Black rats (Rattus rattus) are native to the Indian subcontinent but have now colonized most continents and islands following human movements and international trade. They are involved in the circulation and transmission to humans of many zoonotic agents as well as in massive damage to food stocks and native biodiversity in the regions they have settled. This study investigates the genetic diversity and possible origins of black rats from Benin, West Africa. We sequenced the complete mitochondrial cytochrome b gene in 90 individuals from nine localities in Benin. These sequences were subsequently compared to 390 other cytochrome b haplotypes from individuals from various European, Asian, American and African localities. Nucleotide polymorphism analysis, haplotype network and maximum likelihood phylogenetic tree reconstructions showed low mitochondrial diversity in black rats from Benin. Our results also suggest at least two distinct introduction events: one introduction probably occurred during the spice trade (15th-17th century) through the Indies Road connecting Europe to Asia. Other introduction events could have occurred more recently following the intensification of globalized trade from the eighteenth century, and onwards.
The aim of the present study was to estimate the genetic diversity of the cestode Echinococcus multilocularis Leuckart, 1863 in Poland based on sequence analysis of the mitochondrial genes of worms isolated from red foxes, Vulpes vulpes (Linnaeus). Overall, 83 adults of E. multilocularis from the same number of foxes in different parts of Poland were used for analysis. Sequences of the three mitochondrial genes, cytochrome b (cob), NADH dehydrogenase subunit 2 (nad2) and cytochrome c oxidase subunit 1 (cox1), were analysed. Seventy-four individual biological samples were successfully sequenced. Combined sequence analysis of these three genes exhibited fifteen Polish haplotypes (EmPL1-EmPL15). Most isolates (n = 29; 39%) were classified to the EmPL1 haplotype, which occurred mainly in the east, north and centre of Poland. Haplotype EmPL4 (n = 14; 19%) and other haplotypes appeared predominantly in the south and west area. Fourteen haplotypes were grouped in the European clade. One Polish haplotype (EmPL9) (n = 7, 10%) was assigned to the Asian clade with haplotypes from Japan and Kazakhstan. This haplotype was found only in northeast Poland and this is the westernmost report of haplotype of E. multilocularis belonging to the Asian clade in Europe. Two species of the genus Gryllus occur in Europe: G. campestris and G. bimaculatus. The first is widely distributed in the north-western Palaearctic, while the second, G. bimaculatus, occurs predominantly in the Mediterranean area. There is a visible pattern in the distribution of G. campestris, the insect being rare and threatened in the western part of its range, whereas it is still abundant in the east. Despite the fact that this species is commonly used in laboratory experiments, its natural populations are poorly characterised. In the present study, we analysed cricket populations from the lower Oder and Vistula River valleys in Poland. Based on the phylogeny of the mtDNA cytochrome b fragment, we found that 17% of the individuals studied had a G. bimaculatus-like mtDNA haplotype. Analyses of 11 autosomal microsatellite loci failed to reveal any clear genetic differentiation between individuals assigned to these two clades. This suggests, along with the spatial distribution of G. bimaculatus-like haplotypes, successful interbreeding of G. bimaculatus with native populations of G. campestris. However, both the nuclear data and additional analyses of two X-chromosome-linked microsatellite loci revealed incomplete introgression. The cotton aphid, Aphis gossypii Glover (Hemiptera: Aphididae), is a serious pest of citrus in northern Iran, both because of the damaged caused by its feeding and as a vector of several viruses. The genetic structure of populations of A. gossypii on citrus trees at eight localities in Iran was surveyed using seven polymorphic microsatellite loci. Of 240 individuals tested, 142 multilocus genotypes (MLGs) were identified. The presence of multicopy genotypes and negative FIS values revealed that the major mode of reproduction in northern Iran is obligate parthenogenesis. The genotypic diversity of populations ranged between 0.24 and 0.93. Considerable genotypic diversity and a high frequency of unique MLGs, confirmed there is some cyclical parthenogenesis in the region. The analysis of molecular variance revealed high intrapopulation and weak interpopulation genetic differentiation (overall FST = 0.036) among the different populations. The UPGMA dendrogram of eight populations based on Nei's genetic distance indicated two clusters: genotypes from West of Mazandaran and Guilan provinces and those from East of Mazandaran. The same results were also obtained from the STRUCTURE analysis of these populations. This information on the genetic diversity of populations of A. gossypii in northern Iran could be useful for improving the Integrated Pest Management of this aphid.
Transforming growth factor beta 1 (TGF-β1) is a pro-fibrotic cytokine with a key role in wound repair and regeneration, including induction of fibroblast-to-myofibroblast transition. Genistein is a naturally occurring selective estrogen receptor modulator with promising anti-fibrotic properties. In the present study we aimed to investigate whether genistein modulates TGF-β1 (canonical and non-canonical) signaling in normal dermal fibroblasts at the protein level (Western blot and immunofluorescence). We demonstrated that TGF-β1 induces the myofibroblast-like phenotype in the studied fibroblast signaling via canonical (SMAD) and non-canonical (AKT, ERK1/2, ROCK) pathways. Genistein induced only ERK1/2 expression, whereas the combination of TGF-β1 and genistein attenuated the ERK1/2 and ROCK signaling. Of note, the other studied pathways remained almost unaffected. Brazilian native meliponines are currently threatened by increased human impacts. The assessment of their genetic variation by microsatellite DNA markers can assist in the conservation of populations and help in the planning and establishment of efficient management strategies. The purpose of this study was to develop the first set of microsatellite markers for Melipona fasciculata, selected from partial genome assembly of Illumina paired-end reads. Primer pairs were designed for each detected locus at their flanking regions. Bee samples were genotyped from two different populations of Northeastern Brazil for marker characterization and validation. A total of 17 microsatellite loci displayed polymorphism. Mean HE and HO heterozygosities were 0.453 and 0.536, respectively. PIC across all loci ranged from 0.108 to 0.714. A genetic diversity analysis revealed high values for population differentiation estimates (FST = 0.194, RST = 0.230, and Dest = 0.162) within the investigated region. PCoA and Bayesian clustering showed a separation of the species into two distinct clusters. These microsatellite markers have demonstrated strong potential for population-level genetic studies. Species of Acanthamoeba Volkonsky, 1931 are the commonest among free-living amoebae that are widespread in different water resources but with lacking phylogenetic data. This study aims at detecting molecular prevalence and genetic diversity of Acanthamoeba isolates in Kafrelsheikh Governorate, Egypt. Forty-eight water samples were collected from 12 swimming pools; four samples during each season over one year. Samples were filtered, cultivated on non-nutrient agar plates and examined microscopically. Polymerase chain reaction (PCR) and sequence analysis of positive samples targeting diagnostic fragment 3 (DF3) of the small subunit rRNA gene were done. Cultivation succeeded to detect 14 (29%) positive samples while PCR missed three positive samples. The obtained sequences were phylogenetically analysed. The phylogenetic tree was constructed for them with sequences of reference species from the NCBI database. The identified species were Acanthamoeba castellanii Douglas, 1930 (T4), A. astronyxis (Ray et Hayes, 1954) (T9) and A. hatchetti Sawyer, Visvesvara et Harke, 1977 (T11). The prevalence of species of Acanthamoeba was higher during summer and fall. Therefore, the control of the presence of Acanthmoeba spp. in swimming pools needs immediate, effective and practical measures to prevent and control infection with species of Acanthamoeba.
The Tongshan intrusion is located at the Guichi ore cluster field along the Lower Yangtze River metallogenic belt (LYRB) in eastern China. Elemental data and major elements of biotite are reported for Late Mesozoic plutons in the Guichi ore cluster in the Middle-Lower Yangtze River Region, attempting to explain their diagenetic age, petrogenesis, and tectonic implications. The Tongshan intrusion is dated at 146.3±3.2 Ma, with zircon εHf(t) average value of -5.23, illustrating that the Tongshan intrusion could be derived from crust mantle mixing magmatite provenance. The two-stage Hf model ages between 1184~1422 Ma is indicated magma source relationship with those of the Mesoproterozoic basement in south Anhui. The Tongshan intrusion is high-K calc-alkaline, and exhibit adakite-like geochemical feature, such as high Al2O3, Sr contents, Sr/Y value, low Y and Yb contents. The geochemical and reexplained published isotopic data indicate that these adakites are most likely from partial melting of subducted oceanic crust, and mixed with enriched lithospheric mantle components in the rising process.
The Mariánské Lázně Fault (western Bohemia, CZE) is a morphologically, geologically and tectonically prominent structure with that is 150 km long with an NNW-SSE orientation. Its tectonic activity, especially in the NW part and in the neighbouring Cheb basin, is well known and has been proven by present-day earthquake swarms, mantle-derived CO2 emanations, geophysical and paleoseismological research. It seems that other parts of the MLF (especially segments of NNW-SSE and N-S orientation) might also have been active during the Pleistocene, possible even in the Holocene. This study provides a robust set of morphometric analyses – mountain-front sinuosity, basin asymmetry, longitudinal stream profiles, SL-index, which assesses the possibility of recent tectonic activity. The results suggest that the activity of the central and southern part of the MLF could have been very young. A reconstruction of the evolution of the stream network of the Mže River, as a result of different timing of the activity of particular segments of the MLF, is also put forward. The first ideas about the evolution of terrain morphology and the stream network are proposed by this study, however subsequent field research (geophysics, paleoseismology) could prove and date the tectonic activity. Archaeological evidence has demonstrated that the Bujang Valley is Malaysia's richest archaeological site and served as the primary coastal centre. A study in the Bujang Valley found monuments related to trading activities and others that functioned as a temple related to the Hindu-Buddhist period. The main purpose of this study was to resolve issues and problems arising from previous studies related to the Bujang Valley civilisation, particularly in terms of iron studies. Geophysics plays a vital role in assisting archaeologists to obtain excellent preliminary results before they proceed with excavation and digging works. Therefore, the 2-D resistivity and ground-penetrating radar (GPR) methods were conducted to locate and map the potential iron smelting site at Site B2 (SB2). Three main characteristics that can be observed on the surface are a mound area, exposed clay bricks and surface finds. Two-D resistivity showed the resistivity values of a possible buried structure, with values > 800 Ωm. Radargram profiles showed the highest amplitude, indicating the reflections uncovered in the location in certain survey lines. This paper presents the first summary of research on the metallurgical sites in the Bujang Valley, the most important site in Malaysia. Geophysical methods, which rely on a physical contrast between buried archaeological features and the properties of the surrounding subsoil, can assist archaeological investigations.
