Cr(VI) significantly reduced rates of net photosynthesis and transpiration and of stomatal conductance. Cr(VI) did not affect the Fv/Fm ratio of chlorophyll fluorescence implying that the primary photochemical processes in photosystem 2 were not affected. However, the efficiency of excitation capture by open PS2 centres, in vivo quantum yield of PS2 photochemistry, and electron transport rate were significantly reduced by Cr(VI). The coefficient of photochemical quenching was reduced with a concomitant increase in coefficient of non-photochemical quenching, suggesting reduced demand for ATP and NADPH due to inhibition of CO2 assimilation. Lipid peroxidation was increased by Cr(VI) and the activities of superoxide dismutase and catalase (CAT) were increased. However, the CAT activity was reduced by high Cr(VI) concentration. The activities of ascorbate peroxidase and glutathione reductase were significantly reduced by Cr(VI) treatment.
Pepper is a thermophilous and heliophilic vegetable. In China, pepper is grown in greenhouse during winter and spring under lower temperature and irradiation. In this study, we investigated the effects of low temperature and low irradiance (LTLI) on the physiological characteristics and the expression of related genes in five pepper species, Capsicum annuum L. (CA), C. baccatum L. (CB), C. chinense Jacquin (CC), C. frutescens L. (CF), and C. pubescens Ruiz & Pavon (CP) in order to screen for greenhouse species that is resistant to such adverse conditions. We observed significant reductions not only in photosynthetic pigments and stomatal conductance but also in proline, total soluble sugar, enzyme activity, and root activity; disordered arrangements of leaf palisade and spongy tissues; and first rising and then falling expression of C-repeat binding factor (CBF3) and cold-regulated genes (CORc410). These results indicate that pepper is not resistant to LTLI. We also found that CP showed significantly higher photosynthetic activity, more proline and total soluble sugar, higher enzyme activity, higher root activity, higher CBF3 and CORc410 expression levels, more tightly packed leaf palisade and spongy tissues, and thicker bundle sheath than the other four species did under LTLI, while CF exhibited the lowest values for these indicators. It demonstrated significant differences in the ability to resist to LTLI among different species, with CP showing the strongest resistance, followed by CB. Therefore, we recommend the introduction of CP and CB to greenhouse cultivation to further screen for low temperature and low light-resistant pepper varieties to increase pepper production by strengthening intervariety hybridization., L. Gas exchange, chlorophyll (Chl) fluorescence, and contents of photosynthetic pigments, soluble proteins (ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO), and antioxidant enzymes were characterized in the fully expanded 6th leaves in rice seedlings grown on either complete (CK) or on nitrogen-deficient nutrient (N-deficiency) solutions during a 20-chase period. Compared with the control plants, the lower photosynthetic capacity at saturation irradiance (Pmax) was accompanied by an increase in intercellular CO2 concentration (Ci), indicating that in N-deficient plants the decline in Pmax was not due to stomatal limitation but due to the reduced carboxylation efficiency. The fluorescence parameters ΦPS2, Fv'/Fm', electron transport rate (ETR), and qP showed the same tendency as Pmax in N-deficient plants. Correspondingly, a higher qN paralleled the rise of the ratio of carotenoid (Car) to Chl contents. However, Fv/Fm was still diminished, suggesting that photoinhibition did occur in the photosystem 2 (PS2) reaction centres. In addition, the activities of antioxidant enzymes on a fresh mass basis were gradually lowered, leading to the aggravation of membrane lipid peroxidation with the proceeding N-deficiency. The accumulation of malonyldialdehyde resulted in the lessening of Chl and soluble protein content. Analyses of regression showed PS2 excitation pressure (1 - qP) was linearly correlated with the content of Chl and inversely with soluble protein (particularly RuBPCO) content. There was a lag phase in the increase of PS2 excitation pressure compared to the decrease of RuBPCO content. Therefore, the increased excitation pressure under N-deficiency is probably the result of saturation of the electron transport chain due to the limitation of the use of reductants by the Calvin cycle. The effects of simulated acid rain on gas exchange, chlorophyll fluorescence, and anti-oxidative enzyme activity in cucumber seedlings (Cucumis sativus L. cv. Jingchun No. 4) were investigated. Acid rain significantly reduced net photosynthetic rate and mainly non-stomatal factors contributed to the decrease of photosynthesis during the experimental period. The reduced photosynthesis was associated with a decreased maximal photochemical efficiency (Fv/Fm) and the average quantum yield of the photosystem 2 (PS2) reaction centres (ΦPS2). Meanwhile, acid rain significantly increased the activities of guaiacol peroxidase (GPX) and superoxide dismutase (SOD), but decreased the activity of catalase (CAT) together with an increased content of malonyldialdehyde (MDA), Hence the changes in photosynthesis in acid rain treatment might be a secondary effect of acidity damage probably due to lipid peroxidation of lipids and proteins in thylakoid membrane rather than direct effect on PS2 reaction centre. and Jing-Quan Yu, Su-Feng Ye, Li-Feng Huang.
Coccidiosis is a parasitic disease caused by protists (apicomplexans) of the genus Eimeria Schneider, 1875 and is considered to be the most important disease faced by rabbit breeders due to its high morbidity. In the present study, the antioxidant status and changes in apoptosis and in the expression of some genes were quantified in rabbits' ilea following infection with Eimeria intestinalis Cheissin, 1948. Rabbits, orally infected with 1 × 105 sporulated oocysts of E. intestinalis, started to shed oocysts in their faeces on 8 days post infection (dpi) and reached maximum excretion on 10 dpi, with approximately 5 million oocysts. This was accompanied by a significant decrease in the live body weight of infected rabbits. Also, malondialdehyde and nitric oxide were significantly increased while catalase and glutathione were significantly decreased in the ileum tissues of the infected rabbits. In addition, a significant increase was observed in the percentages of apoptotic cells in the ilea of the infected rabbits. Furthermore, interleukin-1β and interleukin-2 mRNA levels were significantly down-regulated and mRNA levels of interleukin-6, interferon gamma and inducible nitric oxide synthase were significantly up-regulated, while those of C-reactive protein remained unchanged. a1_Reactive oxygen species (ROS) are common products of the physiological metabolic reactions, which are associated with cell signaling and with the pathogenesis of various nervous disorders. The brain tissue has the high rate of oxidative metabolic activity, high concentration of polyunsaturated fatty acids in membrane lipids, presence of iron ions and low capacity of antioxidant enzymes, which makes the brain very susceptible to ROS action and lipid peroxidation formation. Membranes of brain cortex show a higher production of thiobarbituric acid-reactive substances (TBARS) in prooxidant system (ADP.Fe3+/NADPH) than membranes from the heart or kidney. Lipid peroxidation influences numerous cellular functions through membrane-bound receptors or enzymes. The rate of brain cortex Na+ ,K+ -ATPase inhibition correlates well with the increase of TBARS or conjugated dienes and with changes of membrane fluidity. The experimental model of short-term hypoxia (simulating an altitude of 9000 m for 30 min) shows remarkable increase in TBARS in four different parts of the rat brain (cortex, subcortical structures, cerebellum and medulla oblongata) during the postnatal development of Wistar rat of both sexes. Young rats and males are more sensitive to oxygen changes than adult rats and females, respectively. Under normoxia or hypobaric hypoxia both ontogenetic aspects and sex differences play a major role in establishing the activity of erythrocyte catalase, which is an important part of the antioxidant defense of the organism. Catalase is an antioxidant enzyme the activity of which is crucial for the protection against damage caused by reactive oxygen species. The –262C>T polymorphism in the promoter region of catalase gene was found to be associated with altered catalase levels. In this study, peripheral blood mononuclear cells catalase activity was measured after H2 O2-induced oxidative stress. C/T and T/T genotypes were associated with the decrease of catalase levels in contrast to C/C donors who had elevated catalase activity in the presence of 0.4 and 0.7 mM H2 O2. We evaluated the growth and development of the medicinal species Pothomorphe umbellata (L.) Miq. under different shade levels (full sun and 30, 50, and 70 % shade, marked as I100, I70, I50, and I30, respectively) and their effects on gas exchange and activities of antioxidant enzymes. Photosynthetically active radiation varied from 1 254 µmol m-2 s-1 at I100 to 285 µmol m-2 s-1 at I30. Stomatal conductance, net photosynthetic rate, and relative chlorophyll (Chl) content were maximal in I70 plants. Plants grown under I100 produced leaves with lower Chl content and signs of chlorosis and necrosis. These symptoms indicated Chl degradation induced by the generation of reactive oxygen species. Stress related antioxidant enzyme activities (Mn-SOD, Fe-SOD, and Cu/Zn-SOD) were highest in I100 plants, whereas catalase activity was the lowest. Hence P. umbellata is a shade species (sciophyte), a feature that should be considered in reforestation programs or in field plantings for production of medicinal constituents. and J. A. Marchese ... Reactive oxygen species (ROS) have been implicated in the mechanism of postischemic contractile dysfunction, known as myocardial stunning. In this study, we examined protective effects of antioxidant enzymes, superoxide dismutase (SOD) and catalase, against ischemia/reperfusion-induced cardiac dysfunction and inhibition of Na+,K+-ATPase activity. Isolated Langendorff-perfused rabbit hearts were subjected to 15 min of global normothermic ischemia followed by 10 min reperfusion. The hearts treated with SOD plus catalase did not show significant recovery of left ventricular (LV) end-diastolic pressure compared with untreated ischemic reperfused hearts. Treatment with antioxidants had no protective effects on developed LV pressure or its maximal positive and negative first derivatives (±LVdP/dt). Myocardial stunning was accompanied by significant loss in sarcolemmal Na+,K+-ATPase activity and thiol group content. Inhibition of enzyme activity and oxidation of SH groups were not prevented by antioxidant enzymes. Net photosynthetic rate (PN) in the mother leaves was higher in the drought tolerant (DT) clones of tea (Camellia sinensis) while liberation of the fixed 14C in light from the mother leaves was higher in the drought susceptible (DS) clones. The DT clones translocated more photosynthates to the crop shoots (three leaves and a bud) from the mother leaf than the DS clones. Concentrations of RuBP carboxylase (RuBPC) or oxygenase (RuBPO) had no relationship with the drought tolerant nature of tea clones but their ratio correlated with the same. DT tea clones had higher catalase activity that could scavenge the hydrogen peroxide formed in the photorespiratory pathway and thereby reduced photorespiration rate (PR). The ratio of RuBPC/RuBPO had a positive correlation with PN and catalase activity.
