5-aminolevulinic acid (ALA) is an essential precursor for the biosynthesis of tetrapyrrols such as heme and chlorophyll (Chl). Previous studies have focused mainly on promotive effects of exogenous ALA on plant growth, while regulatory mechanisms affecting Chl biosynthesis have been only partially discussed. In the present study, the ameliorative role of exogenous ALA was investigated on Chl and endogenous ALA biosynthesis in six-day-old etiolated oilseed rape (Brassica napus L.) cotyledons during the de-etiolation stage. We showed that exogenously applied ALA of a low dosage enhanced Chl and ALA accumulation in cotyledons, while 600 µM ALA treatment inhibited the accumulation of Chl and ALA severely. However, the gene expression levels of glutamyl-tRNA reductase (HEMA) and glutamate-1-semialdehyde aminotransferase (GSA) were not affected under either low or high ALA concentrations. Furthermore, water deficit induced by polyethylene glycol 6000 (PEG) suppressed the Chl and ALA accumulation in cotyledons, while the inhibition was partially alleviated in the cotyledons pretreated with ALA. The decrease in Chl biosynthesis induced by PEG stress was assumed to be related to downregulation of HEMA and Mg-chelatase ChlH (ChlH), and upregulation of ferrochelatase (FC) genes. Moreover, exogenously applied ALA did not show any effect on the expression of Chl synthesis-related genes under the PEG treatment. These results showed a difference in suppressing ALA synthesis due to the high concentration of ALA and PEG. Exogenously applied ALA did not affect the expression of HEMA and GSA, thus exogenous ALA regulated Chl synthesis not via the regulation of transcriptional level in ALA biosynthesis. The influence of a cytokinin, 6-benzylaminopurine (BAP), on chloroplast structure was studied using biochemical methods and electron microscopy. The average degree of thylakoid stacking was determined by digitonin fractionation and differential centrifugation of chloroplasts from tobacco plantlets after treatment with different concentrations of BAP in agar medium during cultivation in viíro. An elevated concentration of BAP in the medium induced an increase in grana stacking. This was in accordance with the lowering of the chlorophyll a/b ratio in these chloroplasts. The relative amoímt of proteins and carotenoids increased in both stromal and (to a lesser extent) granal chloroplast thylakoid fractions with the BAP concentration. The electron microscopic studies revealed nearly the same volume density of thylakoid membranes within chloroplasts of BAP treated plantlets and control ones. In the BAP treated plantlets the chloroplasts were smaller and had a profound accumulation of starch inclusions and a more flattened shape than the chloroplasts of control plantlets. The effects of polyamines (PAs) on salt stress in Bakraii (Citrus reticulata × Citrus limetta) seedlings were studied. Foliar treatments by putrescine (Put), spermidine (Spd), and spermine (Spm) (0, 0.5, and 1 mM) were applied during the salinity period
(0 and 75 mM of NaCl). PA-treated seedlings showed a lower content of Na+ and Cl- in leaves. Application of PAs increased net photosynthetic rate in salt-stressed plants and it contributed to the enhanced growth parameters. PAs application considerably induced growth improvement in Bakraii seedlings which was found to be associated with reduced electrolyte leakage, increased relative water content, chlorophyll fluorescence parameters, activities of key antioxidant enzymes, as well as increased photosynthetic pigment concentration under saline regime. Salicylic acid (SA) is a common, plant-produced signal molecule that is responsible for inducing tolerance to a number of biotic and abiotic stresses. Our experiment was therefore conducted to test whether the application of SA at various concentrations (0, 0.10, 0.50, and 1.00 mM) as a foliar spray would protect citrus seedlings (Valencia orange/Bakraii) subjected to salt stress (0, 25, 50, and 75 mM NaCl). Growth parameters, leaf chlorophyll (Chl) content, relative water content (RWC), maximal quantum yield of PSII photochemistry (Fv/Fm), and gas-exchange variables were negatively affected by salinity. In addition, leaf electrolyte leakage (EL) and proline content increased by salinity treatments. Application of SA increased net photosynthetic rate and proline content in salt stressed plants and may have contributed to the enhanced growth parameters. SA treated plants had greater Chl content and RWC compared with untreated plants when exposed to salt stress. Fv/Fm ratio and stomatal conductance were also significantly higher in SA treated plants under saline stress conditions. SA application reduced EL compared to untreated plants, indicating possible protection of integrity of the cellular membrane. It appeared that the best ameliorative remedies of SA were obtained when Valencia orange/Bakraii seedlings were sprayed by 0.50 and 1.00 mM solutions. The effects of gemfibrozil (GFZ), an antihyperlipidemic agent, on the anionic transport of the human red blood cells (RBC) during the oxygenation-deoxygenation cycle were examined. Gemfibrozil clearly plays a role in the modulation of the anionic flux in erythrocytes; in fact it causes a strong increment of anions transport when the RBCs are in the high-oxygenation state (HOS). Such an effect is remarkably reduced in the low-oxygenation state (LOS). With the aim of identifying the dynamics of fibrate action, this effect has been investigated also in human ghost and chicken erythrocytes. These latter, in fact, are known to possess a B3 (anion transporter or Band 3) modified at the cytoplasmic domain (cdb3) which plays a significant role in the metabolic modulation of red blood cells. The results were analyzed taking into account the well-known interactions between fibrates and both conformational states of hemoglobin i.e. the T state (deoxy-conformation) and the R state (oxy-conformation). Surgical Plethysmographic Index (SPI), calculated from pulse photo-plethysmographic amplitude oscillations, has been proposed as a tool to measure nociception anti-nociception balance during general anesthesia, but it is affected by several confounding factor that alter the autonomic nervous system (ANS) modulation. We hypothesized that SPI may be mainly affected by sympathetic stimulation independently from nociception. We studied the effects of two sympathetic stimuli on SPI, delivered through passive head-up tilt at 45 and 90 degrees angles, in nine awake healthy adults. The sympathetic modulation was assessed by means of heart rate variability (HRV) analysis. Mean (SD) SPI significantly increased from baseline to 45 degrees [from 38.6 (13.7) to 60.8 (7.6), p<0.001)] and to 90 degrees angle tilt [82.3 (5.4), p<0.001]. The electrocardiographic mean R-to-R interval significantly shortened during both passive tilts, whereas systolic arterial pressure did not change during the study protocol. HRV changed significantly during the study protocol towards a predominance of sympathetic modulation during passive tilt. Gravitational sympathetic stimulation at two increasing angles, in absence of any painful stimuli, affects SPI in awake healthy volunteers. A low birth weight is a new risk factor for the development of premature atherosclerosis. The effect of intrauterine undernutrition on hypercholesterolemia in later life was studied in an experimental model using the Prague Hereditary Hypercholesterolemic (PHHC) rat. Compared to animals in the control group (Wistar rats), animals with an increased sensitivity to high-cholesterol diet (PHHC rats) display hypercholesterolemia. Only in PHHC animals, individuals undernourished in their intrauterine life (hypotrophic group, HG) had a significantly higher total cholesterol, compared with individuals without food restriction in pregnancy (eutrophic group, EG). Restricted food intake in pregnancy led to smaller nests and a decreased number of pups in each litter. We found no significant diferences in birth weight between HG and EG. In spite of similar birth weights in PHHC and Wistar rats, intrauterine undernutrition caused an increase in cholesterolemia in the HG group of the PHHC rats. The effect of intrauterine undernutrition on the development of hypercholesterolemia will most likely play a role in individuals with geneticaly determined increased susceptibility to a high-cholesterol diet. Java citronella suffers from chlorosis of the younger leaves, particularly in tropical climatic conditions. An Fe-efFicient genotype of this species was grown in Solutions containing 0 to 44.80 g(Fe) m'^ in controlled glasshouse conditions. In comparison with normál Fe nutrition (5.60 g m"^), low (0 - 1.40 g m'^) or high (22.40 - 44,80 g m*^) iron nutrition influenced fresh and dry matter yield, plant height, total essential oil and citronellol contents, chlorophyll (Chl) content, net photosynthetic and transpiration rates, and Fe uptake. Largest differences were found in saccharide and Chl a and h contents and Chl a/h ratio, and stomatal resistance. Significarít positive correlations were observed between fresh matter, citronellol content, dry and fresh matter yields and total essential oil content.
In healthy subjects, the right ventricular filling pattern estimated from tricuspid valve inflow is highly load-dependent. This can be clearly demonstrated by changes of Doppler inflow tracings recorded during tidal breathing at rest. The aim of our study was to test the magnitude of tricuspid inflow changes during more pronounced load changes induced by specific maneuvers. In 31 apparently healthy subjects (16 men and 15 women, mean age 36±16 years) we recorded pulsed Doppler tracings of tricuspid inflow during forced inspiration, elevation of lower extremities (increased preload) and handgrip exercise (increased afterload). The obtained values were compared with end-expiratory phase of tidal breathing at rest. We found a significantly larger increase in the early and late filling velocities (for E and A p<0.001) under the conditions of increasing preload (elevation of legs) and less pronounced, but still significant changes with isometric exercise (for E p<0.001 and for A p<0.01). We conclude that the right ventricular filling pattern in healthy humans is highly load-sensitive and for this reason the effect of any intervention (e.g. pharmacological) must be studied under strict and well-defined resting conditions., E. Influence of different phosphorus concentrations was studied in four rice varieties (Akhanphou, MTU1010, RP BIO 226, and Swarna) differing in their tolerance to low phosphorus. There was an increase in shoot and root dry mass with the increase in phosphorus concentration. At the low phosphorus concentration at both tillering and reproductive stages, Swarna, followed by Akhanphou, recorded maximum biomass for both roots and shoots, while the minimum was observed in RP BIO 226. Reduction in photosynthetic rate, stomatal conductance, transpiration rate, and internal CO2 concentration at low phosphorus concentrations were observed at both tillering and reproductive stages in all the genotypes. In low phosphorus, maximum photosynthetic rate was found in Swarna followed by Akhanphou. Phosphorus deficiency did not alter the maximum efficiency of PSII photochemistry, however, there was a reduction in effective PSII quantum yield, electron transport rate, and coefficient of photochemical quenching, while the coefficient of nonphotochemical quenching was higher in the low phosphorus-treated plants. Prolonged exposure to excessive energy and failure to utilize the energy in carbon-reduction cycle induced the generation of reactive oxygen species, which affected PSII as indicated by the fluorescence traits. The reduction was less severe in case of Swarna and Akhanphou. The activities of superoxide dismutase, peroxidase, and catalase increased in roots under low phosphorus concentration indicating that photoprotective mechanisms have been initiated in rice plants in response to phosphorus deficiency. Comparatively, Swarna and Akhanphou exhibited a higher biomass, higher photosynthetic rate, and better reactive oxygen species-scavenging ability which conferred tolerance under low phosphorus conditions., N.
