The objective of this study was to investigate the relative salt tolerance of four eggplant cultivars (Solanum melongena L.) by studying chlorophyll (Chl) fluorescence parameters during the vegetative growth stage under increasing salinity levels. The plants were grown in pots filled with peat under controlled conditions and were subjected to the salt stress ranging from 0 (control), 20, 40, 80, and 160 mM NaCl for 25 days. The results showed that the increasing NaCl concentration affected hardly the maximum quantum yield of photosystem (PS) II. The quantum yield of PSII (ΦPSII) decreased significantly in ‘Adriatica’ and ‘Black Beauty’ under the salt stress. The photochemical quenching decreased in ‘Black Beauty’ and nonphotochemical quenching increased in ‘Adriatica’ under the salt stress. The Chl fluorescence parameters did not change significantly under the salt stress in ‘Bonica’ and ‘Galine’, revealing their tolerance to salinity. After 25 days of the salt stress, the plant growth was reduced in all cultivars, however, this decline was more pronounced in ‘Adriatica’ and ‘Black Beauty’. Additionally, a significant correlation between the biomass and ΦPSII was observed in ‘Adriatica’ and ‘Black Beauty’. Our results suggest that ΦPSII can be used as a diagnostic tool to identify salt-tolerant egg-plant cultivars., S. Hanachi, M. C. Van Labeke, T. Mehouachi., and Obsahuje bibliografii
The light-induced nonphotochemical quenching (NPQ) can safely dissipate excess of absorbed light to heat. Here we describe an application of spectrally resolved fluorescence induction (SRFI) method for studying spectral variability of NPQ. The approach allows detection of spectrally-resolved nonphotochemical quenching (NPQλ) representing NPQ dependency on fluorescence emission wavelength in the whole spectral range of fluorescence emission. The experimental approach is briefly described and NPQλ is studied for the cryptophyte alga Rhodomonas salina and for green alga Chlorella sp. We confirm presence of NPQλ only in membrane-bound antennae (chlorophyll a/c antennae) and not in phycobiliproteins in lumen in cryptophyte and show that NPQλ is inhibited in the whole spectral range by NPQ inhibitors in Chlorella sp. We discuss variability in the quenching in the particular spectral ranges and applicability of the NPQλ parameter to study quenching locus in vivo., R. Kaňa., and Obsahuje bibliografické odkazy
Based on the examination and quantitative comparison of the approaches used to assess the energy partitioning in photosystem II, the unified method was proposed to calculate the contribution of the components of nonphotochemical quenching. and D. Kornyeyev, C. R. Guadagno, N. D’Ambrosio.
Differences in maximal yields of chlorophyll variable fluorescence (Fm) induced by single turnover (ST) and multiple turnover (MT) excitation are as great as 40%. Using mutants of Chlamydomonas reinhardtii we investigated potential mechanisms controlling Fm above and beyond the QA redox level. Fm was low when the QB binding site was occupied by PQ and high when the QB binding site was empty or occupied by a PSII herbicide. Furthermore, in mutants with impaired rates of plastoquinol reoxidation, Fm was reached rapidly during MT excitation. In PSII particles with no mobile PQ pool, Fm was virtually identical to that obtained in the presence of PSII herbicides. We have developed a model to account for the variations in maximal fluorescence yields based on the occupancy of the QB binding site. The model predicts that the variations in maximal fluorescence yields are caused by the capacity of secondary electron acceptors to reoxidize QA-., O. Prášil, Z. S. Kolber, P. G. The effects of actinic light (AL) intensity on the age dependence of nonphotochemical fluorescence quenching (qN) and effective quantum yield in PSII (ΦPSII) were studied in continuously illuminated wheat leaves of the upper tier. Regular changes were revealed in both age dependence of qN at elevated AL intensities and light curves of qN. These changes are related to alterations in strategies of redistribution and use of absorbed light energy by the photosynthetic apparatus at different stages of wheat leaf development. Unlike ΦPSII, qN as a parameter was more sensitive to the differences in the leaf age at a certain range of light intensities. At the same time, the stability of qN at moderate light intensities may serve as an indication of leaf maturity., T. V. Nesterenko, V. N. Shikhov, A. A. Tikhomirov., and Obsahuje seznam literatury
Excessive cadmium (Cd) content in soil leads to a number of phytotoxic effects and challenges agricultural production. Aim of this study was to investigate different responses of two maize inbreds and their hybrid to an elevated Cd content in soil by measuring photosynthetic and biochemical activity and to identify a Cd tolerance mechanism. Antioxidant statusrelated parameters varied significantly between inbreds and treatments. Dry mass increased in both inbreds, but remained unchanged in hybrid. After the Cd treatment, parameters of chlorophyll a fluorescence varied between inbreds and hybrid performance was similar to inbred B84. We concluded that inbred B84 is Cd-sensitive compared to Os6-2, which did not appear to be negatively affected by Cd treatment at this growth stage studied. We suspect that due to a dilution effect in the hybrid, there was no or very weak Cd stress detected by biochemical parameters, although stress was detected by chlorophyll a fluorescence., M. Acclimation to excess light is required for optimizing plant performance under natural environment. The present work showed that the treatment of Arabidopsis leaves with exogenous H2O2 can increase the acclimation of PSII to excess light. Treatments with H2O2 also enhanced the capacity of the mitochondrial alternative respiratory pathway and salicylic acid (SA) content. Our work also showed that the lack in alternative oxidase (AOX1a) in AtAOX1a antisense line and the SA deficiency in NahG (salicylate hydroxylase gene) transgenic mutant attenuated the H2O2-induced acclimation of PSII to excess light. It indicates that the
H2O2-induced acclimation of PSII to excess light could be mediated by the alternative respiratory pathway and SA., Q. Z. Hou, Y. P. Wang, J. Y. Liang, L. Y. Jia, H. Q. Feng, J. Wen, N. Ehmet, J. Y. Bai., and Obsahuje bibliografii
Salicylic acid (SA) is a common, plant-produced signal molecule that is responsible for inducing tolerance to a number of biotic and abiotic stresses. Our experiment was therefore conducted to test whether the application of SA at various concentrations (0, 0.10, 0.50, and 1.00 mM) as a foliar spray would protect citrus seedlings (Valencia orange/Bakraii) subjected to salt stress (0, 25, 50, and 75 mM NaCl). Growth parameters, leaf chlorophyll (Chl) content, relative water content (RWC), maximal quantum yield of PSII photochemistry (Fv/Fm), and gas-exchange variables were negatively affected by salinity. In addition, leaf electrolyte leakage (EL) and proline content increased by salinity treatments. Application of SA increased net photosynthetic rate and proline content in salt stressed plants and may have contributed to the enhanced growth parameters. SA treated plants had greater Chl content and RWC compared with untreated plants when exposed to salt stress. Fv/Fm ratio and stomatal conductance were also significantly higher in SA treated plants under saline stress conditions. SA application reduced EL compared to untreated plants, indicating possible protection of integrity of the cellular membrane. It appeared that the best ameliorative remedies of SA were obtained when Valencia orange/Bakraii seedlings were sprayed by 0.50 and 1.00 mM solutions. Overall, the adverse effects of salt stress could be alleviated by exogenous application of SA., D. Khoshbakht, M. R. Asgharei., and Obsahuje seznam literatury
Increasing human and industrial activities lead to heavy metal pollution. Heavy metal chromium (Cr) is considered to be a serious environmental contaminant for the biota. Phytotoxic effects of Cr were studied in wheat plants. Growth parameters were largely inhibited as a result of disturbances in the plant cell metabolism in response to Cr toxicity. Chromium toxicity led to decline in a number of active reaction centres of PSII, rate of electron transport, and change in PSII heterogeneity. Chromium did not cause any change in heterogeneity of the reducing side. A significant change in antenna size heterogeneity of PSII occurred in response to Cr toxicity. Chromium seems to have extensive effects on the light harvesting complex of PSII., S. Mathur, H. M. Kalaji, A. Jajoo., and Obsahuje seznam literatury
To investigate the photoprotection of energy dissipation and water-water cycle, a C3 euhalophytic herb, Suaeda salsa L., was exposed either to chilling temperature (4°C) accompanied by moderate irradiance (600 μmol m-2 s-1) (CM) and/or to chilling temperature (4°C) accompanied by low irradiance (100 μmol m-2 s-1) (CL). During chilling stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and the oxidizable P700 decreased in S. salsa leaves either under CM or CL, which indicated the severe photoinhibition. Relative to Fv/Fm, the oxidizable P700 decreased markedly under CL, which indicated that PSI was more sensitive to CL treatment than PSII. Initial fluorescence, number of closed PSII centers, and nonphotochemical quenching increased under CM, but more markedly under CL in S. salsa leaves. Activity of superoxide dismutase and ascorbate peroxidase was higher under CM than that under CL. The production of reactive oxygen species (ROS) decreased first and then increased under both treatments, but the content of O2.- and H2O2 was higher under CL than that under CM after 12 h of chilling stress. These results suggested that photoinhibition in S. salsa might be related to the accumulation of reactive oxygen species (ROS) induced by excess energy. The water-water cycle could not dissipate energy efficiently under CL, which caused the great accumulation of ROS., N. Sui., and Obsahuje bibliografii
