Postnatal heart development is characterized by critical periods of heart remodeling. In order to characterize the changes in the lipophilic fraction induced by free radicals, fatty acids and t heir oxidized products, lipofuscin -like pigments (LFP), were investigated. Fatty acids were analyz ed by gas chromatography and LFP were studied by fluorescence techniques. A fluorophore characterized by spectral methods was further resolved by HPLC. Major changes in the composition of fatty acids occurred immediately after birth and then during maturation. Fluorescence of LFP changed markedly on postnatal days 1, 4, 8, and 14, and differed from the adult animals. LFP comprise several fluorophores that were present since fetal state till adulthood. No new major fluorophores were formed during development, just the abundances of individual fluorophores have been modulated which produced changes in the shape of the spectral arrays. HPLC resolved the fluorophore with excitation maximum at 360 nm and emission maximum at 410 nm. New chromatographically distinct species appeared immediately on postnatal day 1, and then on days 30 and 60. Non-photochemical quenching of chlorophyll fluorescence (qN) and its three components (qNf, qNm, and qNs) in the flag leaves of wheat grown in the field were studied by a fluorometer PAM-2000 on clear days. The diurnal variation patterns of qN in just fully extended (JFEL) and aging leaves (AL) were similar, but qNm declined markedly in JFEL while it remained at a relatively high level in AL under strong sunlight at noon. Furthermore, at midday qNf was higher than qNs in JFEL, but much lower in AL. The results show the relative contributions of different mechanisms in preventing the photosynthetic apparatus from photodamage change during leaf development. and S.-S. Hong ... [et al.].
By using a wild-type rice (Oryza sativa L. cv. Norin No. 8) and the chlorophyll (Chl) b-deficient mutant derived from Norin No. 8 (chlorina 11), the present study monitored the oxygen evolution, contents of Chl a and b, β-carotene, and lutein in leaf and the contents of cytochrome f, and the reaction centres of photosystem I (PSI) and photosystem II (PSII) in thylakoids. The oxygen evolution, maximal quantum yield of PSII (Fv/Fm) and Chl concentration remained constant in both Norin No. 8 and chlorina 11 under 5 and 2% of full sunlight for six days. On the other hand, on the thylakoid level, the PSII reaction centre of chlorina 11 was more stable even under high irradiance, while approximately 40% decrease in levels of the PSII reaction centre occurred under 2% of full sunlight for six days. However, under such conditions, by regulating the stoichiometry of active PSII and PSI centres, the light absorption balance in both rice types was adjusted between the two photosystems. The present study attempted to examine whether the light absorption balance between PSII and PSI is altered to effectively conduct photosynthesis in the wild-type and Chl b-deficient mutant rice seedlings. and J. Changes in the pools of carotenoids and protochlorophyll(ide) were investigated in etiolated cucumber cotyledons treated with norflurazon (NF) and an experimental herbicide KC 6361 (KC). Both the NF- and the KC-treated tissues considerably accumulated the colourless carotenes phytoene and phytofluene with a concomitant depletion of the coloured carotenoids lutein and β-carotene in darkness. However, the profiles of changes in chlorophylls (Chls) and carotenoids were different for the two herbicides. The plants were also influenced by the photosynthetic photon flux densities (PPFD's), with a more pronounced decline of Chl under high PPFD than under low PPFD. The ratios of protochlorophyll (PChl)/protochlorophyllide (PChlide) were greatly altered due to a decrease and an increase of PChl in the NF- and the KC-treated etiolated tissues, respectively, whereas the PChlide content was not significantly influenced by the inhibitors. Large increase of PChls in the KC-treated tissues seems to derive from the binding of accumulated geranylgeraniol (GG) equivalents, through carotenogenic inhibition, to PChlide. Therefore, the alterations of PChl and PChlide occurring under disturbed carotenogenesis may suggest an interaction between the biosynthetic pathways of Chls and carotenoids. Free radicals and proinflammatory cytokines from phagocytes have been implicated in the pathogenesis of endotoxic shock, a disease with high mortality caused by Gram-negative bacterial endotoxin. In the present study, male BALB/c and Swiss mice received intraperitoneally lipopolysaccharide (LPS) at 100 mg/kg and 150 mg/kg, respectively, that led to a lethal endotoxic shock (100 % of mortality before 30 h). Swiss mice injected with 100 mg/kg, that did not show lethal endotoxic shock, were also studied. Peritoneal macrophages were obtained from animals at 2, 4, 12 or 24 h after injection of LPS or saline (control) solutions. Superoxide anion and tumor necrosis factor (TNFα) production were determined in these cells as well as other functions such as adherence capacity, chemotaxis and phagocytosis. The increase in superoxide anion production after endotoxin injection was higher in cells from mice with lethal shock than in those with non-lethal shock. However, the enhancement of TNFα production was similar in all cases, although in Swiss mice the highest levels of TNFα were observed at 1.5 h after endotoxin injection, while in BALB/c mice they occurred at 2 h after LPS injection. This oxidative stress was also revealed by the other functions analyzed, since adherence to substrate and phagocytosis were stimulated and chemotaxis was decreased after endotoxin injection as compared to controls, the differences being even more significant in animals with lethal shock. A comparison between maximum quantum yield of PSII photochemistry (Fv/Fm) and chlorophyll fluorescence decrease ratio (Rfd) for low and high temperature resistance was assessed in a seasonal study of the acclimation in Pterocephalus lasiospermus. Analyzing the regression adjustment of both parameters and the lethal temperatures (LT50), Rfd resulted in being a more sensitive indicator for low and high temperature treatments, since the thermic resistance estimated with Rfd parameter was never higher than those estimated with Fv/Fm. Furthermore, the use of Fv/Fm led to an overestimation of the acclimation phenomena, with 6ºC of a maximum difference between both parameters. Using Rfd as the indicator parameter, P. lasiospermus acclimated to low temperatures but it kept on being a sensitive species (the lowest LT50 values only achieved -9.9 ± 0.3ºC). However, no heat acclimation was observed (LT50 around 43.5ºC). French bean (Phaseolus vulgaris L.) cotyledons lost most of their reserve substances during several early days of germination and turned green. In cotyledon mesophyll cells of one-week-old seedlings, plastids were represented predominantly by amyloplasts (starch grains) and chloroamyloplasts, and the cells appeared to be metabolically highly active. Cell heterogeneity associated with distance of the cells from cotyledon vascular bundles was evident. Only mesophyll cells near to the bundles were rich in plastids. In two-weeks-old intact bean plants, the cotyledons were yellow and shrunken, and their cells were nearly "empty". The plastids in them were represented by senescent plastids (gerontoplasts) only. In the gerontoplasts as well as freely in cytosol, fluorescent lipoid inclusions were accumulated. This cotyledon development was more or less independent of irradiance. Water availability is a major limiting factor in desert ecosystems. However, a winter snowfall role in the growth of biological soil crusts is still less investigated. Here, four snow treatments were designed to evaluate the effects of snow depth on photosynthesis and physiological characteristics of biological soil crusts. Results showed that snow strongly affected the chlorophyll fluorescence properties. The increased snow depth led to increased contents of photosynthetic pigments and soluble proteins. However, all biological soil crusts also exhibited a decline in malondialdehyde and soluble sugar contents as snow increased. Results demonstrated that different biological soil crusts exhibited different responses to snow depth treatment due to differences in their morphological characteristics and microhabitat. In addition, interspecies differentiation in response to snow depth treatment might affect the survival of some biological soil crusts. Quantitative and qualitative changes of serum proteins, apart from glycation, have not been sufficiently studied in streptozotocin-induced diabetic rats (D), the most common experimental model for diabetes. Thus, we decided to analyze the serum of diabetic rats by concanavalin A-blotting in comparison with rats with acute inflammation induced by fermented yeast (Y), in which characteristic alterations of serum proteins have been described. Two months after the streptozotocin treatment, the blood glucose levels were highly elevated (456± 24 vs. 124± 10 mg/dl, p<0.001, n=12), the body weight was significantly lower than normal (279± 10 vs. 392± 6 g, p<0.001, n=12), and serum proteins appeared to be highly glycated (p<0.001) when analyzed by the fructosamine assay, without any significant change in the total serum protein concentration. Analysis by concanavalin A-blotting, revealed a significant decrease of a1-inhibitor-3 (a1-I3, p<0.05) and an increase of the b chain of haptoglobin (b-Hp, p<0.05) in both D and Y rats (n=3) compared with control animals. However, acute inflammation caused a marked rise of two prominent acute phase proteins, a2-macroglobulin and hemopexin, which did not change appreciably in diabetic rats. Further work will be necessary to evaluate the physiopathological significance of these phenomena which could result from changes of both concentration and glycosylation of the aforementioned proteins., L. Specific neuronal populations are known to express calcium binding proteins (CBP) such as calbindin (CB), parvalbumin (PV) and calretinin (CR). These CBP can act as calcium buffers that modify spatiotemporal characteristics of intracellular calcium transients and affect calcium homeostasis in neurons. It was recently shown that changes in neuronal CBP expression can have significant modulatory effect on synaptic transmission. Spinothalamic tract (STT) neurons form a major nociceptive pathway and they become sensitized after peripheral inflammation. In our experiments, expression of CBP in STT neurons was studied in a model of unilateral acute knee joint arthritis in rats. Altogether 377, 374 and 358 STT neurons in the segments L3-4 were evaluated for the presence of CB, PV and CR. On the contralateral (control) side 11 %, 9 % and 47 % of the retrogradely labeled STT ne urons expressed CB, PV and CR, respectively. On the ipsilateral (arthritic) side there was significantly more CB (23 %) and PV (25 %) expressing STT neurons, while the number of CR positive neurons (50 %) did not differ. Our results show increased expression of fast (CB) and slow (PV) calcium binding proteins in STT neurons after induction of experimental arthritis.
