Huntingtonova nemoc (HD) je neurodegenerativní porucha způsobená elongací CAG repetic v genu kódující protein huntingtin (Htt). U pacientů jsou v postižených tkáních přítomny vedle monomerní formy hlavně N‑koncové fragmenty, oligomery a polymery mutovaného huntingtinu (mtHtt), oproti tomu samotná monomerní forma mtHtt je exprimována v podstatě ve všech buňkách. Nejvíce postižené tkáně jsou bazální ganglia a mozková kůra. V této studii jsme analyzovali přítomnost N‑koncových fragmentů a oligomerů Htt v různých tkáních 24- a 36měsíčních transgenních (TgHD) miniprasat exprimujících N‑koncovou část lidského mutovaného huntingtinu a jejich zdravých sourozenců. Zjistili jsme, že mozková kůra a varlata na rozdíl od svalu a srdce TgHD miniprasat obsahují kromě monomerní formy i N‑koncové fragmenty a oligomerní smíry. Ve svalech z 36 měsíčních TgHD miniprasat však již začíná mírná fragmentace. Tato zjištění napodobují časnou progresi onemocnění u lidí, a proto miniprase poskytuje slibný model pro terapeutické testování HD. In patients, in addition to the monomeric form of huntingtin, N‑terminal fragments, oligomers, and polymers are present mostly in the affected tissues, even though the mutated huntingtin (mtHtt) is expressed basically in all cells. The most affected tissues are basal ganglia and cerebral cortex. In this study we analyzed the presence of N‑terminal fragments and oligomers of Htt in different tissues of 24 and 36 months old experimental animals. This was done in our large animal model of HD, which uses transgenic (TgHD) minipigs expressing N‑terminal part of human mtHtt. Among all the tissues tested, we found cortex and testes to contain N‑terminal fragments as well as oligomeric smears in TgHD minipigs compared to wild type siblings. On the other hand, we did not detect any fragments or oligomers in muscle and heart of TgHD minipigs, only starting fragmentation in muscles of 36 months old animals. Huntingtonova nemoc (HN) je autozomálně dominantní neurodegenerativní onemocnění charakterizované poškozením volních a mimovolních pohybů, poruchami chování a zhoršením kognitivních funkcí. Společně s hlavními motorickými příznaky byly poruchy hlasu a řeči pozorovány u většiny pacientů s HN. Zvířecí prasečí model je často používán pro výzkum v preklinických studiích. I přes zjevné rozdíly v anatomii artikulačních orgánů mezi prasaty a lidmi lze očekávat stejné trendy u patofyziologických mechanizmů s ohledem na chrochtání i lidskou fonaci. Hlavním cílem této studie bylo proto navržení vhodného experimentu, který umožní získání dostatečně dlouhého záznamu chrochtání od co největšího počtu prasátek. Dalším cílem studie bylo zrealizovaní výsledné verze experimentu na celé databázi a vyhodnocení množství a kvality získaných nahrávek. Databáze použitá pro studii zahrnovala 17 HN transgenních miniprasátek a 16 zdravých sourozenců ze stejných vrhů. Testované varianty experimentu, provedené na části databáze zahrnující čtyři prasnice, byly rozděleni do čtyř podskupin: (a) pozitivní – krmení, (b) pozitivní – zvuková stimulace, (c) negativní – bránění v pohybu, (d) negativní – nepříjemné doteky. Hodnocení kvality získaných nahrávek bylo provedeno s pomocí audio softwaru, ve kterém bylo izolováno čisté prasečí chrochtání a všechny akustické artefakty vymazány. Nejlepších výsledků bylo dosaženo s použitím experimentu při kterém: (i) je záznamové zařízení upevněno na tělo prasátka, (ii) prasátko je ponecháno několik minut o samotě v místnosti, aby se uklidnilo a (iii) osoba vstoupí do místnosti a snaží se nabízet prasátku krmivo zatímco před ním couvá. V důsledku tohoto jednání prasátko následuje osobu s krmením, což je doprovázeno chrochtáním. Dostatečně dlouhé (20 chrochtnutí a více) a čisté nahrávky byly získány od 24 z 33 prasátek (73 %). Závěrem lze tedy říci, že experiment je proveditelný. Besides the main motor symptoms, voice and speech disorders have been documented in a large majority of patients with HD. The animal model of pigs is often used in preclinical studies. Although there are obvious differences in the anatomy of the articulation organs between pigs and humans, the same trends in pathophysiological mechanisms can be expected in both grunting and human phonation. The main aim of the study was therefore to design a suitable experiment that would allow for acquisition of a sufficiently long recording of grunting from as many pigs as possible. The second goal was to perform the final version of the experiment in all available pigs and to evaluate the amount and quality of the acquired recordings. The database consists of 17 HD transgenic minipigs and 16 healthy siblings. Tested variants of the experiment, performed on subgroup of four sows, were divided into four subgroups: (a) positive – feeding, (b) positive – sound stimulation, (c) negative – hindering in movement, (d) negative – unpleasant touch. The evaluation of the quality of the elicited recording was performed using audio software where pure pig grunting was selected and all acoustic artefacts deleted. The best results were reached using the experiment in which: (i) a recording device is put on the pig’s body, (ii) the pig is left alone for few minutes in the pen in order to calm down, and (iii) a person enters the room and tries to offer the pig food while walking backwards. As a result, the pig follows the person and grunts. Sufficiently long (20 single grunts or more) and clear recordings were received from 24 out of 33 pigs (73%). The realisation of the experiment is therefore possible. Zvířecí modely představují klíčový nástroj pro studium Huntingtonovy nemoci (HN). V posledním desetiletí byla snaha vytvářet velké zvířecí modely této choroby z důvodu zlepšení znalostí o patologických změnách v mozku, který se co nejvíce podobá lidskému. Transgenní miniprase exprimující N‑terminální část lidského mutovaného huntingtinu se 124 CAG/CAA repeticemi se zdá být velmi nadějným modelem pro studium HN. Jelikož předchozí charakterizace tohoto modelu prokázala rozličné fenotypy postihující subcelulární, buněčnou stejně tak i orgánově‑systémovou úroveň. Cílem této studie bylo detekovat a analyzovat patologický vzorec ve fyzické aktivitě transgenních (TgHD) kanců ve věku 3 let pomocí telemetrie. Do experimentu bylo zařazeno pět TgHD a pro srovnání pět netransgenních (WT) kanců. Fyzická aktivita byla u kanců měřena pomocí telemetrického systému rodentPACK2, přičemž transmiter byl umístěn do límce na krku. Na základě analýzy bylo zjištěno signifikantní snížení totálního zrychlení reprezentujícího fyzickou aktivitu u TgHD kanců mezi 4:40 a 5:30 ráno (po nočním spánku – před ranním krmením) v porovnání s WT jedinci, které může být vysvětleno narušeným energetickým metabolizmem. Telemetrická analýza bude jistě hrát v budoucnosti významnou roli ve studii fyzické aktivity a biopotenciálů důležitých pro podrobnější charakterizaci preklinické a klinické fázi HN u velkých zvířecích modelů. During the last decade large animal models of HD have been established to improve knowledge of HD under large brain conditions. Transgenic minipig expressing N‑terminal part of human huntingtin with 124 CAG/CAA repeats seems to be very promising HD model. Its previous characterization has shown various phenotypes affecting subcellular, cellular as well as organ systems level. The goal of this study was to detect and analyze a pathological pattern in physical activity of transgenic (TgHD) boars at the age of three years using telemetry approach. Into the study we have included five TgHD and five wild type (WT) animals for comparison. The physical activity was measured by the telemetric system rodentPACK2 whereas transmitter was placed into the collar. The analysis have shown significant decrease of total acceleration representing physical activity in TgHD boars between 4:40 and 5:30 a. m. (after night sleep – before morning feeding) in comparison with WT ones which could be explained with disturbed energy metabolism. AIMS: Authors studied potential side effects of fetal calf serum (FCS) in cultivation media on human dental pulp stem cells (DPSC) during long-term cultivation. METHODS: Two lines of DPSC obtained healthy donors (male 22 years, female 23 years) were used. Both lines were cultivated under standard cultivation conditions in four different media containing 10% or 2% FCS and substituted with growth factors. During long-term cultivation proliferation ability, karyotype and phenotype of DPSC were measured. RESULTS: Both lines of DPSC cultivated in a media containing 2% FCS and ITS supplement showed the highest number of population doublings. On the other hand the proliferation rate of DPSC cultivated in a media with 2% FCS without ITS supplement was slowest. Proliferation rate of DPSC cultivated in 10% FCS media with or without FGF-2 was comparable. DPSC cultivated in a media with 10% FCS showed a significantly higher amount of chromosomal aberrations. These chromosomal aberrations do not seem to be clonal but surprisingly we found large amounts of tetraploid cells in the 9th passage in both media containing 10% FCS. CONCLUSIONS: Our study proved that cultivation of DPSC in media containing higher concentration of FCS has critical side effects on cell chromosomal stability. and J. Suchánek, TS. Kleplová, M. Současné studie naznačují možnou důležitou úlohu melatoninu v Huntingtonově nemoci (HN) a jeho možné terapeutické využití při léčbě této nemoci. HN je dědičné neurodegenerativní onemocnění, které doprovází snižování hladiny melatoninu s postupem onemocnění. U normálních (nenádorových) buněk působí melatonin antiapoptoticky díky svým antioxidačním vlastnostem a schopnosti zabránit aktivaci proteinu p53. Dále melatonin zvyšuje expresi BDNF (brain derived neurotrophic factor) a dalších neuroprotektivních faktorů. Cílem této studie bylo stanovit netoxickou dávku melatoninu pro primární kožní fibroblasty izolované z transgenních miniprasat pro N‑koncovou část lidského mutovaného huntingtinu (TgHD) a popsat efekt tohoto ošetření na tyto buňky vystavené genotoxickému stresu. Buňky byly kultivovány v médiu obohaceném různými dávkami melatoninu. Analýzou proliferačních křivek získaných mikroskopováním živých buněk v pravidelných časových intervalech jsme stanovili efekt různých koncentrací melatoninu.Ukázali jsme, že vyšší dávky melatoninu jsou pro primární prasečí buňky toxické. Je zajímavé, že TgHD buňky byly oproti kontrolním buňkám více citlivé k tímto dávkám melatoninu. Stanovili jsme efektivní dávku melatoninu a současně jsme ukázali její efekt na proliferaci u buněk vystavených genotoxickému stresu. HD, the inherited neurodegenerative disorder, is accompanied by gradual melatonin reduction as it progresses. Melatonin in normal cells (non‑tumor) has the anti‑apoptotic ability due to its antioxidant property and its ability to prevent the activation of p53. Furthermore, melatonin increases the expression of BDNF (brain derived neurotrophic factor) and other neuroprotective factors. The aim of this study was to evaluate the nontoxic dose of melatonin for primary skin fibroblasts isolated from minipigs transgenic for the N‑terminal part of human mutated huntingtin (TgHD), and the effect of melatonin treatment to these cells exposed to genotoxic stress. Cells were cultured in medium supplemented with different doses of melatonin. Using time lapse microscopy, we estimated the effect of decreasing melatonin concentrations by analyzing the proliferation curves. We show that higher doses of melatonin are toxic for primary porcine fibroblasts. Interestingly, TgHD cells were more sensitive to these doses of melatonin treatment than wild type cells.
