Severe meconium aspiration sy ndrome (MAS) in newborns is often treated by exogenous surfac tant. Because its efficacy is reduced by meconium-induced inflammation, glucocorticoid budesonide was added into surfac tant preparation Curosurf to enhance efficacy of the surfactant therapy in experimental model of MAS. Oxygen-ventilated rabbits were intratracheally given meconium (25 mg/ml, 4 ml/kg) to induce respiratory failure. Thirty minutes later, animals were treated by intratracheal budesonide (0.25 mg/kg) ; or surfactant lung lavage (10 ml/kg, 5 mg phospholipids/ml) repeated twice, followed by undiluted Curosurf (100 mg phospholipids/kg) ; or by the above mentioned surfactant treatment with the last surfactant dose fortified with budesonide (0.25 mg/kg) ; or were untreated. Animals were ventilated for additional 5 hours and respiratory parameters were measured regularly. After sacrificing animals, wet-dry lung weight ratio was evaluated and plasma levels of interleukins (IL)-1beta, -6, -8, and TNF-alpha were measured by ELISA method. Efficacy of the given therapies to enhance lung functions and to diminish lung edema formation and in flammation increased from budesonide-only and surf actant-only therapy to surfactant+budesonide therapy. Combined therapy improved gas exchange from 30 min of administration, and showed a longer- lasting effect than surfactant-only therapy. Ruminants are often fed a high-concentrate (HC) diet to meet lactating demands, yet long-term concentrate feeding induces subacute ruminal acidosis (SARA) and leads to a decrease in milk fat. Buffering agent could enhance the acid base buffer capacity and has been used to prevent ruminant rumen SARA and improve the content of milk fat. Therefore, we tested whether a buffering agent increases lipid anabolism in the livers of goats and influences of milk fat synthesis. Twelve Saanen-lactating goats were randomly assigned to two groups: one group received a HC diet (Concentrate: Forage=60:40, Control) and the other group received the same diet with a buffering agent added (10 g sodium butyrate, C4H7NaO2; 10 g sodium bicarbonate, NaHCO3; BG) over a 20-week experimental period. Overall, milk fat increase (4.25±0.08 vs. 3.24±0.10; P<0.05), and lipopolysaccharide levels in the jugular (1.82±0.14 vs. 3.76±0.33) and rumen fluid (23,340±134 vs. 42,550±136) decreased in the buffering agent group (P<0.05). Liver consumption and release of nonesterified fatty acid (NEFA) into the bloodstream increased (P<0.05). Phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT) and ribosomal protein S6 kinase (p70S6K) up-regulated significantly in the livers of the buffering agent group (P<0.05). It also up-regulated expression of the transcription factor sterol regulatory element binding protein-1c (SREBP-1c) and its downstream targets involved in fatty acid synthetic, including fatty acid synthetase (FAS), stearoyl-CoA desaturase (SCD-1) and acetyl-CoA carboxylase 1 (ACC1) (P<0.05). The BG diet increased insulin levels in blood (19.43±0.18 vs. 13.81±0.10, P<0.05), and insulin receptor was likewise elevated in the liver (P<0.05). Dairy goats are often fed a high-concentrate (HC) diet to meet their lactation demands; however, long-term concentrate feeding is unhealthy and leads to milk yield and lactose content decreases. Therefore, we tested whether a buffering agent is able to increase the output of glucose in the liver and influence lactose synthesis. Eight lactating goats were randomly assigned to two groups: one group received a HC diet (Concentrate : Forage = 6:4, HG) and the other group received the same diet with a buffering agent added (0.2 % NaHCO3, 0.1 % MgO, BG) over a 19-week experimental period. The total volatile fatty acids and lipopolysaccharide (LPS) declined in the rumen, which led the rumen pH to become stabile in the BG goats. The milk yield and lactose content increased. The alanine aminotransferase, aspartate transaminase, alkaline phosphatase, pro-inflammatory cytokines, LPS and lactate contents in the plasma significantly decreased, whereas the prolactin and growth hormone levels increased. The hepatic vein glucose content increased. In addition, pyruvate carboxylase (PC), phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6- phosphatase (G6PC) expression in the liver was significantly up-regulated. In the mammary glands, the levels of glucose transporter type 1, 8, 12 as well as of sodium-glucose cotransporter 1 increased. Cumulative buffering agent treatment increased the blood concentrations of glucose via gluconeogenesis and promoted its synthesis in the liver. The 1990s signified the beginning of Taiwan´s "era of localism". Local places in Taiwan and the idea of Taiwan as a place were to gain unprecedented status in both political narratives and social practice. These place-centered acts gradually converged into the state-led Integrated Community-Making Program, in operation since 1994. Amid the prevailing phenomena, the past of a place was frequently perceived as representing a utopia which was rapidly fading or had been already lost as the result of development. The heritage sites, as "sites of memory", have thus not only served as "memory tactics" in rebuilding the sense and identity of a place, but are also expected to mediate the construction of a better future in terms of locality production. This paper examines the substantial components of locality in Taiwan within three frame works: globalization, machizukuri and community-building, which comprise the significance of locality in Taiwan. It concludes with case studies, which examine the social practice of place-making through the development of controversial sites of memory. It attempts to reveal the dialectical relationship between state-propaganda and the local practice of community-building, as well as the dilemma generated from the uses and abuses of global/local discourse.
Partnerships have a long history in European social housing with a mixed degree of success. They are an emerging model in post-socialist countries driven by budgetary constraints, rapid privatisation of public housing, and pragmatic efforts to respond to a complex housing affordability crisis. This article evaluates the challenges and opportunities of a new partnership model implemented in Albania to provide social rental housing. The project, launched in 2009, involves a legally defined partnership between central and local governments, the private sector, and an international financial institution. It has doubled the amount of municipal rental housing, addressing the needs of low- and mid-income households in Albania through the construction of 1,138 rental apartments for 4,300 people in eight cities. The allocation process, although politically charged, has been targeted. The partnership has capitalised on efficiencies, sound fiscal management, and cost and quality control. Despite some construction delays and potential concerns related to future sustainability, we argue that the partnership model is effective and has an important learning and innovation role for the future provision of social housing in Albania as well as in other post-socialist countries in South-East Europe facing similar challenges.
A global decline in pollinator abundance and diversity has demanded increased research attention to the ecology and genetics of bumblebees. However, as progressively more restrictions are placed on sampling for insects, researchers are increasingly obliged to use archival specimens collected for purposes other than genetic analyses. In this study we assessed the suitability, for population genetic studies, of popular, low-cost methods for preservation and storage of bumblebee specimens. Specimens of Bombus terrestris L. were held under six storage regimes for up to two years. DNA was extracted from the samples using three extraction protocols and the quality of the DNA was examined using PCR amplification of a mitochondrial and a nuclear gene. All extraction and storage methods provided sufficient DNA for successful PCR amplification. However, samples preserved in acetone or at freezing temperatures yielded the highest DNA concentrations. DNA yields from pinned specimens at room temperature declined over time, particularly when using standard extraction techniques. DNA concentrations were significantly lower from specimens preserved in 70% ethanol compared to all other extraction techniques and declined linearly over the two years of storage. These results indicate that two of the most popular insect storage methods (pinning and storage in ethanol) should be avoided for the long-term preservation of genetic material for future studies.
