This paper proposes an offline gradient method with smoothing L1/2 regularization for learning and pruning of the pi-sigma neural networks (PSNNs). The original L1/2 regularization term is not smooth at the origin, since it involves the absolute value function. This causes oscillation in the computation and difficulty in the convergence analysis. In this paper, we propose to use a smooth function to replace and approximate the absolute value function, ending up with a smoothing L1/2 regularization method for PSNN. Numerical simulations show that the smoothing L1/2 regularization method eliminates the oscillation in computation and achieves better learning accuracy. We are also able to prove a convergence theorem for the proposed learning method.
We investigated the effects of telmisartan, the blocker of angiotensin II receptor 1, on the regulation of systolic blood pressure (SBP) and oxidative stress through endothelial nitric oxide (NO) release in spontaneously hypertensive rats (SHRs). SHRs randomly received placebo, oral feeding of telmisartan (5 mg/kg or 10 mg/kg) every day and Wistar-Kyoto rats (WKYs) served as normotensive control. The SBP of rat was measured before and weekly thereafter. After a total of 8-week treatment, rats were killed for experimental measurements. Parameters that subject to measurements in isolated aorta endothelial cells include: NO concentration, protein expression levels of angiotensin II receptor 1, nitrotyrosine, 8-isoprostane, SOD, PI3K, Akt, AMPK and eNOS. In addition, L-NMMA, a general inhibitor of nitric oxide synthase, was also applied to test the inhibition of NO concentration. We found that SBPs were significantly lower in telmisartan therapy group than in placebo treated hypertensive rats and WKYs (p<0.05). The NO concentration was significantly higher in telmisartan-treated group with increased activity of the PI3K/Akt pathway and activated eNOS signaling. Blockade of Akt activity reversed such effects. Activation of AMPK also contributed to the phosphorylation of eNOS. L-NMMA treatment reduced less NO concentration in SHR rats than the telmisartan co-treated groups. Oxidative stress in SHRs was also attenuated by telmisartan administration, shown by reduced formation of nitrotyrosine, 8-isoprostane, and recovered SOD protein level. In the past decades, it has become clear that superoxide radical (O2 .-) can be generated from photosystem II (PSII) during photosynthesis. Depending on the extent of its accumulation, O2 .- plays an important role in plant physiology and pathology. The photoinhibition/repair cycle is a typical process in PSII which is mainly responsible for the survival of plants under the photoinihibition condition. It is therefore of significant importance to determine O2 .- production in this cycle, and then explore how O2 .- is controlled by PSII within a normal physiological level. With this in mind, we herein investigate the variation of the O2 .- levels in PSII under Mn-depleted and photoactivated conditions mimicking the photoinhibition/repair cycle in vitro. The effect of intrinsic SOD-like component on the O2 .- levels was also studied. Results show that PSII has the ability to regulate the O2 .- levels in these two processes by simultaneously modulating the O2 .- generation activity and intrinsic SOD-like activity. Ca2+ is an important factor mediating many biotic and abiotic stress responses in plants. In this study, we measured the chlorophyll (Chl) fluorescence of transgenic rice with increased or decreased expression of a calcium-sensing receptor (OsCaS) gene during water deficit caused by polyethylene glycol to prove our hypothesis that increased Ca2+ in combination with increased OsCaS could enhance the drought resistance of transgenic rice. Transcript abundance (evaluated by RT-PCR) was significantly lower in OsCaS antisense line 766 (AS766) than that in the wild type, while the overexpression line 777 (O777) showed four times higher amount than that in the wild type. Chl fluorescence showed that the photochemical quantum yield of PSII in the light increased due to addition of Ca2+ in the O777, but dropped in the AS766. Nonphotochemical quenching increased under stress in both transgenic lines and in the wild type, but less in the O777. Nonregulatory quantum yield of energy dissipation showed no significant change under drought stress. Photochemical quenching was significantly higher in the O777 than those in the AS766 and in the wild type after the Ca2+ treatment. In the absence of stress, the electron transport rate (ETR) was significantly higher in the O777 than in both the AS766 and the wild type. In contrast, the ETR of the wild type and both transgenic lines decreased under drought stress, while the effect of polyethylene glycol was partially alleviated by Ca2+ addition in the O777. In summary, excitation energy conversion and dissipation by PSII were regulated by Ca2+ in the O777. Glutamate is a well-characterized excitatory neurotransmitter in the central nervous system (CNS). Recently, glutamate receptors (GluRs) were also found in peripheral tissues, including the heart. However, the function of GluRs in peripheral organs remains poorly understood. In the present study, we found that N-methyl-D-aspartate (NMDA) could increase intracellular calcium ([Ca2+]i) level in a dose-dependent manner in cultured neonatal rat cardiomyocytes. NMDA at 10-4 M increased the levels of reactive oxygen species (ROS), cytosolic cytochrome c (cyto c), and 17-kDa caspase-3, but depolarized mitochondrial membrane potential, leading to cardiomyocyte apoptosis. In addition, NMDA treatment induced an increase in ba x mRNA but a decrease in bcl-2 mRNA expression in the cardiomyocytes. The above effects of NMDA were bloc ked by the NMDA receptor antagonist (+)-5-methyl- 10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-imine hydrogen maleate (MK-801), and by ROS scavengers glutathione (GSH) and N-acetylcystein (NAC). These results suggest that stimulation of NMDA receptor in the cardiomyocyte may lead to apoptosis via a Ca2+, ROS, and caspase-3 mediated pathway. In the central nervous system (CNS), monocarboxylate transporter 1 (MCT1) is expressed in astrocytes and endothelial cells but also in oligodendroglia. Oligodendroglia support neurons and axons through lactate transportation by MCT1. Limited information is available on the MCT1 expression changes in candidate cells in the developing rat brain, especially in corpus callosum which is the most vulnerable area in demyelinating diseases. In the present study, we investigated the expression pattern of MCT1 during postnatal development in the rat corpus callosum using immunofluorescene staining, Western blotting analysis and RT-PCR. We reported that MCT1 gene and protein were consistently expressed in the rat corpus callosum from birth to adult. MCT1/CNPase and MCT1/GFAP immunofluorescence staining demonstrated that most of MCT1 positive cells were co-labeled with cyclic nucleotide 3′ phosphodiesterase (CNPase) in rat corpus callosum from P7 to adult, whereas MCT1+/GFAP+ cells preserve the dominate position before P7. Moreover, there were significant associations between the expression of MCT1 protein and the expression of myelin basic protein (MBP) (correlation coefficient: r=0.962, P=0.009) from P7 to adult. Similarly, the MCT1 mRNA expression was also significantly associated with MBP mRNA expression (r=0.976, P=0.005). Our results are proposing that in the developing brain white matter, MCT1 is predominately expressed in oligodendrocyte though it mainly expressed in astrocyte in early postnatal, which indicate that MCT1 may involve in the oligodendrocyte development and myelination., F. We studied the potassium channel in the basolateral membrane of the rat proximal convoluted tubule as affected by cyclosporine A. Proximal convoluted tubules were dissected from the rat kidney under a stereoscopic microscope, without a preliminary enzyme treatment. The standard configuration for single-channel tight seal patch-clamp technique
was used to record channel currents. A small conductance, stretch-sensitive potassium channel could be observed spontaneously in most of the cell-attached patches as the gigaohm seal was formed. In the inside-out configuration, channel activity was diminished. The K+
channel appeared to be an inward rectifier. The limiting inward slope
conductance was 28.3±1.7 pS (Vp was between 40 mV and 80 mV, n=6) and the outward chord conductance was 5.6±0.3 pS (Vp was between -40 and -60 mV, n=5). The open dwell time constants of the potassium channel were 0.524 ms and 5.087 ms, while the closed dwell time constants were 1.029 ms and 16.500 ms. The opening probability of the
channel decreased when the extracellular fluid was acidified. Cyclosporine A had no significant effect on the potassium channel of the proximal tubular cell in the basolateral membrane at concentrations of 10 and 50 μg/ml, while at 100 μg/ml, it decreased the opening probability.
Plasma endothelin-1 (ET-1) levels are elevated in spinal cord injury (SCI), and ET-1 may be involved in the pathophysiology of this condition. However, its effects on contractile function of the heart of SCI rats are still unknown. To define more clearly the possiblel role of ET-1 following SCI, we investigated the effect of ET-1 on the contraction, calcium transients and L-type calcium current (ICa,L) in the cardiomyocytes of control and SCI rats. Sixteen Sprague-Dawley male rats aged 80-100 days and weighing 250-350 g were randomized into control and SCI groups. Fourteen days following compression injury to the spinal cord, effects of ET-1 on the contraction, calcium transients and ICa,L were studied in the cardiomyocytes of control and SCI rats by the technique of simultaneous measurement of intracellular Ca2+ and contraction and by whole-cell configuration of the patch-clamp technique. In myocytes from control rats, ET-1 significantly increased contraction, the magnitude of Ca2+ transients and the peak amplitude of ICa,L. However, ET-1 had little effect on the amplitude of contraction, calcium transients and ICa,L in myocytes from SCI rats. These results suggest that the positive inotropic effects of ET-1 on control myocardial contraction may be altered in pathological states such as SCI., Y.-F. Guo ... Leaves under stressful conditions usually show downregulated maximum quantum efficiency of photosystem II [inferred from variable to maximum chlorophyll (Chl) a fluorescence (Fv/Fm), usually lower than 0.8], indicating photoinhibition. The usual method to evaluate the degree of photoinhibition in winter red leaves is generally by measuring the Fv/Fm on the red adaxial surface. Two phenotypes of overwintering Buxus microphylla ‘Wintergreen’ red leaves, with different measuring site and leaf thickness, were investigated in order to elucidate how red pigments in the outer leaf layer affected the Chl a fluorescence (Fv/Fm) and photochemical reflectance index. Our results showed that the Fv/Fm measured on leaves with the same red surface, but different leaf thickness, exhibited a slightly lower value in half leaf (separated upper and lower layers of leaves by removing the leaf edge similarly as affected by winter freezing and thawing) than that in the intact leaf (without removing the leaf edge), and the Fv/Fm measured on the red surface was significantly lower than that on the inner or backlighted green surface of the same thickness. Our results suggest that the usual measurement of Fv/Fm on red adaxial surface overestimates the actual degree of photoinhibition compared with that of the whole leaf in the winter., J. G. Peng, X.
