The aim of this study was to evaluate the role of endogenous histamine in the regulation of reactive hyperaemia (RH) and coronary autoregulation in isolated rat hearts. The basal release of cardiac histamine (perfusion pressure 60 cm H2O) amounted to 100-200 pmol/min/g wL During the first 15 s following 30 s of coronary occlusion, the release of histamine increased about three times and returned to basal levels after approximately 90 s, paralleling the changes of coronary flow (CF). Blockade of Hi-receptors increased basal CF by 23±2 %, significantly reduced blood flow debt and prolonged the duration of RH. Blockade of H2- and H3-receptors produced a significant decline of CF, decreased RH flow and diminished RH by 40±3 %. Blockade of all three classes of histamine receptors indicated that endogenous histamine exerts predominantly vasodilatory effects (mediated by H2- and H3-receptors) on coronary circulation. Hydrogen peroxide injected into the inflow cannula of isolated ventilated rat lungs produced a dose-dependent vasoconstriction in the range 0.25-10 mM, with maximum response between 2 - 5 mM. The effects of H2O2 can be influenced by ionophores or specific inhibitors of ionic channels or pumps. A key role is played by sodium ions which govern the subsequent inflow or outflow of calcium, an ion mediating the vasoconstriction. A physiological role for H2O2 generated by NADPH oxidase is postulated.
1_The young larvae of insects living on dry food produce large amounts of water by the metabolic combustion of dietary lipids. The metabolic production of water needed for larval growth, previously known as hypermetabolic responses to juvenile hormone (JH), is associated with a 10- to 20-fold increase in the rate of O2 consumption (10,000 µl O2/g/h in contrast to the usual rate of 500 µl O2/g/h). Growing and moulting larvae are naturally hypermetabolic due to the endogenous release of JH from the corpora allata. At the last, larval-pupal or larval-adult moult there is no JH and as a consequence the metabolic rate is much lower and the dietary lipid is not metabolized to produce water but stored in the fat body. At this developmental stage, however, a hypermetabolic response can be induced by the exogenous treatment of the last larval instars with a synthetic JH analogue. In D. vulpinus, the JH-treated hypermetabolic larvae survive for several weeks without moulting or pupating. In T. castaneum and G. mellonella, the JH-treated hypermetabolic larvae moult several times but do not pupate. All these larvae consume dry food and the hypermetabolic response to JH is considered to be a secondary feature of a hormone, which is produced by some subordinated endocrine organ., 2_The organ is most probably the controversial prothoracic gland (PG), which is a typical larval endocrine gland that only functions when JH is present. According to our hypothesis, PG activated by JH (not by a hypothetical PTTH) releases an adipokinetic superhormone, which initiates the conversion of dietary lipid into metabolic water. This type of metabolic combustion of dietary lipid produces large quantities of endothermic energy, which is dissipated by the larvae in the form of heat. Thermovision imaging revealed that the body of hypermetabolic larvae of G. mellonella can be as hot as 43°C or more. In contrast, the temperature of "cold" normal last instar larvae did not differ significantly from that of their environment. The role of L-DOPA in spinal nociceptive reflex activity has been re-evaluated. In high spinal ca ts, with supraspinal loops being excluded, the onset of reflex facilitation induced by noxious radiant heat is delayed after injection of L-DOPA by 4 to 10 s, i.e. the early component of nociceptive reflex facilitation is blocked, while the late component persisted. Further investigations have shown that the early component of reflex facilitation induced by noxious radiant heat is mediated by Aδ-fibres and the late component by C-fibres. Therefore, it can be assumed that L-DOPA, like opioids, preferentially blocks the transmission in nociceptive reflex pathways from Aδ-fibres., E. D. Schomburg, P. Dibaj, H. Diglyphus isaea Walker (Hymenoptera: Eulophidae) is an important ectoparasitic wasp of many leaf miners. Ability of D. isaea to find hosts placed in artificial mines and for its larvae to pupate when the larva is not in a mine was studied. Artificial mines consisted of slits cut in index card sandwiched between two cover slips. Almost 80% of the neonate larvae of D. isaea located host larvae in artificial mines compared to only 50% of those not in a mine. Mature larvae removed from mines did not construct normal pupal chambers. Nonetheless, they pupated and emerged successfully. Larvae of Liriomyza sativae Blanchard (Diptera: Agromyzidae) in mines are more likely to be attacked than those not in mines. Moreover, when close to a host larva this parasitoid can use vibrational cues to locate the larva in a mine. In addition, this parasitoid also did not always use volatile and gustatory cues for short range location of hosts. However, adult females of D. isaea more quickly located L. sativae larvae in the presence of the odour of juice extracted from an uninfested host plant. We conclude by proposing that the host mine is the medium by which the vibrations generated by the host larva are transmitted, which are probably the most important cue used by female D. isaea searching for hosts. That is this parasitoid first perceives mines not host larvae. Aim: to improve treatment results of colorectal cancer, complicated carcinomatosis of abdominal cavity, by associated using of endolymphatic chemotherapy (ELCT) and local extremely high frequency (EHF) therapy. Igroup 21 people (33.3%) performed colostomy, then neoadjuvant systemic chemotherapy by the scheme FOLFOX4. It was held two cycles, after two cycles their were received cytoreductive surgery; II group 26 people (41,3%) received cytoreductive operations with early postoperative intraabdominal chemotherapy by oxaliplatin (200 mg/m(2) at day 1 and 5FU 650 mg/m(2) from days 1 to 4. III group 16 people (25,4%) also performed colostomy, then used 2 courses of ELCT with local EHF therapy and all patients were received cytoreductive operations. ELCT was carried out injecting oxaliplatin100 mg/m(2) a day during 12 hours and then 5 FU in doze of 600 mg/m2 a day during 72 hours by the instrumentality of the dozer and LV200 mg/m(2) (2h intravenous infusion). During ELCT patients received local EHF therapy in abdominal cavity for an hour. After chemotherapy the partial regress of tumor observed in 1st group in 4,8 % and in 3rd 37,5 %, stabilization of process noted in 28,6 and 50% cases, progressing 66,7 and 12,5% cases correspondingly to groups. Histomorphologic study of malignant foci after ELCT+EHF therapy showed reduction of specific area of parenchyma cancer cell from 57 till 39% simultaneous growth of stroma from 40 till 58% and in necrosis area from 1,8 till 2,5%. Mitotic activity of tumor cells in the 1st group decreased (average in 22 un.), but in 3rd group it decreased until 11 units. Analysis of life span showed that in 1st group of patients 2year survival rate was 66,7%. In 2nd group of patients 2year survival rate was 42,3%. ELCH+EHFtherapy has high clinic efficacy and promotes to decrease terms of progression of tumor process, increases life span from 0 till 24% and quality of life of the patients. ELCH+EHFtherapy brings to reduction of specific area of parenchyma in cancer cells from 57 till 39% with simultaneous growth of stroma area from 40 till 58% and necrosis area from 1,8 till 2,5%. In the subcontinental, semiarid lowland region of Central Bohemia (Czech Republic), continuous human impact acting together with diverse natural environmental conditions resulted in the present extraordinarily complex pattern of vegetation. Three radiocarbon-dated pollen diagrams for the area indicate that this complexity results from past vegetation development. During prehistory, places suitable for settlement (with respect to climate, geology, hydrology, etc.) were colonized and transformed first. This resulted in a diachrony in vegetation development due to human activity starting in the first half of the Holocene. This caused an increase in diversity in the region as plant species persisting from previous periods, along with those associated with different agricultural practices, increased. Local abiotic factors affected not only the chronology of human impact but also its specific effects on the ecosystem. Anthropogenic pressure may have had different effects under different conditions. Human population pressure was the mediator between the abiotic diversity and selectively transformed vegetation suitable for the respective habitats. Differences in the chronology of human impact, mixed oak woodland degradation, and the chronology of beech, silver fir and hornbeam expansion are documented for the different ecological zones of the study area. These differences shed light on the mechanisms resulting in some of the important changes in Holocene vegetation. In the absence of man, the decline in mixed oak woodlands, typical of the Middle Holocene in Central Bohemia, would have been probably much slower and less extensive. Unlike in the uplands and mountains, the expansion in the area of beech, silver fir and hornbeam would have been insignificant. The present vegetation resulted to a large extent from management during High Middle Ages. There is almost no continuity in vegetation from the late prehistory to the present.
Gastric mucus plays an important role in gastric mucosal protection. Apart from its “barrier” function, it has been demonstrated that mucus protects gastric epithelial cells against toxic oxygen metabolites derived from the xanthine/xanthine oxidase system. In this study, we investigated the effect of malotilate and sucralfate (mucus production stimulators) and N-acetylcysteine (mucolytic agent) on ischemia/reperfusion-induced gastric mucosal injury. Gastric ischemia was induced by 30 min clamping of the coeliac artery followed by 30 min of reperfusion. The mucus content was determined by the Alcian blue method. Sucralfate (100 mg/kg), malotilate (100 mg/kg), and N-acetylcysteine (100 mg/kg) were given orally 30 min before surgery. Both sucralfate and malotilate increased the mucus production in control rats. On the other hand, N-acetylcysteine significantly decreased mucus content in control (sham) group. A significant decrease of mucus content was found in the control and the N-acetylcysteine pretreated group during the period of ischemia. On the other hand, sucralfate and malotilate prevented the decrease the content of mucus during ischemia. A similar result can be seen after ischemia/reperfusion. In the control group and N-acetylcysteine pretreated group a significant decrease of adherent mucus content was found. However, sucralfate and malotilate increased mucus production (sucralfate significantly). Sucralfate and malotilate also significantly protected the gastric mucosa against ischemia/reperfusion-induced injury. However, N-acetylcysteine significantly increased gastric mucosal injury after ischemia/reperfusion. Nitric oxide plays an important role in the control of basal coronary tone and mediation of reactive hyperaemic flow response following short-term coronary occlusion. The results presented in this report indicate that NO is involved in the modulation of coronary autoregulation in isolated rat hearts. Isolated rat hearts exhibit autoregulation of coronary flow (CF) between 50 and 80 cm H2O of coronary perfusion pressure (CPP). Within this autoregulatory range NO release (measured as nitrite) varies from
1.7±0.3 to 2.2±0.7 nmol/min/g wL Below the autoregulatory range it decreases slightly, while above this there is more than a twofold increase. Changes of NO release are accompanied by directly proportional changes of cGMP release. The release of hypoxanthine + xanthine shows a reciprocal relationship to CF values. The inhibition of NO synthesis showed a reciprocal relationship with CF values. Inhibition of NO synthesis by L-NAME (30 /¿mol/l) significantly reduces CF over the entire range of CPP changes (20-120 cm H2O), but much less at lower than at higher pressure values. Therefore, the autoregulatory range is significantly widened to CPP of 40-100 cm H2O. Theophylline (30 yumol/l) reduces CF by 15-25 % throughout the entire range of CPP changes. Hence, the CPP-CF curve is shifted downwards without significant changes of the autoregulatory range. Theophylline-induced reduction of NO release is CPP-dependent: as greater as CPP lower. When L-NAME is coadministered with theophylline, CF is additionally reduced while widened autoregulatory range is shifted to the right
