Ionizing radiation and somatostatin analogues are used for acromegaly treatment to achieve normalization or reduction of growth hormone hypersecretion and tumor shrinkage. In this study, we investigated a combination of somatostatin (SS14) with ionizing radiation of 60Co and its effect on reparation of radiation-induced damage and cell death of somatomammotroph pituitary cells GH3. Doses of γ-radiation 20-50 Gy were shown to inhibit proliferation and induce apoptosis in GH3 cells regardless of somatostatin presence. It has been found that the D0 value for GH3 cells was 2.5 Gy. Somatostatin treatment increased radiosensitivity of GH3 cells, so that D0 value decreased to 2.2 Gy. We detected quick phosphorylation of histone H2A.X upon irradiation by the dose 20 Gy and its colocalization with phosphorylated protein Nbs-1 in the site of double strand break of DNA (DSB). Number of DSB decreased significantly 24 h after irradiation, however, clearly distinguished foci persisted, indicating non repaired DSB, after irradiation alone or after combined treatment by irradiation and SS14. We found that SS14 alone triggers phosphorylation of Nbs1 (p-Nbs1), which correlates with antiproliferative effect of SS14. Irradiation also increased the presence of p-Nbs1. Most intensive phosphorylation of Nbs1 was detected after combined treatment of irradiation and SS14. The decrease of the number of the DSB foci 24 h after treatment shows a significant capacity of repair systems of GH3 cells. In spite of this, large number of unrepaired DSB persists for 24 h after the treatment. We conclude that SS14 does not have a radioprotective effect on somatomammotroph GH3 cells., M. Řezáčová, J. Sophoridine is a type of alkaloid extract derived from the Chinese herb Sophora flavescens Ait (kushen) and possess a variety of pharmacological effects including anti- inflammation, anti - anaphylaxis, anti - cancer, anti - arrhythmic and so on. However, the effect of sophoridine on heart failure has not been known yet. In this study, the effect of sophoridine on heart failure was investigated using Sprague -Dawley (SD) rat model of chronic heart failure. Morphological results showed that in medium and high dose group, myofilaments were arranged orderly and closely, intermyofibrillar ly sis disappeared and mitochondria contained tightly packed cristae compared with heart failure group. We investigated the Ca 2+ induced Ca 2+ transients and assessed the expression of ryanodine receptor (RyR2) and L-type Ca 2+ channel (dihydropyridine receptor, DHPR). We found that the cytosolic Ca 2+ transients were markedly increas ed in amplitude in medium ( ΔF/F 0 =43.33±1.92 ) and high dose groups ( ΔF/F 0 =47.21 ±1.25 ) compared with heart failure group ( ΔF/F 0 =16.7±1.29, P <0.0 1), Moreover, we demonstrated that the expression of cardiac DHPR was significantly increased in medium - and high dose -group compared with heart failure rats. Our results suggest that sophoridine could improve heart failure by ameliorating cardiac Ca 2+ induced Ca 2+ transients, and that thi s amelioration is associated with upregulation of DHPR., S.-T. The aim of the present study was to investigate changes in the activity of branched-chain a-keto acid dehydrogenase (BCKAD) in skeletal muscle and the heart during brief and prolonged starvation. Fed control rats and rats starved for 2, 4 and 6 days were anesthetized with pentobarbital sodium before heart and hindlimb muscles were frozen in situ by liquid nitrogen. Basal (an estimate of in vivo activity) and total (an estimate of enzyme amount) BCKAD activities were determined by measuring the release of 14CO2 from a-keto[1-14C]isocaproate. The activity state of BCKAD complex was calculated as basal activity in percentages of total activity. Both basal and total activities and the activity state of the BCKAD were lower in skeletal muscles than in the heart. In both tissues, starvation for 2 or 4 days caused a decrease in the basal activity and activity state of BCKAD. On the contrary, in the heart and muscles of animals starved for 6 days a marked increase in basal activity and activity state of BCKAD was observed. The total BCKAD activity was increasing gradually during starvation both in muscles and the heart. The increase was significant in muscles on the 4th and 6th day of starvation. The demonstrated changes in BCKAD activity indicate significant alterations in branched-chain amino acid (BCAA) and protein metabolism during starvation. The decreased BCKAD activity in skeletal muscle and heart observed on the 2nd and 4th day of starvation prevents the loss of essential BCAA and is an important factor involved in protein sparing. In order to evaluate the effect of static magnetic field (SMF) on morphological and physiological responses of soybean to water stress, plants were grown under well-watered (WW) and water-stress (WS) conditions. The adverse effects of WS given at different growth stages was found on growth, yield, and various physiological attributes, but WS at the flowering stage severely decreased all of above parameters in soybean. The result indicated that SMF pretreatment to the seeds significantly increased the plant growth attributes, biomass accumulation, and photosynthetic performance under both WW and WS conditions. Chlorophyll a fluorescence transient from SMF-treated plants gave a higher fluorescence yield at J-I-P phase. Photosynthetic pigments, efficiency of PSII, performance index based on absorption of light energy, photosynthesis, and nitrate reductase activity were also higher in plants emerged from SMF-pretreated seeds which resulted in an improved yield of soybean. Abundant evidence indicates that ATP and adenosine act as neurotransmitters or co-transmitters, influencing nerve cell physiology in various ways. Therefore, regulation of ATP-metabolizing enzymes is essential for the normal development and function of neuronal tissue. In the present study we have examined the effect of gonadal (OVX) or adrenal (ADX) steroid hormone deprivation on the activity and expression of synaptic membrane ecto-ATPase in three extrahypothalamic brain areas of female rats, primarily not associated with reproductive function. It was shown that OVX significantly increased ecto-ATPase activity and the relative abundance of this enzyme in the hippocampal (Hip) and caudate nucleus (CN), but not in brain stem (BS) membrane preparations. ADX was followed by an upregulation of the enzyme activity and its relative abundance in all the brain areas investigated. The highest enzyme activity and the most profound effects of OVX and ADX were detected in the CN. The results obtained indicate that ADX and OVX upregulate the expression of ecto-ATPase, potentiating the production of adenosine in synaptic cleft thus modulating the activity of numerous neurotransmitter systems in distinct areas of the CNS., N. Activation of sublobule IX-b of the cerebellar vermis evokes hypotension, bradycardia and decrease of the phrenic nerve activity in the anesthetized animal. Cardiac performance during the isovolumic phases of systole and relaxation can be evaluated by dP/dt max, Vpm, dP/dt/DP40 and τ, respectively. In the present study, we evaluated the changes on cardiac function evoked by the stimulation of sublobule IX-b. New Zealand white rabbits were anesthetized, paralyzed and artificially ventilated. A posterior craniotomy was made to reveal and stimulate the cerebellar uvula (4 s train; 50 Hz; 1 ms; 20 μA). The femoral artery and veins were cannulated and a Swan-Ganz catheter was advanced in the upper abdominal aorta to control afterload when inflating the balloon. The left ventricle was catheterized with a Millar catheter. Blood pressure, heart rate, left ventricular pressure were monitored. Results showed a significant decrease on sublobule IX-b stimulation of all the indices of systolic function and an increase of τ indicating a decrease in the speed of the relaxation. These data provide the first evidence of the influence of sublobule IX-b on cardiac function. This study investigates the effects of long-term treatment with sulodexide (SLX) on norepinephrine (NE)-induced contractions, acetylcholine(Ach)-induced relaxations, acute cyclooxygenase blockade by diclofenac (DIC) in isolated femoral arteries (FA) and the parameters of oxidative phosporylation in liver mitochondria. 15-weeks old Wistar rats were divided into four groups: control (C; injected with saline solution), treated control (C+SLX), diabetic (DM) and treated diabetic (DM+SLX). Diabetes was induced with a single i.v. dose of streptozotocin (STZ) 45 mg.kg-1. SLX was administered i.p., at dose 100 IU.kg-1 daily for 5 weeks. Vascular responses of isolated femoral arteries were measured using Mulvany-Halpern myograph. Respiratory function of the mitochondria was determined using voltamperometric method on oxygraph Gilson. In diabetic rats the amplitude of maximal response to NE was elevated. DIC pretreatment decreased the amplitudes of NE-induced contractions in all groups of rats. SLX treatment decreased sensitivity of FA to NE and caused higher relaxatory responses to Ach in C and DM. Oxygen consumption and phosphorylation rates ([QO2(S3)], [QO2(S4)] and (OPR)) and respiratory control ratio (RCR) were decreased in the mitochondria of DM rats. Mitochondria of C rats were not affected with SLX treatment. Administration of SLX in DM rats was associated with increase of RCR, other parameters were not affected. A numerical-analytical model has been developed to investigate the effect of the coverage density of metallic nanoparticles on the interplay between Raman gain and attenuation of guided mode in SERSactive optical fibers. The model was applied to different types of SERS-active fibers: a single-mode suspended-core photonic crystal fiber with silver nanoparticles immobilized on the walls of cladding channels and multi-mode unclad sapphire fiber with silver nanoparticles on its outer cylindrical surface. The predicted Raman intensity curves as functions of nanoparticle coverage density, covered fiber length and excited modes are in a good agreement with the published experimental results for the contrasting types of SERS-active fibers. and Numericko-analytický model byl vyvinut pro vyšetření vlivu hustoty pokrytí kovových nanočástic na souhru Ramanova zesílení a útlumu vedeného vidu v SERS-aktivních optických vláknech. Tento model byl aplikován na dva odlišné typy SERS-aktivních vláken: na jednovidové fotonické krystalové vlákno typu ''suspended-core'' se stříbrnými nanočásticemi immobilizovaných na stěnách plášťových kanálů a na mnohovidové bezplášťové safírové vlákno se stříbrnými nanočásticemi na jeho válcovém povrchu. Předpovězené křivky Ramanovy intenzity jako funkce hustoty pokrytí nanočástic, délky vlákna pokryté nanočásticemi a excitovaných vidů jsou v dobré shodě s publikovanými experimentálními výsledky pro uvedené kontrastní typy SERS-aktivních vláken.

The empirical conversion equations available from literature are reviewed. The methods of converting the pump characteristics at clear water to those which apply for slurries have been incorporated into a straightforward, user-friendly computer program which can be run on a PC. The program may be adopted in the design of hydro-transportation systems involving hydrodynamic dredge pumps. In the Orinoco lowlands, savannas have been often replaced by pastures composed of the C4 grass, Brachiaria decumbens Stapf. We addressed following questions: (1) How does the replacement of the native vegetation affect CO2 exchange on seasonal and annual scales? (2) How do biophysical constraints change when the landscape is transformed? To assess how these changes affect carbon exchange, we determined simultaneously the CO2 fluxes by eddy covariance, and the soil CO2 efflux by a chamber-based system in B. decumbens and herbaceous savanna stands. Measurements covered a one-year period from the beginning of the dry season (November 2008) to the end of the wet season (November 2009). During the wet season, the net ecosystem CO2 exchange reached maximum values of 23 and 10 μmol(CO2) m-2 s-1 in the B. decumbens field and in the herbaceous savanna stand, respectively. The soil CO2 efflux for both stands followed a temperature variation during the dry and wet seasons, when the soil water content (SWC) increased above 0.087 m3 m-3 in the latter case. Bursts of CO2 emissions were evident when the dry soil experienced rehydration. The carbon source/sink dynamics over the two canopies differed markedly. Annual measurements of the net ecosystem production indicated that the B. decumbens field constituted a strong carbon sink of 216 g(C) m-2 y-1. By contrast, the herbaceous savanna stand was found to be only a weak sink [36 g(C) m-2 y-1]. About 53% of the gross primary production was lost as the ecosystem respiration. Carbon uptake was limited by SWC in the herbaceous savanna stand as evident from the pattern of water-use efficiency (WUE). At the B. decumbens stand, WUE was relatively insensitive to SWC. Although these results were specific to the studied site, the effect of land use changes and the physiological response of the studied stands might be applicable to other savannas., J. San José, R.
