The first trimester of pregnancy is characterized by continuous proliferation, invasion and differentiation of cytotrophoblasts. These processes are precisely controlled both, in space and time by molecules such as endothelin-1 (ET-1). ET-1 is expressed in human first trimester trophoblast and is known to stimulate cytotrophoblast proliferation through endothelin A and B receptor subtypes (ETA and ETB), and cytotrophoblast invasion through ETB. However, temporal changes of the ET system during the first trimester of pregnancy have not been previously studied. This study tested the hypothesis that ET-1 release, ETA and ETB expression are increased towards the end of the first trimester of pregnancy (weeks 10-12 vs. weeks 6-9), resulting in increased cytotrophoblast proliferation and invasion. Tissue samples were obtained from 17 surgical pregnancy interruptions (week 6-9: n=9; week 10-12: n=8). After cytotrophoblast isolation, the invasive and proliferative phenotypes were immune-separated by an α6-integrin antibody. Both proliferative and invasive cytotrophoblasts were cultured separately on plastic or Matrigel for 24 h. ET-1 release into the culture medium of both cytotrophoblast subtypes was measured by radioimmunoassay. ETA and ETB mRNA expression was measured by RT-PCR, and the ET-1 effect on cytotrophoblast proliferation and invasion was determined using proliferation and invasion assays, respectively. ET-1 release increased from early to late first trimester of pregnancy in both proliferative (1.8-4.5 fold) and invasive cytotrophoblasts (9.3-28 fold), especially when cultured on Matrigel. This was paralleled by less ETB mRNA on invasive cytotrophoblasts independent of the time period in first trimester, whereas ETA expression was similar on proliferative an invasive cytotrophoblasts. Proliferation and invasion of cytotrophoblasts under control conditions decreased from early to late first trimester. The ET-1/ET-receptor system changes between weeks 6-9 and 10-12 in pregnancy. Our data suggest an autocrine and endocrine ET-1 effect, which is stronger in late than in early first trimester of pregnancy paralleled by different stimulatory effects on trophoblast invasion and proliferation. Protižidovské násilí na poválečném Slovensku, především pogrom v Topolčanech v září 1945, představuje dnes relativně probádané téma. Vícero autorů a autorek detailně popsalo průběh a spektrum násilí vůči Židům na osvobozeném Slovensku. Některé novější práce pak protižidovské demonstrace, výtržnosti a pogromy proti Židům na Slovensku, kteří přežili holokaust, zkoumaly v širším kontextu přetrvávajícího antisemitismu v poválečné Evropě. A přestože nejednou přitom poukazují na spojitost části slovenské společnosti s tamním válečným režimem a její častou neochotu vrátit přivlastněný židovský majetek jako na dva důležité tehdejší faktory, nálady a postoje obyvatel vůči Židům v poválečném období zůstávají většinou stranou pozornosti historiků. Protižidovské násilí proto autorce v této studii slouží zejména jako východisko k analýze smýšlení a postojů slovenské společnosti ke zbytku židovské menšiny. Ukazuje v ní, že Židé byli ve slovenském veřejném mínění té doby stále vnímáni jako ''cizí'' element, jako Maďaři a maďarizátoři, jejichž zájmy se rozcházejí se ''slovenskou věcí''. K zažitým stereotypům patřilo přesvědčení, že profitují z ''černého obchodu'' a žijí si nad poměry, když využívají úředně jim poskytované výhody. Naopak v tehdejších úředních zprávách i tisku na Slovensku, pokud se týkaly Židů, se téměř nevyskytují zmínky o holokaustu a kolaboraci většinového obyvatelstva s Tisovým režimem, který na něm participoval., Anti-Jewish violence in post-war Slovakia, in particular the pogrom in Topoĺčany in September 1945. has by now been quite will researched by scholars. Several have considered the course and the scope of the violence against Jews in liberated Slovakia. More recent works consider the anti-Jewish demonstrations, provocations, and pogroms in Slovakia against Jewish Shoah survivors in Slovakia in the wider context of persistent antisemitism in post-war Europe. Though several scholars have pointed out the link between part of Slovak society and the local wartime regime and its frequent post-war reluctance to return confiscated Jewish property as two important factors, public moods and attitudes toward the Jews in the post-war period have mostly been ignored by historians. Anti-Jewish violence is therefore the author´s starting point for her her analysis of Slovak society´s thinking and attitudes towards the remainder of the Jewish minority. She demonstrates that the perception of the Jews in Slovak public opinion at the time was still that of a ''foreign'' element (as it was regarding the Magyars and Magyarizers) whose interests differed from those of the ''Slovak cause''. Among the well-established stereotypes was the belief that the Jews profited from black marketeering and lived beyond their means, taking advantage of the officially provided benefits. They also allegedly shunned work on the reconstruction of Slovakia, as they had also allegedly avoided taking part in the Uprising during the war. a1_Článek se zabývá pamětí a komemorací ozbrojeného násilí spáchaného v rámci občanské rezistence vůči komunistickému režimu v jeho „zakladatelském období“ po únoru 1948. Soustřeďuje se na to, jaká paměť se kolem této menšinové formy protikomunistického odporu konstituuje, zejména prostřednictvím pamětních míst v procesu jejich utváření v době před listopadem 1989 i po něm. V první části se autorka věnuje ozbrojeným konfliktům na železné oponě, tedy na západních hranicích komunistického Československa. Ukazuje, že způsob nahlížení na překonávání státní hranice lidmi, kteří se pokoušeli odejít ze země na Západ, je dosud výrazně ovlivňován pamětí a komemoračními aktivitami veteránů z řad bývalých pohraničníků, mezi nimiž dominuje obraz těchto uprchlíků jako vnitřních nepřátel státu. Druhá část je věnována případům takzvaných politických vražd, tj. aktům násilí na komunistických představitelích, jež jsou spojeny zejména s vesnickým prostředím. Většinou jde o příběhy upadající do zapomenutí, které si společnost nechce znovu připomínat. It focuses on how memory is constituted around this minority form of anti-Communist resistance, particularly by means of memorial sites in the process of their formation in the period before the Changes that began in mid-November 1989 and also afterwards. In the first part of the article, the author looks at armed conflicts at the edge of the Iron Curtain, that is, on the western borders of Communist Czechoslovakia. She seeks to demonstrate that the way of looking at border crossings by people fleeing to the West is still considerably influenced by the memory and commemorative activities of veterans of the former border guards, amongst whom dominates the image of these refugees as internal enemies of the State. The second part of the article is devoted to instances of so-called ‘political murder’, that is, acts of violence against Communist politicians, which are connected particularly with villages. Most of these stories are gradually being forgotten; society does not want to recall them. An exception, however, is the memory of the sad events in the village of Babice, in the Bohemian-Moravian uplands, in 1951, which has repeatedly been used by politicians. In the third part of the article, the author considers the social discourse about the ethical dimension of armed anti-Communist resistance, which is almost exclusively focused on the atypical case of the group led by the Mašín brothers, and the process of forming the memory of the three resistances (the first, against Austria-Hungary during the Great War; the second, against the German occupying forces during the Second World War; and the third, against the Communist régime during the Cold War)., b2_She describes the commemorative activities of the Confederation of Political Prisoners as part of the strategy to bolster the social standing of the third, anti-Communist resistance, and she points to certain analogies between the unchallenged memories of political prisoners and the memories of the former border guards in contemporary historiography., and Markéta Devátá.
Autism spectrum disorders (ASD) are neurodevelopmental conditions characterized by impairment in social communication and presence of stereotyped/restricted behaviors. Children with ASD very often demonstrate co-morbid psychiatric problems, problems known to be affected by testosterone in neurotypical populations. However, there are few reports investigating relationships between testosterone and psychiatric conditions in children with ASD. The aim of this study was to determine the relationship between plasmatic levels of testosterone and behavioral/emotional problems in pre-pubertal boys with ASD. The study sample consisted of 31 pre-pubertal boys (ages 3-10) with ASD. Parents completed the Nisonger Child Behavior Rating Form (NCBRF) to assess specific behavioral/emotional problems as observed in the previous 2 months. Plasmatic testosterone levels were determined in boys according to standardized procedures. It was found that there were positive correlations between testosterone levels and the conduct problems subscale (p=0.034, rs=0.382) of NCBRF and also between testosterone levels and the hyperactive subscale (p=0.025, rs=0.402) of NCBRF. Findings in this study are in line with research conducted in the neurotypical population. This is the first large study investigating testosterone and emotional/behavioral problems in ASD and warrants further research in this field in order to clarify the etiopathogenesis of psychiatric co-morbidities and improve their treatment., A. Součástí tektonických změn, kterými dnes prochází akademické prostředí napříč obory, je specifický - a na první pohled triviální - rys. Vědci publikují čím dál více odborných článků a knih. Globální akademické publikace rostou meziročně o osm až devět procent, což znamená, že se jejich objem zdvojnásobí asi každých devět let. Tento trend je znatelný i v České republice. V roce 2013 byla druhým největším celostátním nakladatelstvím, co se počtu vydaných titulů týče, Univerzita Palackého v Olomouci (vydala v daném roce skoro 500 titulů), Univerzita Karlova byla pátým největším (asi 360) a Masarykova univerzita sedmým (310 knih). V současném systému hodnocení a financování vědy není příliš ojedinělým jevem, když humanitně a společenskovědně orientovaní akademici produkují pět až šest článků a jednu až dvě knihy ročně - aby si udrželi zaměstnání, dosáhli na rozumný plat, splnili (grantový) plán. Kdo jsou však čtenáři těchto textů? Zatímco autorů a autorek jsou dnes pod tlakem systému hodnocení a financování výzkumu přehršle, čtenářů, recenzentů a diskutujících je málo a akademická půda se tak stává souborem monologů, případně specializovaných, málo propojených diskusí. and Tereza Stöckelová, Filip Vostal.
