The effect of adding 5 % powdered oyster mushroom (Pleurotus ostreatus) during 12 weeks on kinetic parameters of cholesterol metabolism was studied in male rats (Wistar, initial body weight 85 g) fed a semisynthetic diet containing 0.3 % of cholesterol. The plasma cholesterol decay curve (examined for the final 29 days of the experiment after a single dose of cholesterol-4- 14C) was evaluated by mathematical analysis using a two-pool model of plasma cholesterol metabolism. The oyster mushroom in the diet reduced the half-times of both exponentials resulting in lower calculated values (by 28 %) of total entry of cholesterol into the body cholesterol pool (absorption + endogenous synthesis) and lower sizes of both pools (with slower and faster cholesterol exchange). The rate of cholesterol exchange between the pools was enhanced and the rate of total clearance of cholesterol from the system (metabolic turnover rate of cholesterol, i.e. the rate of degradation and excretion of cholesterol from the organism) was enhanced by 50 %. The oyster mushroom diet effectively prevented the progress of hypercholesterolaemia (decrease by 38 %) and cholesterol accumulation in liver
(decrease by 25 %) that were induced by the cholesterol diet.
A highly significant negative correlation (r= -0.981, p< 0.001) between the amount of oyster mushroom (Pleurotus ostreatus) in the diet and cholesterol levels in the serum has been found in male Wistar rats fed shortly after weaning by a a diet with 0.3 % cholesterol. The addition of 1.0, 2.5 and 5.0 % of oyster mushroom to the diet reduced the levels of serum cholesterol by 11, 31 and 46 %, respectively. The diet containing 5 % of oyster mushroom suppressed cholesterol accumulation in the liver and increased the fraction of cholesterol carried by high-density lipoproteins.
The aim of this work was to study the effect of the daily ingestion of a purified anthocyanin extract from red grape skin on rat serum antioxidant capacity (ORAC) and its safety for the intestinal epithelium. The study was carried out in rats orally administered with the extract for 10 days in either normal physiological conditions or exposed to a pro-oxidant chemical (CCl4). The oral administration of the extract significantly (P<0.05) enhanced the ORAC va lue of the deproteinised serum of about 50 % after 10 days of ingestion. Anthocyanin administration was also able to reverse completely the decrease in the serum ORAC activity induced by the CCl4 treatment. Experiments with Ussing chamber mounted intestine allowed to exclude any toxicity of the extract for the intestinal epithelium. In conclusion, our results demonstrate that the purified anthocyanin extract from red grape skin e nhances the total antioxidant capacity of the serum in either normal physiological condition or during oxidative stress induction, revealing a protective role against the decrease in the serum antioxidant capacity induced by a pro-oxidant compound., M. Iodine is essential in the biosynthesis of thyroid hormones that affect metabolic processes in the organism from the prenatal state to the elderly. The immediate indicator of iodine intake is the concentration of iodine in urine, but the indicator of iodine intake in the longer term of several months is thyroglobulin (Tg). Tg negatively correlated with increasing intake of iodine in population that do not suffer from thyroid disease, while a more than adequate to excessive iodine intake leads to an increase in Tg. The dependence of Tg on iodine can be described by a U-shaped curve. Thyroglobulin in serum is elevated in thyroid disease mainly in hyperthyroidism (diagnosis E05 of WHO ICD-10 codes) and in goiter (diagnosis E04 of WHO ICD-10 codes). Tg values decrease below 20 µg/l after effective treatment of patients with thyroid disease. The effects of octadecylamide of alginic acid (amidated alginate) and tetrahydrolipstatin on serum and hepatic cholesterol, and the faecal output of fat and sterols, were investigated in rats. Amidated alginate is a sorbent of lipids, tetrahydrolipstatin is an inhibitor of pancreatic lipase. Rats were fed diets containing cholesterol and palm fat at 10 and 70 g/kg, respectively. Palm fat was provided by coconut meal. Amidated alginate at 40 g/kg diet significantly decreased serum total cholesterol, low-density lipoprotein and hepatic cholesterol, and hepatic lipids and increased the faecal output of fat and coprostanol. Tetrahydrolipstatin at 300 mg/kg diet significantly decreased low-density lipoprotein cholesterol and hepatic lipids and increased the faecal output of fat. The intake of feed was not significantly influenced; however, the weight gains in rats fed amidated alginate were lower than in rats of the control group. Both amidated alginate and tetrahydrolipstatin modified the fatty acid profile in excreta lipids. Concentrations of saturated fatty acids were decreased and those of unsaturated fatty acids increased. Despite different modes of action, amidated alginate and tetrahydrolipstatin were equally efficient in removing the dietary fat from the body.
