Isolation and characterisation of Plasmodium falciparum (Welch, 1897) soluble antigens from infected patient plasma, Western blotting, thermal stability and ELISA assays using hyperimmune IgG-antimalaria antibodies was the main objective of this work. A circulating antigen of approximately Mr 33-35 kDa with good specificity and antigenicity, in the plasma of malarial patients was shown. Heating at 100°C did not destroy its antigenicity. When fractions highly enriched in the 33-35 kDa proteins were used in ELISAs, a seroreactivity in plasma obtained from primary-infected individuals was found. Controls from normal patients were always negative. The antigenic characteristics suggest that it may be included within the group of new described Plasmodium soluble antigens.
Films of aliphatic polyurethane-silica nanocomposite containing up to 27.3 mol. % of silica were obtained by the parallel synthesis of both composite components in one common process. This method permits to obtain the nanocomposite materials with the uniform distribution of nanoparticles in the polymer matrix volume. The rise of the silica concentration in the nanocomposite film was shown to lead from one hand to the progressive fall of both the Young’s modulus and yield stress and from another hand to the rise of the ultimate deformation and thermal expansion coefficient values. The effect of heat stress (35 to 50 °C) on photosynthesis was investigated in heat tolerant (N 22) and heat sensitive (IR 8) cultivars of rice {Oryza sativa L.). The net photosynthetic rate showed greater thermal stability in N 22 than in IR 8. The relative dechne of the rate of whole chain electron transport and photosystem 2 (PS2) activity was more pronounced in IR 8 than N 22. In both cultivars photosystem (PSI) activity was stimulated by thermal treatment. Chlorophyll (Chl) a fluorescence transient arising ffom PS2 showed inhibition in both cultivars at 45 and 50 °C. Maximum fluorescence decreased more in IR 8 than in N 22 by high temperature treatment.
The peptide surfactants are amphiphilic peptides which have a hydrophobic tail and a hydrophilic head, and have been reported to stabilize and protect some membrane proteins more effectively than conventional surfactants. The effects of a class of peptide surfactants on the structure and thermal stability of the photosynthetic membrane protein lightharvesting complex II (LHCII) in aqueous media have been investigated. After treatment with the cationic peptide surfactants A6K, V6K2, I5K2 and I5R2, the absorption at 436 nm and 470 nm decreased and the absorption at 500-510 nm and 684-690 nm increased. Moreover, the circular dichroism (CD) signal intensity in the Soret region also decreased significantly, indicating the conformation of some chlorophyll (Chl) a, Chl b, and the xanthophyll molecules distorted upon cationic peptide surfactants treatment. The anionic peptide surfactants A6D and V6D2 had no obvious effect on the absorption and CD spectra. Except for A6D, these peptides all decreased the thermal stability of LHCII, indicating that these peptides may reconstitute protein into a less stable conformation.
