We feature an article this month on a research expedition to Svalbard, an archipelago between the Arctic Ocean, Barents Sea, Greenland Sea and Norwegian Sea. The first Czech expedition by the University of South Bohemia and of the Academy of Sciences of the Czech Republic established a research station in the vicinity of Petuniabukta, in the northern part of Billefjorden, Isfjorden, in central Svalbard, which is the northernmost part of Norway. Our study was aimed to characterize the phenotype and functional endpoints of local microwave hyperthermia (LHT, 42 °C) on tumor infiltrating and spleen leukocytes. The effectiveness of LHT applied into the tumor of B16F10 melanoma-bearing C57/BL6 mice was compared with anesthetized and non-treated animals. Subpopulations of leukocytes were analyzed using the flow cytometry, and the cytotoxic activity of splenocytes against syngeneic B16F10 melanoma and NK-sensitive YAC-1 tumor cell lines was evaluated in 51 Cr-release assay. Similarly, the in vitro modification of the heat treatment was performed using healthy and melanoma-bearing splenocytes. We found a 40 % increase of activated monocytes (CD11b+CD69+) infiltration into the tumor microenvironment. In the spleen of experimental animals, the numbers of cytotoxic T lymphocytes (CTLs-CD3+CD8+) and NK cell (CD49b+NK1.1+) raised by 22 % and 14 %, respectively, while the NK1.1+ monocytes decreases by 37 %. This was accompanied by an enhancement of cytotoxic effector function against B16F10 and YAC-1 targets in both in vivo and in vitro conditions. These results demonstrate that LHT induces better killing of syngeneic melanoma targets. We have separated 2b myosin heavy chain (MyHC) isoform from the rat extensor digitorum longus muscle by SDS-PAGE and analyzed it by two subsequent mass spectrometry techniques. After tryptic digestion, the obtained peptides were identified by Matrix-Assisted Laser Desorption/Ionisation reflectron Time of Flight mass spectrometry (MALDI-TOF MS) and sequenced by Liquid chromatography tandem mass spectrometry (ESI LC/MS/MS). The analyzed peptides proportionally covered 30 % of the 2b MyHC isoform sequence. The results suggest that the primary structure is identical with the highest probability to a NCBI database record ref|NP_062198.1|, representing the last updated record of rat 2b isoform. Tato studie se věnuje rané fázi mikroskopického zkoumání přírody Antoni van Leeuwenhoeka (1632-1723), které představil ve své korespondenci členům Royal Society. Studie se zaměřuje na období od navázání styku s Royal Society (1673) až do roku 1680, kdy byl Leeuwenhoek zvolen členem společnosti. Z metodologického hlediska studie uplatňuje na Leeuwenhoekovy dopisy členům Royal Society přístup, který představili autoři Steven Shapin a Simon Schaffer v knize Leviathan and the Air-Pump (1985). Pokouší se zjistit, jestli se v Leeuwenhoekových dopisech objevují tři strategie, (materiální, literární a sociální), které Shapin a Schaffer přisoudili Robertu Boylovi. Studie potvrzuje, že závěry britských autorů platí i pro jiné vědce., The aim of the study is to discuss the early microscopic examination of nature, introduced by Antoni van Leeuwenhoek (1632-1723) in his letters to the Royal Society. The paper focuses on period from the establishing of contact with the Royal Society (1673) to the electing of Leeuwenhoek the member of the Society in 1680. From the methodological point of view the study uses the approach which Steven Shapin a Simon Schaffer presented in their book Leviathan and the Air-Pump (1985). The study tries to find out to what extent it is possible to understand Leeuwenhoek's letters as expressing three technologies (material, literary, and social) proposed by the British authors. The paper shows that the Shapin's and Schaffer's understanding of experimental science of Robert Boyle is transferrable to other scientists., and Monika Špeldová.
Aquatic microbial ecology group at Institute of Hydrobiology (Biology Center AS CR, v. v. i.) primarily deal with carbon and nutrient flows through microbial food webs in systems of different trophic status. In particular, members of this group have begun to study relative interaction of organisms and their function in nutrient cycling in the impounding reservoirs and the lakes. and Jaroslav Vrba.
