Photoinhibition of photosynthesis was investigated in Vitis berlandieri and Vitis rupestris leaves under field conditions at different sampling time in a day. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the photochemical efficiency of PS2, Fv/Fm, markedly declined, F0 increased significantly in leaves of V. berlandieri, while F0 did not increase in V. rupestris leaves. Isolated thylakoids of leaves of V. berlandieri showed significant inhibition of whole chain and PS2 activities at midday. A smaller inhibition was observed for V. rupestris. Later, the leaves reached maximum PS2 efficiencies similar to those observed early in the morning during sampling at evening. The artificial exogenous electron donor Mn2+ failed to restore PS2 activity in both species, while DPC and NH2OH significantly restored PS2 activity in V. rupestris midday leaf samples. Quantification of the PS2 reaction centre protein D1 and 33 kDa protein of water splitting complex following midday exposure of leaves showed pronounced differences between V. berlandieri and V. rupestris leaves. The marked loss of PS2 activity noticed in midday samples was mainly due to the marked loss of D1 protein in V. berlandieri while in V. rupestris it was the 33 kDa protein. and M. Bertamini, N. Photoinhibition under irradiance of 2 000 µmol m-2 s-1 (HI) was studied in detached control (C) and water deficit (WD) leaves of grapevine (Vitis vinifera L.) plants. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem (PS) 2, Fv/Fm, marginally declined under HI in WD-leaves without significant increase of F0. In contrast, Fv/Fm ratio declined markedly with significant increase of F0 in C-leaves. In isolated thylakoids, the rate of whole chain and PS2 activity under HI were more decreased in C-than WD-leaves. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in both C-and WD-leaves. Thus HI operates at the acceptor side of PS2 in both leaf types. Quantification of the PS2 reaction centre protein D1 following HI exposure of leaves showed pronounced differences between C-and WD-leaves. Exposure of plants to irradiation, in excess to saturate photosynthesis, leads to reduction in photosynthetic capacity without any change in bulk pigment content. This effect is known as photoinhibition. Photoinhibition is followed by destruction of carotenoids (Cars), bleaching of chlorophylls (Chls), and increased lipid peroxidation due to formation of reactive oxygen species if the excess irradiance exposure continues. Photoinhibition of photosystem 2 (PS2) in vivo is often a photoprotective strategy rather than a damaging process. For sustainable maintenance of chloroplast function under high irradiance, the plants develop various photoprotective strategies. Cars perform essential photoprotective roles in chloroplasts by quenching the triplet Chl and scavenging singlet oxygen and other reactive oxygen species. Recently photoprotective role of xanthophylls (zeaxanthin) for dissipation of excess excitation energy under irradiance stress has been emphasised. The inter-conversion of violaxanthin (Vx) into zeaxanthin (Zx) in the light-harvesting complexes (LHC) serves to regulate photon harvesting and subsequent energy dissipation. De-epoxidation of Vx to Zx leads to changes in structure and properties of these xanthophylls which brings about significant structural changes in the LHC complex. This ultimately results in (1) direct quenching of Chl fluorescence by singlet-singlet energy transfer from Chl to Zx, (2) trans-thylakoid membrane mediated, ΔpH-dependent indirect quenching of Chl fluorescence. Apart from these, other processes such as early light-inducible proteins, D1 turnover, and several enzymatic defence mechanisms, operate in the chloroplasts, either for tolerance or to neutralise the harmful effect of high irradiance. and N. To investigate the photoprotection of energy dissipation and water-water cycle, a C3 euhalophytic herb, Suaeda salsa L., was exposed either to chilling temperature (4°C) accompanied by moderate irradiance (600 μmol m-2 s-1) (CM) and/or to chilling temperature (4°C) accompanied by low irradiance (100 μmol m-2 s-1) (CL). During chilling stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and the oxidizable P700 decreased in S. salsa leaves either under CM or CL, which indicated the severe photoinhibition. Relative to Fv/Fm, the oxidizable P700 decreased markedly under CL, which indicated that PSI was more sensitive to CL treatment than PSII. Initial fluorescence, number of closed PSII centers, and nonphotochemical quenching increased under CM, but more markedly under CL in S. salsa leaves. Activity of superoxide dismutase and ascorbate peroxidase was higher under CM than that under CL. The production of reactive oxygen species (ROS) decreased first and then increased under both treatments, but the content of O2.- and H2O2 was higher under CL than that under CM after 12 h of chilling stress. These results suggested that photoinhibition in S. salsa might be related to the accumulation of reactive oxygen species (ROS) induced by excess energy. With untransformed rice cv. Kitaake as control, the characteristics of carbon assimilation and photoprotection of a transgenic rice line over-expressing maize phosphoenolpyruvate carboxylase (PEPC) were investigated. The PEPC activity in untransformed rice was low, but the activity was stimulated under high irradiance or photoinhibitory condition. PEPC in untransformed rice contributed by about 5-10 % to photosynthesis, as shown by the application of the specific inhibitor 3,3-dichloro-2-(dihydroxyphosphinoylmethyl)propenoate (DCDP). When maize PEPC gene was introduced into rice, transgenic rice expressed high amount of maize PEPC protein and had high PEPC activity. Simultaneously, the activity of carbonic anhydrase (CA) transporting CO2 increased significantly. Thus the photosynthetic capacity increased greatly (50 %) under high CO2 supply. In CO2-free air, CO2 release in the leaf was less. In addition, PEPC transgenic rice was more tolerant to photoinhibition. Treating by NaF, an inhibitor of phosphatase, showed that in transgenic rice more phosphorylated light-harvesting chlorophyll a/b-binding complexes (LHC) moved to photosystem 1 (PS1) protecting thus PS2 from photo-damage. Simultaneously, the introduction of maize PEPC gene could activate or induce activities of the key enzymes scavenging active oxygen, such as superoxide dismutase (SOD) and peroxidase (POD). Diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem 2 (PS2) as well as H2O2 content were analyzed in Reaumuria soongorica (Pall.) Maxim., a perennial semi-shrub. The rate of photorespiration was estimated by combined measurement of gas exchange and Chl fluorescence. The rate of photorespiration increased with the increasing drought stress (DS). The ratio of carboxylation electron flow to oxygenation electron flow (Jc/Jo) and the maximal photochemical efficiency of PS2 (variable to maximum fluorescence ratio, Fv/Fm) decreased with the increasing DS. Fv/Fm in isonicotinic acid hydrazide (INH)-sprayed plants was lower than that in normal plants under moderate DS, but no significant difference was observed under severe DS. H2O2 content in INH-sprayed plants was significantly lower than that in normal plants under severe DS. Taken together, photorespiration in R. soongorica consumed excess electrons and protected photosynthetic apparatus under moderate DS, whereas it accelerated H2O2 accumulation markedly and induced the leaf abscission under severe DS. and J. Four grapevine cultivars, i.e. Cabernet Sauvignon (a member of the Western Europe cultivar group), Rizamat (a member of the East cultivar group), Red Double Taste (a hybridized cultivar from Vitis vinifera L. and V. labrusca L.), and 1103Paulsen (a hybridized rootstock), were treated by three severity orders of drought stress for 25 d. Then net photosynthetic rate (PN), maximal photochemical efficiency (Fv/Fm), actual photochemical efficiency (ΦPS2) of photosystem 2, total electron transport rate (JT), and electron transport flows used in carboxylation (JC) and in oxygenation (JO) reactions catalysed by ribulose-1,5-bisphosphate carboxylase/oxygenase were determined. PN was determined again after re-watering for 2 d by gas exchange measurement. Along with the increase in severity of drought stress, PN, Fv/Fm, ΦPS2, JT, and JC in all four cultivars decreased. The range of decrease differed among cultivars. JO expressed various trends from cultivar to cultivar. In Rizamat that received slight and moderate drought stress, PN evidently decreased, but JO markedly increased, thus maintaining high values of JT and ΦPS2. Prior to the moderate drought stress, the Fv/Fm was high in Rizamat, indicating that the photodamage had not happened ahead of the moderate drought stress given. Under the severe drought stress, the photorespiration rate in Rizamat decreased by 70 %, and JT, ΦPS2, and Fv/Fm also dropped to very low values, i.e. the photodamage of photosynthetic apparatus has taken place. This suggested that the photorespiration has consumed the excessive assimilatory power and the photo-protective function of photorespiration is very important for Rizamat. When Cabernet Sauvignon grew under drought stress, its JO decreased in a small range, thus maintaining higher values of JC, JT, ΦPS2, and Fv/Fm; hence no serious photodamage occurred. Despite of the fact that PN of cv. Red Double Taste decreased markedly under the slight drought stress, JO still increased under the severe drought stress. This suggests that photorespiration is important in photoprotection under drought stress. JO in cv. 1103Paulsen markedly decreased under slight stress. Accordingly, PN, Fv/Fm, ΦPS2, JT, and JC decreased to extremely low values. Thus photorespiration effectively protects the photosynthetic apparatus from photo-damage under drought, assists in maintaining a relatively high ΦPS2, and helps PN to be rapidly recovered after re-watering. and X. The influence of viral infection caused by two different potyviruses, Potato virus Y (PVY) and Potato virus A (PVA) on plant metabolism and photosynthetic apparatus of Nicotiana tabacum L. cv. Samsun and cv. Petit Havana SR1 was studied. The main stress was focused on the activities of phosphoenolpyruvate carboxylase (PEPC), NADP-malic enzyme (NADP-ME), and pyruvate phosphate dikinase (PPDK). The analysis of the presence of viral proteins, enzyme activities, and different photosynthetic parameters showed the time dependent progress of viral infection and NADP-ME and PEPC activities. PVY caused significant response, while PVA affected both tobacco cultivars only slightly. Viral infection, namely PVY, affected more negatively photosynthetic apparatus of cv. Available water stored in deep soil layers could increase the photosynthetic capacity of cotton. It was hypothesized that the photosynthesis of cotton would be enhanced by changing the fertilizer application depth under different deep-layer water conditions. We examined two deep-layer water levels, i.e.,
well-watered (W80) and not watered (W0), combined with surface application (F10) and deep application (F30) of basal fertilizer. Compared to W0, W80 resulted in increased leaf area (LA), photosynthetic pigment contents, maximal PSII efficiency (Fv/Fm), effective quantum yield of PSII (YII) and PSI (YI), electron transport rate of PSII (ETRII) and PSI (ETRI). W80 also increased the aboveground and root dry mass by 39 and 0.6%, respectively, and decreased the root/shoot ratio by 40-73%. Under the W0 condition, higher values of Fv/Fm, YII, YI, ETRII, and ETRI were measured for F10 compared to F30 after 69 d from emergence. Under the W80 condition, cotton plants with F10 showed higher LA, Fv/Fm, YII, YI, ETRII, and ETRI, but there were no significant differences in the photosynthetic pigments compared to F30. Our results suggest that sufficient water in deeper soil layers and the surface application of basal fertilizer could increase photosynthetic activity and efficiency, which promoted aboveground dry mass accumulation and partitioning towards reproductive organs., Z. Increase in both atmospheric CO2 concentration [CO2] and associated warming are likely to alter Earths' carbon balance and photosynthetic carbon fixation of dominant plant species in a given biome. An experiment was conducted in sunlit, controlled environment chambers to determine effects of atmospheric [CO2] and temperature on net photosynthetic rate (P N) and fluorescence (F) in response to internal CO2 concentration (C i) and photosynthetically active radiation (PAR) of the C4 species, big bluestem (Andropogon gerardii Vitman). Ten treatments were comprised of two [CO2] of 360 (ambient, AC) and 720 (elevated, EC) µmol mol-1 and five day/night temperature of 20/12, 25/17, 30/22, 35/27 and 40/32 °C. Treatments were imposed from 15 d after sowing (DAS) through 130 DAS. Both F-P N/Ci and F-P N/PAR response curves were measured on top most fully expanded leaves between 55 and 75 DAS. Plants grown in EC exhibited significantly higher CO2-saturated net photosynthesis (Psat), phosphoenolpyruvate carboxylase (PEPC) efficiency, and electron transport rate (ETR). At a given [CO2], increase in temperature increased P sat, PEPC efficiency, and ETR. Plants grown at EC did not differ for dark respiration rate (RD), but had significantly higher maximum photosynthesis (P max) than plants grown in AC. Increase in temperature increased Pmax, RD, and ETR, irrespective of the [CO2]. The ability of PEPC, ribulose-1,5-bisphosphate carboxylase/oxygenase, and photosystem components, derived from response curves to tolerate higher temperatures (>35 °C), particularly under EC, indicates the ability of C4 species to sustain photosynthetic capacity in future climates. and V.
