Spinal deformities such as scoliosis and kyphosis are incurable, and can lead to decreased physical function, pain, and reduced quality of life. Despite much effort, no clear therapies for the treatment of these conditions have been found. Therefore, the development of an animal model for spinal deformity would be extremely valuable to our understanding of vertebral diseases. In this study, we demonstrate that mice deficient in the mitochondrial enzyme isocitrate dehydrogenase 2 (IDH2) develop spinal deformities with aging. We use morphological analysis as well as radiographic and micro-CT imaging of IDH2-deficient mice to characterize these deformities. Histological analysis showed increased abnormalities in IDH2-deficient mice compared to wild type mice. Taken together, the results suggest that IDH2 plays a critical role in maintaining the spinal structure by affecting the homeostatic balance between osteoclasts and osteoblasts. This indicates that IDH2 might be a potent target for the development of therapies for spinal deformities. Our findings also provide a novel animal model for vertebral disease research., U. Our experiment was carried out in order to explore effects of plant growth regulators (PGR; thidiazuron, paclobutrazol, and ascorbic acid) on physiological traits of wheat genotypes under water surplus and deficit conditions. Study revealed that relative water content, membrane stability index, chlorophyll content, photosynthetic rate (PN), and maximal quantum yield of PSII improved with PGRs application across the genotypes both under irrigation and water stress. The response of HD 2733 genotype was more positive toward PGRs treatment as compared to other genotypes under water stress. Higher PN and chlorophyll contents were observed in HD 2987 followed by C 306 genotype under water-stress conditions. Moreover, Rubisco small subunit (SSU) expression was lower in wheat genotypes under water stress as compared to irrigated conditions. Application of PGRs led to upregulation of SSU under water stress, while no significant change was found in Rubisco level and activity under irrigated condition in dependence on PGRs treatments. Yield-related traits showed also significant reduction under water-stress conditions, while application of PGRs enhanced the yield and its components. Jedním z nejspornějších problémů v současné biologii a medicíně je existence plasticity kmenových buněk. Termínem plasticita se míní fenotypový potenciál kmenových buněk, který je širší než jsou fenotypy diferencovaných buněk jejich původních tkání. Mnoho laboratoří již podalo důkaz o existenci plasticity kmenových buněk, ale vznikla celá řada námitek k prezentovaným výsledkům. Prezentujeme některé z námitek zpochybňujících údaje o plasticitě kmenových buněk. Tyto sporné otázky uvedeme do kontextu s tradičním pohledem na potenciál kmenových buněk a diverzifikaci buněčných fenotypu u dospělých a stárnoucích buněčných fenotypech. Vzhledem k malému prostoru se nebudeme zabývat plasticitou embryonálních a nádorových kmenových buněk. Naopak chceme v našem sdělení poukázat na některé otázky a problémy spojené s plasticitou kmenových buněk v dospělém organizmu a na úzký vztah k procesům regenerace a stárnutí. Z dosavadních experimentálních výsledků můžeme říci, že kmenové buňky pravděpodobně mají klíčovou úlohu v těchto procesech. The term plasticity, as it is understood, is the potential of stem ceU fenotypes that is much broader that fenotypes of differentiated ceUs of their original tissues. Many laboratories have documented the existence of stem cell plasticity, however, there are still many objections to the reported results. Here, we present some of these objections to the reported results. Here, we present some of these objections questioning the data on stem cell plasticity in the context with the traditional view on stem cell potential and diversihcation in adult and aging cell fenotypes. As there is not enough space, we will not discuss the plasticity of embryonal and tumour stem ceUs. Instead, we wish to point out some problems associated with plasticity of stem ceUs in an adult organism and show their close relation to processes of regeneration and aging. The aging process is associated with a decline in mitochondrial functions. Mitochondria dysfunction is involved in initiation and progression of many health problems including neuromuscular, metabolic and cardiovascular diseases. It is well known that endurance exercise improves mitochondrial function, especially in the elderly. V tomto článku předkládáme své zkušenosti s moderními přístupy k léčbě chronické rány u geriatrických pacientů v následné péči. Uvedené kazuistiky demonstrují význam komplexní péče, která je předpokladem úspěšné léčby chronických ran. Onemocnění štítné žlázy je ve starším věku diagnostikováno až u 25 % osob, především se nachází uzlová přestavba nejen ve strumě, ale i v nezvětšené štítné žláze. Tyto změny, často jako důsledek degenerativnich změn, bývají náhodně diagnostikovány při vyšetření krku ultrazvukem při vyšetřování karotid. Překvapivě často je při cíleném vyšetření starší populace diagnostikována porucha tyreoidální funkce, častěji hypotyreóza, a to až u 10-15 % žen starších 65 let. Jak subklinické, tak i hypotyreózy s klinickými projevy unikají pozornosti díky nenápadnému a pozvolnému průběhu, často imitujícími příznaky stárnutí. Příslušná substituční léčba je nezbytná, vyžaduje však zpočátku dobrou spolupráci s pacientem a častější kontroly zvláště u osob s postižením srdce. Tyreotoxikóza je u starších osob méně častá, nicméně při nálezu nodózní strumy a dysrytmií je (sub)klinická tyreotoxikóza pravděpodobná. Negativní dopad nedostatečně léčené tyreotoxikózy především na oběhový aparát a na skelet je prokázán, proto je zapotřebí tyto osoby léčit definitivně, tj. operad nebo radiojodem. Vyvolání poruchy tyreoidální funkce (tyreotoxikózy nebo hypotyreózy) podáváním amiodaronu je až u 10 % takto léčených, léčba cytokiny (interferon) z onkologické či jiné indikace má obdobné důsledky. Doporučujeme proto u léčených starších osob monitorování tyreoidální funkce. Vzhledem k častější incidenci tyreopatií u osob s diabetes mellitus I. typu má být vyšetření tyreoidální funkce provedeno opakovaně. O doporučení screeningu dysfunkce tyreoidey u starší populace při pravidelných prohlídkách u praktických lékařů se nyní vedou diskuse., About 25 % of people are diagnosed with thyroid disease in older age-groups, most frequently nodular changes are to be found, not only in patients with goitre but in those with the thyroid gland of normal size .These changes - which are often the result of degenerative process - are usually diagnosed by chance during ultrasound of the neck for carotid examination. Ssurprisingly frequent is the diagnosis of thyroid dysfunction during deliberate examination in the older populaťion; hypothyroidism is the most common, in up to 10-15 % older females 65+. Like subclinical forms, even hypothyroidism with clinical symptoms is underdiagnosed in elderly because of its inconspicuous and gradual development which mimics the symptoms of ageing. Appropriate substitution therapy is necessary. However, in older people more frequent check-ups particularly in those with cardiac disease and monitoring of patient's compliance is recommended. Thyrotoxicosis is less frequent among older people. Nevetheless when nodal goiter and dysrhythmia are discovered, it is also likely that (sub)clinical thyrotoxicosis is present. The negative impact of insufficiently treated thyrotoxicosis particularly on the circulatory system and bones has been proven. Therefore it is necessary to treat these persons thoroughly, i.e., by operation or radioiodine. Problems with the thyroid function (thyrotoxicosis or hypothyrosis) due to amiodaron treatment develop in up to 10% of treated patients; treatment with cytokins (e.g. interferon) in oncological or other indilarly in older age-groups. In view of the more frequent incidence of thyropathy in persons with diabetes mellitus 1. type, examination of the thyroid function should be carried out repeatedly in older diabetics. An ontogenetic study of ecto-ATPase activity and the content of enzyme proteins was assessed in the caudate nucleus and hippocampal synaptic plasma membranes isolated from rats at various ages (15, 30, 90, 180 and 365 days). The ontogenetic profile revealed that the enzyme activities in both brain areas were the highest on day 30 and 365, while the ecto-ATPase protein abundance was the highest on day 15 after birth. Possible explanation for obtained ontogenetic profile and the discrepancy between activity and abundance may reside in the fact that ecto-ATPase during development could exert additional roles other than those related to metabolism of ATP. Na osteoporózu, průvodní problém postmenopauzálního období je třeba pohlížet spíše jako na „perignózu“ a chápat ji komplexněji v souvislosti se sarkopenií jako projev stárnutí organizmu. Tuto skutečnost se snaží postihnout nový termín „syndrom dysmobility“. Záměr. Studie shrnuje poznatky o použití české verze Dotazníku proaktivního zvládání (Proactive Coping Inventory, PCI) u seniorské populace. V práci je srovnáván vztah mezi jednotlivými škálami dotazníku PCI, paměťovým výkonem a koncentrací.
Soubor a procedura. Soubor tvořilo 257 samostatně žijících dospělých ve věku 55 až 92 let z Prahy a okolí. Při výzkumu byl využit Dotazník proaktivního zvládání. Paměťový výkon byl měřen pomocí Paměťového testu učení (AVLT) a subtestu Logická paměť z WMS-III. Kromě kognitivních testů byly administrovány dotazníky Cognitive Failure Questionnaire (CFQ), Schwartz Outcomes Scale (SOS-10) a doplňkové škály Implicitního asociačního testu zaměřeného na sebehodnocení (IAT).
Statistická analýza. Nejprve byla provedena psychometrická analýza škál dotazníku PCI, kde autoři zjišťovali vnitřní konsistenci škál a korelace jednotlivých položek v rámci škály. V druhém kroku byly zjišťovány vztahy mezi vzděláním, věkem a jednotlivými škálami PCI pomocí korelací (Spearmanův koeficient korelace). Pomocí korelační analýzy byl srovnáván vztah jednotlivých škál PCI s výsledky ostatních nástrojů.
Výsledky. Srovnání ukázalo, že paradoxně horší paměťový výkon souvisí s vyšší preferencí řešení situací, které zahrnují reflexi a plánování (škály reflektivní řešení a strategické řešení situací). Tento jev je interpretován jako kompenzace. Psychometrická analýza potvrdila, že česká verze dotazníku má při použití u starších osob srovnatelné vlastnosti s původním nástrojem. U seniorů byly nalezeny nižší hodnoty Proaktivního zvládání.
Omezení studie. Sledovaný soubor není reprezentativní, převažují v něm ženy, a úroveň vzdělání je nadprůměrná. and Proactive coping and cognitive performance in elderly
Objectives. The study reports the results of Czech version of Proactive Coping Inventory (PCI) in the elderly. The results of PCI subscales were compared to the cognitive performance in memory and attention domains.
Subjects and setting. The sample included 257 community-dwelling senior citizens from Prague region. The Czech version of Proactive Coping Inventory was administered. The memory performance was assessed using AVLT and Logical Memory subtest from WMS-III. Besides the cognitive tests Cognitive Failure Questionnaire (CFQ), Schwartz Outcomes Scale (SOS-10) and explicit measures from self-esteem Implicit Association Test were administered. Statistical analysis. The psychometric properties of PCI subscales were calculated (Cronbach alpha, item-scale correlations). The relationship of PCI subscales to demographic factors (age, education), cognitive performance and other measures was investigated in correlation analysis.
Results. Contrary to prior expectations worse cognitive performance is related to higher scores in Reflective Coping and Strategic Planning subscales, which refer to the use of reflection and planning. This effect is interpreted as a compensation mechanism. The psychometric analysis confirmed similar psychometric properties of the questionnaire in elderly compared to the original study. Lower Proactive Coping scores were found in this study.
Study limitations. The sample is not representative, with majority of women and above-average education level.
