- Alterations in mitochondrial parameters are an important hallmark of Huntington’s disease (HD). The ubiquitous expression of mutant huntingtin raises the prospect that mitochondrial disturbances can also be detected and monitored through buccal epithelial cells. In a group of 34 patients with Huntington’s disease and a group of 22 age-related healthy volunteers, respiratory complex I and IV protein quantities in buccal epithelial cells were measured using the dipstick immunocapture assay. The protein quantity of respiratory complex I correlates with age (r = 0.427, P = 0.026, FWE-P = 0.156) in the patient group, but not in the group of healthy subjects. Our non-invasive approach allows us to obtain valuable information for the studies of mitochondrial biochemical parameters in patients with neurodegenerative diseases and could also be useful in epidemiological studies. and Corresponding author: Hana Hansíková

- Buckling and postbuckling behaviour of a delaminated plate subjected to shear loading has been studied by means of the finite element analysis. The plate was assumed to contain either one or two circular delamintions of various diameteers and of various out-of-plane and in-plane positions. The size of the plate corresponded to the size of laminate panels used in aircraft structures. - Severe meconium aspiration sy ndrome (MAS) in newborns is often treated by exogenous surfac tant. Because its efficacy is reduced by meconium-induced inflammation, glucocorticoid budesonide was added into surfac tant preparation Curosurf to enhance efficacy of the surfactant therapy in experimental model of MAS. Oxygen-ventilated rabbits were intratracheally given meconium (25 mg/ml, 4 ml/kg) to induce respiratory failure. Thirty minutes later, animals were treated by intratracheal budesonide (0.25 mg/kg) ; or surfactant lung lavage (10 ml/kg, 5 mg phospholipids/ml) repeated twice, followed by undiluted Curosurf (100 mg phospholipids/kg) ; or by the above mentioned surfactant treatment with the last surfactant dose fortified with budesonide (0.25 mg/kg) ; or were untreated. Animals were ventilated for additional 5 hours and respiratory parameters were measured regularly. After sacrificing animals, wet-dry lung weight ratio was evaluated and plasma levels of interleukins (IL)-1beta, -6, -8, and TNF-alpha were measured by ELISA method. Efficacy of the given therapies to enhance lung functions and to diminish lung edema formation and in flammation increased from budesonide-only and surf actant-only therapy to surfactant+budesonide therapy. Combined therapy improved gas exchange from 30 min of administration, and showed a longer- lasting effect than surfactant-only therapy. - Programy externího hodnocení kvality očekává celá řada změn. Ty jsou podmíněny zejména dvěma faktory. Prvním z nich je změna akreditační normy pro organizátory programů EHK. Namísto dřívějších ISO Guide 34 a ILAC-G-13 nastupuje modernější nová norma ISO 17043. Pro praxi EHK to znamená rozšíření pracovního záběru z dosavadních dominantních analytických mezilaboratorních srovnávání také na hodnocení preanalytických a postanalytických fází. Dále se počítá s významným zvýšením edukačního charakteru programů a s používáním podrobnějších kritických komentářů výsledků. Koncepčně vychází novelizované pojetí EHK z metrologie, a tedy z akcentu na metrologickou návaznost a nejistoty měření, filosoficky ze snahy učinit z programů EHK nástroj zlepšené péče o pacienta a z výsledků EHK jeden z pracovních nástrojů managementu rizika zdravotní péče. Proces novelizace programů EHK se neobejde bez prověření stavu cílových hodnot, kontrolních limitů a komutability používaných kontrolních materiálů., In near future we can expect some significant changes in performance of external quality assessment programs. These changes are conditioned namely by two factors. Firstly, by introducing the new standard ISO 17043 into practice use, and secondly by the change of quality control from classical control to tool of patients risk management. What does it all means for performance of EQA? Introducing the preanalytical and postanalytical as part of EQA programs. Intensification of their educational potential is expected. Much larger application of two essential parts of metrology-traceability and uncertainty is involved in EQA programs. - Zinkové proteiny jsou nejpočetnější skupinou metaloproteinů lidského genomu. Jejich struktura je tak rozličná, jak pestré jsou funkce, které v organismu plní – od biokatalyzátorů, přes transportéry, detoxikanty až po transkripční faktory. Proteiny spojené se zinkem je možno formálně rozdělit na zinkové enzymy a zinkové neenzymové proteiny. Mezi nejpočetnější skupinu zinkových enzymů patří zinek dependentní metaloproteinázy. Zástupci této skupiny enzymů se účastní procesů, jako je embryonální vývoj, formace kostí, reprodukce, artritida nebo nádorové procesy. Pro přehlednost a zařazování proteináz byly vytvořeny různé klasifi kační systémy. V této práci je použit systém MEROPS, který rozděluje proteinázy na základě strukturních podobností. V přehledu biologických funkcí vybraných zinkových metaloproteináz u člověka jsou uvedeny významné skupiny enzymů, jakými jsou aminopeptidáza N, neprilysin, karboxypeptidázy, ADAM enzymy a zvláště matrixiny. Mezi zinkové neenzymové proteiny patří proteiny pro distribuci zinku, proteiny se zinkovými prsty a metalothioneiny. V současnosti jsou známy dvě rodiny proteinů zodpovědných za transport zinku, ZnT proteiny a Zip proteiny. Jakákoliv porucha regulace transportu zinku prostřednictvím jejich specifi ckých transportérů je většinou spojena se specifi ckým onemocněním. Také ve skupině proteinů se zinkovými prsty, které většinou fungují jako transkripční faktory, byla prokázána přímá souvislost mezi proteinem a určitým onemocněním. Příkladem je Wilmsův nádor nebo některé neurodegenerativní poruchy. Metalothioneiny, superrodina neenzymatických peptidů s malou molekulovou hmotností a s unikátní aminokyselinovou sekvencí, mají mnoho biologických funkcí. Mezi hlavní patří funkce imunoregulační, neuroprotektivní, metaloregulační a detoxikační. Zájem o tyto molekuly vzrůstá díky tomu, že jsou to nadějné tumorové markery a že jsou příčinou chemorezistence v léčbě nádorových onemocnění. Také o jiných zinkových metaloproteinech se uvažuje jako o perspektivních diagnostických markerech. V laboratorní medicíně se tak otevírají nové možnosti diagnostického využití zinkových metaloproteinů., Zinc proteins are the most numerous group of metalloproteins in the human genome. Their structure is as diverse as their function in organism, which ranges from biocatalysts, transporters, detoxicants to transcription factors. It is common to divide zinc proteins formally into group of zinc enzymes and group of zinc non-enzymatic proteins. The most abundant zinc enzymes are zinc-dependent metalloproteinases. Members of this group of enzymes are implicated in processes including embryonic development, bone formation, reproduction and arthritis or tumor growth. For lucidity and proteinases organising were created various one-term systems. In this work we used the system MEROPS, where proteinases are assigned to families on the basis of structure similarities. In this overview of biological functions of selected human zinc metalloproteinases are listed important enzyme groups such as aminopeptidase N, neprilysin, carboxypeptidases, ADAM enzymes, and especially matrixins. Proteins for zinc transport, zinc fi ngers proteins, and metallothioneins belong between zinc non-enzymatic proteins. Two protein families have now been implicated in zinc transport, ZnT proteins and Zip proteins. Any dysregulation in zinc transport via the specifi c protein transporters have been linked to specifi c diseases. Direct connection between protein and specifi c disease was proofed also in the group of zinc fi ngers proteins functioned as transcription factors. The examples are Wilm’s tumour or some neurodegenerative disorders. Metallothioneins, the superfamily of non-enzymatic peptides with low molecular mass and with unique sequence of amino acids, play many important biological roles, including immunoregulation, neuroprotection, metalloregulation, and detoxifi cation. The interest in these molecules has been increased because they were found as promising tumour markers. They are also responsible for platinum cytostatic chemoresistence in treatment of tumour diseases. Also other zinc metalloproteins are hoped to be perspective diagnostic markers. 3770. Buffering agent via insulin-mediated activation of PI3K/AKT signaling pathway to regulate lipid metabolism in lactating goats

- Ruminants are often fed a high-concentrate (HC) diet to meet lactating demands, yet long-term concentrate feeding induces subacute ruminal acidosis (SARA) and leads to a decrease in milk fat. Buffering agent could enhance the acid base buffer capacity and has been used to prevent ruminant rumen SARA and improve the content of milk fat. Therefore, we tested whether a buffering agent increases lipid anabolism in the livers of goats and influences of milk fat synthesis. Twelve Saanen-lactating goats were randomly assigned to two groups: one group received a HC diet (Concentrate: Forage=60:40, Control) and the other group received the same diet with a buffering agent added (10 g sodium butyrate, C4H7NaO2; 10 g sodium bicarbonate, NaHCO3; BG) over a 20-week experimental period. Overall, milk fat increase (4.25±0.08 vs. 3.24±0.10; P<0.05), and lipopolysaccharide levels in the jugular (1.82±0.14 vs. 3.76±0.33) and rumen fluid (23,340±134 vs. 42,550±136) decreased in the buffering agent group (P<0.05). Liver consumption and release of nonesterified fatty acid (NEFA) into the bloodstream increased (P<0.05). Phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT) and ribosomal protein S6 kinase (p70S6K) up-regulated significantly in the livers of the buffering agent group (P<0.05). It also up-regulated expression of the transcription factor sterol regulatory element binding protein-1c (SREBP-1c) and its downstream targets involved in fatty acid synthetic, including fatty acid synthetase (FAS), stearoyl-CoA desaturase (SCD-1) and acetyl-CoA carboxylase 1 (ACC1) (P<0.05). The BG diet increased insulin levels in blood (19.43±0.18 vs. 13.81±0.10, P<0.05), and insulin receptor was likewise elevated in the liver (P<0.05). Cumulatively, the BG diet increased plasma concentrations of NEFA by INS-PI3K/AKTSREBP- 1c signaling pathway promoting their synthesis in the liver., L. Li, M. L. He, K. Wang, Y. S.
