Příspěvek seznamuje s výskytem nadváhy a obezity u dětí a dospívajících v současné Velké Británii a s některými poznatky z britských výzkumů, zaměřených na roli psychosociálních a environmentálních proměnných při vzniku, rozvoji a udržování nadváhy a obezity. Velká Británie patří k zemím s etnicky pestřejším složením obyvatel, což vždy vedlo k pochybnostem odborníků o hodnotě srovnávaných údajů o nadváze a obezitě mezi původními obyvateli a obyvateli z řad jednotlivých etnik, která ve Velké Británii žijí. Tomuto problému a jeho současnému způsobu řešení v této zemi je věnována jedna z částí příspěvku. Dále příspěvek seznamuje s výzkumy, zabývajícími se vztahy mezi nadváhou a obezitou příslušníků původních britských obyvatel i jednotlivých etnik a spolupůsobícími psychosociálními a environmentálními proměnnými, které jsou vždy tvořeny, doprovázeny nebo se navzájem liší charakteristickým způsobem mezilidské komunikace. Britská odborná veřejnost se prostřednictvím britských vládnoucích institucí vyjádřila k problému nadváhy a obezity řadou projektů. Tvůrci projektu Foresight (Butland et al., 2007) navrhují řešení stoupajícího výskytu nadváhy a obezity z celonárodní a déledobé budoucí perspektivy, kdy jsou v jednotlivých resortech prostřednictvím odpovědných institucí zaváděny změny, které by lépe vyhovovaly (psycho) biologii člověka a tak přirozeně vedly ke zdravějšímu životnímu stylu. and The paper introduces the prevalence of overweight and obesity in children and adolescents in the current UK and some knowledge of British researches focused on the role of psychosocial and environmental variables in the creation, development and maintenance of overweight and obesity. The United Kingdom is country with ethnically more diverse composition, that has always raised doubts about the value of the compared data of overweight and obesity, that has been thus obtained in different ethnic groups living here. This problem and its solution in the current country is devoted one part of this contribution. Further part introduces the researches concerned with the relationships between overweight and obesity of British native population and various ethnic groups and their psychosocial and environmental variables, that are always formed, accompanied or mutual differ in a characteristic way of interpersonal communication.The conclusion of the paper is devoted to the strategies, that are currently applied to address the problem of overweight and obesity in some administrative areas of this country. Creators of project Foresight (Butland et al., 2007) propose solutions of increasing incidence of overweight and obesity, in which are involved various ministries and that could be realised through the responsible institutions. These changes would better fit the (psycho) human biology and so could more naturally lead to a healthier lifestyle.
Neuromedin beta (NMB) is a member of the bombesin-like peptide family expressed in brain, gastroin testinal tract, pancreas, adrenals and adipose tissue. The aim of our study was to compare the frequency of P73T polymorphism in overweight and obese patients (37 men: age 50.6±11.7 years, BMI 41.1±7.8 kg/m2; 255 women: age 49.0±11.9 years, BMI 37.9±6.8 kg/m2) with that of healthy normal weight subjects (51 men: age 28.2±7.1 years, BMI 22.3±2.0 kg/m2; 104 women: age 29.1±9.1 years, BMI 21.5±1.9 kg/m2) and to investigate the polymorphism’s influence on anthropometric, nutritional and psychobehavioral parameters in overweight/obese patients both at the baseline examination and at a control visit carried out 2.5 years later, regardless of the patient́s compliance with the weight reduction program. No significant differences in the genotype distribution were demonstrated between normal weight and overweight/obese subjects. Male T allele non-carriers compared to T allele carriers had higher energy (p=0.009), protein (p=0.018) and fat (p=0.002) intakes and hunger score (p=0.015) at the beginning of treatment. Male T allele non-carriers had a more favorable response to weight management at the follow-up, as they exhibited a significant reduction in waist circumference, energy intake and depression score as well as a significant increase in dietary restraint. No significant differences between carriers and non-carriers were demonstrated in women at the baseline examination. Both female T allele carriers and non-carriers demonstrated similar significant changes in nutritional parameters and in restraint score at the follow-up. Associations between different infectious agents and obesity have been reported in humans for over thirty years. In many cases, as in nosocomial infections, this relationship reflects the greater susceptibility of obese individuals to infection due to impaired immunity. In such cases, the infection is not related to obesity as a causal factor but represents a complication of obesity. In contrast, several infections have been suggested as potential causal factors in human obesity. However, evidence of a causal linkage to human obesity has only been provided for adenovirus 36 (Adv36). This virus activates lipogenic and proinflammatory pathways in adipose tissue, improves insulin sensitivity, lipid profile and hepatic steatosis. The E4orf1 gene of Adv36 exerts insulin senzitizing effects, but is devoid of its pro-inflammatory modalities. The development of a vaccine to prevent Adv36- induced obesity or the use of E4orf1 as a ligand for novel antidiabetic drugs could open new horizons in the prophylaxis and treatment of obesity and diabetes. More experimental and clinical studies are needed to elucidate the mutual relations between infection and obesity, identify additional infectious agents causing human obesity, as well as define the conditions that predispose obese individuals to specific infections., V. Cílem studie bylo zjistit, zda typ osobní příčinné orientace (OPO) může predikovat hmotnostní úbytek u účastnic kurzů snižování nadváhy s aplikací kognitivně-behaviorální terapie. Sledování probíhalo 12 týdnů a respondentky (N = 333) byly během této doby dvakrát vyzvány k vyplnění českého standardizovaného tříškálového „Dotazníku osobní příčinné orientace“ (DOPO). Absolventky kurzu (ženy, které se zúčastnily 11-12 lekcí) v průběhu kurzu signifikantně (p < 0,001) snížily svoji tělesnou hmotnost o 7,6 %. Ve studii bylo u skupiny absolventek zjištěno několik významných korelací, které poukázaly na existenci vztahu mezi OPO a změnou tělesné hmotnosti. Byla zjištěna negativní korelace mezi vnější příčinnou orientací (EO) a vstupní hmotností a jejím úbytkem (p < 0,05) a pozitivní korelace mezi vnitřní příčinou orientací (IO) a vstupní hmotností (p < 0,05). Míra vnější OPO a hodnota vstupní hmotnosti mohou predikovat velikost hmotnostního úbytku, avšak pouze u absolventek kurzu, a nikoliv u celé skupiny žen vstupujících do kurzů snižování nadváhy.
Parabens are a group of chemicals used as preservatives in the food, cosmetic and pharmaceutical industries. They are known to possess estrogenic effects, and therefore have been classified as endocrine disruptors. In addition to the classical endocrine organs, other tissues have endocrine activity, including adipose tissue. Several chemicals are known to cause obesogenic effects, and parabens are currently being studied in this context. The aim of this study was to investigate the possible connections of paraben exposure and obesity. Blood plasma from 27 healthy women was collected during their menstrual cycle. Basal anthropometric measures, levels of parabens (methylparaben, ethylparaben and propylparaben), adipokines (adiponectin, adipsin, leptin, resistin and visfatin) and hormones affecting energy balance and metabolic health (c-peptide, ghreline, GIP, GLP-1, glucagon, insulin, PAI-1) were measured. A Kolmogorov- Smirnov test showed higher methylparaben and propylparaben levels in women with BMI 25-34.9 compared to those with BMI 18.5-24.9. Plasma levels of methylparaben as well as the sum of parabens were positively associated with the plasma adipsin levels. Negative associations for methylparaben were found for glucagon, leptin and PAI-1. Clusterin is a heterodimeric glycoprotein with wide range of functions. To further explore its possible regulatory role in energy homeostasis and in adipose tissue, we measured plasma clusterin and its mRNA expression in subcutaneous adipose tissue (SCAT) of 15 healthy lean women, 15 obese women (OB) and 15 obese women with type 2 diabetes mellitus (T2DM) who underwent a 2-week very low-calorie diet (VLCD), 10 obese women without T2DM who underwent laparoscopic sleeve gastrectomy (LSG) and 8 patients with T2DM, 8 patients with impaired glucose tolerance (IGT) and 8 normoglycemic patients who underwent hyperinsulinemic euglycemic clamp (HEC). VLCD decreased plasma clusterin in OB but not in T2DM patients while LSG and HEC had no effect. Clusterin mRNA expression in SCAT at baseline was increased in OB and T2DM patients compared with controls. Clusterin mRNA expression decreased 6 months after LSG and remained decreased 12 months after LSG. mRNA expression of clusterin was elevated at the end of HE C compared with baseline only in normoglycemic but not in IGT or T2DM patients. Serum adipocyte fatty acid-binding protein (FABP-4) concentrations are linked to human obesity and other features of metabolic syndrome. Patients with Cushing's syndrome (CS) develop numerous features of metabolic syndrome due to chronic cortisol excess. Here we tested the hypothesis that chronically increased cortisol levels in CS patients may alter circulating levels of FABP-4. Fourteen patients with CS, 19 patients with simple obesity (OB) and 36 healthy control subjects (C) were included in the study. Serum FABP-4 concentrations were significantly higher in both CS and OB patients relati ve to C group, but they did not differ between CS and OB groups. In a combined population of all groups, serum FABP-4 levels correlated positively with BMI, body fat content, serum glucose, triglycerides, HbA1c and HOMA index and were inversely relate d to HDL-cholesterol, resting energy expenditure and freeT3 levels. We conclude that FABP-4 levels are significantly increased in both patients with simple obesity and obese patients with Cushing's syndrome. We suggest that increased FABP-4 concentrations in CS patients are rather due to their excessive fat accumulation and related metabolic abnormalities than due to a direct effect of cortisol on FABP-4 production., V. The objective of this study was to measure plasma fibroblast growth factor 21 and 19 (FGF21 and FGF19) levels in patients with Cushing's syndrome (CS) and to compare it with those of lean control subjects (C) and patients with obesity (OB). Fourteen untreated patients with CS, 19 patients with OB and 36 controls were included in the study. Plasma FGF21 and FGF19 levels were measured by ELISA kits, other hormonal and biochemical parameters were measured by standard laboratory methods. Plasma FGF19 did not significantly differ among the studied groups. Plasma FGF21 levels were significantly higher in both CS and OB groups relative to C group but they did not differ between CS and OB groups. In a combined population of all three groups FGF21 levels positively correlated with BMI, waist circumference and percentage of total and truncal fat mass. Less prominent inverse relationship with these parameters was found for FGF19. Neither FGF21 nor FGF19 were significantly related to cortisol concentrations. Increased FGF21 concentrations in both patients with CS and OB relative to lean subjects suggest that excessive body fat and/or related metabolic abnormalities rather than direct effects of cortisol are responsible. Obestatin is a recently discovered peptide produced in the stomach, which was originally described to suppress food intake and decrease body weight in experimental animals. We investigated fasting plasma obestatin levels in normal weight, obese and anorectic women and associations of plasma obestatin levels with anthropometric and hormonal parameters. Hormonal (obestatin, ghrelin, leptin, insulin) and anthropometric parameters and body composition were examined in 15 normal weight, 21 obese and 15 anorectic women. Fasting obestatin levels were significantly lower in obese than in normal weight and anorectic women, whereas ghrelin to obestatin ratio was increased in anorectic women. Compared to leptin, only minor differences in plasma obestatin levels were observed in women who greatly differed in the amount of fat stores. However, a negative correlation of fasting obestatin level with body fat indexes might suggest a certain role of obestatin in the regulation of energy homeostasis. A significant relationship between plasma obestatin and ghrelin levels, independent of anthropometric parameters, supports simultaneous secretion of both hormones from the common precursor. Lower plasma obestatin levels in obese women compared to normal weight and anorectic women as well as increased ghrelin to obestatin ratio in anorectic women might play a role in body weight regulation in these pathologies., H. The aim was to find the differences in ketogenesis initiation in the early period after the exercise in obese patients and to find if these changes may predict the weig ht loss during the physical activity program. 96 females were enrolled. A clamped heart rate test (CHR) was performed to establish comparable exercise intensity. Blood samples for beta hydroxybutyrate (BOHB) assessment were collected prior, immediately after and 60 min after the test. Patients underwent a three month fitness program. Anthropometric measurements (fat mass and biochemical parameters) were measured. An energy intake was monitored and comparable in all subjects. A significant increase of BOHB was found in 60 th minute after the test, when compared with initiation levels (BOHB1 vs. BOHB3; p=0.03). This increase correlates with % fat mass (R=0.196; p=0.02) and negatively with age (R= –0.147; p=0.05) and with weight reduction during the three-month program (R= -0.299; p=0.03). Serum BOHB increase after the single exercise may detect individuals with an ability to induce lipolysis in three-month program of physical activity for obese patients., M. Matoulek, S.
