formerly placed in the Euryphoridae.
The sicklefin redhorse, Moxostoma sp. (Cypriniformes: Catostomidae), is an innominate imperiled catostomid endemic to the Hiwassee and Little Tennessee river basins, which has been restricted to a few tributaries of these systems by impoundments. During collections to propagate sicklefin redhorse for reintroduction, a myxozoan, described herein, was observed infecting sicklefin redhorse in the Little Tennessee River Basin, North Carolina. Myxobolus naylori Ksepka et Bullard sp. n. infects the stratum spongiosum covering the scales of sicklefin redhorse. Myxospores of the new species differ from all congeners by the combination of having a mucous envelope, intercapsular process, and sutural markings as well as lacking an iodinophilic vacuole in the sporoplasm. and A phylogenetic analysis of the 18S rDNA gene recovered the new species in a polytomy with Myxobolus marumotoi Li et Sato, 2014 and a clade comprised of species of Myxobolus Bütschli, 1882; Thelohanellus Kudo, 1933, and Dicauda Hoffman et Walker, 1973. Histological sections of infected sicklefin redhorse skin revealed myxospores within a plasmodium in the stratum spongiosum dorsal to scales, encapsulated in collagen fibres, and associated with focal erosion of scales directly beneath the plasmodium; in some instances, the scale was perforated by the plasmodium. The specificity of the new species to sicklefin redhorse may make it a useful biological tag to differentiate sicklefin redhorse from morphologically similar species. The new species is the first parasite reported from sicklefin redhorse, a species of concern to the United States Fish and Wildlife Service. No species of Myxobolus has been reported from species of Moxostoma in the Southeast United States. As it was observed that Myxobolus minutus Rosser, Griffin, Quiniou, Alberson, Woodyard, Mischker, Greenway, Wise et Pote, 2016 is a primary junior homonym of Myxobolus minutus Nemeczek, 1911, we propose the replacement name Myxobolus diminutus (Rosser, Griffin, Quiniou, Alberson, Woodyard, Mischker, Greenway, Wise et Pote, 2016).
Monozoic cestodes of the recently amended genus Promonobothrium Mackiewicz, 1968 (Cestoda: Caryophyllidea), parasites of suckers (Cypriniformes: Catostomidae) in North America, are reviewed, with information on their host specificity, distribution and data on the scolex morphology of seven species studied for the first time using scanning electron microscopy (SEM). Evaluation of type and voucher specimens from museum collections and newly collected material of most species indicated the following valid nominal species: Promonobothrium minytremi Mackiewicz, 1968 (type species); P. ingens (Hunter, 1927); P. hunteri (Mackiewicz, 1963); P. ulmeri (Calentine et Mackiewicz, 1966); P. fossae (Williams, 1974) and P. mackiewiczi (Williams, 1974). Rogersus Williams, 1980 with its only species R. rogersi is transferred to Promonobothrium based on morphological and molecular data. Promonobothrium currani sp. n. and P. papiliovarium sp. n. are described from Ictiobus bubalus (Rafinesque) and Ictiobus niger (Rafinesque), and Erimyzon oblongus (Mitchill), respectively. The newly described species can be distinguished from the other congeners by the morphology of the scolex, the position of the anteriormost vitelline follicles and testes, the presence of postovarian vitelline follicles and the shape of the ovary. Molecular phylogenetic analyses of six species based on sequences of the small and large subunits of the nuclear ribosomal RNA genes (ssrDNA, lsrDNA) confirmed the monophyletic status of the genus and supported the validity of the species analysed. Two new microleafhopper genera of Empoascini within the subfamily Typhlocybinae (Hemiptera: Cicadellidae), Condensella Xu, Dietrich & Qin gen. n., based on the type species C. filamenta Xu, Dietrich & Qin sp. n., and Endogena Xu, Dietrich & Qin gen. n., based on the type species E. flava Xu, Dietrich & Qin sp. n., are described from southern China and Thailand. Male habitus photos and illustrations of male genitalia of the two new species are provided. Comparative notes on related genera are provided. A morphological type of Sarcocystis cysts found in one of two examined great black-backed gull, Larus marinus (Linnaeus) (Laridae), is considered to represent a new species for which the name Sarcocystis lari sp. n. is proposed and its description is provided. The cysts are ribbon-shaped, very long (the largest fragment found was 6 mm long) and relatively narrow (up to 75 μm). Under a light microscope the cyst wall reaches up to 1 μm and seems to be smooth. Using a computerized image analysis system, knolls, which resemble protrusions on the wall surface, are visible. Lancet-shaped cystozoites measure in average 6.9 × 1.4 μm (range 6.3-7.9 μm × 1.2-1.5 μm) in length. Observed using Transmission electron microscopy (TEM), the cyst wall is wavy and measures up to 1.2 μm in thickness. The parasitophorous vacuolar membrane has regularly arranged small invaginations. Cyst content is divided into large chambers by septa. Sarcocystis lari sp. n. has type-1 tissue cyst wall and is morphologically indistinguishable from other bird Sarcocystis species characterized by the same type of the wall. On the basis of 18S rRNA gene, 28S rRNA gene and ITS-1 region sequences, S. lari is a genetically distinct species, being most closely related to avian Sarcocystis species whose definitive hosts are predatory birds.
The scydmaenine tribe Eutheiini is recorded from Australia for the first time. Paraneseuthia carltoni sp. n. and P. booloumba sp. n. are described and illustrated, both from Queensland. In a parsimony-based phylogenetic analysis using adult morphological characters including genital features, the Australian species together with the Melanesian type species of Paraneseuthia Franz, P. peckorum Franz, were found to be more closely related to East Palearctic congeners than to most of the Paraneseuthia in the Sunda-Papuan area. The topology of the tree and biogeographic data suggest a Sundaland origin of this genus, with three major dispersal routes from a center located in present-day Sumatra: (i) north-eastern colonization of the Palearctic Far East, via a continental or island-arc route; (ii) south-eastern dispersal to East Australia; and (iii) eastern dispersal to Melanesia, possibly via the Quaternary Outer-Melanesian Arc. The important role of dispersal in the evolution of Paraneseuthia is supported by the presence of this genus on isolated volcanic islands, such as the southern Moluccas and Fiji, which were never connected to larger land masses. and Paweł Jałoszyński.
Concordant differences in morphology, phenology and RAMS markers, as well as in sequenced mtDNA (COI, COII, cytb) and nuclear DNA (ITS2) fragments, indicate that Dolerus asper Zaddach, 1859 and Dolerus brevicornis Zaddach, 1859 are valid species. On the basis of morphology, molecular markers, and distributional records, both species are distinct from Dolerus gibbosus Hartig, 1837 (= Dolerus planatus Hartig, 1837). Taxonomy of the species is clarified and the neotypes of Dolerus asper Zaddach, 1859 and Dolerus brevicornis Zaddach, 1859 are designated. The synonymies of Dolerus asper Zaddach, 1859, to Dolerus planatus Hartig, 1837 and Dolerus derzavini Malaise, 1931, spec. rev. to D. asper Zaddach, 1859 are abandoned. Dolerus carbonarius Zaddach, 1859 and Dolerus fumosus Zaddach, 1859 are considered to be species inquirendae. Phylogenetic analyses of the ITS2 fragment and fragments of ITS2 + COI and ITS2 + COII yielded the topology [D. asper, (D. brevicornis, D. gibbosus)], while those of all other markers and their combinations resulted in the topology [D. brevicornis, (D. asper, D. gibbosus)]. In the latter hypothesis the clade asper + gibbosus is also supported by structural synapomorphies.
Two differently coloured strains of the genus Chroococcus were isolated from a cyanobacterial assemblage collected from the stony littoral of a backwater of the Danube River in southern Slovakia. When grown after isolation, both subcultures were similar morphologically and their growth parameters did not differ substantially, but their pigment content (PC: PE and carotenoid ratios), details in their morphology during their life cycles and slime production were different. Identical and different characters of both morphotypes remained stable during cultivation on both agarized and liquid media, even when the cultivation parameters were changed. Both of the subcultures were studied using electron microscopy and almost their complete 16S rRNA genes were sequenced, which showed that in terms of their genetic relationship there was a 96.4% sequence similarity and certain taxonomic interspecific differences between both subcultures were confirmed. The various chromatic modifications recorded in cyanobacteria and their ecological consequences are discussed. The results yielded further data on the changes that occur during the cyanobacterial differentiation processes and their genetic stabilization.
This paper is the first attempt to resolve relationships among the Ceratocanthinae: Ceratocanthini pill scarab beetles using DNA sequences. It is focused on the Philharmostes group of seven Afrotropical genera: Baloghianestes (3 spp.), Callophilharmostes (1 sp.), Carinophilharmostes (1 sp.), Chaetophilharmostes (1 sp.), Cryptophilharmostes (3 spp.), Petrovitzostes (1 sp.) and Philharmostes (31 spp.). A phylogenetic analysis of 46 terminals and alignment of 2,913 bp from one mitochondrial and two nuclear fragments corroborates monophyly of this group, but rejects that of Philharmostes, the largest genus. The latter is paraphyletic with respect to at least four other smaller genera and consists of at least three distantly related clades. One of them, formed by Philharmostes ballerioi sp. n. from the Tanzanian Nguru (the type locality) and Kaguru Mountains, is sister to the rest of the entire Philharmostes group. The nominal genus Philharmostes is, therefore, a waste-basket taxon for accommodating members of this group that lack the distinct characters of the smaller genera. Pending further research, the phylogenetically inadequate generic taxonomy of the Philharmostes group is not modified. Molecular clock analysis estimates separation of the mitochondrial lineages of two known populations of the new species at about 2.2 Ma, which corresponds with recurring shrinkage and expansion of African rainforest caused by climatic fluctuations during the Pleistocene. Adults of all nominal ingroup genera are illustrated along with male and female body parts of the new species. Diagnostic and/or synapomorphic morphological characters of the Philharmostes group of genera are revised. Effort was made to identify Naegleria strains isolated from organs of fish, using phylogenetic analyses of SSU rDNA and ITS sequences. Eighteen fish-isolated strains studied enlarged substantially the so far available set of Naegleria strains characterized by both molecular markers. The phylogenetic analyses of separate and concatenated SSU rDNA and ITS sequences revealed phylogenetic relationships of strains under study; however, they failed to solve classification of fish-isolated strains into species. The sequence similarity of strain-representatives of Naegleria species as well as data obtained on intragenomic variation of ITS sequences discouraged the authors from the definition of new species. The results of the present study provide evidence of a need to re-evaluate the current practice of setting boundaries between species of the genus Naegleria.
