Over last decades, several studies have been focused on
short-term high light stress in lichens under laboratory conditions. Such studies reported a strong photoinhibition of photosynthesis accompanied by a partial photodestruction of PSII, involvement of photoprotective mechanisms, and resynthetic processes into gradual recovery. In our paper, we applied medium [800 μmol(photon) m-2 s-1] light stress to induce negative changes in PSII funcioning as well as pigment and glutathione (GSH) content in two Antarctic fruticose lichen species. Chlorophyll (Chl) fluorescence parameters, such as potential and effective quantum yield of photosynthetic processes and fast transients (OJIP) recorded during high light exposition and recovery, revealed that Usnea antarctica was less susceptible to photoinhibition than U. aurantiaco-atra. This might be supported by a more pronounced high light-induced reduction in Chl a and b contents in U. aurantiaco-atra compared with U. antarctica. In both experimental species, total GSH showed an initial increase during the first 30-40 min of high light treatment followed by a decrease (60 min) and an increase during dark recovery. Full GSH recovery, however, was not finished in U. aurantiaco-atra even after 5 h indicating lower capacity of photoprotective mechanisms in the species. OJIP curves showed high light-induced decrease in both species, however, the recovery of the OJIPs shape to pre-photoinhibitory values was faster and more apparent in U. antarctica than in U. aurantiaco-atra. The effects of phosphate concentration on plant growth and photosynthetic performance were examined in leaves of Zizania latifolia. Plants were grown for four weeks in a solution containing 0, 0.16, 0.64, and 2.56 mM orthophosphate. The results showed that the highest net photosynthetic rate (P N) was achieved at 0.64 mM orthophosphate, which corresponded to the maximum content of organic phosphorus in leaves. Low phosphorus (low-P) content in the culture solution inhibited plant growth, affecting plant height, leaf length, leaf number, tiller number, and fresh mass of leaf, sheath, culm, root, and total plant. In addition, we observed that low-P (0.16 mM) did not hinder the growth of roots but increased the root:shoot ratio, and significantly decreased the chlorophyll content, P N, stomatal conductance, and transpiration rate, but increased the intercellular CO2 concentration. Additionally, low-P significantly decreased the maximum carboxylation rate of Rubisco, the maximum rate of ribulose-1,5-bisphosphate regeneration, the effective quantum yield of PSII photochemistry, photochemical quenching coefficient, and electron transport rate, but increased the nonphotochemical quenching. However, the maximal quantum yield of PSII photochemistry was not significantly affected by low-P. High phosphorus (2.56 mM) caused only a slight decrease in gas-exchange parameters. A comparison between maximum quantum yield of PSII photochemistry (Fv/Fm) and chlorophyll fluorescence decrease ratio (Rfd) for low and high temperature resistance was assessed in a seasonal study of the acclimation in Pterocephalus lasiospermus. Analyzing the regression adjustment of both parameters and the lethal temperatures (LT50), Rfd resulted in being a more sensitive indicator for low and high temperature treatments, since the thermic resistance estimated with Rfd parameter was never higher than those estimated with Fv/Fm. Furthermore, the use of Fv/Fm led to an overestimation of the acclimation phenomena, with 6ºC of a maximum difference between both parameters. Using Rfd as the indicator parameter, P. lasiospermus acclimated to low temperatures but it kept on being a sensitive species (the lowest LT50 values only achieved -9.9 ± 0.3ºC). However, no heat acclimation was observed (LT50 around 43.5ºC). Water availability is a major limiting factor in desert ecosystems. However, a winter snowfall role in the growth of biological soil crusts is still less investigated. Here, four snow treatments were designed to evaluate the effects of snow depth on photosynthesis and physiological characteristics of biological soil crusts. Results showed that snow strongly affected the chlorophyll fluorescence properties. The increased snow depth led to increased contents of photosynthetic pigments and soluble proteins. However, all biological soil crusts also exhibited a decline in malondialdehyde and soluble sugar contents as snow increased. Results demonstrated that different biological soil crusts exhibited different responses to snow depth treatment due to differences in their morphological characteristics and microhabitat. In addition, interspecies differentiation in response to snow depth treatment might affect the survival of some biological soil crusts. The purpose of the study was to check whether hypoxia of corneal tissue increases the collagenolytic activity due to release of reactive oxygen and nitrogen species. Rats were exposed to hypoxia 10 % O2 for 4, 14, and 21 days. The radical tissue injury was measured by the level of nitrotyrosine and changes in the lipoperoxide-related fluorophores. Collagen protein composition was analyzed by slab gel electrophoresis. The activity of gelatinolytic enzymes was studied using the zymography. The vascularization of the corneas was measured. We found no differences in the corneal tissue in the gel electrophoretic profile of collagenous proteins and gelatinolytic activity between normoxic and hypoxic rats. We did not find any sign of radical tissue injury. There were no changes in the vascularization of corneas after exposition to hypoxia. Six types of sphaeractinomyxon are reported from the coelomic cavity of oligochaetes collected from the Minho River estuary in northern Portugal. Four new types are morphologically and molecularly described from freshwater species belonging to the genera Psammoryctides Hrabě and Potamothrix Vejdovský et Mrázek in the upper estuary, thus significantly increasing the number of known freshwater sphaeractinomyxon. In the lower estuary, sphaeractinomyxon types 8 and 10 of Rangel et al. (2016) are recorded infecting the marine oligochaete Tubificoides pseudogaster (Dahl). A single specimen of T. pseudogaster further displayed infection by one of the four new types found in the upper estuary, suggesting the involvement of sphaeractinomyxon in the life cycles of myxosporean species that infect migratory fish hosts. The acquisition of these second hosts is proposed to have allowed the myxosporean counterparts of sphaeractinomyxon to cross environmental barriers and conquer new habitats. Phylogenetic analyses of the SSU rRNA gene reveal the four new types clustering within the monophyletic clade of mugiliform-infecting myxobolids, strengthening the previously proposed involvement of the sphaeractinomyxon collective group in the life cycles of this specific group of myxosporeans. Endocapsa types also cluster within the latter clade, having actinospores that differ from those of sphaeractinomyxon only in the presence of valvular swellings that do not change when in contact with water. In this study, however, one type was found displaying actinospores with and without valvular swellings in the same oligochaete specimen. Growth of the remnant embryonic kidney (the mesonephros), as expressed by wet weight, was more rapid in the chick embryos with experimentally induced unilateral renal agenes is compared to controls. The difference was significant between embryonic days 8-12, when the doubled weights of remnant kidneys were increased compared with the weights of paired control kidneys. The excessive growth of the mesonephros ceased on day 14, when the normal physiological regression of the embryonic kidney begins. In the definitive kidney, the metanephros, no significant differences in weights of the control vs. remnant metanephros were found on days 10-14. The characteristics of increased mesonephric growth were evaluated by determination of DNA/protein ratios in homogenates of the kidneys. Significant cellular hypertrophy was found in both the mesonephros and metanephros of the embryos with URA on day 10. Additionally, a non-significant cellular hyperplasia was also revealed in the remnant mesonephros on day 8. Serpin is a broadly distributed superfamily of proteins that have a crucial role in regulating various immune reactions. Herein we identified a serpin-10 gene from Antheraea pernyi that encodes a 1557 amino acid residue protein with a predicted molecular weight of 58.76 kDa. Recombinant Apserpin-10 protein was expressed in a prokaryotic expression system (Escherichia coli) and the purified protein was used to prepare rabbit anti-Apserpin-10 polyclonal antibodies. Quantitative real-time polymerase chain reaction and western blot analysis indicate that Apserpin-10 was transcribed in all the tissues examined, including haemolymph, malpighian tubules, fat body, silk gland, integument and mid gut; the greatest expression level of Apserpin-10 was recorded in the fat body and haemocytes. The comparison of different developmental stages showed that Apserpin-10 transcript level was highest in 5th instar larvae, while the lowest expression was recorded at the egg stage. We also investigated the expression patterns of Apserpin-10 in fat body and haemocyte samples, following administration of heat-inactivated gram-positive bacteria (Micrococcus luteus), gram negative bacteria (Escherichia coli), a fungus (Beauveria bassiana) and virus (nuclear polyhedrosis virus, NPV). A substantial up-regulation of Apserpin-10 expression was recorded following pathogen challenge in both the tissues tested. Further the knock down of Apserpin-10 led to down regulation of antimicrobial peptide genes. Satellite DNAs are the major repetitive DNA components in eukaryotic genomes. Although satellite DNA has long been called "parasite DNA" there is substantial evidence that it could be associated with some functions of chromosome biology. Ladybird beetles (Coccinellidae) are one of the largest and most important groups of beetles. Many ladybirds are of economic interest as biological control agents because they eat some agricultural pests such as aphids and scale insects. However, other species are phytophagous and can damage crops. Despite the ecological importance of the latter group there are no studies on their satellite DNA. A satellite DNA family was isolated and characterized in the ladybird Henosepilachna argus. This satellite DNA is organized in tandem repeats of 658 bp and is A + T rich (67.3%). The recorded high sequence conservation of the monomers together with the detection of putative gene conversion processes indicate concerted evolution. Reverse transcription polymerase chain reaction (RT-PCR) revealed that this satellite DNA is transcribed and in situ hybridization its location in the subtelomeric regions of all chromosomes except the long arm of the X chromosome. The presence of this satellite DNA in other species of the genus Henosepilachna and Epilachna was also tested using PCR. A novel panel of 16 microsatellite markers, obtained by pyrosequencing of enriched genomic libraries, is reported for the flightless European bushcricket Ephippiger diurnus (Dufour) (Orthoptera: Tettigoniidae). Five multiplex and one simplex PCR protocols were optimized, and the polymorphism at the 16 loci was assessed in two natural populations from southern France. The mean allele number and (expected mean heterozygosity) were 8.94 (0.71) and 6.57 (0.70), respectively, in each population. Several loci were at Hardy-Weinberg disequilibrium (HWD), possibly due to the incidence of null alleles. The occurrence of null alleles has been previously reported for this species, and it is a common feature of microsatellite loci in Orthoptera. Cross-amplification tests demonstrated the transferability of some of these loci to other ephippigerine species. The microsatellite loci reported here substantially increase the number of available loci for this species and will afford an accurate picture of E. diurnus phylogeography, the genetic structure of its populations, and an improved understanding of the evolution of male song and other sexually-selected traits in this highly variable species., Yareli Esquer-Garrigos, Michael D.
