The transfer of light energy from phycobilisomes (PBS) to photosystem II (PSII) reaction centers is vital for photosynthesis in cyanobacteria and red algae. To investigate the relationship between PBS and PSII and to optimize the energy transfer efficiency from PBS to PSII, isolation of the PBS-PSII supercomplex is necessary. SPC (sucrose/phosphate/citrate) is a conventional buffer for isolating PBS-PSII supercomplex in cyanobacteria. However, the energy transfer occurring in the supercomplex is poor. Here, we developed a new buffer named SGB by adding 1M glycinebetaine and additional sucrose to SPC buffer. Compared to SPC, the newly developed SGB buffer greatly enhanced the associated populations of PBS with thylakoid membranes and PSII and further improved the energy transfer efficiency from PBS to PSII reaction centers in cyanobacteria in vitro. We have developed a simple and an effective method for the isolation of photochemically active broken chloroplasts from conifer needles that can be applied for a wide variety of conifer species with needle-like leaves. The utilisation of this method in photosynthetic studies offers a possibility to examine the efficiency of almost any component of thylakoid electron-transport chain and to disclose information about individual parts of primary photosynthetic processes that would be otherwise difficult to obtain. The objective of this study was to investigate the relative salt tolerance of four eggplant cultivars (Solanum melongena L.) by studying chlorophyll (Chl) fluorescence parameters during the vegetative growth stage under increasing salinity levels. The plants were grown in pots filled with peat under controlled conditions and were subjected to the salt stress ranging from 0 (control), 20, 40, 80, and 160 mM NaCl for 25 days. The results showed that the increasing NaCl concentration affected hardly the maximum quantum yield of photosystem (PS) II. The quantum yield of PSII (ΦPSII) decreased significantly in ‘Adriatica’ and ‘Black Beauty’ under the salt stress. The photochemical quenching decreased in ‘Black Beauty’ and nonphotochemical quenching increased in ‘Adriatica’ under the salt stress. The Chl fluorescence parameters did not change significantly under the salt stress in ‘Bonica’ and ‘Galine’, revealing their tolerance to salinity. After 25 days of the salt stress, the plant growth was reduced in all cultivars, however, this decline was more pronounced in ‘Adriatica’ and ‘Black Beauty’. Additionally, a significant correlation between the biomass and ΦPSII was observed in ‘Adriatica’ and ‘Black Beauty’. The light-induced nonphotochemical quenching (NPQ) can safely dissipate excess of absorbed light to heat. Here we describe an application of spectrally resolved fluorescence induction (SRFI) method for studying spectral variability of NPQ. The approach allows detection of spectrally-resolved nonphotochemical quenching (NPQλ) representing NPQ dependency on fluorescence emission wavelength in the whole spectral range of fluorescence emission. The experimental approach is briefly described and NPQλ is studied for the cryptophyte alga Rhodomonas salina and for green alga Chlorella sp. We confirm presence of NPQλ only in membrane-bound antennae (chlorophyll a/c antennae) and not in phycobiliproteins in lumen in cryptophyte and show that NPQλ is inhibited in the whole spectral range by NPQ inhibitors in Chlorella sp. In the past decades, it has become clear that superoxide radical (O2 .-) can be generated from photosystem II (PSII) during photosynthesis. Depending on the extent of its accumulation, O2 .- plays an important role in plant physiology and pathology. The photoinhibition/repair cycle is a typical process in PSII which is mainly responsible for the survival of plants under the photoinihibition condition. It is therefore of significant importance to determine O2 .- production in this cycle, and then explore how O2 .- is controlled by PSII within a normal physiological level. With this in mind, we herein investigate the variation of the O2 .- levels in PSII under Mn-depleted and photoactivated conditions mimicking the photoinhibition/repair cycle in vitro. The effect of intrinsic SOD-like component on the O2 .- levels was also studied. Results show that PSII has the ability to regulate the O2 .- levels in these two processes by simultaneously modulating the O2 .- generation activity and intrinsic SOD-like activity. Differences in maximal yields of chlorophyll variable fluorescence (Fm) induced by single turnover (ST) and multiple turnover (MT) excitation are as great as 40%. Using mutants of Chlamydomonas reinhardtii we investigated potential mechanisms controlling Fm above and beyond the QA redox level. Fm was low when the QB binding site was occupied by PQ and high when the QB binding site was empty or occupied by a PSII herbicide. Furthermore, in mutants with impaired rates of plastoquinol reoxidation, Fm was reached rapidly during MT excitation. In PSII particles with no mobile PQ pool, Fm was virtually identical to that obtained in the presence of PSII herbicides. We have developed a model to account for the variations in maximal fluorescence yields based on the occupancy of the QB binding site. The influence of various concentrations of imazapic residues (0-800 μg kg-1) on the growth, chlorophyll content, and photosynthetic characteristics of maize seedlings was studied in a greenhouse pot experiment. Plant height, root length, shoot dry mass, root dry mass, and total dry mass of maize declined with the increase of imazapic residue concentrations. The root/shoot ratio initially decreased and then increased in presence of imazapic, which indicated that the effects of imazapic residues on plant height and root length might differ in maize seedlings. Lowered chlorophyll content and net photosynthetic rate were observed in leaves of maize seedlings in all treatments and indicated a dose-response relationship to imazapic concentrations. Intercellular carbon dioxide concentration, transpiration rate, and stomatal conductance also declined to varying extents, but the chlorophyll a/b ratio increased gradually together with the increase of imazapic residue concentrations. Generally, the maize seedlings were negatively affected by the imazapic residues in soil. The effects of actinic light (AL) intensity on the age dependence of nonphotochemical fluorescence quenching (qN) and effective quantum yield in PSII (ΦPSII) were studied in continuously illuminated wheat leaves of the upper tier. Regular changes were revealed in both age dependence of qN at elevated AL intensities and light curves of qN. These changes are related to alterations in strategies of redistribution and use of absorbed light energy by the photosynthetic apparatus at different stages of wheat leaf development. Unlike ΦPSII, qN as a parameter was more sensitive to the differences in the leaf age at a certain range of light intensities. Effects of root treatment with 5-aminolevulinic acid (ALA) on leaf photosynthesis in strawberry (Fragaria ananassa Duch.) plants were investigated by rapid chlorophyll fluorescence and modulated 820 nm reflection using 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU) and methyl viologen (MV). Our results showed that ALA treatments increased the net photosynthetic rate and decreased the intercelluar CO2 concentration in strawberry leaves. Under DCMU treatment, trapping energy for QA reduction per PSII reaction center increased greatly, indicating DCMU inhibited electron transfer from QA−. The maximum photochemical efficiency of PSII (Fv/Fm) decreased under the DCMU treatment, while a higher Fv/Fm remained in the ALA-pretreated plants. Not only the parameters related to a photochemical phase, but also that one related to a heat phase remained lower after the ALA pretreatment, compared to the sole DCMU treatment. The MV treatment decreased PSI photochemical capacity. The results of modulated 820 nm reflection analysis showed that DCMU and MV treatments had low
re-reduction of P700 and plastocyanin (PSI). However, the strawberry leaf discs pretreated with ALA exhibited high re-reduction of PSI under DCMU and MV treatments.
