The moths belong to the best-studied groups of insects in the Podyjí NP. In total, 2250 species of butterflies and moths (Lepidoptera) have hitherto been recorded here. Many species are closely associated with rocky steppe and heeathland, the most valuable natural habitats in this terriotory. Concurrently, the Podyjí NP is located on the western distribution border of many eastern and southern species. The Podyjí NP has very diverse vegetation which results in the occurrence of rich butterfly communities. To date, 142 species of butterflies have been recordeed in this area, of which 19 species (13 %) are considered regionally extinct and three species (2 %) are only migrants, not native to the Czech fauna. The main cause of the decline in butterflies is the abandonment of traditional techniques of habitat management. The Northern pine processionary moth, Thaumetopoea pinivora (Treitschke, 1834) shows a highly scattered distribution with fragmented populations across Europe. A previous study exploring the postglacial history of T. pinivora defined it as a cold-tolerant relict species and concluded that a progressive reduction of suitable habitats after the postglacial expansion from refugia in the southern Iberian peninsula best explained the distribution and genetic structure of populations of this species. However, recent records, both by us and others, challenge this view. Surprisingly, some of the newly found populations from southern Spain use black pine, Pinus nigra J.F. Arnold as a host plant despite the fact that the typical host of the species, Scots pine, Pinus sylvestris L. occurs in the area. We provide genetic data for one of these recently found southern populations where the larvae feed on P. nigra, and compare this with previously published data on individuals collected on P. sylvestris. This data reveals that populations from different host trees are no more genetically differentiated than populations sharing the same host plant. The findings of a wider diet breadth open the way to widen the search for the still unidentified glacial refugium of T. pinivora, and as such may contribute to a better understanding about how the species has spread across Europe., José A. Diapause is a common dormancy strategy exhibited by many species of invertebrates and insects to temporarily avoid seasonally recurring unfavourable conditions for their development, most usually in winter. Less frequently, a prolonged diapause lasting two or more years is described in species living in unpredictable environments where it is adaptive, but with significant costs. In this paper we examine the occurrence of prolonged diapause in the lycaenid butterfly Tomares ballus. Pupae of this species undergo an obligate diapause from mid-May to late January the following year. However, during our rearing experiments (from 2009 to 2016) the emergence of adults occurred sequentially and a fraction of the pupae remained in diapause for up to seven years. The annual percentage emergence after the first year of diapause was 45.6%, and only barely exceeded 50.0% in 2015. Remarkably, 12 pupae (11.4% of the initial brood) remained in diapause in their eighth year. The negative exponential equation fitted to the emergence data suggests that further emergences may occur within the next five years. Therefore, the potential for successful prolonged diapause of T. ballus pupae may be more than 10 years. The adaptive value of this strategy is discussed in relation to the effects of adverse and unpredictable weather during the flight period of the butterfly, intra-guild competition, parasitoids and changes in habitat quality. Allozyme polymorphism was investigated in adult males of the stenotopic peat bog (tyrphobiontic) noctuid moth, Coenophila subrosea Stephens, from three isolated peat bog localities in Austria (Styria) and Czech Republic (South and North Bohemia). Of the eighteen enzyme loci examined, twelve were polymorphic and six monomorphic. Significant deviations of genotype frequencies from Hardy-Weinberg equilibrium were observed at about one third of polymorphic loci within the populations. The average heterozygosities for the populations from three geographically distinct localities ranged from 0.192 to 0.245, and 61% of the loci were polymorphic. The FST mean value of 0.0675 was higher than that found in most other Lepidoptera. The genetic distances based on allozyme heterozygosity ranged from 0.019 to 0.051, with the population from South Bohemia being the most distant. The genetic distances and FST values do not reflect the geographic distances between the populations. Morphometric analysis revealed a difference between the Austrian Purgschachen Moor and Bohemian populations. Light trapping is the most widely used tool for determining the diversity of nocturnal Lepidoptera, but UV LEDs have yet to be used as light sources for the large-scale monitoring of Lepidoptera. We assessed the efficiency of this novel light source for sampling moths using a Heath type moth equipped with a strip of 150 high brightness UV LEDs (emission peak 398 nm, ~ 15 W) powered by a 12 V battery. We compared the number of individuals, the number of species and the Geometridae / Noctuidae ratio recorded for the samples collected using UV LED traps with those collected in two monitoring programs carried out in the same geographic region using two different light sources: a 200 W incandescent lamp (Rothamsted trap) and a 160 W mercury vapour lamp (manual catch). The total catch consisted of 61,120 individuals belonging to 699 species. The species richness rarefaction curves revealed that the Rothamsted trap collected fewer species and individuals than UV LED traps. Furthermore, the median numbers of species and individuals caught by UV LED traps fell within the range of those caught by mercury vapour lamp traps. In addition, the community composition recorded using incandescent lamps and UV LEDs was similar. The data obtained using UV LED traps, in absolute terms and in comparison with the other light sources and different sampling methods, clearly reveal that this light source is suitable for sampling macro-moth communities. Duponchelia fovealis Zeller is a polyphagous insect that has been recently reported attacking strawberry plants (Fragaria x ananassa Duchesne). Despite its economic importance there are few studies on this pest because it is difficult to rear it in the laboratory. With a constant supply of insects, studies on alternative methods of pest control can be conducted. This study aimed at developing an artificial diet for rearing D. fovealis with biological characteristics similar to those reared on their natural diet. This study was carried out in a climate-controlled room (25°C ± 2°C, RH 70% ± 10%, and 14L : 10D). The natural diet consisted of 'San Andreas' strawberry leaves (D1), while the artificial diet (D2) was developed in which beans, casein, soy protein, yeast and wheat germ are used as sources of protein. Five instars were identified. D. fovealis completed its life cycle in 38 and 40 days when fed D1 and D2, respectively. Survival was highest for the larvae fed the artificial diet. Females fed D1 lay a mean of 300.2 ± 62.3 eggs, while those fed D2, 220.3 ± 41.8 eggs. The artificial diet is suitable for the continuous rearing of D. fovealis in the laboratory., Maria A. C. The term cellular immune response refers to haemocyte-mediated responses, including phagocytosis, nodulation, and encapsulation. In the present study, we identified five types of circulating haemocytes in larvae of the haemolymph of the Asian corn borer, Ostrinia furnacalis (Guenée), including granulocytes, oenocytoids, plasmatocytes, prohaemocytes, and spherulocytes. The relative number of total free haemocytes per larva decreased significantly 0.5, 24, and 36 h after the injection of Beauveria bassiana conidia. Upon conidia challenge, both phagocytosis and nodulation were observed in the collected haemolymph from O. furnacalis larvae. In addition, plasma was found to be necessary for both phagocytosis and nodulation. Therefore, we here confirm that phagocytosis and nodulation are involved in O. funacalis larvae during their fight against infection by B. bassiana, and further, that the cellular immune response of O. furnacalis helps eliminate the invading organisms despite the fact that not all the fungal conidia are killed., Dongxu Shen, Miao Li, Yuan Chu, Minglin Lang, Chunju An., and Obsahuje bibliografii
Serpin is a broadly distributed superfamily of proteins that have a crucial role in regulating various immune reactions. Herein we identified a serpin-10 gene from Antheraea pernyi that encodes a 1557 amino acid residue protein with a predicted molecular weight of 58.76 kDa. Recombinant Apserpin-10 protein was expressed in a prokaryotic expression system (Escherichia coli) and the purified protein was used to prepare rabbit anti-Apserpin-10 polyclonal antibodies. Quantitative real-time polymerase chain reaction and western blot analysis indicate that Apserpin-10 was transcribed in all the tissues examined, including haemolymph, malpighian tubules, fat body, silk gland, integument and mid gut; the greatest expression level of Apserpin-10 was recorded in the fat body and haemocytes. The comparison of different developmental stages showed that Apserpin-10 transcript level was highest in 5th instar larvae, while the lowest expression was recorded at the egg stage. We also investigated the expression patterns of Apserpin-10 in fat body and haemocyte samples, following administration of heat-inactivated gram-positive bacteria (Micrococcus luteus), gram negative bacteria (Escherichia coli), a fungus (Beauveria bassiana) and virus (nuclear polyhedrosis virus, NPV). A substantial up-regulation of Apserpin-10 expression was recorded following pathogen challenge in both the tissues tested. Further the knock down of Apserpin-10 led to down regulation of antimicrobial peptide genes. The complete mitochondrial genome (mitogenome) of Spilarctia robusta (Lepidoptera: Noctuoidea: Erebidae) was sequenced and analyzed. The circular mitogenome is made up of 15,447 base pairs (bp). It contains a set of 37 genes, with the gene complement and order similar to that of other lepidopterans. The 12 protein coding genes (PCGs) have a typical mitochondrial start codon (ATN codons), whereas cytochrome c oxidase subunit 1 (cox1) gene utilizes unusually the CAG codon as documented for other lepidopteran mitogenomes. Four of the 13 PCGs have incomplete termination codons, the cox1, nad4 and nad6 with a single T, but cox2 has TA. It comprises six major intergenic spacers, with the exception of the A+T-rich region, spanning at least 10 bp in the mitogenome. The nucleotide composition of the genome is greatly A+T biased (81.09%), with a negative AT skewness (-0.007), indicating the presence of fewer As than Ts, similar to other Noctuoidea. The A+T-rich region is 343 bp long, and contains some conserved regions, including an "ATAGA" motif followed by a 19 bp poly-T stretch, a microsatellite-like (AT)9 and a poly-A element, a characteristic shared with other lepidopteran mitogenomes.
