RNA gel hybridization showed that the expression of monodehydroascorbate reductase (MDHAR) in the wild type (WT) tomato was decreased firstly and then increased under salt- and polyethylene glycol (PEG)-induced osmotic stress, and the maximum level was observed after treatment for 12 h. WT, sense transgenic and antisense transgenic tomato plants were used to analyze the antioxidative ability to cope with osmotic stresses. After salt stress, the fresh mass (FM) and height of sense transgenic lines were greater than those of antisense lines and WT plants. Under salt and PEG treatments, sense transgenic plants showed a lower level of hydrogen peroxide (H2O2) and malondialdehyde (MDA), a higher net photosynthetic rate (PN), and the maximal photochemical efficiency of PSII (Fv/Fm) compared with WT and antisense transgenic plants. Moreover, sense lines maintained higher ascorbate peroxidase (APX) activity than WT and antisense plants under salt- and PEG-induced osmotic stress. The purpose of this study was to determine the role of lipotoxicity in vascular smooth muscle (VSM). C1-BODIPY 500/510 C12 used to assess the ability of VSM A7r5 cells to transport long-chain fatty acids showed that lipid transport did not appear to limit metabolism. Thin layer chromatography revealed that storage of transported fatty acid occurred primarily as mono- and diglycerides and fatty acids but not as triglycerides. We used lipid-induced apoptosis as a measure of lipotoxicity and found that 1.5 mM palmitate (6.8:1) bound to albumin resulted in a 15-fold increase in the number of apoptotic cells compared to the control at 24 hours. This apoptosis did not seem to be due to an increase in reactive oxygen species (ROS) since VSM cells incubated in palmitate showed less ROS production than cells incubated in albumin only. Similar exposure to oleate did not significantly increase the number of apoptotic cells compared to the control. Oleate actually significantly attenuated the apoptosis induced by palmitate, suggesting that unsaturated fatty acids have a protective effect on cells undergoing palmitate-induced apoptosis. Zinc is a critical mineral nutrient that protects plant cells from salt-induced cell damage. We tested whether the application of Zn at various concentrations [0, 5, 10, or 20 mg kg-1(soil)] would protect almond (Prunus amygdalus) seedlings subjected to salt stress (0, 30, 60, or 90 mM NaCl). All concentrations of Zn, particularly the application of 10 and 20 mg kg-1, increased the net photosynthetic rate, stomatal conductance, the maximal efficiency of PSII photochemistry, and a proline content in almond seedlings grown under salt stress; 20 mg(Zn) kg-1 was the most effective concentration. The activity of superoxide dismutase showed a significant increase under salinity stress and Zn application. The catalase activity decreased in the salt-treated seedlings, but recovered after the Zn treatment.
