The maize weevil, Sitophilus zeamais (Motschulsky), is a major pest of stored grain kernels. Irradiation is an established technique for controlling insects in stored grain and is a major stress factor affecting these insects. Since heat shock protein (hsp) genes respond to this stress, we proposed that hsps may be associated with irradiated stress tolerance in S. zeamais. The responses of the maize weevil to exposure to ultraviolet-C (UV-C) and microwave irradiation were assessed at four developmental stages: egg, larva, pupa and adult. The results revealed that exposure to UV-C (254 nm, < 8 h) did not affect the survival of the maize weevils; however, Szhsp70, Szhsc70 and Szhsp90 mRNA levels significantly increased during the first 1 h of UV-C exposure. The median lethal time (LT50) of exposure to microwave irradiation indicated that the adult stage was more tolerant of microwave irradiation than the other developmental stages. Microwave irradiation enhanced the expression of the three hsps, but the intensity of up-regulation differed among the three genes, with Szhsp70 the most highly up-regulated. Our experiments revealed that UV-C and microwave irradiation influenced the expression profile of hsp genes in S. zeamais. Traps baited with plant volatiles and/or bark beetle pheromones have been used to survey for exotic and potentially invasive bark and wood-boring beetles in North America since the mid-1990s. Recent discoveries of sex and aggregation pheromones in the Cerambycidae offer means of improving detection rates of longhorn beetles, but little is known of their effects on detection of bark and ambrosia beetles in the subfamily Scolytinae. Our objectives were to determine the efficacy of host volatile trap lures for survey and detection of Scolytinae species and whether that efficacy was affected by the addition of longhorn beetle pheromones. More than 12,000 specimens and 36 species of Scolytinae were collected in two field trapping bioassays conducted in the Russian Far East in 2009 and 2010. The lure combination of spruce blend (a blend of racemic α-pinene, (-) β-pinene, (+)-3-carene, (+)-limonene, and α-terpinolene) and ethanol significantly increased detection rates and mean catches of Hylastes brunneus Erichson, Hylastes obscurus Chapuis, Ips typographus (L.), and Dryocoetes striatus Eggers compared with unbaited traps. The addition of the longhorn beetle pheromones, E-fuscumol, or E-fuscumol acetate, to traps baited with spruce blend and ethanol, slightly reduced mean catches of D. striatus but otherwise did not affect catch of any Scolytine species. Baiting traps with ethanol significantly increased mean catches of Anisandrus apicalis (Blandford), Anisandrus dispar (Fabr.), Anisandrus maiche (Kurenzov), Xyleborinus attenuatus (Blandford), Xyleborinus saxesenii (Ratzeburg), Xylosandrus germanus (Blandford), Scolytoplatypus tycon Blandford, and Trypodendron lineatum (Olivier). By themselves, the longhorn beetle pheromones, racemic hydroxyhexan-2-one and racemic hydroxyoctan-2-one, were not attractive to any Scolytine species. However, when added to ethanol-baited traps, hydroxyhexan-2-one lures significantly increased mean catch of S. tycon, hydroxyoctan-2-one lures significantly reduced mean catches of A. maiche and X. attenuatus, and lures of either hydroxyketone significantly reduced mean catch of T. lineatum. The lure treatments that detected the greatest number of species per sampling effort were spruce blend plus ethanol in 2009 (16 Scolytinae species and 13 species of Cerambycidae combined in an eight-trap sample) and hydroxyhexan-2-one plus ethanol in 2010 (20 Scolytinae species and 7 species of Cerambycidae combined in an eight-trap sample). A novel method was used to study dispersal in the spruce bark beetle, Ips typographus (L.), under epidemic conditions (rapidly increasing population density) in the Šumava National Park. Infested spruce logs were coated with a fine fluorescent powder and the passively marked emerging beetles were captured in pheromone baited traps located at various distances from these logs. The number of marked beetles captured decreased exponentially with increasing distance from the release point. The sex ratio of the bark beetles was more female biased the further they were recaptured from the logs, being 57% and 60% at distances of up to 50 and 100 m, respectively. The maximum distance flown by a marked beetle recorded in this experiment was 1094 m. A model fitted to the data on dispersal indicates that 10% of the spruce bark beetles dispersed over distances of 55 m and 4 m in spring (overwintered parental generation) and summer (first filial generation), respectively. Coccinellids are effective predators and a key component of the predator guild in rice ecosystems. In order to enhance their efficacy, a study was undertaken to assess the seasonal movement of coccinellids into rice fields and the role of the surrounding flora on their colonization. The seasonal abundance of coccinellids and their prey was recorded on the rice crop and the surrounding flora at fortnightly intervals from 2012 to 2015. Coccinellid prey range was assessed using PAGE electrophoresis. The herbivorous insects associated with weeds were Aphis gossypii Glover, Aphis craccivora (Koch), Cicadulina bipunctata (Melichar), Schizaphis graminum (Rondani), Sitobion sp., Thaia oryzivora Ghauri and Zygina maculifrons Matsumura. Of the species of coccinellids recorded in rice fields, Harmonia octomaculata (Fabricius), Micraspis discolor (F.), Propylea dissecta (Mulsant), Coccinella transversalis Fabricius, Cheilomenes sexmaculata (Fabricius), Scymnus nubilus Mulsant and Brumoides suturalis (Fabricius) were also recorded on weeds. The esterase profiles indicated that the leafhoppers and aphids on the weeds were the prey of the coccinellids before they colonized the rice fields. The coccinellids recorded on the weeds showed bands corresponding to the insects present on the weeds. Beetles collected from rice fields had different bands, some of which corresponded to the green leafhopper (GLH) Nephotettix virescens Distant, the brown planthopper (BPH) Nilaparvata lugens Stal and white backed planthopper (WBPH) Sogatella furcifera Hovarth infesting rice. In addition, some bands corresponded to hoppers and aphids that were present on the surrounding flora. Conifer bark beetles are well known to be associated with fungal complexes, which consist of pathogenic ophiostomatoid fungi as well as obligate saprotroph species. However, there is little information on fungi associated with Ips acuminatus in central and eastern Europe. The aim of the study was to investigate the composition of the fungal communities associated with the pine engraver beetle, I. acuminatus, in the forest-steppe zone in Ukraine and to evaluate the pathogenicity of six associated ophiostomatoid species by inoculating three-year-old Scots pine seedlings with these fungi. In total, 384 adult beetles were collected from under the bark of declining and dead Scots pine trees at two different sites. Fungal culturing from 192 beetles resulted in 447 cultures and direct sequencing of ITS rRNA from 192 beetles in 496 high-quality sequences. Identification of the above revealed that the overall fungal community was composed of 60 species. Among these, the most common were Entomocorticium sp. (24.5%), Diplodia pinea (24.0%), Ophiostoma ips (16.7%), Sydowia polyspora (15.1%), Graphilbum cf rectangulosporium (15.1%), Ophiostoma minus (13.8%) and Cladosporium pini-ponderosae (13.0%). Pathogenicity tests were done using six species of ophiostomatoid fungi, which were inoculated into Scots pine seedlings. All ophiostomatoid fungi tested successfully infected seedlings of Scots pine with varying degrees of virulence. Ophiostoma minus was the only fungus that caused dieback in inoculated seedlings. It is concluded that I. acuminatus vectors a species-rich fungal community including pathogens such as D. pinea and O. minus. The fungal community reported in the present study is different from that reported in other regions of Europe. In the last two decades a huge amount of research has focused on the invasive harlequin ladybird, Harmonia axyridis, particularly on potential or actual deleterious effects that have arisen after it has colonised new regions. A focus of this work has been real or anticipated declines in native ladybird abundance since the introduction of H. axyridis, for which it is deemed responsible. Scientists have generally painted a very bleak picture of the effects of H. axyridis on native species: in this paper I argue that the picture painted is often too bleak. I use the case of the 2-spot ladybird, Adalia bipunctata, the species most often invoked as threatened by H. axyridis, to illustrate my point. While there is little question that H. axyridis has led to a decline in A. bipunctata populations in Europe, it seems likely that prior to the invasive ladybird's arrival A. bipunctata occurred in artificially high numbers in the urban environments in which it was typically studied. Pollution in towns and cities led to enhanced numbers of prey aphids on plants there which initially favoured A. bipunctata, and later H. axyridis. Thus one species, A. bipunctata, that has benefitted from an association with humans has been replaced by another, H. axyridis, just as brown rats replaced black rats in Europe and North America. Viewed with a longer perspective, A. bipunctata has more likely declined back to pre-industrial levels: the artificially high level from which it has declined recently was not a 'natural' one, and thus its decline from this level does not imply that it is now threatened or endangered. More broadly, we need a wider perspective, encompassing other ladybirds, longer timeframes and better comparisons with other (non-ladybird) invasive species to more clearly assess whether H. axyridis really poses as much of a threat as is often proposed., John J. We deployed branch traps in an ash (Fraxinus) plantation to investigate how Agrilus planipennis behavior is associated with Fraxinus pennsylvanica condition and dispersal patterns. Data were collected from traps with or without the presence of beetle visual decoys, and from a yearly survey of exit holes. The traps were placed on trees that were either clearly declining, with most foliage arising from epicormic sprouting, or on apparently healthy trees, with little evidence of damage or decline. We calculated correlations of exit holes among neighboring tree rings and also between exit holes and male trap captures. The damaged trees the traps were hung upon had more cumulative exit holes observed than the corresponding healthy trees. However, there was otherwise no evidence that the experiment was biased by differences in exit hole patterns of the surrounding trees. Male captures were greater on decoy-baited traps than controls and this decoy effect was most clearly apparent late in the season when traps were placed on healthy trees. There were also patterns of correlations between male captures and exit hole numbers that may be indicative of short-range mate finding-and dispersal behaviors. Female captures were sparser, but were positively affected by decoys on healthy and declining trees early in the season. Thus, the results suggest that the placement of such traps on healthier trees will maximize detection, and the branch traps also show promise for further use in dispersal studies., Michael J. Urban gardens can harbour a high diversity of insects, which are critically important components of urban ecosystems. In this work, we investigate the richness and diversity of a major taxon of economic and ecological importance, the aphids (Hemiptera: Aphididae), and their main insect predators, the hoverflies (Diptera: Syrphidae) and ladybirds (Coleoptera: Coccinellidae), in urban gardens. We examined how variation in environmental factors associated with urbanisation (garden host plant abundance, garden plant richness, garden size, proportion of impervious surfaces in the surrounding area) directly and indirectly (via prey and predator abundance) influence the local diversity and abundance of aphids, ladybirds, and hoverflies. Sixty-seven domestic gardens located in southern England were surveyed during the peak period of aphid abundance, and the numbers and identity of aphids and their predators were recorded. We observed 45 aphid species (179917 individuals in total), 15 hoverfly species (494 individuals) and 8 ladybird species (173 individuals). We found that aphid species richness and abundance were positively associated with utilised host plant abundance and garden plant species richness. Hoverfly abundance was positively correlated with garden plant richness. The abundance of ladybirds was positively correlated with aphid abundance and garden plant species richness, and negatively associated to the proportion of impervious surfaces in the surrounding environment. The difference in responses between the two major taxa of aphid predators may reflect differences in their behaviour and natural history. Our results indicate that overall increases in urban land cover are not favourable for ladybirds as a group, and that fine scale habitat variables that are determined by garden owners have the potential to greatly affect the diversity of aphids and their primary predators., Elise A. The European stag beetle, Lucanus cervus, is recognised as a flagship species for biodiversity conservation. Although the species is widely distributed across Europe declines have led to it being granted protected or endangered status in a number of countries and regarded as "near threatened" by the IUCN. The integration of genetic approaches into conservation efforts is urgently needed but has been impeded to date by the lack of appropriate genetic markers. To provide such a resource the development of the first microsatellite loci for stag beetle is described. Loci were identified using two methods (i) enriched library cloning (ELC) and (ii) Restriction enzyme Associated DNA sequencing (RAD-Seq). Inefficient microsatellite detection using the ELC method suggests that RAD-Seq, or other Next Generation Sequencing based methods, may ultimately be more cost effective for obtaining informative suites of markers for this and other coleopteran species. 18 loci were characterised by genotyping 42 UK specimens collected as prey leftover/roadkill. All loci produced unambiguous genotypes and were polymorphic. Though preliminary, estimates of genetic variability suggest UK populations may be genetically depauperate.
