Pioglitazone (PIO) is a thiazolidindione antidiabetic agent which improves insulin sensitivity and reduces blood glucose in experimental animals and treated patients. At the cellular level the actions of PIO in diabetic heart are poorly understood. A previous study has demonstrated shortened action potential duration and inhibition of a variety of transmembrane currents including L-type Ca2+ current in normal canine ventricular myocytes. The effects of PIO on shortening and calcium transport in ventricular myocytes from the Goto-Kakizaki (GK) type 2 diabetic rat have been investigated. 10 min exposure to PIO (0.1-10 μM) reduced the amplitude of shortening to similar extents in ventricular myocytes from GK and control rats. 1 μM PIO reduced the amplitude of the Ca2+ transients to similar extents in ventricular myocytes from GK and control rats. Caffeine-induced Ca2+ release from the sarcoplasmic reticulum and recovery of Ca2+ transients following application of caffeine and myofilament sensitivity to Ca2+ were not significantly altered in ventricular myocytes from GK and control rats. Amplitude of L-type Ca2+ current was not significantly decreased in myocytes from GK compared to control rats and by PIO treatment. The effects of pleuran, ß-1,3 glucan isolated from Pleurotus ostreatus, were studied in a model of acute colitis induced by intracolonic administration of acetic acid. There was a reduction of the colonic damage score, colonic wet weight and wet/dry weight ratio 48 h after single luminal 2 % pleuran suspension pretreatment. Similar results were obtained after repeated intraperitoneal administration of pleuran in doses of 30 and 100 mg/kg. Pleuran given orally as a 10 % food component over 4 weeks was effective in reducing the extent of mucosal damage, but did not prevent the increase of myeloperoxidase in the injured colonic segment. In the segment without macroscopic evidence of inflammation, myeloperoxidase activity was significantly lower as documented by histological examination. Potato tubers, Solanum tuberosum L. cv. Bintje were stored at 4 or 20 °C. A segment of parenchyma with a bud was cultívated in vitro, in order to study the growth of plantlets according to the storage temperature and the atmosphere composition in vessels. The conservation temperature had no effect on the shoot length, the leaf area and the leaf number. On the other hand, the atmosphere composition in vessels modifíed the morphology. The plantlets cultívated in non-sealed vessels were more developed than the plantlets cultívated in hermetically dosed tubes. The photosynthetic potentíals were 4.7 times higher for the plantlets coming from the tubers stored at 20 °C than for the plantlets coming from the tubers stored at 4 °C. These potentíals were 1.7 times higher when the plantlets were cultívated in non- hermetícally dosed tubes than in hermetically dosed ones. So a conservation of tubers at 20 °C and a culture in non-enclosed vessels seemed to be the suitable conditions for the micropropagation of potatoes in viiro. Moreover, this culture type could improve the acdimatízatíon.
The purpose of this study was to examine how oxygen uptake (V.o2) in decrement-load exercise (DLE) is affected by changing rate of decrease in power output. DLE was performed at three different rates of decrease in power output (10, 20 and 30 watts ・min-1: DLE10, DLE20 and DLE30, respectively) from power output corresponding to 90 % of peak V.o2. V.o2 exponentially increased and then decreased, and the rate of its decrease was reduced at low power output. The values of V.o2 in the three DLE tests were not different for the first 2 min despite the difference in power output. The relationship between V.o2 and power output below 50 watts was obtained as a slope to estimate excessive V.o2 (ex-V.o2) above 50 watts. The slopes were 10.0±0.9 for DLE10, 9.9±0.7 for DLE20 and 10.2±1.0 ml ・min-1 ・ watt-1 for DLE30. The difference between V.o2 estimated from the slope and measured V.o2 was defined as ex-V.o2. The peak value of ex-V.o2 for DLE10 (189±116 ml ・min-1) was significantly greater than those for DLE20 and for DLE30 (93±97 and 88±34 ml ・min-1). The difference between V.o2 in DLE and that in incremental-load exercise (ILE) below 50 watts (ΔV.o2) was greater in DLE 30 and smallest in DLE10. There were significant differences in ΔV.o2 among the three DLE tests. The values of ΔV.o2 at 30 watts were 283±152 for DLE10, 413±136 for DLE20 and 483±187 ml ・min-1 for DLE30. Thus, a faster rate of decrease in power output resulted in no change of V.o2 at the onset of DLE, smaller ex-V.o2 and greater ΔV.o2. Background: Few data are available on the specific care giving-related problems of stroke patient’s caregivers and factors that influence the burden of these caregivers. Aim: To study the influences of the active rehabilitation process on anxiety, depression, care burden and perceived social support level of stroke patients caregivers. Design: A prospective clinical trial. Setting: Patients and caregivers entering a rehabilitation program at a university hospital in Turkey. Populations: Ninety patients with a first episode of stroke and 90 caregivers responsible for their care were recruited for our study. Methods: Patients and caregivers were assessed before and after the active rehabilitation process. The functional disability level of the patients was assessed by Functional Independence Measure (FIM). The Beck Anxiety Scale (BAS) and the Beck Depression Scale (BDS) were used for anxiety and depression assessment, the Zarit Care Burden Scale (ZCBS) for care burden assessment and the Multi-Dimensional Scale of Perceived Social Support (MDSPSS) for perceived social support assessment. Results: A statistically significant rise is observed in the special person sub-assessment of MDSPSS in both female and male caregivers. Also, a significant decrease in care burden, anxiety and depression levels of caregivers was noted after the rehabilitation program (p < 0.05). Conclusion: Caregivers accept the rehabilitation period as important social support in addition to the support provided by family and friends. Also, our positive results were associated with an improvement in the patients’ functional level and an increase in the acquisition of knowledge and skill required of caregivers in order to provide care during rehabilitation. Clinical Rehabilitation Impact: The rehabilitation team should be aware of the fact that the perceived care burden may be greater due to the lack of knowledge concerning available resources and due to the inability to cope with stress effectively. and A. Arterial compliance (AC) is an index of the elasticity of large arteries. Endothelial dysfunction has been reported to result in reduced arterial compliance, which represents increased arterial stiffness. A reduction in AC is elicited by high-intensity resistance training, however the mechanisms are obscure. Because a single bout of resistance exercise causes a transient increase in circulating plasma endothelin-1 in humans, some vasoconstrictors may play a role in the mechanisms. The present study aimed to investigate whether resistance training-induced decrease in AC is associated with changes in circulating vasoconstrictors levels in young men. Young sedentary men were assigned to control (n=5) or training (n=9) groups. The training group performed four-week high-intensity resistance training (weight training exercise; three sessions/week). We measured AC and plasma levels of endothelin-1, angiotensin II, and norepinephrine before and after intervention. Resistance training significantly decreased AC, whereas the changes in plasma levels of neither endothelin-1, nor angiotensin II, nor norepinephrine were significantly different between the control and the training groups. Moreover, we found no significant correlations between changes in circulating plasma levels (endothelin-1, angiotensin II, and norepinephrine) and in the AC. Despite of no alteration of the resting circulating plasma levels (endothelin-1, etc.), we cannot exclude a possibility that the tissue/local concentrations of vasoconstrictors (endothelin-1, etc.) around the vessels might be increased and also involved in a reduction of AC in the training group. Acute lung injury (ALI) is associated with det erioration of alveolar-capillary lining and transmigration and activation of inflammatory cells. Whereas a selective phosphodiesterase-4 (PDE4) inhibitor roflumilast has exerted potent anti-inflammatory properties, this study evaluated if its intravenous delivery can influence inflammation, edema formation, and respiratory parameters in rabbits with a lavage-induced model of ALI. ALI was induced by repetitive saline lung lavage (30 ml/kg). Animals were divided into 3 groups: ALI without therapy (ALI), ALI treated with roflumilast i.v. (1 mg/kg; ALI+Rofl), and healthy ventilated controls (Control), and were ventilated for following 4 h. Respiratory parameters (blood gases, ventilatory pressures, lung compliance, oxygenation indexes etc.) were measured and ca lculated regularly. At the end of experiment, animals were overdosed by anesthetics. Total and differential counts of cells in bronchoalveolar lavage fluid (BAL) were estimated microscopically. Lung edema was expressed as wet/dry lung weight ratio. Treatment with roflumilast reduced leak of cells (P<0.01), particularly of neutrophils (P<0.001), into the lung, decreased lung edema formation (P<0.01), and improved respiratory parameters. The invasion of Spartina alterniflora along the coasts of China has allowed this C4 grass to outcompete often much of the native, salt marsh vegetation, such as Phragmites australis (C3 grass), in the Yangtze Estuary. In this study, native grass, P. australis, and
non-native grass, S. alterniflora, were grown in fresh and saline water (moderate salinity of 15‰ and high salinity of 30‰) to compare the effects of salinity on photosynthetic and biochemical parameters in combination with measurement temperatures. The C4 grass, S. alterniflora, showed a greater CO2 assimilation rate than P. australis, across the tested temperatures. The net photosynthetic rate declined significantly with increasing salinity as a result of inhibited stomatal conductance together with a greater decrease in the maximum rate of electron transport (Jmax). In P. australis, salt treatments shifted the optimum temperatures for the maximum rate of carboxylation by Rubisco (Vcmax) and J max to lower temperatures. S. alterniflora showed a greater salt tolerance to moderate stress than that of the native grass, with lower sensitivity of V cmax, Jmax, and the maximum rate of phosphoenolpyruvate carboxylation. Both moderate and high stress decreased significantly stomatal conductance of S. alterniflora; high salinity reduced significantly photosynthetic efficiency and Jmax. Our findings indicated that the combination of stomatal conductance, enzyme activity, and electron transport affected the photosynthetic performance of the plants in response to salt treatments. The success of S. alterniflora could be probably attributed to its C4 photosynthetic pathway and the tolerance to moderate salinity. The effects of NaCl treatment were analysed in two species of considerably different resistance. In glycophyte, the content of ascorbate decreased but lipophilic antioxidants (α-tocopherol, plastochromanol, and hydroxy-plastochromanol) increased due to 150 mM NaCl. In halophyte, 300 mM NaCl caused a significant increase in hydrophilic antioxidants (ascorbate, total glutathione) but not in the lipophilic antioxidants. The redox states of plastoquinone (PQ) and P700 were also differently modulated by salinity in both species, as illustrated by an increased oxidation of these components in glycophyte. The presented data suggest that E. salsugineum was able to avoid a harmful singlet oxygen production at PSII, which might be, at least in part, attributed to the induction of the ascorbate-glutathione cycle. Another important cue of a high salinity resistance of this species might be the ability to sustain a highly reduced states of PQ pool and P700 under stress, which however, drastically affect the NADPH yield., M. Plant essential oils (EOs) have been reported to have health benefit properties and their preventive and therapeutic use in animals is expected to increase in the future. We evaluated the influence of five essential oils obtained from plant species which are known to have positive antimicrobial, antioxidative and anti-inflammatory effects – sage EO from Salvia officinalis L. (Lamiaceae), oregano EO from Origanum vulgare L. (Lamiaceae), thyme EO from Thymus vulgaris L. (Lamiaceae), clove EO from Syzygium aromaticum L. (Myrtaceae) and cinnamon EO from Cinnamomum zeylanicum Blume (Lauraceae) on the growth and development of mouse preimplantation embryos in vivo. Essential oils were added to commercial diet at concentrations of 0.25 % for sage EO, thyme EO, clove EO, cinnamon EO and 0.1 % for oregano EO, and fed to ICR female mice for 2 weeks ad libitum. Females were then mated with males of the same strain. Embryos obtained on Day 4 of pregnancy at the blastocyst stage were stained by morphological triple staining (Hoechst, PI, Calcein-AM) and evaluated using fluorescent microscopy. The effects of essential oils were estimated by the viability of embryos, number of nuclei and distribution of embryos according to nucleus number. Cinnamon EO significantly decreased the number of nuclei and the distribution of embryos according to nucleus number was significantly altered. Sage EO negatively influenced the distribution of embryos according to nucleus number. Clove and oregano EOs induced a significantly increased rate of cell death. Only thyme EO had no detectable effects on embryo development. In conclusion, none of the essential oils had any positive effect on embryo development, but some of them reduced the number of cells and increased the incidence of cell death., M. Domaracký, P.
