The cell culture became an invaluable tool for studying cell behaviour, development, function, gene expression, toxicity of compounds and efficacy of novel drugs. Although most results were obtained from cell cultivation in two-dimensional (2D) systems, in which cells are grown in a monolayer, three-dimensional (3D) cultures are more promising as they correspond closely to the native arrangement of cells in living tissues. In our study, we focused on three types of 3D in vitro systems used for cultivation of one cell type. Cell morphology, their spatial distribution inside of resulting multicellular structures and changes in time were analysed with histological examination of samples harvested at different time periods. In multilayered cultures of WRL 68 hepatocytes grown on semipermeable membranes and non-passaged neurospheres generated by proliferation of neural progenitor cells, the cells were tightly apposed, showed features of cell differentiation but also cell death that was observable in short-term cultures. Biogenic scaffolds composed of extracellular matrix of the murine tibial anterior muscle were colonized with C2C12 myoblasts in vitro. The recellularized scaffolds did not reach high cell densities comparable with the former systems but supported well cell anchorage and migration without any signs of cell regression. and H. Cell-penetrating compounds are substances that enhance the cellular uptake of various molecular cargoes that do not easily cross the cellular membrane. The majority of cell-penetrating compounds described in the literature are cell-penetrating peptides (CPPs). This review summarizes the various structural types of cell-penetrating compounds, with the main focus on CPPs. The authors present a brief overview of the history of CPPs, discuss the various types of conjugation of CPPs to biologically active cargoes intended for cell internalization, examine the cell-entry mechanisms of CPPs, and report on the applications of CPPs in research and in preclinical and clinical studies., E. The thymus plays a critical role in establishing and maintaining the peripheral T-cell pool. It does so by providing a microenvironment within which T-cell precursors differentiate and undergo selection processes to create a functional population of major histocompatibility complex-restricted, self-tolerant T cells. These cells are central to adap tive immunity. Thymic T-cell development is influenced by loca lly produced soluble factors and cell-to-cell interactions, as well as by sympathetic noradrenergic and endocrine system signalling. Thymic lymphoid and non- lymphoid cells have been shown not only to express β - and α 1 - adrenoceptors (ARs), but also to synthesize catecholamines (CAs). Thus, it is suggested that CAs influence T-cell development via both neurocrine/endocrine and autocrine/ paracrine action, and that they serve as immunotransmitters between thymocytes and nerves. CAs acting at multiple sites along the thymocyte developmental route affect T-cell generation not only numerically, but also qualitatively. Thymic CA level and synthesis, as well as AR expression exhibit sex steroid-mediated sexual dimorphism. Moreover, the influence of CAs on T-cell development exhibits glucocorticoid-dependent plasticity. Summary The aim of this study was to characterize an in vitro modulating effect of three commensal Lactobacillus strains on cellular differentiation of non-transformed crypt-like rat small intestinal cell line IEC-18. IEC-18 was grown on extracellular matrix, with or without presence of Lactobacillus strains. Gene expression of IEC-18 bacterial detection system - such as Toll-like receptors TLR-2, TLR-4, signal adapter MyD88, cytoplasmic NOD2 receptor, inflammatory cytokines IL-18, IL-1β, chemokine IL-8 and enzyme caspase-1 - was evaluated using real-time PCR. Expression and localization of TLR-2, TLR-4, IL-18 and caspase-1 proteins was demonstrated by Western blotting and immunofluorescent staining. Secretion of IL-18 to apical and basolateral surfaces was assayed by ELISA. Our results suggested that L. casei LOCK0919 accelerated differentiation of IEC-18 by stimulating TLR-2, TLR-4, MyD88, IL-18, caspase-1 mRNAs and proteins. L. casei LOCK0919 increased expression and transfer of villin and β-catenin from cytoplasm to cell membrane. Presence of L. rhamnosus LOCK0900 resulted in detachment of IEC-18 layer from extracellular matrix leading to induction of IL-1β, of TLR-2 and IL-8 mRNAs and stimulation of MyD88, caspase-1 and cytosolic receptor NOD2 mRNAs. The term cellular immune response refers to haemocyte-mediated responses, including phagocytosis, nodulation, and encapsulation. In the present study, we identified five types of circulating haemocytes in larvae of the haemolymph of the Asian corn borer, Ostrinia furnacalis (Guenée), including granulocytes, oenocytoids, plasmatocytes, prohaemocytes, and spherulocytes. The relative number of total free haemocytes per larva decreased significantly 0.5, 24, and 36 h after the injection of Beauveria bassiana conidia. Upon conidia challenge, both phagocytosis and nodulation were observed in the collected haemolymph from O. furnacalis larvae. In addition, plasma was found to be necessary for both phagocytosis and nodulation. Therefore, we here confirm that phagocytosis and nodulation are involved in O. funacalis larvae during their fight against infection by B. bassiana, and further, that the cellular immune response of O. furnacalis helps eliminate the invading organisms despite the fact that not all the fungal conidia are killed., Dongxu Shen, Miao Li, Yuan Chu, Minglin Lang, Chunju An., and Obsahuje bibliografii

Examination of a total of 581 fish specimens of 15 species from 39 cenotes (sinkholes) in the Yucatan Peninsula, southeastern Mexico, revealed the presence of 10 species of adult trematodes. These were as follows: Saccocoelioides sogandaresi Lumsden, 1963, Saccocoelinides sp. (family Haploporidae), Cichlasotrema ujati Pineda et Andrade, 1989 (Angiodictyidae), Crassicutis cichlasomae Manter, 1936 (Homalometridae), Magnivitellinum simplex Kloss, 1966 (Macroderoididae), Stunkardiel-la minima (Stunkard, 1938) (Acanthostomidae), Oligogonotylus manieri Watson, 1976 (Cryptogonimidae), Genarchella tropica (Manter, 1936), G. astyanactis (Watson, 1976), and G. isabellae (Lamothe-Argumedo, 1977) (Derogenidae). Saccocoelioides sogandaresi is reported from Mexico for the first time. Poecilia velifera and P. latipunctata for S. sogandaresi, Cichlasoma octo-fasciatum for C. cichlasomae, Cichlasoma friedrichslahli and C. meeki for O. manieri, and C. meeki, C. octofasciatum and Go-hiomorus dormitor for C. isabellae represent new host records. The aim of this study was to compare the central and peripheral components of cardiorespiratory fitness during incremental to maximal exercise between older men who were either recreational athletes (RA) or leisurely active (LA) men, i.e., those who fall between trained and untrained. This was a crosssectional study in which all subjects completed an exercise test on a cycle ergometer. Maximal oxygen consumption (VO2max) and ventilatory threshold (VT) were assessed using gas analysis, and central components of VO2max were assessed using a non-invasive thoracic bio-impedance device. VO2max (RA: 45.1±4.8 ml/kg/min; LA: 32.2±4.6 ml/kg/min, p≤0.001) and SV at maximal exercise (RA: 133.5±24.96 ml/beat; LA: 107.9±17.6 ml/beat, p=0.005) were higher in the RA group compared to the LA group. A plateau in SV occurred between 30-45 % of maximal exercise capacity in the RA group. No differences in SV were observed across workloads in the LA group. No differences in the calculated arterio-venous oxygen difference ((a-v)O2diff) were observed between groups. In conclusions, training volume appears to influence central components of cardiorespiratory fitness among a matched sample of older men who are neither trained nor untrained.
